Activities of Antioxidation and AChE Inhibition of Extract from Hericium erinaceus

  • It has been known that the novel diterphenoids, hericenone and erinacine isolated from the fruiting body and cultured mycelia of Hericium erinaceus showed potent stimulating activity of nerve growth factor (NGF)-synthesis. To investigate the biological activities of extracts from fruiting body, cultured mycelium and cell-free broth of H. erinaceus, the activity experiments of antioxidation and AChE inhibition were carried out. Sephadex G-10 gel filtration followed by HPLC on a ${\mu}Bondapak$ $C_{18}$ column of EtOAc extract from cultured mycelium showed a biological activity.

  • The resistance inhibitory activities of 54 odorant mixtures (essential oil) from 41 Korean aromatic herbs were tested against multi-drug resistant Staphylococcus aureus SA2, which has resistances to 10 usual antibiotics including chloramphenicol. As results, combinations of 28 kinds of samples from 21 herbs and chloramphenicol have resistance inhibitory activities in dose dependent manner.

Comparison of Biological Activities of Extracts from Different Parts and Solvent Fractions in Cornus kousa Buerg (산딸나무의 부위별 추출물 및 용매 분획물의 생리활성 비교)

  • Several extracts of different parts and solvent fractions of Cornus kousa were obtained and their functional material contents, antioxidant activities and tyrosinase inhibition effects were determined. Content of total polyphenols and flavonoids contents in flower were 169.638 $mg{\cdot}g^{-1}$ and 25.418 $mg{\cdot}g^{-1}$, respectively, which were much higher than those of other parts. Also, flower extracts showed the strongest effects on DPPH and ABTS radicals scavenging and ferrous ion chelating. In flower, leaf, and stem extracts, inhibition effects on peroxidation of linoleic acid determined by ferric thiocyanate(FTC) method were higher than a synthetic antioxidant, BHT. Tyrosinase inhibition activities were shown only in flower extract. Flower and leaf extracts, showing high biological activities in various system, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. Total polyphenol contents of water fractions were higher than any other solvent fractions in both flower and leaf, 67.006 $mg{\cdot}g^{-1}$ and 67.739 $mg{\cdot}g^{-1}$, respectively. But total flavonoid contents were higher in ethyl acetate fraction for flower extract and butanol fraction for leaf extract. Among the solvent fractions, the highest efficiency of free radical scavenging activities was obtained in ethyl acetate fraction for flower extract and n-butanol fraction for leaf extract. Tyrosinase inhibition activities were higher in water fraction for both flower and leaf extracts, 49.24% and 31.8%, respectively.

Antioxidant and Anti-Diabetic, Anti-Alzheimer Activities of Stem from Opuntia ficus-indica var. saboten Cultivated in Jeju at Harvest Time (채취시기별 보검선인장 줄기의 항산화, 항당뇨 및 항알츠하이머 활성평가)

  • In this study, we assessed antioxidant, anti-diabetic, and anti-Alzheimer activities of Opuntia ficus-indica var. saboten (OFI) at harvest time. OFIs were cultivated December 2015~November 2016 in Jeju island. The 70% ethanol extracts of OFI were used to investigate total polyphenol and flavonoid contents, antioxidant(DPPH and ABTS radical scavenging assay), anti-diabetic(yeast ${\alpha}$-glucosidase and rat ${\alpha}$-glucosidase inhibition assay), and anti-Alzheimer(Acetylcholinesterase and butyrylcholinesterase inhibition assay) activities. Total polyphenol and flavonoid contents of OFIs were $17.40{\sim}23.11{\mu}g$ garlic acid/mg Ex and 2.17~6.22 ug (+)-catechine/mg Ex, respectively. DPPH and ABTS radical scavenging activity of OFIs were 131.98~184.90 mg ascorbic acid(AA) eq/100 g and 63.60~101.83 mg AA eq/100 g, respectively. In the anti-diabetic and anti-Alzheimer activities, 70% ethanol extracts of OFI exhibited moderate inhibition activity, compared to control (acarbose and beberine). Total polyphenol and flavonoid contents, antioxidant, anti-diabetic, and anti-Alzheimer activities were no significant differences by season, respectively. Therefore, information on comparative biological evaluations of OFI may be a beneficial in exploring functional food and drug development.

Synthesis, Urease and Acetylcholine Esterase Inhibition Activities of Some 1,4-Disubstituted Thiosemicarbazides and their 2,5-Disubstituted Thiadiazoles

  • A new series of 2,5-disubstituted-1,3,4-thiadiazoles 6a-i was synthesized by overnight stirring various 1,4-disubstituted thiosemicarbazides 5a-i in polyphosphoric acid followed by neutralization. The structures of newly synthesized compounds 5a-i and 6a-i were characterized by IR, $^1H$ and $^{13}C$ NMR, elemental analysis and mass spectrometric studies. All the synthesized compounds were evaluated for their urease and acetylcholine esterase inhibition activities. Thiosemicarbazides 5a-i are found to possess excellent potential for urease inhibition, more than the standard drug. Thiosemicarbazides 5a-i are more potent urease inhibitor than their cyclic analogues thiadiazoles 6a-i. Almost all of the compounds are excellent inhibitors of acetylcholine esterase. The inhibition of acetylcholine esterase of compounds 5a, 5c, 5d, 5g, 5i, 6e, 6f, 6g, and 6i is much more than that of standard drug.

Temperament, Parenting, and Behavioral Inhibition of Korean Toddlers (걸음마기 아동의 행동억제 : 아동의 기질 및 어머니의 양육행동과의 관계)

  • Mothers of 113 Korean toddlers completed Toddler Behavior Assessment Questionnaire(TBAQ) and Child Rearing Practice Report(CRPR). Observations of mother-child dyads in novel situations involving unfamiliar settings and adults assessed child's behavioral inhibition. Nonsocial inhibition were measured by the amount of time each child spent in physical contact with his/her mother in free-play episodes. Adult-social inhibition was based on child's behavior when an unfamiliar adult requested that the child approach her as she presented them with toys or activities. Results showed that TBAQ Social Fearfulness was positively associated with nonsocial and adult-social inhibition. TBAQ Pleasure was correlated negatively and TBAQ Anger Proneness positively correlated with adult-social inhibition. An interaction effect showed that child's social fearfulness and mother's overprotective behaviors affected adult-social inhibition. The regression model explained 31% of the variance.

The Comparative Evaluation of Fermented and Non-fermented Soybean Extract on Antioxidation and Whitening

  • The present study was performed to compare the antioxidative and whitening activities of fermented soybean extract (FSB) and non-fermented soybean extract (SB). Antioxidative and whitening activities of FSB and SB were evaluated by the determination of DPPH, superoxide radical and hydroxyl radical scavenging activities, linoleic acid inhibition activity, and tyrosinase inhibition activity. FSB showed the higher effect than SB in the antioxidative activities. Also FSB showed the better effect than SB in whitening activity. These results demonstrated that the fermentation played a more excellent role than the non-fermentation in antioxidation and whitening. Therefore, this study suggested that FSB could be a useful cosmetic ingredient for antioxidation and skin whitening.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

Bioassay-Guided Isolation and Identification of Compounds from Arecae Pericarpium with Anti-inflammatory, Anti-oxidative, and Melanogenesis Inhibition Activities

  • This study describes the anti-inflammatory, anti-oxidant, and melanogenesis inhibition activities of methanol extract and various organic solvent fractions of Arecae Pericarpium. We examined the inhibition of lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW 264.7 cells, 1,1-diphenyl-2-picrylhydrazine (DPPH) scavenging activity, mushroom tyrosinase inhibition activity and melanin contents. The study showed that, among all tested fractions, methylene chloride fraction showed the strongest inhibition of LPS-induced NO production in RAW 264.7 cells ($IC_{50}$ value $8.89{\mu}g/mL$) and DPPH radical scavenging activity ($EC_{50}$ value $21.39{\mu}g/mL$). Methylene chloride and ethyl acetate fractions similarly inhibited mushroom tyrosinase activity. Methanol extract exhibited strongest reduction of melanin content in B16F10 melanoma cells. Based on the bioactivity assay results, methylene chloride and ethyl acetate fractions were further separated. Eight phenolic compounds were isolated, which are dimeric syringol (1), catechol (2), 4-hydroxybenzaldehyde (3), vanillin (4), 4-hydroxyacetophenone (5), apocynin (6), protocatechuic acid (7) and 4-hydroxybenzoic acid (8). Among the isolated compounds tested, catechol showed the strongest inhibition of LPS-induced NO production in RAW 264.7 cells. Catechol also showed the concentration-dependent NF-${\kappa}B$ inhibition activity. Arecae Pericarpium might have potentials to be developed as anti-inflammatory agent or dermatological product for skin-whitening agent.

Effect of Ssanghwa-tang Extract on Antioxidant and Anti-aging Enzyme Activities (쌍화탕 추출물이 항산화효소 및 항노화관련 효소 활성에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of Ssanghwa-tang (Shu$\bar{a}$nghu$\bar{a}$-g$\bar{e}$ng) on oxidation/reduction reaction-related and aging-related enzymes $in$ $vitro$. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase (XO) inhibition assay. Results : The 50% ethanol (EtOH) extract of Ssanghwa-tang (SHT) showed 55% inhibition of collagenase activity, and 42% inhibition of elastase activity at 1 mg/ml concentration. Also it's treatment showed 18% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of 50% ethanol extract of SHT. Antioxidant activities were determined by DPPH radical scavenging, XO inhibiting activity and SOD-like activity. These scavenging, XO-inhibiting and SOD-like activities were measured in 80%, 75%, and 28% inhibitions, respectively, at a 1 mg/ml treated dose, compared to those of control. The inhibitory effects of 50% EtOH extract on aging and oxidation-related enzyme activities were higher than those of water extract and 95% EtOH extract. Conclusions : Taken together, our findings suggest that the SHT has potential and applicable benefits for development of cosmetics to have anti-aging (anti-wrinkle and whitening) and anti-oxidation functions.
