고온 조건에서 사료 내 생균제의 생존성 및 오염미생물의 생장 억제 효과 (Viability of Probiotics in Feed under High Temperature Conditions and Their Growth Inhibitory Effect on Contaminant Microbes)

  • 여름철의 고온으로의 온도 상승에 따른 사료빈 내의 생균제 L. plantarum, S. cerevisiae 및 B. subtilis의 생존성을 실험실의 가상 조건에서 분석하였다. 상온인 $25^{\circ}C$에서 멸균 사료와 비멸균 사료에 단일 혹은 혼합 균주 첨가 시 pH 변화와 생균제들의 생장을 상호 비교하였다. pH는 멸균 사료와 비멸균 사료 모두에서 4일째에 가장 감소한 것은 같았으나 비멸균 사료에서는 2일째까지는 상승하는 변화의 양상을 보여주었다. 멸균 여부 혹은 혼합 여부와 관계없이 S. cerevisiae와 B. subtilis의 생균수는 일정하게 유지되었지만 L. plantarum의 경우에는 그 수가 모두 줄어드는 것을 확인할 수 있었다. 따라서 3종의 혼합 생균제는 상호 길항작용은 없는 것으로 나타났다. 멸균 및 비멸균 사료에 생균제를 첨가한 후 $60^{\circ}C$의 고온 환경에서 사료의 pH 와 단일접종 생균제의 생존성을 조사하였다. 멸균 및 비멸균 사료 사이에 뚜렷한 pH의 변화는 관찰되지 않았으며 B. subtilis의 pH가 가장 낮게 관찰되었다. 고온 하에서 L. plantarum과 S. cerevisiae 균주는 생존할 수 없었으며, 내열성을 가진 B. subtilis 균주는 생존하면서 사료에 자연적으로 생존하는 오염세균의 증식을 억제하였다. 또 B. subtilis를 접종한 비멸균 사료에서 2일째부터 오염 곰팡이가 관찰되지 않았다. 따라서 내열성이 강한 B. subtilis 균주를 사용하면, 여름철 사료빈 내에 병원성 세균과 곰팡이의 오염을 억제할 수 있는 것으로 나타났다.

패혈증 환자에서 NF-κB 활성화에 의한 호중구 아포프토시스의 억제 (Decreased Neutrophil Apoptosis in Patients with Sepsis is Related to the Activation of NF-κB)

  • 연구배경 : 호중구에 의해 매개되는 염증반응은 호중구의 수명이 매우 짧기 때문에 대부분 자연 종결된다. 패혈증에서는 이러한 호중구의 아포프토시스가 감소되어 수명이 연장되어 있어서 지속적인 염증반응이 일어나게 된다. 호중구의 수명 연장을 유도하는 많은 염증 매개 물질들이 nuclear factor-${\kappa}B$ 전사인자에 의해 조절되기 때문에 이 연구에서는 nuclear factor-${\kappa}B$가 패혈증 환자에서 관찰되는 호중 구의 아포프토시스 억제와 연관이 있을 것으로 가정하였다. 방법 : 건강한 정상인과 패혈증 환자의 호중구를 정맥혈로부터 신선하게 분리하여 실험하였다. 호중구의 아포프토시스는 특징적인 아포프토시스의 형태를 보이는 세포를 광학현미경으로 세거나 Annexin V를 이용한 유세포분석법으로 정량하였다. Nuclear factor-${\kappa}B$의 활성도는 면역형광 엽색법 또는 electrophoretic mobility shift assay로 판단하였다. 항아포프토시스 단백인 X-linked inhibitor of apoptosis의 발현 정도는 western blot으로 평가하였다. 결과 : 패혈증 환자에서 자발적 아포프토시스가 감소되어 있음을 확인하였다. 패혈증 환자의 호중구에 cycloheximide를 처치하였을 때 아포프토시스가 유의하게 증가하여 아포프토시스 감소가 새로운 단백 합성에 의존적임을 관찰하였다. 패혈증 환자의 호중구에서는 정상인과는 달리 기저상태에서도 nuclear factor-${\kappa}B$가 핵 내로 이동되어 활성화되어 있었고 nuclear factor-${\kappa}B$의 활성을 억제하였을 때 아포프토시스의 억제가 반전되었다. 또한 nuclear factor-${\kappa}B$에 의존적인 X-linked inhibitor of apoptosis 단백의 발현 수준이 정상인의 호중구에서는 24시간 동안 배양하면서 점차 감소하였지만 패혈증 환자의 호중구에서는 지속적으로 발현 수준이 유지되었다. 결론 : 패혈증 환자에서 관찰되는 호중구의 아포프토시스 억제에는 nuclear factor-${\kappa}B$ 전사인자의 활성화에 의한 생존 단백의 유도가 관여할 것으로 생각하였다.

신안군(新安郡) 낙도(落島)에서 발생(發生)한 괴질(怪疾)의 원인(原因)에 관한 역학적(疫學的) 조사(調査)(I) (Epidemiologic Investigation for the Etiology of an Epidemic Ocurred among Animals and Humans in an Isolated Island, Korea(I))

  • This is preliminary report on anthrax epidemic occurred in an island with about 100 residents. Since 1982 there had been sudden deaths among all kinds of domestic animals including cattle, dogs, ducks, chicken and goat but only a few among cats in an isolated island about three hours distance away by ferry boat from Mokpo city. From 1986 through 1988 nine human deaths and four patients occurred, which made the government intervene for investigation on June 25 1988. The epidemiological investigation consisted of interview survey and medical examination, medical record analysis, laboratory work to isolate the pathogens under the direction of hypothesis derived from the study and further confirmation of the pathogens by international institute. The summarized results are as followings: 1. According to the interview survey there were many deaths among domestic animals usually in cold and dry season such as January through March and September through November; 36 heads of cattle leaving one head, more than 40 hogs(all), hundreds of chicken leaving few alive, goats that had taken home from mountain and two or three cats out of around 40 had sudden deaths from 1982 till 1985, when the residents stopped to purchase and take them into the island anymore. Also there were eleven persons who had experienced the similar syndrome complex to those of admitted and expired patients and four of them revealed typical chest X-ray findings; from one of these four patients(Rho) B. anthracis is isolated. 2. Medical record on patients who had been admitted, showed common characteristics of the disease course. On admission they had either gastrointestinal or upper respiratory infection symptoms which invariably progressed to septicemic nature with pulmonary interstitial infiltration and mediastinal widening/bulging, and then to deadly acute respiratory distress syndrome. At the end stage chest X-ray revealed multiple bullous emphysema. One of another characteristics was oral ulceration with bleeding occurred in about 50% of the patients. Laboratory test results in common were leukocytosis with left shift and abnormal liver and kidney functions, particularly at the later stage of the illness. 3. Epidemiological characteristics was striking in that both mortality and incidence rates were high: the mortality rate was 8.7% average, male being three times higher than females but there was no distictive clustering by age group. The incidence rate for both sexes was 28.2% and there was no sex difference although a tendency of higher incidence among older ages was noticed. The highest mortality and incidence were observed in Won village where the first death of animal occurred and with the highest frequency among three villages of the island. 4. Among twelve bacilli species isolated from various specimens, two strains, one from patient and the other from soil where the recently died cow is hurried, were confirmed as B. anthracis by Pasteur Institute and CDC of USA(strain from soil). CDC reported that the strain did not produce capsule in bicarbonate media but reacted with the bacteriophage and one of five sera taken from the patients. Mode of transmission as well as incubation period of the agent has not been established yet, which needs further investigation in relation to the antigenic structure of the variant when it is confirmed.

Low temperature plasma deposition of microcrystalline silicon thin films for active matrix displays: opportunities and challenges

  • The spectacular development of AMLCDs, been made possible by a-Si:H technology, still faces two major drawbacks due to the intrinsic structure of a-Si:H, namely a low mobility and most important a shift of the transfer characteristics of the TFTs when submitted to bias stress. This has lead to strong research in the crystallization of a-Si:H films by laser and furnace annealing to produce polycrystalline silicon TFTs. While these devices show improved mobility and stability, they suffer from uniformity over large areas and increased cost. In the last decade we have focused on microcrystalline silicon (${\mu}c$-Si:H) for bottom gate TFTs, which can hopefully meet all the requirements for mass production of large area AMOLED displays [1,2]. In this presentation we will focus on the transfer of a deposition process based on the use of $SiF_4$-Ar-$H_2$ mixtures from a small area research laboratory reactor into an industrial gen 1 AKT reactor. We will first discuss on the optimization of the process conditions leading to fully crystallized films without any amorphous incubation layer, suitable for bottom gate TFTS, as well as on the use of plasma diagnostics to increase the deposition rate up to 0.5 nm/s [3]. The use of silicon nanocrystals appears as an elegant way to circumvent the opposite requirements of a high deposition rate and a fully crystallized interface [4]. The optimized process conditions are transferred to large area substrates in an industrial environment, on which some process adjustment was required to reproduce the material properties achieved in the laboratory scale reactor. For optimized process conditions, the homogeneity of the optical and electronic properties of the ${\mu}c$-Si:H films deposited on $300{\times}400\;mm$ substrates was checked by a set of complementary techniques. Spectroscopic ellipsometry, Raman spectroscopy, dark conductivity, time resolved microwave conductivity and hydrogen evolution measurements allowed demonstrating an excellent homogeneity in the structure and transport properties of the films. On the basis of these results, optimized process conditions were applied to TFTs, for which both bottom gate and top gate structures were studied aiming to achieve characteristics suitable for driving AMOLED displays. Results on the homogeneity of the TFT characteristics over the large area substrates and stability will be presented, as well as their application as a backplane for an AMOLED display.

Hypoxia Inducible Factor-1α Directly Regulates Nuclear Clusterin Transcription by Interacting with Hypoxia Response Elements in the Clusterin Promoter

  • Differential transcription of the clusterin (CLU) gene yields two CLU isoforms, a nuclear form (nCLU) and a secretory form (sCLU), which play crucial roles in prostate tumorigenesis. Pro-apoptotic nCLU and anti-apoptotic sCLU have opposite effects and are differentially expressed in normal and cancer cells; however, their regulatory mechanisms at the transcriptional level are not yet known. Here, we examined the transcriptional regulation of nCLU in response to hypoxia. We identified three putative hypoxia response elements (HREs) in the human CLU promoter between positions -806 and +51 bp. Using a luciferase reporter, electrophoretic gel mobility shift, and chromatin immunoprecipitation assays, we further showed that hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) bound directly to these sites and activated transcription. Exposure to the hypoxia-mimetic compound $CoCl_2$, incubation under 1% $O_2$ conditions, or overexpression of HIF-$1{\alpha}$ enhanced nCLU expression and induced apoptosis in human prostate cancer PC3M cells. However, LNCaP prostate cancer cells were resistant to hypoxia-induced cell death. Methylation-specific PCR analysis revealed that the CLU promoter in PC3M cells was not methylated; in contrast, the CLU promoter in LNCap cells was methylated. Co-treatment of LNCaP cells with $CoCl_2$ and a demethylating agent promoted apoptotic cell death through the induction of nCLU. We conclude that nCLU expression is regulated by direct binding of HIF-$1{\alpha}$ to HRE sites and is epigenetically controlled by methylation of its promoter region.
