Study on the Promotion Effect of Ionic Liquid on CH4 Hydrate Formation (이온성 액체를 이용한 메탄 하이드레이트 생성 촉진효과 연구)

  • In this study, we investigated the kinetics of gas hydrate formation in the presence of ionic liquid (IL). Hydroxyethyl-methyl-morpholinium chloride (HEMM-Cl) was chosen as a material for the promotion effect test. Phase equilibrium curve for $CH_4$ hydrate with aqueous IL solution was obtained and its induction time and consumed amount of $CH_4$ gas were also measured. Aqueous solutions containing 20~20,000 ppm of HEMM-Cl was prepared and studied at 70 bar and 274.15 K. To compare the measured results to those of the conventional promoter, sodium dodecyl sulfate was also tested at the same condition. Result showed that the hydrate equilibrium curve was shifted toward higher pressure and lower temperature region. In addition, the induction time on $CH_4$ hydrate formation in the presence of IL was not shown. The amount of consumed $CH_4$ was increased with the whole range of tested concentration of IL and the highest consumption of $CH_4$ happened at 1,000 ppm of HEMM-Cl. HEMM-Cl induced and enhanced the $CH_4$ hydrate formation with a small amount of addition. Obtained result is expected to be applied for the development of technologies such as gas storage and transport using gas hydrates.

Analysis of Degradation Behaviors of Geomembrane by Accelerated Test under UV Exposure Conditions (자외선 노출조건 하에서 가속시험에 의한 지오멤브레인의 분해거동 해석)

  • In this paper the effect of UV (ultraviolet) exposure on HDPE (high density polyethylene)-smooth and f-PP (flexible polypropylene) geomembranes is evaluated under UVB-313 (ultraviolet wavelength 290-315 nm) exposure. Tensile property, melt flow index (MFI), oxidation induction time (OIT), both standard-OIT and high pressure-OIT and Fourier transform infrared spectroscopy/attenuated total reflectance (FTIR/ATR) results are discussed. Although tensile properties of the exposed geomembrane samples remained unchanged, the depletion of antioxidants was found higher for f-PP than for HDPE geomembrane. Arrhenius model by extrapolation was used on the data to predict the antioxidant lifetime to a typical site temperature of $20^{\circ}C$. There was no significant difference between the MFI value of the virgin and UV exposed HDPE geomembrane samples but a decrease in MFI was found in f-PP geomembrane that signifies that crosslinking has occurred. From FTIR spectra, the small peak (near $1750\;cm^{-1}$) observed in the spectrum of UV exposed sample corresponds to a carbonyl (C=O) linkage, which suggests that oxidation has occurred in the polymer structure, and another new band for f-PP between 3100 and $3500\;cm^{-1}$ is attributed to a hydroxyl bond and/or hydroperoxide bond.

Response to Anther and Tissue Cultures of Corn, Pearl Millet and Buckwheat Genotypes (옥수수, 진주조, 메밀의 약 및 조직배양 반응)

  • Anther and/or tissue culture of cross pollinated crops would be very important because it can result in the direct use of haploids or doubled haploids for developing superior hybrids or varieties. The objective of the study was to investigate the response frequencies in anther and/or tissue-cultured hybrids of corn. pearl millet and buckwheat to identify agronomically acceptable germplasm of the crops. 27 crosses of corn inbred lines were evaluated by plating their anthers on N6. MS and Yu-Pei media. Two genotypes of FR1l41/FR16 hybrid cultured on N6 medium and Fla 2BT73/S6013 hybrid cultured on N6 medium responded with one anther producing calli when plated after 5$^{\circ}C$ low temperature treatment for one week. Immature embryos of corn hybrid Suwon 19 responded producing calli that were regenerated to plants at a 8.6 percent success rate. Of the 20 corn hybrids. immature tessels of FR1l41/FR16. B68/A1l6N//KS15. KS16/KS17. GA209/DB578 and SDB126/GA209 crosses responded at a relatively higher success rate producing calli that were regenerated to plants. In tissue culture of elongating culms of pearl millet x Napier grass interspecific hybrid. 2.5-4.0mm long pieces of the culm were good for callus induction resulting in higher success rate. The epicotyl of buckwheat was very good for tissue culture. and the node produced the plants regenerated directly without callus induction on the B5 medium containing I ppm BA and 0.05 ppm IBA. There were great differences in response to anther and/or tissue culture of corn, pearl millet and buckwheat due to genotype x medium and environment interactions.

  • Room-temperature operating CdZnTe(CZT) material is an innovative radiation detector which could reduce the patient dose to one-tenth level of conventional CT (Computed Tomography) and mammography system. The pixel and pixel pitch in the imaging device determine the conversion efficiency of incident X-or gamma-ray and the cross-talk of signal, that is, image quality of detector system. The weighting potential is the virtual potential determined by the position and geometry of electrode. The weighting potential obtained by computer-based simulation in solving Poisson equation with proper boundaries condition. The pixel was optimized by considering the CIE (charge induced efficiency) and the signal cross-talk in CT detector system. The pixel pitch was 1-mm and the detector thickness was 2-mm in the simulation. The optimized pixel size and inter-pixel distance for maximizing the CIE and minimizing the signal cross-talk is about $750{\mu}m$ and $125{\mu}m$, respectively.

Regeneration and the Maturation Induction of Free-Living Gametophytes of a Kelp Saccharina sculpera (Phaeophyceae) (갈조류 개다시마 유리배우체의 재생 및 성숙 유도)

  • Investigation of optimal temperature, irradiance and photoperiod conditions for free-living gametophytes of Saccharina sculpera for natural resources conservation and mass cultivation of endangered species in the eastern coast of Korea. Induction of regeneration and maturation of the free-living gametophytes of S. sculpera were cultured at temperatures (5, 10, 15, 20 and $25^{\circ}C$), irradiance (10, 20, 40, 60 and $80{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and photoperiods (14:10, 12:12 and 10:14 h L:D). The female gametophyte were actively regenerated without reaching sexual maturity under $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D) conditions. In contrast, the conditions for male gametophytes were slightly different at $15^{\circ}C$, $10{\mu}mol\;photons\;m^{-2}\;s^{-1}$ and 12:12 h (L:D). The sexual maturation of female and male gametophytes took place under $15^{\circ}C$, $40{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 14:10 h (L:D) and $10^{\circ}C$, $20{\mu}mol\;photons\;m^{-2}\;s^{-1}$, 10:14 h (L:D), respectively. These results provide basic information for controlling the regeneration and maturation of free-living gametophytes for conservation and utilization of S. sculpera.

Mushroom growth and cultivation environment at cultivation house of vinyl bag cultivation Shiitake mushroom on high-temperature period (고온기 표고 톱밥재배용 재배사 내의 환경 제어시스템과 버섯생육 온도)

  • Although sawdust cultivation of shiitake (Lentinula edodes) is becoming more common, it is insufficiently competitive in spring and autumn, the best time to breed shiitake. Thus, it is urgently needed to develop a technique for all year round cultivation of shiitake using mushroom growing beds. In the present study, the temperature changes according to the location of shiitake cultivation facilities were investigated. We confirmed that a refrigerator, an air conditioner, triple membranes, shiitake cultivation beds, fog nozzles which were installed in the shiitake cultivation facilities play an important role in keeping the low temperature. Bag cultivation of shiitake was tested in temperature variation from $14^{\circ}C$ to $29^{\circ}C$ with a $3^{\circ}C$ interval to know its cultivating temperature range in hot summer season. In summary, the sawdust cultivation of shiitake is possible when the temperature difference between top and bottom is maintained below $1^{\circ}C$. And the temperature of the shiitake cultivation facilities should be maintained below $23^{\circ}C$ in the induction period for fruitbody formation.

Physiological Function of Isoflavones and Their Genetic and Environmental Variations in Soybean (콩 Isoflavone의 생리활성 기능과 함량 변이)

  • Soyfoods have potential roles in the prevention and treatment of chronic diseases, most notably cancer, osteoporosis, and heart disease. There is evidence that carcinogenesis are supressed by isolated soybean derived products in vivo such as a protease inhibitor, phytic acid, saponins and isoflavones. It is believed that supplementation of human diets with soybean products markedly reduces human cancer mortality rates. Especially, recent papers recognize the potential benefit of soybean isoflavone components for reducing the risk of various cancers. Isoflavones exhibit a multitude of medicinal effects that influence cell growth and regulation, which may have potential value in the prevention and treatment of cancer. In addition to potential biological effects, soybean isoflavones have the important physiological functions such as the induction of Bradyrizobium japonicum nod genes and the responses of soybean tissues to infection by Phytophthora megasperma as well as biochemical activities such as antifungal and antibacterial actions. Genistin, daidzin, glycitin and their aglycone (genistein, daidzein, glycitein) are the principal isoflavones found in soybean. Malonyl and acetyl forms have also been detected but they are thermally unstable and are usually transformed during the processing in glucoside form. Most soy products, with the exception of soy sauce, alcohol-extracted soy protein concentrate, and soy protein isolate, have total isoflavone concentrations similar to those in the whole soybean. Soybean-containing diets inhibit mammary tumorigenesis in animal models of breast cancer, therefore, it is possible that dietary isoflavones are an important factor accounting for the lower incidence and mortality from breast cancer. Of the total soybean seed isoflavones, $80\~90\%$ were located in cotyledons, with the remainder in the hypocotyls. The hypocotyls had a higher concentrations of isoflavones on a weight basis compared with cotyledons. Isoflavone contents were influenced by genetics, crop years, and growth locations. The effect of crop year had a greater impact on the isoflavone contents than that of location. The climate condition might be the attribution factor to variation in isoflavone contents. Also, while the isoflavone content of cotyledons exhibited large variations in response to high temperature during seed development, hypocotyls showed high concentration in isoflavone content. So, it is concluded that one of the factors affecting isoflavone content in soybean seeds is temperature during seed development. High temperature, especially in maturity stage, causes lower isoflavone content in soybean seed. It is also suggested that there may exist a different mechanism to maintain isoflavone contents between cotyledon and seed hypocotyls. In a conclusion, soy foods may be able to have a significant beneficial impact on public health.

Characteristics of Mayonnaise Prepared with Palm Oil (팜유의 사용에 의한 마요네즈의 품질 특성)

  • To know the availability of double fractionated palm olein (DFPL) for vegetable oil in commercial mayonnaise preparation, oxidative stability and stability to cold test of DFPL, soybean oil and blended oils (blended soybean oil with DFPL) were tested. Mayonnaises with these oils were prepared and then emulsion stability at low temperature $(-5^{\circ}C)$ were compared. The oxidative stability of vegetable oil by Rancimat test showed that induction time of DFPL (26.9 hr) was longer when compared with soybean oil (13.4 hr), and became longer with increase of DFPL ratio in the blended oil. Emulsion stability of mayonnaises at low temperature $(-5^{\circ}C)$ was decreased with the increase of DFPL ratio in the blended oil. But, mayonnaise with blended oil of below 20% DFPL was comparable to that with soybean oil only. Among quality characteristics of mayonnaises with soybean oil and blended oil (soybean oil 85% plus DFPL 15%) the latter showed stronger oxidative stability and less flavor reversion during high temperature treatment. This result suggested that the possibility of DFPL to substitute for vegetable oil in the preparation of commercial mayonnaise.

Seed Germination and Seedling Emergence of Indian jointvetch (Aeschynomene indica L.) in Different Conditions (자귀풀 종자의 발아 및 출아에 미치는 환경요인)

  • Several experiments were conducted in growth chambers and a greenhouse to determine the influence of various environmental factors on seed germination and seedling emergence of Indian jointvetch. Fully matured seeds of Indian jointvetch germinated only 42%. The germination percent increased as the storage temperature increased with time. More than 90% seeds germinated when the seeds were kept at $40^{\circ}C$ for seven months, but germination was 58.9 and 55.1% when kept at 25 and $4^{\circ}C$, respectively. Non-dormant seeds of Indian jointvetch germinated 91.1 and 92.4% at 30 and $30/25^{\circ}C$, respectively. Germination percent increased with increasing both prethermal temperature and time. The prethermal temperature of $90^{\circ}C$ for 40 minutes was the best for maximum germination (94.5%). Germination and growth of Indian jointvetch tended to decrease slightly until -0.3 MPa osmotic potential (water stress induction) and then declined drastically and the seeds did not germinate at below -0.5 MPa osmotic potential. Indian jointvetch seems to grow well in moist and flooding conditions since emergence and growth of seedling increased with increasing soil moisture content and the water level.

Influence of the Differences in Altitude during Raising Seedlings on Daughter Plant Characteristics and Subsequent Strawberry Production (육묘 기간중의 고도 차이가 자묘의 특성 및 딸기 수량에 미치는 영향)

  • The nursery plant quality and flower bud induction of new strawberry cultivars, 'Maehyang' and 'Seolhyang' in forcing culture were evaluated in the highland and lowland region. In order to produce daughter plants, the new cultivars were grown in the open field located at both highland (Daekwallyung, above 800 m sea level) and lowland (Gangneung, above 20 m sea level) region, respectively. The average air temperature at highland during nursery plant propagation period was $5.3^{\circ}C$ lower than those at lowland. The number of daughter plants produced at lowland was 2 times as high as those of highland, presumably due to the higher air temperature. Anthracnose incidence rates of runner plants produced at lowland were 4 to 7% as high as those in highland. Mean temperature in the highland ($23.5^{\circ}C$) during flower bud differentiation treatment was $5.1^{\circ}C$ lower than that in the lowland ($28.6^{\circ}C$). Seedlings produced in highland showed higher C/N ratio and lower T/R ratio compared to those produced in lowland. The average flower bud formation date of the daughter plants grown in highland were advanced by 30 days compared to the date in lowland. Accordingly, highland was supposed to be appropriate region for raising seedlings in forcing culture of the new strawberry cultivars of 'Maehyang' and 'Seolhyang'.
