어류 전염성훼장괴사증의 면역조직화학적 진단 (Immunohistochemical diagnosis of infectious pancreatic necrosis)

  • This experiment was carried out to establish the immunohistochemical diagnostic method for infectious pancreatic necrosis in the monolayers of CHSE-214 cell cultures and paraffin-embedded tissue sections from rainbow trout infected with infectious pancreatic necrosis virus(IPNV). Specific identification of IPNV antigens was often demonstrated in the pancreatic exocrine cells, and less in the intestinal mucous epithelia and the renal hemopoietic tissues by the use of monoclonal antibodies against capsid protein VP2. The specific reaction was seen as a distinct red cytoplasmic color, often as small granules of various sizes. The result showed that streptavidin alkaline phosphatase immunohistochemisry specifically identified IPNV antigens in both infected cell cultures and tissue sections.

In vitro Antiviral Activities of Korean Marine Algae Extracts against Fish Pathogenic Infectious Hematopoietic Necrosis Virus and Infectious Pancreatic Necrosis Virus

  • To investigate the antiviral activity of marine algae against fish pathogenic viruses, which are often the causes of viral disease in aquaculture, the 80% methanolic extracts of 21 species collected from the coast of Korea were screened for their in vitro antiviral activities on infectious hematopoietic necrosis virus (IHNV) and infectious pancreatic necrosis virus (IPNV), using a flounder spleen (FSP) cell-line. Among them, Monostroma nitidum (10 ${\mu}g/mL$) exhibited the strongest inactivation on IHNV, showing a 2 log reduced virus titre as compared to the control in the determination of direct virucidal activity. In addition, Polysiphonia morrowii (100 ${\mu}g/mL$) remarkably reduced the virus titres of treated cells by 2-2.5 log, for both IHNV and IPNV, in the determination of cellular protective activity, implying the existence of substances that may modulate innate host defense mechanisms against viral infections. These results reveal that some marine algae could be promising candidates as sources of antiviral agents or as health-promoting feeds for aquaculture.

  • Infectious pancreatic necrosis(IPN) virus was known as a causative agent of newly recognized viral disease of young rainbow trout characterized by highly contagious, high mortality and necrosis of pancreas. Several strains of IPN viruses were recovered from young rainbow trout that have been shown a typical cinical sign of infectious pancreatic necrosis disease. The field isolate produced cytopathic effect, and multiplied up to $10^{6.0}$ to $10^{6.5}$ $TCID_{50}/0.1ml$ in BT cell culture. In the indirect immunofluorescent assay with trout anti-IPN virus IgG and goat anti-trout IgG FITC conjugate, these isolates were proved to be a IPN virus that were closely related with VR277 strain of IPN virus antigenically.

  • A survey was conducted to determine the prevalance of infectious pancreatic necrosis virus (IPNV) on fish farms in Korea and the epidemiology of IPNV infection in the farmed rainbow trout. In total, 43 pools of rainbow trout with apparent signs of viral infection from five provinces were obtained and analyzed. Evident cytopathic effects, including karyopycnosis and cell destruction, were observed in CHSE (chinook samlmon embyro)-214 cells infected with the virus isolates. Of these, ten viral isolates were assumed to be IPNV based on biophysical properties. RNA analysis revealed that the isolates contained two-segmented RNA genomes, further indicating that the viral isolates are IPNV. Antigenic comparison of the IPNV isolates identified three distinct serological groups separable by the cross-neutralization test. Of the ten IPNV isolates, six could be classified as strain DRT, two as strain Ab, and two as strain VR299. We were not able to isolate new strain of IPNV or any isolate serologically similar to the standard strain Sp.poraceae and families of the Agaricales, they are genetically more related to the Polyporaceae. These results are consistent with morphological characters observed in those mushrooms. However, it is premature to conclude taxonomic status Ganoderma species in the present study employing small sample size.

PT-PCR 법에 의한 Infectious Pancreatic Necrosis Virus의 조기진단 (Rapid and Sensitive Detection of Infectious Pancreatic Necrosis Virus (IPNV) by Revers Transcription-Polymerase Chain Reaction (RT-PCR))

  • Infectious pancreatic necrosis virus (IPNV)는 치어에 감염되어 치명적인 질병을 유발하는, 양식산업에 있어 중요한 어류 병원체이다. 본 연구에서는 IPNV를 신속, 정확하게 진단하는 방법을 개발하고자 IPNV의 항원성 단백질인 VP2 유전자 부분에서 선택한 primers를 이용하여 역전사-중합효소연쇄반응법(Reverse Transcription-Polymerase Chain Reaction, RT-PCR)을 실시하였다. RT-PCR 증폭법으로 순수분리도니 IPNV dsRNA 40 ng 정도의 적은 양도 확인 할 수 있었으며, IHNV와 같은 다른 어류 병원체의 게놈을 RT-PCR templates로 사용하였을 경우는 어떠한 PCR 산물도 검출되지 않는 특이성을 보였다. 특히 유전자의 분리없이 조직 그 자체를 대상으로 RT-PCR을 행하는 in situ RT-PCR 방법으로 IPNV가 감염된 넙치 (Paralichthy olivaceus) 치어의 조직에서 IPNV 감염을 신속하게 확인할 수 있었다. 따라서 RT-PCR 및 in situ RT-PCR 방법은 IPNV를 신속, 정확하게 진단하는데 활용될 수 있을 것으로 생각된다.

Terminal Nucleotide Sequences in the Double-stranded RNA Genome Segments of Infectious Pancreatic Necrosis Virus DRT Strain

  • The terminal regions of the double-stranded RNA (dsRNA) genome segments of infectious pancreatic necrosis virus (IPNV) DRT strain were sequenced. The dsRNAs, which were $^{32}P$-labelled at their 3'-termini by incubation with [$^{32}P$]pCp and T4 RNA ligase, were separated by 5$%$ polyacrylamide gel electrophoresis, and the segments A and B of IPNV-DRT were sequenced by two-dimensional gel electrophoresis. The 5'-terminal sequences of the IPNV-DRT plus strand from two genome segments were found to have the same conserved nucleotide (5'-CGG(C/A)A-), but the 3'-terminal sequences -CCCCAGGCG-3' and -CGGACCCCG-3' were found in the plus strand from segments A and B, respectively. The inverted oligonucleotide sequences of 3'-terminal of between segments A and B were found and they differ from those of other IPNVs.

Nucleotide Sequence Analysis of the RNA-dependent RNA Polymerase Gene of Infectious Pancreatic Necrosis Virus DRT Strain

  • To determine the nucleotide sequence of the ds RNA segment B containing the RNA dependent RNA polymerase (RdRp) gene of the DRT strain of infectious pancreatic necrosis virus (lPNV), the cDNA of the ds RNA segment B of the DRT strain of IPNV was synthesized using the reverse transcriptase (RT)-polymerase chain reaction (PCR) and its cDNA nucleotide sequence was determined. The DRT segment B was 2, 783 bp long and contained only a single long open reading frame (ORF) of 2, 535 bp in length. This ORF nucleotides encoded the VPl protein, the putative RdRp of IPNV. The VPl protein comsisted of 845 amino acids. The molecular weight of the RdRp, as deduced from the nucleotide sequence, is 94, 426. The nucleotide sequence of the ORF of the DRT showed 89.7% homology to the Jasper strain, but 80.8% to the Sp strain. The amino acid sequence of the ORF of the DRT sho.wed 97.6% homology to the Jasper strain, but 88.7% to the Sp strain. The conserved GTP-binding motif was detected in VPl protein.

해수사육 무지개송어 (Oncorhynchus mykiss)에 미치는 어류 병원성 바이러스의 영향 (Effect of Fish Pathogenic Viruses on Mariculture of Rainbow Trout (Oncorhynchus mykiss))

  • 본 연구에서는 무지개송어의 해수면 양식장으로의 이동에 따라 나타날 수 있는 어류 병원성 바이러스의 영향을 알아보기 위해, 담수 무지개송어 유래 병원체인 infectious hematopoietic necrosis virus (IHNV)와 infectious pancreatic necrosis virus (IPNV)를 사용하여 주요 해산 양식 어종인 넙치, 조피볼락, 돌돔, 참돔 및 능성어에 주사한 후 병원성 및 감염 여부를 조사하였으며, 또한 해산 양식 어종 유래 병원체인 marine birnavirus (MABV), hirame rhabdovirus (HIRRV) 및 nervous necrosis virus (NNV)를 사용하여 무지개송어에 대한 병원성 및 감염 여부를 조사하였다. IHNV와 IPNV는 주요 해산 양식 어종에 감염되는 것으로 확인되었으며, 또한 무지개송어는 해산 유래 위해 병원체인 MABV와 NNV에 감염되거나 HIRRV에 의해 폐사되는 것으로 확인되었다. 이상의 연구 결과, 해수사육 무지개송어는 어류 병원성 바이러스들에 의해 영향 받을 수 있을 것으로 사료되었다.

Infectious hematopoietic necrosis virus (IHNV)에 대한 단클론 항체 생산 (Production of monoclonal antibodies against infectious hematopoietic necrosis virus (IHNV))

  • Infectious hematopoietic necrosis virus (IHNV) is s significant viral pathogen affecting cultured rainbow trout (Oncorhynchus mykiss) in Korea. In this study, five monoclonal antibodies (mAbs) (IHNV-1, 2, 3, 4, and 5) were produced using purified IHNV. Reactivities of these mAbs were analyzed by western blot (WB), enzyme-linked immunosorbent assay (ELISA), and indirect fluorescent antibody test (IFAT). These mAbs recognized glycoprotein (69 kDa, IHNV-1), nucleocapsid protein (39 kDa, IHNV-3, 4, and 5), or phosphoprotein (27 kDa, IHNV-2) of IHNV by WB analysis. ELISA results indicated that these five mAbs were specific to IHNV without showing any cross-reactivity against other fish viruses (hirame rhabdovirus, infectious pancreatic necrosis virus, and viral hemorrhagic septicemia virus). IFAT demonstrated specific fluorescence signals of IHNV-infected epithelioma papulosum cyprini (EPC) cells, whereas no reactivity of normal EPC cells was observed. These mAbs can be very useful for immuno-diagnosis of IHNV infection.
