Inhibition of growth and biofilm formation of Staphylococcus aureus by corosolic acid (Corosolic acid에 의한 Staphylococcus aureus의 생장 및 생물막 형성 저해)

  • Staphylococcus aureus is a pathogenic bacterium that causes food poisoning, exhibits a strong capacity to form biofilm, and is highly resistant to antimicrobial agents. The purpose of this study was to investigate the antimicrobial characteristics of corosolic acid against S. aureus. S. aureus showed high susceptibility to corosolic acid in a concentration-dependent manner. The minimum inhibitory concentration and colony-forming ability determined by the broth microdilution method showed that corosolic acid had strong antimicrobial activity against the bacteria. The diameters of the inhibition zone and numbers of colony forming units at each concentration of corosolic acid were also measured. In addition, corosolic acid displayed potent biofilm inhibition activity against S. aureus at concentrations below its minimum inhibitory concentration. These results suggest that corosolic acid can be used to effectively prevent biofilm formation by S. aureus, thereby making S. aureus more susceptible to the action of antimicrobials.

Characteristics and Inhibition of Polyphenol Oxidase from Fuji Apples (후지 사과 Polyphenol Oxidase의 특성 및 활성억제)

  • As a basic research for inhibition of enzymatic browning of apples during dehydration or processing, polyphenol oxidase was extracted from Fuji apples to investigate heat inactivation, chemical inhibition and other properties. Polyphenol oxidase showed the highest activity at $20^{\circ}C$ and pH 5.5 with catechol as substrate, and the Michaelis constant of 0.14 M under the same condition of substrate and pH. The thermal inactivation followed pseudo first-order kinetics to have activation energy of 23.0 kcal/mol and z value of $19.7^{\circ}C$. As for substrate specificity the polyphenol oxidase showed high affinity toward the o-diphenolic compounds, particularly chlorogenic acid. Neither the m- and p-dihydroxy phenols nor monophenols were attacked. Browning by polyphenol oxidase was completely inhibited at the concentrations of 10mM for potassiummetasulfite and thiourea and 1mM for L-cysteine, ascorbic acid and sodium diethyldithiocarbamate.

  • Weissella spp. from traditional Korean foods of Kimchi were isolated and characterized against food-borne pathogenic Listeria monocytogens. The isolates were identified as W. cibaria 0D17 and W. confusa 0D23 from Kimchi by the biochemical characteristics and 16S DNA sequencing. The culture solutions of the isolates adjusted to pH 7.0 showed L. monocytogens inhibition. To analyze the quantitative detection of L. monocytogenes, real-time PCR was performed according to the SYBR Green I method. The isolates grew well and L. monocytogens did not grow during the co-culture with those strains at $37^{\circ}C$. Therefore, W. cibaria 0D17 and W. confusa 0D23 might be the candidates as the functional lactic acid bacteria for improving food safety.

The Probiotic Characteristics of Lactobacillus acidophilus Isolated from Infant Feces (신생아분변 유래 Lactobacillus acidophilus의 프로바이오틱으로서의 특성)

  • This study was conducted to obtain a good probiotic strain of L. acidophilus from infant feces which have the acid and bile tolerance. The selection criteria for the strain included antimicrobial activity, serum cholesterol reduction, resistance to the hydrogen peroxide, angiotensin converting enzyme (ACE) inhibition activity and iron solubility. To this end, five probiotic Lactobacillus strains have been isolated from infant feces. Especially, L. acidophilus SD 105 had strong antimicrobial activity against Listeria sp., high deconjugation activity in the medium which contained 0.5% of glycocholate (GCA) and high resistance to the hydrogen peroxide. L. acidophilus SD 102 showed the highest ACE inhibition activity among the tested cultures and L. acidophilus SD 103 showed iron solubility of more than 70%.

Population Pharmacokinetics of Midazolam in Healthy Koreans: Effect of Cytochrome P450 3A-mediated Drug-drug Interaction (건강한 한국인에서 미다졸람 집단약동학 분석: CYP3A 매개 약물상호작용 평가)

  • Objective: Midazolam is mainly metabolized by cytochrome P450 (CYP) 3A. Inhibition or induction of CYP3A can affect the pharmacological activity of midazolam. The aims of this study were to develop a population pharmacokinetic (PK) model and evaluate the effect of CYP3A-mediated interactions among ketoconazole, rifampicin, and midazolam. Methods: Three-treatment, three-period, crossover study was conducted in 24 healthy male subjects. Each subject received 1 mg midazolam (control), 1 mg midazolam after pretreatment with 400 mg ketoconazole once daily for 4 days (CYP3A inhibition phase), and 2.5 mg midazolam after pretreatment with 600 mg rifampicin once daily for 10 days (CYP3A induction phase). The population PK analysis was performed using a nonlinear mixed effect model ($NONMEM^{(R)}$ 7.2) based on plasma midazolam concentrations. The PK model was developed, and the first-order conditional estimation with interaction was applied for the model run. A three-compartment model with first-order elimination described the PK. The influence of ketoconazole and rifampicin, CYP3A5 genotype, and demographic characteristics on PK parameters was examined. Goodness-of-fit (GOF) diagnostics and visual predictive checks, as well as bootstrap were used to evaluate the adequacy of the model fit and predictions. Results: Twenty-four subjects contributed to 900 midazolam concentrations. The final parameter estimates (% relative standard error, RSE) were as follows; clearance (CL), 31.8 L/h (6.0%); inter-compartmental clearance (Q) 2, 36.4 L/h (9.7%); Q3, 7.37 L/h (12.0%), volume of distribution (V) 1, 70.7 L (3.6%), V2, 32.9 L (8.8%); and V3, 44.4 L (6.7%). The midazolam CL decreased and increased to 32.5 and 199.9% in the inhibition and induction phases, respectively, compared to that in control phase. Conclusion: A PK model for midazolam co-treatment with ketoconazole and rifampicin was developed using data of healthy volunteers, and the subject's CYP3A status influenced the midazolam PK parameters. Therefore, a population PK model with enzyme-mediated drug interactions may be useful for quantitatively predicting PK alterations.

Preparation of 5-fluorouracil-loaded Nanoparticles and Study of Interaction with Gastric Cancer Cells

  • Aims: To prepare 5-fluorouracil (5-Fu) nanoparticles with higher encapsulation efficiency and drug loading, and then investigate interaction with the SGC-7901 gastric cancer cell line. Materials and Methods: Prescription was optimized by orthogonal experiments, the encapsulation efficiency and loading capacity were tested by high-performance liquid chromatography, and inhibition of proliferation by 5-Fu nanoparticles and 5-Fu given to cells for 24, 48 and 72 hours was investigated by methyl thiazolyl tetrazolium assay (MTT). In addition, 5-Fu nanoparticles were labeled by fluorescein isothiocyanate (FITC), and absorption into cells was tested by flow cytometry. Results: The optimal conditions for preparation were concentrations of 5-Fu of 5mg/ml, of $CaCl_2$ of 60 mg/ml and of chitosan of 2 mg/ml. With a stirring speed of 1200rpm, encapsulation efficiency of 5-Fu nanoparticles was $55.4{\pm}1.10%$ and loading capacity was $4.22{\pm}0.14%$; gastric cancer cells were significantly inhibited by 5-Fu nanoparticles in a time and concentration dependent manner, and compared to 5-Fu with slower drug release, in a certain concentration range, inhibition with 5-Fu nanoparticles was stronger. 5-Fu nanoparticles were absorbed by the cells in line with the concentration. Conclusions: 5-Fu nanoparticles can inhibit growth of gastric cancer cells in vitro to a greater extent than with 5-Fu with good adsorption characteristics, supporting feasibility as a carrier.

Characteristics of Tyrosinase Inhibitory Extract from Ecklonia stolonifera

  • Tyrosinase inhibitory activities of 14 kinds of seaweed, Ecklonia stolonifera, Ecklonia cava, Undaria pinnatiflda, Laminaria japonica, Sargassum fulvellum, Sargassum miyabei, Sargassum thunbergii, Porphyra yezoensis, Gracilaria verrucosa, Carpopeltis affinis, Pachymeniopsis elliptica, Gelidium amansii, Codium fragile and Ulva pertusa were determined using commercially available mushroom tyrosinase in an in vitro assay system. The $1\%$ (w/v) methanol extract from E. stolonifera showed the highest tyrosinase inhibitory activity of $79.0\%$, electron donating activity of $79.0\%$ and total phenol content of 3.75 mg/100g. Ethyl acetate-methanol-water (7 : 2 : 0.2, v/v) fraction $(0.5\%,\;w/v)$ isolated from the methanol extract showed tyrosinase inhibitory activity of $75.9 \%$, electron donating activity of $88.1 \%$ and total phenol content of 4.38 mg/100g. Tyrosinase inhibitory activity was closely associated with total phenol content (R = 0.99) and electron donating activity (R=0.99). Maximum absorption wavelength of the fraction was 218nm and that of phenolic compounds showed about a range from 210 to 220nm. The inhibition mode of the fraction was noncompetitive inhibition.

  • Recently, a method for detecting animal tinnitus objectively, gap prepulse inhibition of acoustic startling (GPIAS) were reported. However, the GPIAS method is difficult to be directly applied to human tinnitus research because measuring repeatedly startling responses evoked by more than 110 dB SPL acoustic stimuli for human is not easy. In this paper, the auditory late latency response (ALLR) measurement system which can measure conveniently evoked potentials involving the information about the brain cortical activity related with auditory psychologic phenomena such as a tinnitus has been implemented. By using the implemented system, 8 persons with normal hearing sense have been experimented to measure N1-P2 amplitudes of ALLRs evoked by gap prepulse based acoustic stimuli. Through the experimental results, the prepulse inhibitions of all the participants' N1-P2 responses have been observed and their characteristics were evaluated. And it is verified that our implemented system can be utilized as a device for researching and evaluating the objective tinnitus detection method in the future study.

Antitumor Effects of Kluyveromyces marxianus TFM-7 Isolated from Kefir

  • The Strain TFM-7, Which has an antitumor effect, was isolated from Kefir and identified based on analysis using the API 50 CHL kit and 265 rDNA sequencing. Strain TFM-7 was confirmed to belong to the genus Kluyveromyces. Analysis of the 265 rDNA nucleotide sequences found strain TFM-7 to be related to Kluyveromyces marxianus. NRRL Y-828IT. K. marxianus. TFM-7 was cultured with potato dektrose broth medium at $27^{\circ}C$ for 72 hr, and its inhibition effects on the proliferation of seven tumor cell lines and a normal cell line were assessed using the MTT assay. The antitumor effects and growth characteristics of K. marxianus TFM-7 were investigated during a culture period of 7 days. By the $3^{rd}\;day$, K. marxianus TFM-7 showed a dry cell weight 2.39 g/L, a pH of 4.39, an ethanol content of 0.89%, and an inhibition effect on the proliferation of seven tumor cell lines above 50%, except for A-549 tumor cell line. K. marxianus TFM-7 was the most effective at inhibiting the growth of Hep-2 cell line among all tumor cell lines tested. Growth inhibition of a normal cell line, NIH/3T3, was less than 35%, suggesting a decreased level of cytotoxicity toward normal cells. These results indicate that K. marxianus TFM-7 may have used as a yeast strain with antitumor activity.

  • Effects of alumlnlum(AA), soil pH and calcium(Cal on growth response and heavy metal accumulation and regulation of nitrogen fixation In Melilotus suaveolens seedlings, a biennial legume plant dominating in the disturbed area, were quantitatively analyzed during the growing periods. Accumulation of metals In each organ of M. suaveojens was Increased UC the lowering of pH. Al contents In leaf and root treat- ed with 30ppm Al at pH 4.2 on the 28th day after treatment were 8 and 11 folds higher than those of control, while the contents with 100ppm AA were 21 and 24 folds as compared to control. The significent inhibition in Al contents was induced by 100ppm AA and Ca at pH 6.5. Increased metals inhibited higher at pH 4.2 and the growth inhibition due to Al was reversed by the addition of Ca, suggesting that growth correlates to the pH value. Chlorophyll contents in leaves increased during growth stave were inhibited by Al treatments. The biomass was decreased UD the lowering of pH and the increase of concentrations. 100ppm Ca treatment resulted in 5.1-5.9% increase of the biomass as compared to that of 100ppm Al. Specific nitrogen fuation activities In nodules In the 100ppm Al at pH 4.2 and pH 6.5 were reduced to 35. 2% and 52.2% of control, on the 28th day after treatment, while the combination of Al and Ca induced reduced by 10% as compared to control with the treatment of 100ppm AA at pH 4.2 due to the inhibition bination of 100ppm Al and Ca and lowering of pH.
