Antitumor activities of a peptide fraction isolated from traditional Korean soy sauce (SSP) were investigated in vitro and in vivo using cancer cell lines and F9 teratocarcinoma-bearing BALB/c mice. SSP exerted a dose-dependent antiproliferative effect on P388D1 mouse lymphoma, F9 mouse teratocarcinoma, and DLD-l human colon cancer cells with $IC_{50}$ values of 11, 50, and $50\mug/ml$, respectively. Tumor growth in F9 teratocarcinoma-bearing BALB/c mice, orally administered with 80 and 200 mg/kg/day of SSPs, was inhibited 10.3% and 52.4%, respectively, and survival days increased by 11.9% and 22.1%, respectively, compared to the control group. The results of antitumor activities exerted by SSP in vitro and in vivo suggest the feasibility of using SSP as an antitumor agent.
Backgroud: Immunotherapy using dendritic cells (DC) loaded with tumor antigens may represent a potentially effective method for inducing antitumor immunity. We evaluated the effectiveness of DC-based antitumor immune response in various conditions. Methods: DC were cultured from peripheral blood mononuclear cells (PBMNC) in myelogenous leukemia (ML) and lysates of autologous leukemic cells are used as tumor antigen. The effectiveness of interleukin-12 (IL-12) and CD40L (CD154) on the antigen presenting function of lysates-loaded DC was analyzed by proliferation, cytokine production, and cytotoxicity tests with activated PBMNC (mainly lymphocytes). For generating antigen-loaded DC, direct fusion of DC with ML was studied. Results: Antigen loaded DC induced significantly effective antitumor immune response against autologous leukemic cells. Administration of IL-12 on the DC based antitumor immune response showed higher proliferation activity, IFN-$\gamma$ production, and cytotoxic activity of PBMNC. Also, fused cell has a potent antitumor immune response. Conclusion: We conclude that lysates-loaded DC with IL-12 may be effectively utilized as inducer of antitumor immune reaction in ML and in vivo application with DC-based antitumor immunotherapy or tumor vaccination seems to be feasible.
본 연구에서는 단 삼(Salvia miltiorrhiza)의 추출물이 인체 결장암세포인 HT-29, 간암세포인 HepG2 및 직장 암세포인 HRT-18의 증식에 미치는 영향을 in vitro에서 확인하였다. 암세포의 배양액에서 단삼의 수용성 추출물과 에탄올 추출물의 암세포 증식억제효과는 농도에 비례하여 에탄올 추출물에서 더 효과적이었다. 이를 토대로 단삼의 에탄올 추출물의 급성독성, 수명연장 및 고형암형 성억제를 살펴보기 위해 in vivo 실험을 하였다. 급성독성실험결과 대조군의 15일 째의 평균체중은 32.3 g 이었고 실험군은 31.6 g으로 정상적인 상태를 유지하였으며, 흰쥐의 육종암세포인 sarcoma-180를 접종한 mouse의 수명연장실험결과 대조군에 비해 61%의 수 명연장 효과가 있음을 관찰하였다. 고형암억제 실험 결과에서도 대조군에 비해 에찬올 추출 물 처리군이 35%의 고형암 형성억제능을 나타내었다. 따라서 단삼의 에탄올 추출물 중에는 in vitro와 in vivo 실험을 통해 항암활성을 갖는 성분이 존재하며, 이성분은 유효한 항암제 로 개발될 수 있으리라 사료된다.
Our previous reports demonstrated that ip. administration of Korean red ginseng acidic polysaccharide (RGAP) exerts antitumor activity In mice. The present study was carried out to compare the effects of ip. and p.o. routes of administration of RGAP on either normal or tumor-bearing BALB/c mice. RGAP was administered either ip. or p.o. at doses of 100, 300, 500, 1000 mg/kg for 1 or 5 weeks. Peritoneal macrophages from mice treated with RGAP p.o. at a dose of 300 mg/kg either for 1 or 5 weeks did not exhibit growth inhibition activity toward WEHI-I64 tumor cells. However, administration of RGAP at a dose of 600 mg/kg for both 1 and 5 weeks increased the antitumor activity of macrophages. Oral administration of RGAP (600 mg/kg) for 5 weeks and ip. administration of RGAP (300 mg/kg) for 1 week resulted in antitumor activities of $40\%$ and $45\%$, respectively, indicating that the effect of i.p. injection is more potent 2 and 5 times than that of p.o. one in terms of dose and duration, respectively. Tumor inhibition rates of RGAP at doses of 300, 500, 1000 mg/kg in mice transplanted with B16-F10 melanoma were 4.4, 12.0, and $45.4\%$, respectively, meaning that p.o. dose higher than 500 mg/kg possess marked antitumor activity. The results above suggests that p.o. administration of RGAP also show antitumor activity in vivo depending on the dose.
Daunirubicin and doxorubicin analogues (5,7,8,9,) in which the natural amino sugar, daunosamine, is replaced by rhamnopyranosyl or 4'-amino rhamnopyranosyl residues have been p repared. The in vitro cytotoxicity of compound 5 or 7 was similar to that of doxorubicin for P388 murine leukemic cell line. But compound 8 or 9 was less cytotoxic than doxorubicin. When administered intravenously on day 1, compound 9 showed antitumor activity comparable to that of doxorubicin against ip-inoculated L1210 murine leukemia and found to be less toxic than doxorubicin. But the in vivo antitumor activity of compound 7 or 8 was inferior to that of doxorubicin.
DW2282, (S)- (+)-4-phenyl -1-[N-(4-am mob enzoyl) -indolin-5- sulfonyl]-4,5- dihydro-2-imidazolone hydrochloride, is a novel anticancer agent thought to have an unique mechanism of action on the inhibition of tumor growth. In this study, we estimated in vivo antitumor activities and pharmacokinetics of Dw2282 depending on various vehicles. The inhibition rate of tumor growth was increased by 50, 100 and 200 mg/kg of Dw2282 in a dose-dependent manner. When Dw2282 dissolved in 4 sorts of vehicles was orally single dosed to rats at 50 mg/kg, Cmax of Dw2282 in 0.5% CMC.Na was a half as high as those in PG, PG+CP and PG+CP+DW. When Dw2282 was orally administered to mice for 5 days, antitumor activity of 130 mg/kg suspended in 0.5% CMC.Na was as effective as that of 65 mg/kg dissolved in the rest of vehicles. Taken together, it is thought that antitumor activities of Dw2282 are resulted from the absorption extent of it and related to the vehicle used.
Immunostimulation effects of the cell wall components isolated from Lactobacillus plantarum were investigated by studying the macrophage s tumorcidal activity, splenocyte proliferation, anticomplementary activity and the inhibition of peritoneal tumor cell growth measured with ICR mice inoculated with sarcoma 180. The immunopotentiating cell wall components were a complex of peptidoglycan and exopolysaccharides. The tumorcidal activity of macrophage against Yacl and B16 tumor cells was enhanced when the cell wall components were added into the macrophage s culture medium. They also stimulated splenocytes to proliferate up to the same level as when the concanavalin A was added into the splenocyte's culture medium. The complementary activity was inhibited by 50% when the cell wall components were incubated with the sheep red blood cells treated with hemolysin and guinea pig complement. This result confirmed that the cell wall components had an antitumor effect, because the anticomplementary activity is usually accompanied by an antitumor activity at the same time. This fact was confirmed again by the inhibition of the growth of sarcoma 180 when the cell wall components were injected intraperitoneally into ICR mice inoculated with sarcoma 180. As a result, it is concluded that the cell wall components isolated from Lactobacillus plantarum had multifunctional immunostimulation effects in vitro and in vivo.
In the previous report, we described the in vivo antitumor activity of PJ-4, a protein-polysaccharide fraction prepared from the culture filtrate of an insect-born fungus, Paecilomyces tenuipes DGUM 32001. In the present study, we elucidated the immunomodulating activity of PJ-4 on the BALB/c mouse splenic lymphocytes using flow cytometrical techniques. As a result, PJ-4 was found to stimulate the lymphocytes not only to form lymphoblasts but also to express CD25 (IL-2 receptor $\alpha$ chain) molecule, which is well known as a T cell activation marker. More interestingly, its T cell stimulatory activity was more strongly exerted on CD8$^{+}$ T cells than on CD4$^{+}$ T cells. All these data suggest that PJ-4 exerts its antitumor activity at least partly through stimulation of T cells which play major roles in the cell-mediated immune system.tem.

1,4-Naphthoquinones are widely distributed in nature and many clinically important antitumor drugs containing a quinone moiety, such as anthracyclines, mitoxantrones and saintopin, show excellent anticancer activity. In this study, 2- or 6-substituted 5,8-dimethoxy-1,4-naphthoquinone (DMNQ) and 5,8-dihydroxy-1,4-naphthoquinone (DHNQ) derivatives were synthesized, and their cytotoxic activity against L1210 and P388 cancer cells was examined. Their antitumor activity was also assessed in mice bearing S-180 cells in the peritoneal cavity. In comparison with the DMNQ derivatives, the DHNQ derivatives exhibited more potent bioactivities than the DMNQ derivatives against both L1210 and P388 cells in vitro and S-180 cells in vivo. The $ED_{50}$ values of the DHNQ derivatives against P388 cells were in the range of 0.18-1.81 ${\mu}g/mL$ whereas those of the DMNQ derivatives were in the range of 0.26-40.41 ${\mu}g/mL$. The T/C ($\%$) values of the DHNQ derivatives, 8, 17, 18, 19, and 20, were found to be comparable to or even better than that of adriamycin. It was also observed that the 2-substituted derivatives (8, 19, 20) showed better antitumor activity than the 6-substituted derivatives (7, 17, 18) in the mice bearing S-180 cells in the peritoneal cavity.
Praecoxin A, an ellagitannin, purified from Alnus hirsuta var.microphlla was evaluated on the antitumor activity. Praecoxin A had the significant cytotoxicity to s ix tumor cell lines: human chronic myelogenous leukemia K-562, human promyelocytic leukemia HL-60, mouse leukemia P388, mouse lymphocytic leukemia L-1210, sarcoma-l8O, mouse lymphoma L5178Y except L-1210. And the most sensitive cell line was K-562 ($ED_{50}=2.43{\mu}g/ml$). The $ED_{50} of praecoxin A against HL-60, P388, L-1210, sarcoma7l8O and L5178Y were 6.28, 8.66, 10.00, 7.01, $9.32{\mu}g/ml$, respectively. Praecoxin A showed the increasing effect in life span by 36.8% on the 1st day after treatment of 10mg/kg in mice bearing sarcoma-180 tumor cells (ascitic form) via NCI (National Cancer Institute, U.S.A.) protocol in vivo assay. As a result, praecoxin A is considered to show the antitumor activity.
