TRAIL Based Therapy: Overview of Mesenchymal Stem Cell Based Delivery and miRNA Controlled Expression of TRAIL

  • Rapidly increasing number of outstanding developments in the field of TRAIL mediated signaling have revolutionized our current information about inducing and maximizing TRAIL mediated apoptosis in resistant cancer cells. Data obtained with high-throughput technologies have provided finer resolution of tumor biology and now it is known that a complex structure containing malignant cells strictly coupled with a large variety of surrounding cells constitutes the tumor stroma. Utility of mesenchymal stem cells (MSCs) as cellular vehicles has added new layers of information. There is sufficient experimental evidence substantiating efficient gene deliveries into MSCs by retroviral, lentiviral and adenoviral vectors. Moreover, there is a paradigm shift in molecular oncology and recent high impact research has shown controlled expression of TRAIL in cancer cells on insertion of complementary sequences for frequently downregulated miRNAs. In this review we have attempted to provide an overview of utility of TRAIL engineered MSCs for effective killing of tumor and potential of using miRNA response elements as rheostat like switch to control expression of TRAIL in cancer cells.

In Vivo Evaluation of Curcumin-loaded Nanoparticles in a A549 Xenograft Mice Model

  • Curcumin (Cum) has been reported to have potential chemo-preventive and chemotherapeutic activity through influencing various processes, inducing cell cycle arrest, differentiation and apoptosis in a series of cancers. However, the poor solubility of Cum limits its further applications in the treatment of cancer. We have previously reported Cum-loaded nanoparticles (Cum-NPs) prepared with amphilic methoxy poly(ethylene glycol)-polycaprolactone (mPEG-PCL) block copolymers. The current study demonstrated superior antitumor efficacy of Cum-NPs over free Cum in the treatment of lung cancer. In vivo evaluation further demonstrated superior anticancer effects of Cum-NPs by delaying tumor growth compared to free Cum in an established A549 transplanted mice model. Moreover, Cum-NPs showed little toxicity to normal tissues including bone marrow, liver and kidney at a therapeutic dose. These results suggest that Cum-NPs are effective to inhibit the growth of human lung cancer with little toxicity to normal tissues, and could provide a clinically useful therapeutic regimen. They thus merit more research to evaluate the feasibility of clinical application.

알루미늄 전극의 용출에 따른 해성점토의 개량에 관한 실험적 연구 (A Experimental Study on Improvement of Marine Clay through the Electrolytic Leaching Effect in Aluminum Electrode)

  • In this study, aluminum electrodes were put in marine clay which was taken from the south coast in Korea to increase the undrained shear strength by inducing the densification and cementation between clay particles and precipitates which were developed by electric decomposition in an electrode. To raise the cementation rate and reduce treatment time, high electric current (2.5A) was applied in each electrode at a semi-pilot scale soil box with marine clay. After the tests, the undrained shear strength was measured at designated points using a static cone penetration test device and sampling was conducted simultaneously in order to measure water content, pH and electric conductivity which would be the key for configuring the cementation effects indirectly. In the results of electric decomposition in aluminum electrode, the measured shear strength was increased considerably compared to the initial shear strength because of the cementation effect between iron ions and soil particles.

대학생의 결혼의식, 원가족 건강성 및 성의식에 대한 조사연구 (Marriage Outlook, Health of the Family of Origin and Sexual Awareness among University Students)

  • Purpose: This study was conducted to determine the predictors of marriage outlook for university students in Korea. Method: A convenience sampling method was used and 438 university students were studied for final analysis. Data collection was conducted through the use of questionnaires which were constructed to include Marriage outlook scale, Family-of-origin Scale-55 and Sexual Knowledge & Attitude Test. Results: Marriage outlook for university students didn't demonstrate a positive point of view. There were positive correlations between marriage outlook, health of the family of origin and sexual awareness. The significant factors influencing marriage outlook of university students were current dating, health of the family of origin, gender, and religion. These factors explained 10.6% of marriage outlook of university students. Conclusions: These results support that special programs are needed in inducing a change in marriage outlook to overcome the low birthrate of Korea. These programs need to consider personal factors including the health of the family of origin, gender and religion.

Stenver's 법을 이용한 인공와우관 환자의 촬영에 관한 연구 (Stenver's Radiographic Assessment of the Multichannel Cochlear Implant)

  • To assess the new multichannel cochlear implant by radiography in Stenver's projection, because MRI generates artifacts, inducing an electrical current and causing device magnetization. CT is relatively expensive and the metal electrodes scatter the image. Multichannel cochlear implant insertion using the multichannel cochlear implant device. Patients underwent postoperative radiography of their implants. The radiographs were obtained in a Stenver's. The insertion depth of the implant was measured on the radiographs and the results were correlated with the surgical results of insertion depth and with audiometric tests. Patients a correct inserted electrode was found, while in patient complications concerning the electrode were noticed. Radiographs in the Stenver's projection are sufficient for the postoperative assessment of the multichannel cochlear implant device and an exact evaluation of the insertion depth.

  • The current vision-based approaches for emotion recognition, such as facial expression analysis, have many technical limitations in real circumstances, and are not suitable for applications that use them solely in practical environments. In this paper, we propose an approach for emotion recognition by combining extrinsic representations and intrinsic activities among the natural responses of humans which are given specific imuli for inducing emotional states. The intrinsic activities can be used to compensate the uncertainty of extrinsic representations of emotional states. This combination is done by using PRMs (Probabilistic Relational Models) which are extent version of bayesian networks and are learned by greedy-search algorithms and expectation-maximization algorithms. Previous research of facial expression-related extrinsic emotion features and physiological signal-based intrinsic emotion features are combined into the attributes of the PRMs in the emotion recognition domain. The maximum likelihood estimation with the given dependency structure and estimated parameter set is used to classify the label of the target emotional states.

Formation of iron oxides from acid mine drainage and magnetic separation of the heavy metals adsorbed iron oxides

  • There are a few thousand abandoned metal mines in South Korea. The abandoned mines cause several environmental problems including releasing acid mine drainage (AMD), which contain a very high acidity and heavy metal ions such as Fe, Cu, Cd, Pb, and As. Iron oxides can be formed from the AMD by increasing the solution pH and inducing precipitation. Current study focused on the formation of iron oxide in an AMD and used the oxide for adsorption of heavy metals. The heavy metal adsorbed iron oxide was separated with a superconducting magnet. The duration of iron oxide formation affected on the type of mineral and the degree of magnetization. The removal rate of heavy metal by the adsorption process with the formed iron oxide was highly dependent on the type of iron oxide and the solution pH. A high gradient magnetic separation (HGMS) system successfully separated the iron oxide and harmful heavy metals.

Mechanistic Studies of Cyclin-Dependent Kinase Inhibitor 3 (CDKN3) in Colorectal Cancer

  • Colorectal cancer is one of the most severe subtypes of cancer, and has the highest propensity to manifest as metastatic disease. Because of the lack of knowledge of events that correlate with tumor cell migration and invasion, few therapeutic options are available. The current study aimed to explore the mechanism of colorectal cancer in hope of identifying the ideal target for future treatment. We first discovered the pro-tumor effect of a controversial cell cycle regulator, cylin-dependent kinase inhibitor 3 (CDKN3), which is highly expressed in colorectal cancer, and the possible related signaling pathways, by bioinformatics tools. We found that CDKN3 had remarkable effects in suppressing colorectal cancer cell proliferation and migration, inducing cell cycle arrest and apoptosis in a colorectal cancer cell line, SW480 cells. Our study, for the first time, provided consistent evidence showing overexpression of cell cycle regulator CDKN3, in colorectal cancer. The in vitro studies in SW480 cells revealed a unique role of CDKN3 in regulating cellular behavior of colorectal cancer cells, and implied the possibility of targeting CDKN3 as a novel treatment for colorectal cancer.

Novel Scanning Tunneling Spectroscopy for Volatile Adborbates

  • Reactive or unstable adsorbates are often difficult to study spectroscopically. They may have, for instance, resonance states lying close to the Fermi level, inducing them to desorb or decompose by the probe itself, low-energy tunneling electrons. In order to overcome this limitation, we developed a novel method, which we call x-ramp scan. The method sweeps the bias voltage, with the simutaneous scan along the imaging direction, in a constant current mode. This mapping yields the tip-height variation as a function of bias, or Z(V), at nominally always fresh surface. We applied this method to the investigation of methanol-induced molecular features, attributed to methoxy, found on NiAl(110) surface. These were produced by methanol molecules deposited by a pulse injection method onto the metallic surface. Our study shows adsorbed methoxy are very reactive to the bias voltage, rendering the standard spectroscopy useless. Our new x-ramp scan shows that the decomposition of adsorbates occurs at the sample bias of 3.63 V, and proceeds with the lifetime of a few milliseconds. 전침자극(電針刺戟)이 염소의 제일(第一) 및 사위운동(四胃運動)에 미치는 영향 (Effects of Electroacupuncture on Motility of the Rumen and Abomasum of Goats)

  • Ruminal and abomasal motility after electroacupuncture was investigated in normal goats and in goats with induced hypocalcaemia. Electroacupuncture was performed with a current of 1 volt and 3Hz at the acupoints of Tien Ping(positive) and Pai Hui(negative) for 30 minutes. The adequate range of electroacupuncture stimulation for goats was 1 to 2 volt and 2 to 3 Hz. After electroacupuncture stimulation in normal goats, rates of rumen and abomasal contractions were not changed or were slightly increased, but amplitudes of rumen and abomasal motility markedly increased. The effects of electroacupuncture stimulation lasted from about 10 minutes to 30 minutes after the end of electroacupuncture. The amplitudes of rumen motility were increased by the administration of bethanechol chlorids in goats. Ruminal contractions were not influenced by electroacupuncture following atropine sulphate. The rates of rumen and abomasal movements were not changed, but its amplitudes were reduced by inducing hypocalcaemia in goats. Motilily of the rumen and abomasum was not accelerated by electroacupuncture in goats with induced hypocalcaemia.
