  • Incubation routine and sex role of Streaked Shearwaters Calonectris leucomelas at Sasudo Island, in Jeju, South Korea, were studied during the incubation period, June to August in 2002. Incubation routine in Procellariiformes represents a sequence of alternating shifts taken in turn by female and male in a species-specific pattern. Hence, coordination of individual incubation rhythms between partners is crucial for successful breeding attempt. In Streaked Shearwaters, incubation routine represents a sequence of alternating shifts taken in turn by male and female. The first incubation shift was made by male after female had laid the egg. The mean incubation period was 50.8 days until hatching. Males had spent on average 26.5 days incubating and females 24.3 days accordingly. The mean duration of incubation shifts decreased progressively from 6th and 7th shift to hatching. Overall, males had spent more time incubating than females during the incubation period, but the mean duration of the incubation shift 5.6 days for males and 5.7 days for females did not differ between males and females. There were no effect of the body size of the breeding pair on incubation performance. For males the mean of body weight decreased during the incubation, whereas for females it remained approximately stable. In Streaked Shearwaters, the duration of incubation shift and subsequent foraging trip are related to loss of body weight during the period of fasting. In addition, coordination of individual incubation rhythms affects their incubation behaviour.

콩물 농도 및 숙성 시간이 Amylase 의 활성과 유과 반죽의 특성에 미치는 영향 (Effect of Bean Water Concentration and Incubation Time on Amylase Activity and Physicochemical Characteristics of Yukwa Paste)

  • 콩물의 ${\alpha}-amylase$ 활성은 콩물 농도에 따른 유의적인 차이가 없었으나, ${\beta}-amylase$와 glucoamylase의 활성은 7% 콩물보다 14% 콩물의 활성이 유의적으로 높았으며, 각각의 효소는 온도 변화 및 기질에 따른 차이를 나타내었고, 생전분보다 호화전분 기질에 대한 활성이 높게 나타났다. 유과 반죽의 pH는 콩물 농도 증가에 따라 증가하였고 incubation 시간 증가에 따라서 감소하였으며, 점도는 콩물 농도가 증가할수록 incubation 시간이 길어질수록 감소하였다. 반죽의 환원당 함량은 생반죽, 호화 반죽 모두 콩물 농도가 증가할수록, incubation 시간이 길어질수록 높게 나타났으며, 특히 호화 반죽에서는 0% 콩물 첨가군보다 7%, 14% 콩물 첨가군의 환원당 함량이 $3{\sim}4$배 이상 높게 나타났다. 반죽의 ${\alpha},{\beta}-amylase$, glucoamylase 역시 콩물 농도가 증가할수록 활성이 증가하였고 생전분보다 호화전분에 대한 활성이 매우 높게 나타났다. 유과 반죽을 동결 건조시켜 미세구조를 관찰한 결과 콩물 무첨가군에 비해 콩물 첨가군에서 incubation 시간이 증가함에 따라 입자의 크기가 작아지고, 입자 표면에 구멍이 관찰되어 콩물 첨가시 전분 분해 효소 작용이 일어났음을 알 수 있었다.

냉각 후 배양시간이 생쥐 난자의 방추체와 염색체에 미치는 영향 (Effect of Incubation Time after Cooling on the Meiotic Spindle and Chromosomes of Mouse Oocytes)

  • 동결 과정 중 필수적인 단계중 하나인 냉각(cooling)과 냉각 후 배양시간이 생쥐 난자의 방추체의 형태와 염색체의 배열에 미치는 영향을 알아봄으로서 냉각 후 손상되었던 난자의 방추체와 염색체가 정상적으로 회복하는데 필요한 최적의 배양시간을 알아보기 위해 본 실험을 실시하였다. 생후 4-6주령의 암컷 B6C3Fl 생쥐를 과배란 처리하여 metaphase II상태의 난자를 회수하여 다음과 같이 처리하였다. 대조군은 난자를 냉각처리하지 않았으며 실험군은 난자를 $0^{\circ}C$에서 30분간 냉각한 후 37$^{\circ}C$에서 가온하여 즉시 일부 난자는 면역형광 염색을 실시하고 나머지 난자는 5% $CO_2$ 37$^{\circ}C$가 유지된 배양기내에서 Ml6 배지에 각각 5분, 15분, 30분, 60분, 120분간 배양한 후 면역 형광염색을 실시하였다. 난자의 방추체와 염색체를 평가하기 위한 면역형광염색은 Zenes 등의 방법(2001)에 준하여 실시하였다. 냉각처리하지 않은 생쥐 난자를 면역형광 염색하여 방추체와 염색체를 관찰한 결과 생쥐 metaphase II 상태의 난자는 대칭성의 원통모양의 방추체 형태를 보였으며 염색체는 metaphase plate위에 분리된 다발모양으로 밀집되어 보였다. 냉각 직후 미세관의 소실에 의한 방추체 형태의 이상과 형광성의 소실이 나타났으며 염색체는 다발모양의 밀집된 형상에서 벗어나 비정상적인 배열상을 보였다. 냉각 처리된 난자를 37$^{\circ}C$에서 가온하고 배양하였을 때 미세관의 재중합이 일어나 미세관의 형광성을 회복하기 시작하였고 방추체는 정상적인 배열상으로 회복되었다. 생쥐 난자를 냉각처리한 후 배양시간에 따른 방추체 미세관의 형광성(FIS), 염색체의 배열, 방추체의 형태를 비교하였다. 배양 5분에서 60분까지 FIS, 정상 염색체 배열을 보인 난자의 비율, 정상 방추체의 형태를 보인 난자의 비율이 점진적으로 증가하였으나 120분 배양에서는 감소하였다(P<0.05). 위의 세 가지 평가를 기준으로 하여 냉각 후 난자의 회복율을 관찰하였을 때 배양 60분에서 최상의 회복율을 나타냈다.

회분식 생물반응기를 이용한 BDOCrapid와 BDOCslow 결정 (Determination of BDOCrapid and BDOCslow using Batch Bio-reactor)

  • In this study, biodegradable organic matter was divided into a rapidly biodegradable fraction($BDOC_{rapid}$) and a slowly biodegradable fraction($BDOC_{slow}$) for various waters with different types of DOC. These fractions($BDOC_{rapid}$ and $BDOC_{slow}$) were defined by using a shaking incubation method modified from Carlson's method. Also, in this study, optimum incubation time and accuracy were investigated to determine $BDOC_{rapid}$ and $BDOC_{slow}$. When suspended bacteria obtained from raw water and BAC effluent, or attached bacteria from BAC was respectively used as an inoculum, the difference in total BDOC($BDOC_{total}$) was minimal. Therefore, total BDOC was determined in 7~8 days by the shaking method, which is comparable with Servais's method by which BDOC was determined in 28 days. In addition, the difference of BDOC between these two methods was within 7%. Although $BDOC_{rapid}$ and $BDOC_{slow}$ were effectively determined by a method defined by Klevens, the difference in optimal incubation time was significant for different water samples. However, when using the shaking method, optimal incubation time for $BDOC_{rapid}$ was found to be 3 days, therefore, the $BDOC_{rapid}$ was defined as the difference between $DOC_0$ and $DOC_{3days}$, and $BDOC_{slow}$ was defined as the difference between $BDOC_{total}$ and $BDOC_{rapid}$. As a conclusion, for determining the fraction of BDOC using the shaking method, the concentrations of an inoculurns and optimal incubation times used in this study were very effective.

반추동물의 에너지 손실을 줄이기 위한 연구; 무 추출물을 이용한 메탄 손실 억제 (Reduce the Energy Loss in Ruminant; Using Raphanus Sativus Extracts to Mitigate Methane Emission)

  • This study was conducted to evaluate Raphanus sativus extracts to methane reduction in rumen. Five different levels of R. sativus extracts were used to investigate the most effective dosing level for the decrease of methane production in the rumen. The rumen fluid was collected from a cannulated one Hanwoo cow ($BW=450{\pm}30kg$) consuming 600 g/kg timothy and 400 g/kg concentrate. On fermentation day, rumen fluid was collected at 2 hr postfeeding R. sativus extracts was dosed to achieve final concentration of 0, 1, 3, 5, 7, and 9% respectively, to fermentation bottles containing the mixture of rumen fluid and McDougall's buffer and 300 mg of timothy was added as a substrate. The fermentation was conducted for 3, 6, 9, 12, 24, 48 and 72 hr incubation time at $39^{\circ}C$ with shaking. In vitro ruminal pH values were measured normal range for ruminal fermentation. Dry matter disappearance was significantly higher (p<0.05) at 3 hr incubation time 1, 3 and 5% doses than that of control. The highest methane reduction was observed in 12 hr incubation time 5, 7 and 9%. The carbon dioxide emission was also significantly (p<0.05) lower than that of control at 12 hr incubation time 5, 7 and 9%. The total volatile fatty acid was no significant difference between control and all doses level at 12 and 24 hr incubation time. At 24 hr incubation time, the result of real-time PCR were indicated that M. archea was significantly lower (p<0.05) at all doses level comparing to that of control. In conclusion, R. sativus extracts were significantly decreased methane emission. R. sativus extracts were significantly lower (p<0.05) than that of control at 12 hr incubation time 5, 7 and 9% and no adversely effect in rumen pH, dry matter disappearance and total VFA.

배양조건에 의한 일과성 저산소상태 후 신경세포회복의 차이 (Difference of Neuronal Recovery by Incubation Condition after Transient Hypoxia)

  • Objective : The transverse hippocampal slice is one of the most commonly studied in vitro models of mammalian brain physiology. However, despite its broad usage, there has been no standardization of slice preparation techniques or recording condition. It is well known that variations in recording conditions can result in profound different effects to neuronal responses. Evoked field potentials, recorded extracellularly, were used to investigate the effects of variations in hippocampal slice preparation protocol on hypoxia responses of CA1 neurones. Material & Methods : Before hypoxic injury, hippocampal slices were incubated for 4 hours. During incubation period, the slices were placed in a incubation chamber($21^{\circ}C$) for recovery from preparation injury and then transferred to recording chamber($34^{\circ}C$) for more recovery and baseline electric recording with current stimulation(0.1Hz). Various time periods in incubation chamber and recording chamber were applied to each experimental group(group 1=60min : 180min, group 2=90min : 150min, group 3=180min : 60min, time in incubation chamber : time in recording chamber) before 10 min hypoxia produced by replacing 95% $O_2$+5% $CO_2$ mixed gas to 95% $N_2$+5% $CO_2$ gas. Calcium, Magnesium ions and several drugs effecting on glutamate receptor also were studied. Recoveries from hypoxic injury of hippocampal slices were estimated by percent recovery of population spike(PS). Statistic analysis of study were performed using paired t-test. Results : The percent recovery of PS after 10min hypoxia was considerably enhanced by increasing the period of current stimulation during incubation period before hypoxic injury. Temperature effect on the result of this experiment was also studied(group 4) but the result from this showed no statistic significance. Low magnesium ion concentration of artificial CSF(Mg-free aCSF) during incubation period enhanced the recovery of PS but low calcium (calcium-free) and high magnesium ion concentration(2mM) reduced it after hypoxic injury. L-glutamate($100{\mu}M$) and AP-5($50{\mu}M$) had no effect on the recovery of PS but CNQX($10{\mu}M$) in artificial CSF during incubation period markedly enhanced the recovery of PS. Co-treatment of AP-5($50{\mu}M$), CNQX($10{\mu}M$) and high magnesium concentration(2mM) enhanced recovery of PS in immediate following period of hypoxic injury but the effect of cotreatment after then decayed rapidly and lost statistic significance. Conclusions : Judging from above results, the condition of baseline recording is important in observing the recovery of population spike after hypoxia, and the time and the condition should be controled more strictly to obtain reliable results.

The "open incubation model": deriving community-driven value and innovation in the incubation process

  • Globalization, increasing technological advancements and dynamic knowledge diffusion are moving our world closer together at a unique scale and pace. At the same time, our rapidly changing society is confronted with major challenges ranging from demographic to economic ones; challenges that necessitate highly innovative solutions, forcing us to reconsider the way that we actually innovate and create shared value. As such the linear, centralized innovation models of the past need to be replaced with new approaches; approaches that are based upon an open and collaborative, global network perspective where all innovation actors strategically network and collaborate, openly distribute their ideas and co-innovate/co-create in a global context utilizing our society's full innovation potential (Innovation 4.0 - Open Innovation 2.0). These emerging innovation paradigms create "an opportunity for a new entrepreneurial renaissance which can drive a Cambrian like explosion of sustainable wealth creation" (Curley 2013). Thus, in order to materialize this entrepreneurial renaissance, it is critical not only to value but also to actively employ this new innovation paradigms so as to derive community-driven shared value that stems from global innovation networks. This paper argues that there is a gap in existing business incubation model that needs to be filled, in that the innovation and entrepreneurship community cannot afford to ignore the emerging innovation paradigms and rely upon closed incubation models but has to adopt an "open incubation" (Ziouvelou 2013). The open incubation model is based on the principles of open innovation, crowdsourcing and co-creation of shared value and enables individual users and innovation stakeholders to strategically network, find collaborators and partners, co-create ideas and prototypes, share their ideas/prototypes and utilize the wisdom of the crowd to assess the value of these project ideas/prototypes, while at the same time find connections/partners, business and technical information, knowledge on start-up related topics, online tools, online content, open data and open educational material and most importantly access to capital and crowd-funding. By introducing a new incubation phase, namely the "interest phase", open incubation bridges the gap between entrepreneurial need and action and addresses the wantpreneurial needs during the innovation conception phase. In this context one such ecosystem that aligns fully with the open incubation model and theoretical approach, is the VOICE ecosystem. VOICE is an international, community-driven innovation and entrepreneurship ecosystem based on open innovation, crowdsourcing and co-creation principles that has no physical location as opposed to traditional business incubators. VOICE aims to tap into the collective intelligence of the crowd and turn their entrepreneurial interest or need into a collaborative project that will result into a prototype and to a successful "crowd-venture".

닭 태자의 직결장에 대한 조직학적 및 면역조직화학적 연구 (Histological and immunohistochemical studies on the colo-rectum of the chicken embryos)

  • Histological changes and ontogeny, distribution and relative frequency of bovine SP-1/chromogranin(BCG)-, serotonin-, human pancreatic polypeptide(HPP)- and glucagon-immunoreactive cells were investigated in the colo-rectum of the chicken embryos. The pseudostratified columnar-like epithelium was observed in 10 days of incubation to 15 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium and goblet cells were detected for the first time on 19 days of incubation. BCG and serotonin-immunoreactive cells were observed for the first time on 15 days of incubation respectively, thereafter these cells were increased with ages. However no HPP and glucagon-immunoreactive cells were found in this study.

New Venture Incubation Framework: An Indian Academic Model

  • Institution-based Technology Business Incubators are on the rise in India, as a means of promoting innovation-based tech start-up ecosystems, due to increased policy initiatives. Against this background, we have traced the origin and process of building a start-up ecosystem in IIT Madras, Chennai of India, based on semistructured interviews held with the stakeholders of the ecosystem. Subsequently, we have ascertained the key components of IIT Madras start-up ecosystem, and the process of incubation comprising pre-incubation, incubation and post-incubation phases. Finally, we have derived the key lessons from the ecosystem development experience and incubation process which enable generation of start-ups from both students and faculty, apart from alumni and ex-industry executives. Though this ecosystem model has emerged over a period of time through learning and experience, the ecosystem is able to generate more than 100 start-ups, majority of them being from students and faculty. Thus, the evolved start-up ecosystem of IIT Madras is able to generate faculty-supported and student-led entrepreneurship successfully.

Effect of dark incubation in germination of indirect date palm somatic embryos and conversion into plantlets

  • All studies on date palm somatic embryogenesis have focused on germination in the presence of light while neglecting germination in darkness, which mimics the germination process of zygotic embryos within seeds. To improve the date palm micropropagation protocol, we investigated the effects of light and darkness incubation on the germination of indirect date palm somatic embryos and their subsequent conversion into plantlets. Darkness incubation emerged as a pivotal factor in the germination of indirect date palm somatic embryos and their successful conversion into plantlets. Darkness incubation significantly decreased the time required for the conversion of indirect somatic embryos into plantlets, halving the duration from 24 weeks to only 12 weeks. The micropropagation protocol was modified, consolidating the previous two distinct stages of germination and elongation under light incubation into a single stage under darkness incubation. These findings modified the protocol and significantly reduced the overall duration of the date palm micropropagation protocol.
