A study on selenium quantification using ICP-MS with oxygen reactive gas in soil of Korea

  • This study evaluates a method for quantifying selenium (Se) concentration in soil using inductively coupled plasma mass spectrometry (ICP-MS), with oxygen as a reaction gas. This approach addresses the challenge of detecting low levels of Se in complex soil matrices and aims to effectively minimize interference problems typically associated with argon plasma in traditional ICP-MS analyses. The analytical method utilizes conditions optimized for minimizing spectral interference and were validated by linearity, accuracy, precision, limit of detection (LOD), and limit of quantification (LOQ). The method demonstrated good linearity, high accuracy (90-97 %), and remarkable sensitivity, achieving detection and quantification limits of 0.15 ㎍/kg and 0.44 ㎍/kg, respectively. Developed analysis method for Se in soil was applied to field samples in the different regions of South Korea and Se concentration ranged from 0.11 to 0.52 mg/kg. Correlation analysis between Se concentration and soil properties showed that Se concentration was significantly correlated with cation exchange capacity (CEC) and available phosphorus among other soil properties.

Investigation on Dissolution and Removal of Adhered LiCl-KCl-UCl3 Salt From Electrodeposited Uranium Dendrites using Deionized Water, Methanol, and Ethanol

  • Deionized water, methanol, and ethanol were investigated for their effectiveness at dissolving LiCl-KCl-UCl3 at 25, 35, and 50℃ using inductively coupled plasma mass spectrometry (ICP-MS) to study the concentration evolution of uranium and mass ratio evolutions of lithium and potassium in these solvents. A visualization experiment of the dissolution of the ternary salt in solvents was performed at 25℃ for 2 min to gain further understanding of the reactions. Aforementioned solvents were evaluated for their performance on removing the adhered ternary salt from uranium dendrites that were electrochemically separated in a molten LiCl-KCl-UCl3 electrolyte (500℃) using scanning electron microscopy with energy dispersive spectroscopy (SEM-EDS). Findings indicate that deionized water is best suited for dissolving the ternary salt and removing adhered salt from electrodeposits. The maximum uranium concentrations detected in deionized water, methanol, and ethanol for the different temperature conditions were 8.33, 5.67, 2.79 μg·L-1 for 25℃, 10.62, 5.73, 2.50 μg·L-1 for 35℃, and 11.55, 6.75, and 4.73 μg·L-1 for 50℃. ICP-MS analysis indicates that ethanol did not take up any KCl during dissolutions investigated. SEM-EDS analysis of ethanol washed uranium dendrites confirmed that KCl was still adhered to the surface. Saturation criteria is also proposed and utilized to approximate the state of saturation of the solvents used in the dissolution trials.

용량분석법을 이용한 화장품 중 티타늄옥사이드의 정량 (A Novel Volumetric Method for Quantitation of Titanium Dioxide in Cosmetics)

  • 자외선차단용 화장품은 기능성화장품으로 분류되어 있으며 매년 다양한 종류의 유기 또는 무기 자외선 차단제를 함유하는 제품이 출시되고 있다. 높은 SPF 수치를 요구하는 제품과 유아 및 민감성 피부를 위한 자외선차단제품에 무기자외선 차단제가 많이 사용되고 있으며 대표적인 것으로 티타늄옥사이드가 있다. 티타늄옥사이드는 피부를 하얗게 표현하는 특성 때문에 착색제로 오랜 기간 사용되어 왔으나 이러한 특성이 자외선 차단제로서의 응용을 제한하여 왔다. 그러나 최근 마이크로 화 기술의 발달로 하얗게 보이는 현상 없이 티타늄옥사이드를 높은 농도로 함유할 수 있게 됨에 따라 티타늄옥사이드의 자외선차단제로서의 응용 연구가 활발히 진행되고 있다. 그러나, 자외선차단 화장품 중 티타늄옥사이드의 함량분석에 대한 연구는 거의 전무한 상황이다. 본 연구에서는 산화환원적정법을 이용한 자외선차단 화장품 중 티타늄옥사이드 함량분석법을 개발하였으며, 대표적인 자외선차단제품인 크림, 메이크업베이스, 파운데이션 및 파우더에 일정량의 티타늄옥사이드를 포함시킨 다음 분석하여 시험법의 회수율이 $96{\sim}105%$임을 확인하였다. 또한, 본 연구의 방법으로 분석한 시판되는 자외선차단제품 7 종의 티타늄옥사이드 함량 분석결과를 ICP-AES (Inductively Coupled Plasma-Mass Spectrometry)를 이용한 기기분석결과와 비교하는 방법으로 시험법을 재검증하였다. 비록 최신 분석기기인 ICP-AES로 정확한 티타늄분석이 가능하지만 고도의 기술이 필요하고 모든 화장품사가 품질관리를 위하여 구비하기에는 분석장비가 고가인 만큼 현실적인 방법이라고는 할 수 없을 것이다. 본 연구의 티타늄옥사이드 정량법은 일반적인 초자기구 만으로 수행할 수 있으므로 현장에서 자외선차단 기능성화장품의 품질관리에 널리 활용될 수 있을 것이다.의하는 점 자체의 식별오차뿐 아니라, 각 점의 위치관계에도 영향을 받는 것을 알 수 있었다. 따라서, 안정적인 3차원 기준 좌표계를 구성하려면 양측 Po과 편측 Or으로 구성되는 평면을 수평기준면으로, 수평면에 수직이고, Foramen Spinosum의 중점과 Nc를 포함하는 평면을 수직기준면으로. 수평면과 수직면에 수직이고. clinoidale나 sella, PNS를 지나는 평면을 전두면으로 설정하는 것이 바람직할 것으로 생각된다. 얻었으며 두 군 사이에 임상적 결과의 차이는 없었다.건측 보다 10도의 굴곡 제한이 있었다. 결론: 자가 슬괵 건을 이용한 전방 십자 재건술 시, 경골 부의 고정 시 슬관절의 굴곡 각도 따른 슬관절의 전방 안정성에 차이는 없었으나, 신전 위에서 고정하는 것이 과도한 이식 건의 장력으로 인한 슬관절 굴곡 구축을 예방하는데 도움이 될 것으로 사료된다.라 증가할 것으로 추정되었으며 이에 대한 연근 추출물 경구 투여가 간 조직을 보호할 수 있는지를 확인하기 위해 분리한 혈청으로부터 ALT 함량을 측정한 결과 대조군에 비하여 유의한 감소를 나타내었다. 또한 연근 추출물이 혈청 내 지질 과산화물의 생성을 억제할 수 있다면 질병의 예방과 치료에 효과적일 것으로 추정할 수 있으므로 그 생성량을 측정하여 보았으나 대조군과의 차이가 나타나지 않았다. 이상의 결과들을 종합하여 보면 스트레스가 부하된 5일 동안 연근(蓮根) 추출물을 함께 투여한 결과 혈청 corticosterone 함량을 유의하게 감소시켰고 뇌 조직내 noradrenaline 함량을 증가시키는 경향을 나타내어 스트레스 해소에 도움이 될 수 있음을 시사하였다. 또한 혈청 내 ALT 함량을 유의하게 감소시켜 스트레스로 인해 발생하는 간 기능의 손상도 어느 정도 억제시키는 것을 확인할 수 있었는데 앞으로 연근(蓮根)의 이러한 작용에 대한 보다 자세한 연구들이

Purification and Characterization of Phocaecin PI80: An Anti-Listerial Bacteriocin Produced by Streptococcus phocae PI80 Isolated from the Gut of Peneaus indicus (Indian White Shrimp)

  • A bacteriocin-producing strain PI80 was isolated from the gut of Penaeus indicus (Indian white shrimp) and identified as Streptococcus phocae PI80. The bacteriocin was purified from a culture supernatant to homogeneity as confirmed by Tricine SDS-PAGE. Reverse-phase HPLC analysis revealed a single active fraction eluted at 12.94 min, and MALDI-TOF mass spectrometry analysis showed the molecular mass to be 9.244 kDa. This molecular mass does not correspond to previously described streptococcal bacteriocins. The purified bacteriocin was named phocaecin PI80 from its producer strain, as this is the first report of bacteriocin production by Streptococcus phocae. The bacteriocin exhibited a broad spectrum of activity and inhibited important pathogens: Listeria monocytogenes, Vibrio parahaemolyticus, and V. fischeri. The antibacterial substance was also sensitive to proteolytic enzymes: trypsin, protease, pepsin, and chymotrypsin, yet insensitive to catalase, peroxidase, and diastase, confirming that the inhibition was due to a proteinaceous molecule (i.e., the bacteriocin), and not due to hydrogen peroxide or diacetyl. Phocaecin PI80 moderately tolerated heat treatment (up to $70^{\circ}C$ for 10 min) and resisted certain solvents (acetone, ethanol, and butanol). A massive leakage of $K^+$ ions from E. coli $DH5\alpha$, L. monocytogenes, and V. parahaemolyticus was induced by phocaecin PI80, as measured by Inductively Coupled Plasma Optical Emission Spectrometry (ICPOES). Therefore, the results of this study show that phocaecin PI80 may be a useful tool for inhibiting L. monocytogenes in seafood products that do not usually undergo adequate heat treatment, whereas the cells of Streptococcus phocae PI80 could be used to control vibriosis in shrimp farming.

  • This study was aimed to establish analytical method of Bi to develop a guideline of the bioequivalence test of tripotassium dicitrato bismuthate (TDB). For this purpose, a simple, specific and sensitive inductively coupled plasma-mass spectrometry (ICP/MS) method were developed and validated in human plasma. Various concentrations of bismuth standard solution (0-25ng/mL) were prepared with distilled water and human blank plasma. To 10mL of the volumetric flasks, 2mL of blank plasma was added with 8ml of distilled water. Bi standard solution was added to prepare the calibration samples and injected into ICP-MS. The plasma samples obtained from volunteers given 3 tablets of bismuth (total 900mg as TDB) were analyzed as described above. As a result, the coefficients of variation were <20% in quantitation limit (0.2 ng/mL) and <15% at the rest of concentrations. The stability test by repeated freezing-thawing cycles showed that the samples were stable only for 24hr. The stability tested for samples with a short-term period of storage at room temperature and pre-treatment prior to the analysis showed very stable over 24hr. In 8 healthy Korean subjects received Denol tablets at the dose of 900mg bismuth, AUC, $C_{max},\;T_{max}$ and half-life $(t_{1/2})$ were determined to be $198.33{\pm}173.78 ng{\cdot}hr/mL,\;64.48{\pm}27.06 ng/mL,\;0.52{\pm}0.21 hr,\;and\;5.15{\pm}2.67 hr$, respectively, from the plasma bismuth concentration-time curves. In conclusion, the method was suitable for the determination of bismuth in human plasma samples and could be applied to bioequivalence test of bismuth tablet.

  • $SrBi_2Ta_2O_9$ thin films were etched at high-density $Cl_2/CF_4/Ar$ in inductively coupled plasma system. The chemical reactions on the etched surface were studied with x-ray photoelectron spectroscopy and secondary ion mass spectrometry. The etching of SBT thin films in $Cl_2/CF_4/Ar$ were chemically assisted reactive ion etching. The maximum etch rate was 1060 Am /min in $Cl_2$(20)/CF_4(20)/Ar(80). The small addition of $Cl_2$ into $CF_4$(20)/Ar(80) plasma will decrease the fluorine radicals and the increase CI radical.   • Hydrogen was produced from water plasma excited in high frequency (HF) inductively coupled tubular reactor. Mass spectrometry was used to monitor gas phase species at various process conditions, Water dissociation rate depend on the process parameters such as ICP power, $H_{2}O$ flow-rate and process pressure, Water dissociation percent in ICP reactor decrease with increase of chamber pressure, while increase with increase of ICP power and $H_{2}O$ flow rate. The effect of $CH_4$ gas addition to a water plasma on the hydrogen production has been studied in a HF ICP tubular reactor. The main roles of $CH_4$ additive gas in $H_{2}O$ plasma are to react with 0 radical for forming $CO_x$ and CHO and resulting additional $H_2$ production. Furthermore, $CH_4$ additives in $H_{2}O$ plasma is to suppress reverse-reaction by scavenging 0 radical. But, process optimization is needed because $CH_4$ addition has some negative effects such as cost increase and $CO_x$ emission.

Safety Profile Assessment and Identification of Volatile Compounds of Krill Eupausia superba Oil and Residues Using Different Extraction Methods

  • In this study, Krill Eupausia superba oil was extracted using different solvents and supercritical carbon dioxide (SC-$CO_2$). During SC-$CO_2$ extraction, the pressure was set at 40 MPa and temperatures ranged from $40^{\circ}C$ to $55^{\circ}C$. We examined the differences in volatile compounds and safety profiles among extraction methods. Volatile compounds were determined using the thermal desorption system integrated with gas chromatography-mass spectrometry (GC-MS). Heavy metal content was analyzed by inductively coupled plasma mass spectrometry (ICP-MS). According to our results 10 volatile compounds were identified in krill sample. After SC-$CO_2$ extraction of oil, the concentrations of volatile compounds decreased, but increased after solvent extraction. In krill, heavy metal concentrations remained within the permissible limit. Moreover, Zn and Fe which have health benefits were detected at high concentrations. During a 90 days storage period at different temperatures, microbial activity was found to be lowest in SC-$CO_2$ extracted residues. Thus, the quality of krill oil and the residues obtained using SC-$CO_2$ extraction was higher and the oil was safer than those obtained using conventional solvent extraction. These results can be applied to the food industry to maintain high quality krill products.

  • The World Health Organization(WHO, 1998) and the United States Environmental Protection Agency (USEPA, 1992) recommended $2{\mu}\;guranium/{\ell}$ in drinking water as a guideline. The Korea Institute for Environmental Research recently reported that the radioactive pollution in ground water was almost negligible In Korea$(1999\~2002)$. Cs were cast into beads(2mm in wet form) and treated with hexamethylene diisocyanate for stability in acidic aqueous solution through cross-linking of the beads surfaces. The removal study was carried out in a static batch system and a flow system. In the static system, a certain amount of sample water was confined in a vessel and beads(dry weight 0.5g) were packed into it in order to adsorb uranium for a certain period of time. Afterwards the remaining uranium in water vessel was determined by inductively coupled plasma mass spectrometry. The effective pH range was 4 to 8. The smaller the size of beads, the better the removal efficiency. Furthemore, the lower the flow rates, the higher the removal efficiency. The results showed that chitosan beads can be effectively used for the removal of uranium contained in water.

  • The solar energy is dramatically increasing as the alternative energy source and the silicon(Si) solar cell are used the most. In this study, the improved process and equipment for the metallurgical refinement of multicrystalline Si were evaluated for the inexpensive solar cell. The planar plane and columnar dendrite aheadof the liquid-solid interface position caused the superior segregation of impurities from the Si. The solidification rate and thermal gradient determined the shape of dendrite in solidified Si matrix solidified by the directional solidification(DS) method. To simulate this equipment, the commercial software, PROCAST, was used to solve the solidification rate and thermal gradient. Si was vertically solidified by the DS system with Stober process and up-graded metallurgical grade or metallurgical grade Si was used as the feedstock. The inductively coupled plasma mass spectrometry (ICP) was used to measure the concentration of impurities in the refined Si ingot. According to the result of ICP and simulation, the high thermal gradient between the two phases wasable to increase the solidification rate under the identical level of refinement. Also, the separating heating zone equipped with the melting and solidification zone was effective to maintain the high thermal gradient during the solidification.
