  • The increased level of glucose, ketone bodies, non-esterified fatty acids and lactate in blood, decrease of glycogen content, phosphofructokinase activities and glucokinase activity and the increased level of glucose-6-phosphatase activity in the liver of streptozotocin injected rats were significantly moderated by ginseng saponin administration. It is not likely, however, that the hypoglycomic action of ginseng saponin might be due to their direct action on enzyme activities, since the saponin effect of servers enzymes in vitro was not enough to explain such an appreciable hypoglycemic activity of the saponin in streptozotocin induced diabetic rats, for which much work have to be done.

  • This study was undertaken to find effects of ethanol extracts from Bombyx mori on the hyperglycemia induced by streptozotocin in rats. Blood glucose level of rats increased depending on experimental days after streptozotocin treatment. But there was no difference in blood glucose level between the rats injected with ethanol extracts of B. mori normal rats. (omitted)

Rutin alleviated lipopolysaccharide-induced damage in goat rumen epithelial cells

  • Objective: Rutin, also called vitamin P, is a flavonoids from plants. Previous studies have indicated that rutin can alleviate the injury of tissues and cells by inhibiting oxidative stress and ameliorating inflammation. There is no report on the protective effects of rutin on goat rumen epithelial cells (GRECs) at present. Hence, we investigated whether rutin can alleviate lipopolysaccharide (LPS)-induced damage in GRECs. Methods: GRECs were cultured in basal medium or basal medium containing 1 ㎍/mL LPS, or 1 ㎍/mL LPS and 20 ㎍/mL rutin. Six replicates were performed for each group. After 3-h culture, the GRECs were harvested to detect the relevant parameters. Results: Rutin significantly enhanced the cell activity (p<0.05) and transepithelial electrical resistance (TEER) (p<0.01) and significantly reduced the apoptosis rate (p<0.05) of LPS-induced GRECs. Rutin significantly increased superoxide dismutase, glutathione peroxidase, and catalase activity (p<0.01) and significantly decreased lactate dehydrogenase activity and reactive oxygen species and malondialdehyde (MDA) levels in LPS-induced GRECs (p<0.01). The mRNA and protein levels of interleukin 6 (IL-6), IL-1β, and C-X-C motif chemokine ligand 8 (CXCL8) and the mRNA level of tumor necrosis factor-α (TNF-α) and chemokine C-C motif ligand 5 (CCL5) were significantly increased in LPS-induced GRECs (p<0.05 or p<0.01), while rutin supplementation significantly decreased the mRNA and protein levels of IL-6, TNF-α, and CXCL8 in LPS-induced GRECs (p<0.05 or p<0.01). The mRNA level of toll-like receptor 2 (TLR2), and the mRNA and protein levels of TLR4 and nuclear factor κB (NF-κB) was significantly improved in LPS-induced GRECs (p<0.05 or p<0.01), whereas rutin supplementation could significantly reduce the mRNA and protein levels of TLR4 (p<0.05 or p<0.01).   • The effect of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) on the intracellular $Ca^{2+}$ level was studied in NIH3T3 fibroblast cells. A reduction of the intracellular $Ca^{2+}$ level was observed after exposure to 300 ${\mu}m$ MNNG. However, the intracellular level of $IP_3$, a well-known regulator of $Ca^{2+}$ release from internal storage, was not changed by MNNG treatment. Instead, a reduction of the intracellular NAD level was observed. NAD as well as $IP_3$ stimulated intracellular $Ca^{2+}$ release from permeabilized cells. The treatment of 3-aminobenzamide, which inhibited the MNNG-induced reduction of the NAD level, also prevented the MNNG-induced decrease of the $Ca^{2+}$ level. Our data suggest that MNNG reduces the intracellular $Ca^{2+}$ level by NAD depletion in NIH3T3 cells.

Creep-permeability behavior of sandstone considering thermal-damage

  • This investigation presented conventional triaxial and creep-permeability tests on sandstones considering thermally-induced damage (TID). The TID had no visible effects on rock surface color, effective porosity and permeability below $300^{\circ}C$ TID level. The permeability enlarged approximately two orders of magnitude as TID increased to $1000^{\circ}C$ level. TID of $700^{\circ}C$ level was a threshold where the influence of TID on the normalized mass and volume of the specimen can be divided into two linear phases. Moreover, no prominent variations in the deformation moduli and peak strength and strain appeared as TID< $500^{\circ}C$ level. It is interesting that the peak strength increased by 24.3% at $700^{\circ}C$ level but decreased by 11.5% at $1000^{\circ}C$ level. The time-related deformation and steady-state creep rate had positive correlations with creep loading and the TID level, whereas the instantaneous modulus showed the opposite. The strain rates under creep failure stresses raised 1-4 orders of magnitude than those at low-stress levels. The permeability was not only dependent on the TID level but also dependent on creep deformation. The TID resulted in large deformation and complexity of failure pattern for the sandstone.

  • Fertility has been declined since 1966 (see Table 1). This fertility reduction was duely caused by age at first marriage, induced abortion and contraceptive practice which has been largely increased in recent years. Although tbe proximate determinant such as induced abortion, age at marriage and breastfeeding can and do have an effect on fertility, the principal cause of the reduction in fertility in Korea during the fertility transition can be supported by correlation between level of fertility and contraceptive prevalence (See Fig. 4). Taking a regression equation between fertility (TER. Y) and prevalence level (X), the total fertility rate in 1984 was estimated as 1.9 and 2.1 based on lenear and expotential function shown as follow; $Y_1$=5.709-0.0549 X and $Y_2$_______80________ 1+e2.433+0.017X ($R^2$=O. 93) ($R^2$=0. 96) Where $Y_1$ and $Y_2$ denote total fertility rates obtained through two equations respectivelly. The peak of contraceptive prevalence was assumdd as 80 percent which is almost upper limit in human society. On the other hand, an observed value of 1984 fertility level obtained from five month period shows 2.1 which is coincident with logistic fitting after the adjustment of response error assumed around 10 to 20 percent, At any rate, fertility of Korean women will have been reached replacement level (2.1) by 1985.Thus policy for family planning program must be reviewed toward the direction of integrated approach particularly with MCH program inasmuch as fertility in Korea has already shown population replacement level that require more good quality of service in family planning and their There must be an advanced level of fertility in Korea because wide use of contraception and induced life abortion and age at marriage will effect modern fertility which shows up and down trend between 2.1 and 1.5 in general.

Hepatoprotective Effects of the Water Extract of Protaetia brevitarsis Larva Against Carbon Tetrachloride-Induced Liver Injury in Rats (사염화탄소에 의해 유도된 흰쥐의 간장해에 미치는 지잠의 보호효과)

  • Hepatoprotective effects of the water extract of Protaetia brevitarsis larva (PB) was investigated in carbon tetrachloride ($CCI_{4}$) treated male Sprague-Dawley rats. PB administration protected rats against ALT, AST and LDH elevations induced by $CCI_{4}$, as well as the severity of liver damage. PB recovered the decrease in serum level of high-density lipoprotein-cholesterol and the increase in serum level of low-density lipoprotein-cholesterol induced by $CCI_{4}$. In histopathological observation, massive fatty change and necrosis in the centrilobular area, degenerative change including pyknosis of nucleus and swelling of parenchymal cell induced by $CCI_{4}$ were clearly protected by PB. These histopathological findings paralleled with the serum biochemical results. The present results demonstrated that the water extract of PB may have the hepatoprotective effect against $CCI_{4}$-induced liver damage in vivo.

Effects of Oxidative Stress Induced by Diquat on Arginine Metabolism of Postweaning Pigs

  • A total of 16 crossbred post-weaning pigs (10.64${\pm}$0.27 kg BW) were individually penned and assigned to one of two treatments to investigate the influences of diquat-induced oxidative stress on performance and arginine metabolism. Pigs in the oxidative stress group were injected intra-peritoneally with 10 mg/kg BW of diquat, while the control group were injected with isotonic saline. All pigs were fed ad libitum. The experiment lasted for 7 days. The results indicated that compared with control treatment, oxidative stress induced by diquat significantly decreased average daily gain, intake and feed conversion. The treatment decreased activities of antioxidant enzymes, increased concentration of malondialdehyde in plasma, increased cationic amino acid transporter-1 mRNA level and activity of ornithine aminotransferase and concentrations of arginine and citrulline in the jejunum, decreased the concentrations of arginine in plasma and kidney, and decreased induced nitric oxide synthase mRNA level. It is concluded that oxidative stress induced by diquat can influence absorption and metabolism of arginine and consequently modify the requirement of arginine for post-weaning pigs.

Influences of Adrenergic Blockades on the Hyperglycemic Action of Clonidine (Clondine의 과혈당작용(過血糖作用)에 미치는 교감신경차단약물(交感神經遮斷藥物)의 영향(影響))

  • In this paper, the influences of adrenergic blockades; propranolol and phenoxybenzamine on the changes of hyperglycemic action, hepatic glycogen content, and brain norepinephrine (NE) content induced by clonidine were investigated in the male mice. The results obtained were summarized as follows: 1) Blood glucose level was significantly increased by clonidine $(30{\mu}g/kg)$. The increase of blood glucose level induced by clonidine was not affect by the propranolol (10mg/kg) pretreatment, but significantly inhibited by the phenoxybenzamine (10mg/kg) pretreatment. 2) Hepatic glycogen content was moderately inhibited by clonidine. The decrease of hepatic glycogen content induced by clonidine was not affected by the propranolol and phenoxybenzamine pretreatment. 3) Brain NE content was significantly increased in 30 minutes and 60 minutes after clonidine treatment. The increase of brain NE content induced by clonidine was significantly inhibited by the phenoxybenzamine pretreatment. The increase of train NE content induced in 90 minutes and 120 minutes after clonidine treatment was more markedly increased by the propranolol pretreatment.

Significance of Feeding Induced Hypovolemia in Feed Intake Control of Goats Fed on Alfalfa Hay

  • The objective of this study was to examine whether feeding induced hypovolemia (decrease in plasma volume) acts on the regulation of feed intake in goats fed on dry forage. In order to prevent feeding induced hypovolemia, a 2 h intravenous infusion (16-18 ml/min) of isotonic mannitol solution was begun 1 h prior to feeding and continued until 1 h after the start of the 2 h feeding period. The intravenous infusion of isotonic mannitol solution (MI) decreased plasma osmolality by 1.0%, plasma total protein concentration by 4.2% and hematocrit by 5.9%, respectively. In comparison with no infusion (NI), MI significantly decreased thirst level by approximately 13%. At the completion of the 2 h feeding period, cumulative feed intake had been increased by 43% by MI. In conclusion, feeding induced hypovolemia in goats fed on dry forage increased thirst level more than the increase in plasma osmolality did. The results demonstrate that feeding induced hypovolemia is one of the factors controlling feed intake in goats fed on dry forage.
