수서생태계 납오염에 대한 생체지표로서 Maceobrachium nipponnese내 Na+/K+ATPase의 활용 (Use of Na+/K+ATPase in Maceobrachium nipponnese as a biomaker of lead pollution in aquatic ecosystem)

  • Lead is one of the most toxic metal and is detectable in practically all phases of environment and in all biological system. Transport, industrial and domestic waste products are the main sources of this pollutant. Ingested lead is rapidly absorbed and widely distributed throughout the body, causing extensive tissue damage. In this study, we chose the freshwater decapods Maceobrachium nipponnese as a sensitive indicator organism for environmental pollution. In order to investigate the possibility in use of $Na^+/K^+ATPase$ activity as a biomarker of lead pollution, we tested the acute toxicity of lead to Maceobrachium nipponnese. The $LC_{50}(96hr)$ value for lead in Maceobrachium nipponnese was found to be $446{\mu}g/L$ with the 95% confidence limits. The lead exposure group at $LC_{50}$ showed a significant $Na^+/K^+ATPase$ inhibition, depending on the exposure time. Comparision of several concentrations of lead revealed that the $Na^+/K^+ATPase$ activity in Maceobrachium nipponnese was significantly decreased in a concentration dependent manner. These results suggest that $Na^+/K^+ATPase$ activity in Maceobrachium nipponnese may possibly be used as a biomarker of lead pollution in aquatic ecosystem.

Kosinostatin, a Major Secondary Metabolite Isolated from the Culture Filtrate of Streptomyces violaceusniger Strain HAL64

  • During a screening program, an actinomycete strain isolated from the Egyptian soil was investigated for its potential to show antimicrobial activity. The identification of this isolate was performed according to spore morphology and cell wall chemo-type, which suggested that this strain is a streptomycete. Further cultural, physiological characteristics and the analysis of the nucleotide sequence of the 16S rRNA gene (1480 bp) of this isolate indicated that this strain is identical to Streptomyces violaceusniger (accession number EF063682) and then designated S. violaceusniger strain HAL64. In its culture supernatant, this organism could produce one major compound strongly inhibits the growth of Gram-positive but the inhibition of Gram-negative indicator bacteria was lower. The antibiotic was separated by silica gel column chromatography and then purified on a sephadex LH-20 column and finally the purity was checked by HPLC. The chemical structure of the purified compound was determined using spectroscopic analyses (molecular formula of $C_{33}H_{32}N_{2}O_{10}$ and molecular weight of 617.21) and found to be identical to the kosinostatin, a quinocycline antibiotic which is known to be produced by Micromonspora sp. TP-A0468 (Igarashi et al., 2002) and to quinocycline B isolated from Streptomyces aureofaciens (Celmer et al., 1958). Although the antibiotic is known, the newly isolated strain was able to produce the antibiotic as a major product providing an important biotechnological downstream advantage.

Inhibitory Effect of Dalbergioidin Isolated from the Trunk of Lespedeza cyrtobotrya on Melanin Biosynthesis

  • Tyrosinase is a key enzyme for melanin biosynthesis, and hyperpigmentation disorders are associated with abnormal accumulation of melanin pigments, which can be reduced by treatment with depigmenting agents. The methanol extract of Lespedeza cyrtobotrya $M_{IQ}$ showed inhibitory activity against mushroom tyrosinase. The active compound was purified from the methanol extract of L. cyrtobotrya, followed by several chromatographic methods, and identified as dalbergioidin (DBG) by spectroscopic methods. The results showed that DBG exhibited tyrosinase inhibitory activity with an $IC_{50}$ of $20\;{\mu}M$. The kinetic analysis of tyrosinase inhibition revealed that DBG acted as a noncompetitive inhibitor. In addition, DBG showed a melanin biosynthesis inhibition zone in the culture plate of Streptomyces bikiniensis that has commonly been used as an indicator organism. Furthermore, $27\;{\mu}M$ DBG decreased more than 50% of melanin contents on the pigmentation using the immortalized mouse melanocyte, melan-a cell.

무안만 초입 인근 갯벌의 대형저서동물 분포 특성 (Distribution of Macrobenthos at the Mudflat in the Mouth of Muan Bay)

  • 대형저서동물은 갯벌에서 우점을 차지하고 있으며, 저서환경을 유지하고 환경을 정화하는데 중요한 역할을 담당하고 있다. 퇴적환경에서 유기물 함량은 저서동물의 서식, 먹이행동, 생존 등에 영향을 미치게 된다. 본 연구에서는 무안만 초입의 목포연안 인근갯벌(목포지역과 압해지역)에서 유기물 함량과 저서동물 분포 특성에 대해서 조사하였다. 목포지역은 압해지역에 비해 높은 유기물 함량을 보였으며, 오염지표종인 Musculista senhousia가 우점을 차지하였다. 무안만 초입 인근 갯벌에서 대형저서동물의 분포는 퇴적층 유기물 함량과 유의적 상관관계를 나타내며 두 지역으로 나누어졌다.

생흔학의 중요성 및 활용 (Importance and Application of Ichnology)

  • 생흔학이란 암석내의 생물들에 의해 만들어진 흔적에 대해 연구하는 학문으로 그들에 대한 설명, 분류 및 해석하는 것을 포함한다. 생흔의 형태, 생물의 행동 및 기질의 특성을 바탕으로 정리된 생흔구조의 조합인 생흔상은 정밀한 퇴적작용과 퇴적환경을 해석하는데 중요한 자료를 제시한다. 생흔학 연구는 퇴적학, 층서학, 석유탐사, 고생물학과 고생태학 등과 같은 퇴적환경학의 전반적인 분야를 연구하는데 다양하게 활용되어져 왔다. 퇴적학 분야에서는 퇴적환경의 물리적인 특성 이해, 퇴적양상에 대한 이해 및 사건층의 인지 등에 직접적인 증거가 될 수 있다. 층서학에서는 순차층서학에서 중요하게 다루어지는 주요 경계면의 인지 및 생층서학적 불연속면의 경계등을 인지하는데 활용된다. 또한 적은 양의 자료로 퇴적환경을 해석하는데에 중요한 정보를 제공함으로서 석유탐사분야에서 비용을 절감하는데 많은 도움이 된다. 이 외에도 화석학적 자료로 사용이 가능하며, 퇴적환경의 고생태를 복원하는 데도 중요한 자료로 활용될 수 있다. 강화도 조간대에서 시범적으로 적용된 생흔상 변화는 조간대의 위치에 따라 상이한 계절변화가 관찰되었다. 생흔상의 조간대의 위치에 따라 상이한 계절변화는 고기퇴적층에서의 계절적 특성, 정밀 퇴적환경, 보존잠재성 등을 파악할 수 있는 좋은 자료가 될 수 있을 것이다. 현생 연안환경에 해당되는 한반도 서해안 조간대에서의 생흔에 대한 연구결과는 퇴적학은 물론 석유지질학에의 활용 가능성이 높은 것으로 판단된다.

국내 수돗물 정수장에서 발견된 깔따구 유충(파리목: 깔따구과)의 유전적-형태적 종 동정 연구 (Morphological and Genetic Species Identification in the Chironomus Larvae (Diptera: Chironomidae) Found in Domestic Tap Water Purification Plants)

  • 깔따구(Diptera: Chironomidae)는 저서성 대형무척추동물로 환경오염 및 수질 모니터링에 이용되는 중요한 지표생물이다. 본 연구에서는 인천 수돗물 정수장에서 발견된 깔따구류의 정밀한 종 동정을 위해 형태적 분류와 미토콘드리아 DNA에서 cytochrome c oxidase subunit I (COI) 유전자의 염기서열을 이용하여 분석하였다. 정수장 6곳의 20개체는 안개무늬날개깔따구(Chironomus kiiensis) 12개체, 노랑털깔따구(Chironomus flaviplumus) 6개체, 등깔따구(Chironomus dorsalis) 1개체, 용산무늬깔따구(Polypedilum yongsanensis) 1개체 등 4종으로 확인되었다. 각 깔따구 종의 형태적 특징은 두부, 하순기절, 대악, 안테나, 발톱의 형태적 특징을 살펴보았다. NCBI Genbank에 등록된 깔따구 17종 21개체의 COI 염기서열을 바탕으로 본 연구에서 조사된 20개체의 계통진화적 분석한 결과 각 4종의 깔따구 COI 염기서열은 등록된 동인 종과 높은 상동성을 보이며 (99~100%) 같은 계통군(clade)으로 나타났다. 이러한 결과는 국내 깔따구의 종 동정을 위한 형태적- 유전적 정보를 통합적으로 제공함으로 담수생태계의 모니터링을 위한 주요한 정보로 활용될 것이다.

한국형 육포제조를 위한 원료 돈육의 미생물 분포 및 병원성 미생물의 확인 (The Distribution of Indicator Organisms and Incidence of Pathogenic Bacteria in Raw Pork Material Used for Korean Pork Jerky)

  • 시중의 정육점 및 백화점 등에서 유통 중인 10종의 돈육 원료에 대한 일반세균, 저온균, 고온균, 혐기성균 및 진균류, 대장균군 등의 미생물학적 분포와 병원성 미생물에 대한 분리동정을 실시하였다. 실험 결과, 원료 돈육에서 중온균은 $3.9{\times}10^2-3.9{\times}10^5cfu/g$으로 높은 분포를 보였고, 저온균은 $1.5{\times}10^3-8.6{\times}10^2cfu/g$, 혐기성균은 중온균, 저온균과 유사한 분포를 보였으나 상대적으로 적게 검출되었고, 고온균은 모든 검체에서 검출되지 않았다. 대장균군 또한 모든 검체에 대해서 검출되지 않았으며, 곰팡이와 효모 등 진균류는 $3.8{\times}10^1-5.1{\times}10^2cfu/g$으로 검출되었다. B. cereus는 돈육 sample B와 J에서 분리되었고, S. aureus의 경우 돈육 sample B에서만 검출되었다. B. cereus는 API kit를 이용한 간이동정에서 99.8%의 상동성을 보였고, S. aureus는 97.8%의 상동성을 보였다.

The Stydy on Application to Cosmetics of Phellinus Iimteus

  • Phellinus linteus was artificially cultivated in kangwon province in Korea. The air-dried phellinus linteus was frozen in liquid nitrogen tank and powdered in jar. 10g of the powder was extracted with each 200g of ethanol, methanol, distilled water and 1,3-butylene glycol/distilled water 4 hours under refluxing and then the liquidextract was concentrated under reduced pressure. As a result of analysis by high performance liquid chromatography and thin layer chromarography, many kinds of sugar and flavonoids were detected. Also we knew that phellinus linteus' extract had a strong UV-ray absorption. In the efficacy test for applying to cosmetics, free radical scavenging effect was confirmed. As a result, 2% of sample was the most potent inhibitory effect and the free radical savenging activity, was 0.31%. This is more effective than any other meterial. In the test of antioxidative activity against lipid autoxidation, phellinus linteus' extract had a good effect by 46% while vitamine E was 42.3%. The immunological activity of phellinus linteus was showed through the activation of macrophage cell. Actually, phellinus linteus activated macrophage function of 1.1-1.8 times including nitrite production compared to control. The whitening effect of phellinus linteus was showed through the inhibition of tyrosinase activity, melanin biosynthesis of S. bikiniensis and B-16 melanoma cells. Phellinus linteus' extract was showed strong mushroom tyrosinase inhibitory activity with IC50 value of 0.5% and inhibited melanin biosynthesis with 28mm inhibition zone at 0.005%/paper disc in S. bikinniensis, a bacterium used as an indicator organism in this work. Also it inhibited melanin biosynthesis in B-16 melanoma cells with a minimum inhibitory concentration of 0.134%.

소음(騷音) Stress에 의한 요(尿)중 Catecholamine의 분비량(分泌量) 변화(變化) (A Study on the Change of Urinary Catecholamine Sexcretion due to Noise Stress)

  • Noise is not only affecting the ear and the auditory cortex locally, but its influence is widely spread throughout the brain structures, e. g., the reticular formation, the brain stem nuclei or the subcortical forebrain area. Hence, any of the organism's activities can be hindered or stimulated by noise. High noise is a stressor and the catecholamine level can be used both as a stress marker and as an indicator of modified sympathetic nervous system activity. Several recent studies have found that the urinary excretion of catecholamines is increased due to high noise intensity, especially unexpectedly high and long lasting noise. The present study was conducted in order to examine the effects of noise stress on urinary excretion of ctecholamines in rats and humans. Rats were exposed to 90 dB noise for 10, 30, and 60 minutes, 3 and 12 hours. 24 hour . urinary samples were collected and the catecholamones were extracted by alumina and analyzed by HPLC-ECD. Catecholamine levels increased with time of exposure up to 60 minutes : norepinephrine concentration at 60 min of noise=1.038 ng/ml, epinephrine=0.636 ng/ml. Urine catecholamines of blue collar workers exposed to 90 dB of noise at the work place were collected between 2 and 4 p.m. and compared to that of white collar workers exposed to 70 dB. Mean norepinephrine level of the blue collar workers was 0.89 ng/ml (${\pm}0.25$), epinephrine 0.24ng/m1 (${\pm}0.09$), and that of the white collar workers 0.48 ng/ml (${\pm}0.12$), epinephrine 0.19 ng/ml(${\pm}0.05$). It was concluded that noise acts as a stressor and increases the catecholamine levels in both rats and humans.

Differential Gene Expression in the Pathogenic Strains of Actinobacillus pleuropneumoniae Serotypes 1 and 3

  • The limited information on differential gene expression in the different serotypes of Actinobacillus pleuropneumoniae has significantly hampered the research on the pathogenic mechanisms of this organism and the development of multivalent vaccines against A. pleuropneumoniae infection. To compare the gene expressions in the A. pleuropneumoniae strains CVCC259 (serotype 1) and CVCC261 (serotype 3), we screened the differentially expressed genes in the two strains by performing representational difference analysis (RDA). Northern blot analyses were used to confirm the results of RDA. We identified 22 differentially expressed genes in the CVCC259 strain and 20 differentially expressed genes in the CVCC261 strain, and these genes were classified into 11 groups: (1) genes encoding APX toxins; (2) genes encoding transferrin-binding protein; (3) genes involved in lipopolysaccharide (LPS) biosynthesis; (4) genes encoding autotransporter adhesin; (5) genes involved in metabolism; (6) genes involved in the ATP-binding cassette (ABC) transporter system; (7) genes encoding molecular chaperones; (8) genes involved in bacterial transcription and nucleic acid metabolism; (9) a gene encoding protease; (10) genes encoding lipoprotein/membrane protein; and (11) genes encoding various hypothetical proteins. This is the first report on the systematic application of RDA for the analysis of differential gene expression in A. pleuropneumoniae serotypes 1 and 3. The determination of these differentially expressed genes will serve as an indicator for future research on the pathogenic mechanisms of A. pleuropneumoniae and the development of a multivalent vaccine against A. pleuropneumoniae infection.
