  • Nitric oxide (NO) produced in large amounts by inducible nitric oxide synthase (iNOS) is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. Inhibitors of iNOS, thus, may be useful candidate for the treatment of inflammatory diseases accompanied by the overproduction of NO. We prepared alcoholic extracts of woody plants and screened the inhibitory activity of NO production in lipopolysaccharide (LPS)-activated macrophages after the treatment of these extracts. LPS로 유도된 RAW 264.7 세포에 대한 조록나무 잎 Biorenovation 추출물의 항염증 활성 (Anti-inflammatory effect of Distylium racemosum leaf biorenovate extract in LPS-stimulated RAW 264.7 macrophages cells)

  • 조록나무는 제주도 및 일본 혼슈 이남, 중국 동남부, 타이완 등에 분포하는 조록나무과의 상록 교목으로, 항산화 및 tyrosinase, elastase의 억제에 효과적인 것으로 알려져 있지만 NO에 대한 억제 효능은 미미한 것으로 보고되었다. 이에 본 연구는 조록나무 잎 추출물(DL)에 biorenovation 생물 전환 기법을 적용하여 항 염증 활성을 증진 시키고자 수행되었다. 이들의 활성은 LPS로 자극된 RAW264.7 염증 모델에서 평가 되었으며 NO, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) 및 전 염증성 사이토카인에 대한 억제 실험이 수행되었다. 그 결과, biorenovation을 적용한 조록나무 잎 추출물(DLB)는 독성이 없는 농도에서 DL대비 향상된 NO와 prostaglandin E2 억제효능을 나타내었으며, 이들의 합성 효소인 iNOS 및 COX-2의 발현에도 유의한 억제 경향을 나타내었다. 또한 대표적인 전 염증성 사이토 카인인 tumor necrosis factor-α, Interleukin 6, Interleukin-1β 에서도 향상된 억제 효능을 확인 하였다. 이러한 결과를 근거로 우리는 biorenovation을 통해 DL의 항염증 효능이 개선될 수 있으며, DLB가 효과적인 천연 항염증 소재로 적용될 수 있음을 제시한다.

  • The nitric oxide (NO) formation and intrinsic nitrosation may be involved in the possible mechanisms of liver fluke-associated carcinogenesis. We still do not know much about the responses of inducible NO synthase (iNOS) induced by Clonorchis sinensis infection. This study was conducted to explore the pathological lesions and iNOS expressions in the liver of mice with different infection intensity levels of C. sinensis. Extensive periductal inflammatory cell infiltration, bile duct hyperplasia, and fibrosis were commonly observed during the infection. The different pathological responses in liver tissues strongly correlated with the infection intensity of C. sinensis. Massive acute spotty necrosis occurred in the liver parenchyma after a severe infection. The iNOS activity in liver tissues increased, and iNOS-expressing cells with morphological differences were observed after a moderate or severe infection. The iNOS-expressing cells in liver tissues had multiple origins.

Yomogin, an Inhibitor of Nitric Oxide Production in LPS-Activated Macrophages

  • In activated macrophages the inducible form of nitric oxide synthase (i-NOS) generates high amounts of toxic mediator, nitric oxide (NO) which contributes to the circulatory failure associated with septic shock. A sesquiterpene lactone compound (yomogin) isolated from medicinal plant Artemisia princeps Pampan inhibited the production of NO in LPS-activated RAW 264.7 cells by suppressing i-NOS enzyme expression. Thus, yomogin may be a useful candidate for the development of new drugs to treat endotoxemia and inflammation accompanied by the overproduction of NO.

말 서골코기관에서 protein kinase C 및 nitric oxide synthase의 면역조직학적 관찰 (Immunohistochemical localization of protein kinase C and nitric oxide synthase in the vomeronasal organ of the horse)

  • The expression of protein kinase C(PKC) isoforms and nitric oxide synthase (NOs) isoforms was studied in the equine vomeronasal organ(VNO), a pheromone receptor organ, using immunohistochemistry. All PKC isoforms including PKC $\alpha$, ${\beta}I$, $\delta$, and $\theta$ were detected in the supporting cells, sensory receptor cells, and basal sensory epithelial cells, while constitutive PKC $\alpha$ and ${\beta}I$ were stained more intensely than novel PKC $\delta$ and ${\theta}$. There was also a varying degree of immunostaining for PKCs in the glandular acini and VNO nerve. Constitutive neuronal and endothelial NOSs, and inducible NOS were detected in the VNO sensory epithelia. There was intense immunoreactivity for endothelial NOS in the VNO sensory epithelia but weak reactivity for neuronal NOS, while inducible NOS showed little immunoreactivity in the adjacent section. These findings suggest that both PKCs and NOSs may be involved in the process of pheromone reception in the horse. Constitutive isoforms of these enzymes may play a more important role in signal trasduction in the VNO of the horse.

  • Objectives : Sam-Myo-Whan(SMW) has been known traditional prescription with anti- anthritis activities. We investigated inhibitory effects of SMW on lipopolysaccharide (LPS)-induced nitric oxide(NO), $TNF-{\alpha}$ and inducible nitric oxide synthase(iNOS) production from RAW264.7 cells and BV-2 Microglia cells. Methods : SMW, which had been extracted with 70% MeOH, concentrated and freeze-dried was used for this experiment. After BV2 mouse brain macrophages and RAW264.7 mouse peritoneal macrophages were pretreated with increasing concentrations of SMW extract for 30min, and then activated with LPS. To investigate cytotoxicity of SMW extract, cell viability was measured by MTT assay. NO production was measured in each culture supernatant by Griess reaction. mRNA expression of iNOS in two type cells was investigated by RT-PCR. $TNF-{\alpha}$ production was measured in each culture supernatant by ELISA. Results : SMW extract significantly inhibited LPS-induced NO and $TNF-{\alpha}$ production in BV2 cells and RAW264.7 cells dose-dependently. SMW extract also greatly suppressed mRNA expression of iNOS in both type cells activated with LPS. Conclusion : These data suggests that SMW extract may have an anti-inflammatory effect through the inhibition of iNOS expression.

Parthenolide Inhibits Ovalbumin-Induced cyclooxygenase-2 and Inducible Nitric Oxide Synthase Expression

  • In recent years, egg allergy has been raised as the most prevalent food hypersensitivity in the pediatric population. One of the major egg allergens is ovalbumin (OVA) which is the major protein in egg white, comprising 54% of its total protein content. Parthenolide isolated from Tanacetum partheniumb has been used to treat many chronic diseases for many years. In the present report, we present biochemical evidence that parthenolide inhibits the nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation induced by OVA. Parthenolide also inhibits OVA-induced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. These data suggest new approaches for the efficient alleviation of the allergic symptoms.

Isoliquiritigenin (ILG) is an active component present in licorice and has been used to treat many chronic diseases for many years. In this study, we present biochemical evidence that ILG inhibits OVA-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation. ILG also inhibits OVA-induced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. These data suggest that ILG has anti-allergy effects.

Inhibition of nitric oxide and TNF-$\alpha$ production by propenone compound through blockaded of NF-$\kappa$B activation in cultured murine macrophages

  • Lipopolysaccharide (LPS)-stimulated macrophages produced a large amounts of nitric oxide (NO) by inducible nitric oxide synthase (iNOS). This is an important mechanism in macrophages-induced septic shock and inflammation. In the present study, we tested a synthetic propenone compound, l-furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) for its ability to inhibit the production of tumor necrosis factor-a (TNF-$\alpha$) and an inducible enzyme, iNOS, in the LPS-stimulated murine macrophage-like cell line, Raw264.7. FPP-3 consistently inhibited nitric oxide (NO) and TNF-$\alpha$ production in a dose dependent manner, with $IC_50$> values of 10.0 and 13.1 $\mu$M, respectively. (omitted)

Anti-inflammatory Activity of Veronica peregrina

  • Veronica peregrina (Scrophylariaceae) has been widely used as a Korean traditional medicine for the treatment of various pathological conditions including infection, hemorrhage and gastric ulcer. In the current study, we investigated the inhibitory effect of methanolic extracts of V. Peregrina (VPM) on the LPS-mediated nitric oxide (NO) production in mouse (C57BL/6) peritoneal macrophages. NO production was significantly down-regulated by the treatment of VPM dose dependently. To evaluate the mechanism of the inhibitory action of VPM on NO production, we performed iNOS enzyme activity assay and checked the change of inducible nitric oxide synthase (iNOS) levels by Western blotting. Although VPM did not affect iNOS enzyme activity, iNOS protein expression was attenuated by VPM indicating VPM inhibits NO production via suppression of iNOS enzyme expression. In addition, VPM attenuated the expression of another pro-inflammatory mediator such as cyclooxygenase-2 (COX-2) in a dose dependent manner. We also found that VPM can reduce trypsin-induced paw edema in mice. Based on this study, we suggest that V. peregrina may be beneficial in diseases which related to macrophage-mediated inflammatory disorders.
