루틴, 퀘르세틴, 식이섬유 혼합물 설계를 이용한 전분소화 지연 효과의 최적화에 대한 융합 연구 (Convergence Study on the Optimization for Suppression of Starch Hydrolysis using Rutin, Quercetin and Dietary Fiber Mixture Design)

  • 본 연구는 루틴, 퀘르세틴, 식이섬유가 전분 소화율에 미치는 영향을 통계적 혼합물 설계방법을 이용하여 확인하고자 하였다. 세 가지 성분들은 모두 농도가 증가함에 따라 전분소화 지연 효과가 나타났으며, 그 중에서도 퀘르세틴과 식이섬유가 함께 섞였을 때 가장 높은 지연 효과를 보였다. 이 혼합물 설계는 simplex-centroid 실험설계법을 이용하였고, 최적 모델은 quadratic 모델에서 나타났다. 이때 얻어진 회귀방정식을 통하여 유의한 상승효과를 확인할 수 있었다. 최적화 통계 방법을 사용하여 혼합비를 분석한 결과 퀘르세틴과 식이섬유가 72: 28 비율에서 전분 소화 지연 효과가 최대로 나타나는 것을 확인하였다. 전분소화에 대한 영향을 3차원 표면 도표로 시각화하여 나타내었으며, 이 결과로 퀘르세틴과 식이섬유의 상호작용으로 인하여 전분 소화를 15% 이상 지연시키는 것을 확인할 수 있었다.

세로무늬 먼지진드기의 실험실적 배양에서 나프탈렌의 효과 (Naphthalene effects on in vitro culture of house dust mite, Dermatophagoides pteronyssinus)

  • 집먼지진드기는 호흡기 알레르기 질환 즉, 소아 천식, 알레르기성 비염, 아토피 피부염 등의 가장 중요한 기인항원으로 알려져 있다. 본 연구는 일반 살충제로 알려진 나프탈렌이 먼지진드기에 대해서도 유의한 살충효과를 나타내는지를 확인하기 위해 실시되었다. 페트리디쉬에 5 g의 혼합배지를 넣고 나프탈렌 농도를 달리한 6개의 그룹을 설정한 후 살아있는 세로무늬 먼지진드기 20마리씩을 분주하여 $25^{\circ}C$, 상대습도 75% 조건에서 4 주간 배양하였다. 그룹별 나프탈렌 농도에 따른 회수 된 먼지진드기의 개체수는 0 mg에서 (대조군) 191.5, 1 mg에서 24.3, 2 mg에서 1.3, 3 mg에서 1.3 이었고, 5 mg 및 10 mg 그룹에서는 살아있는 먼지진드기가 회수되지 않았다. 이상으로 나프탈렌은 실험실적 조건에서 세로무늬 먼지진드기에 대한 살충효과가 있는 것으로 확인되었다.

Extracts of Grifola frondosa inhibit the MAPK signaling pathways involved in keratinocyte inflammation and ameliorate atopic dermatitis

  • BACKGROUND/OBJECTIVES: Grifola frondosa, commonly referred to as the maitake mushroom, has been studied extensively to explore its potential health benefits. However, its anti-inflammatory effects in skin disorders have not been sufficiently elucidated. This study aimed to elucidate the anti-inflammatory role of the ethanol extract of G. frondosa in atopic dermatitis (AD) using in vivo and in vitro models. MATERIALS/METHODS: We investigated its impact on skin and spleen inflammatory responses in Dermatophagoides farinae extract (DFE)/1-chloro-2,4 dinitrochlorobenzene (DNCB)-induced AD-like skin lesions in a mouse model. Additionally, we determined the immunosuppressive response and mechanism of G. frondosa by inducing atopic-like immune reactions in keratinocytes through tumor necrosis factor (TNF)-α/interferon (IFN)-γ stimulation. RESULTS: Our study revealed that G. frondosa ameliorates clinical symptoms in an AD-like mouse model. These effects contributed to the suppression of Th1, Th2, Th17, and Th22 immune responses in the skin and spleen, leading to protection against cutaneous inflammation. Furthermore, G. frondosa inhibited the production of antibodies immunoglobulin (Ig)E and IgG2a in the serum of AD mice. Importantly, the inhibitory effect of G. frondosa on inflammatory cytokines in TNF-α/IFN-γ-stimulated AD-like keratinocytes was associated with the suppression of MAPK (Mitogen Activated Protein Kinase) pathway activation. CONCLUSIONS: Collectively, these findings highlight the potential of G. frondosa as a novel therapeutic agent for AD treatment and prevention.

  • Role of metabolism by intestinal bacteria in arbutin-induced immunotoxicity was investigated in splenocyte cultures. Following an incubation of arbutin with 5 different intestinal bacteria for 24 hr, its aglycone hydroquinone could be produced and detected in the bacterial culture media with different amounts. Toxic effects of activated arbutin by intestinal bacteria on lymphoproliferative response were tested in splenocyte cultures from normal mice. Lipopolysaccharide and concanavalin A were used as mitogens for B- and T-cells, respectively. When bacteria cultured medium with arbutin was treated into the splenocytes for 3 days, the medium cultured with bacteria producing large amounts of hydroquinone induced suppression of lymphoproliferative responses, indicating that metabolic activation by intestinal bacteria might be required in arbutin-induced toxicity. The results indicated that the present testing system might be applied for determining the possible role of metabolism by intestinal bacteria in certain chemical-induced immunotoxicity in animal cell cultures.

Silencing MR-1 attenuates atherosclerosis in ApoE-/- mice induced by angiotensin II through FAK-Akt -mTOR-NF-kappaB signaling pathway

  • Myofibrillogenesis regulator-1 (MR-1) is a novel protein involved in cellular proliferation, migration, inflammatory reaction and signal transduction. However, little information is available on the relationship between MR-1 expression and the progression of atherosclerosis. Here we report atheroprotective effects of silencing MR-1 in a model of Ang II-accelerated atherosclerosis, characterized by suppression focal adhesion kinase (FAK) and nuclear factor kappaB ($NF-{\kappa}B$) signaling pathway, and atherosclerotic lesion macrophage content. In this model, administration of the siRNA-MR-1 substantially attenuated Ang II-accelerated atherosclerosis with stabilization of atherosclerotic plaques and inhibited FAK, Akt, mammalian target of rapamycin (mTOR) and NF-kB activation, which was associated with suppression of inflammatory factor and atherogenic gene expression in the artery. In vitro studies demonstrated similar changes in Ang II-treated vascular smooth muscle cells (VSMCs) and macrophages: siRNA-MR-1 inhibited the expression levels of proinflammatory factor. These studies uncover crucial proinflammatory mechanisms of Ang II and highlight actions of silencing MR-1 to inhibit Ang II signaling, which is atheroprotective.

Suppression of Melanose Caused by Diaporthe citri on Citrus Leaves Pretreated with Bio-sulfur

  • Melanose, caused by Diaporthe citri, is one of severe diseases in citrus, a major economic resource in Jeju island. To reduce the usage amount of organic synthetic fungicide, bio-sulfur was tested as an alternative chemical to control citrus melanose in the present study. Direct antifungal activity of bio-sulfur against D. citri was determined through in vitro experiment using artificial nutrient media. Disease severity of melanose on bio-sulfur pretreated citrus leaves was lower than that on untreated ones. To illustrate the mechanism of disease suppression by bio-sulfur, infection structures were observed with a fluorescent microscope and a scanning electron microscope. In fluorescent microscopic observation, most conidia rarely germinated. In addition, hyphal growth on leaves pretreated with bio-sulfur was inhibited compared to that on untreated ones. In scanning electron microscope images of bio-sulfur pretreated leaves, surfaces of most conidia were shrunk while hyphae were morphologically changed and frequently branched. Such microscopic observations were also found for leaves pretreated with a commercial fungicide Dithianon. These results suggest that bio-sulfur may be used to control citrus melanose as an environment friendly alternative to organic synthetic fungicides

Beneficial Effects of Fluorescent Pseudomonads on Seed Germination, Growth Promotion, and Suppression of Charcoal Rot in Groundnut (Arachis hypogea L.)

  • Rhizobacteria are used as inoculants to enhance crop yield and for biological control of fungal pathogens. Fluorescent pseudomonads isolated from the rhizosphere of groundnut showed suppression of the phytopathogen Macrophomina phaseolina that causes charcoal rot of groundnut, an economically important agroproduct. Two strains of fluorescent pseudomonads, designated as PS1 and PS2, were selected as a result of in vitro antifungal activity. After 5 days of incubation at $28{\pm}1^{\circ}C$, both PS1 and PS2 caused clear inhibition zones in dual cultures, restricting the growth of M. phaseolina by 71 % and 74%, respectively. Both the strains were capable of producing siderophores, indole acetic acid, and hydrocyanic acid, and causing phosphate solubilization under normal growth conditions. These strains, when used as inoculants in groundnut, enhanced germination up to 15% and 30% with subsequent increase in grain yield by 66% and 77%, respectively. Conversely, when the pathogen alone was tested 57% decrease in yield was recorded. Thus the studies revealed the potential of the two pseudomonads not only as biocontrol agents against M. phaseolina, but also as a good growth promoter for groundnut.

Autolysis of Pseudomonas aeruginosa Quorum-Sensing Mutant Is Suppressed by Staphylococcus aureus through Iron-Dependent Metabolism

  • Microorganisms usually coexist as a multifaceted polymicrobial community in the natural habitats and at mucosal sites of the human body. Two opportunistic human pathogens, Pseudomonas aeruginosa and Staphylococcus aureus commonly coexist in the bacterial infections for hospitalized and/or immunocompromised patients. Here, we observed that autolysis of the P. aeruginosa quorum-sensing (QS) mutant (lasRmvfR) was suppressed by the presence of the S. aureus cells in vitro. The QS mutant still displayed killing against S. aureus cells, suggesting the link between the S. aureus-killing activity and the autolysis suppression. Independent screens of the P. aeruginosa transposon mutants defective in the S. aureus-killing and the S. aureus transposon mutants devoid of the autolysis suppression revealed the genetic link between both phenotypes, suggesting that the iron-dependent metabolism involving S. aureus exoproteins might be central to both phenotypes. The autolysis was suppressed by iron treatment as well. These results suggest that the interaction between P. aeruginosa and S. aureus might be governed by mechanisms that necessitate the QS circuitry as well as the metabolism involving the extracellular iron resources during the polymicrobial infections in the human airway.

Efficacy of Fluazinam and Iprodione+Propineb in the Suppression of Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii, the Causal Agents of Seed Decay in Soybean

  • Seed decay of soybean caused by Diaporthe phaseolorum, Colletotrichum truncatum and Cercospora kikuchii is a serious disease when soybean is harvested under warm and wet weather conditions. Benomyl has been used for controlling the disease, however, benomyl application may be limited due to common occurrence of resistance. The efficacy of 21 fungicides against the pathogens was evaluated in vitro. Among the fungicides tested, benomyl, carbendazim, fluazinam, iprodione+propineb, thiophanate-methyl, and triflumizole were found effective and were evaluated for their ability to control the seed pathogens. Fluazinam completely inhibited mycelial growth at a concentration of 100 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for D. phaseolorum; and at a concentration of 500 $\mu\textrm{g}$/$\textrm{m}{\ell}$ for C. truncatum and C. kikuchii. $EC_90$ values of fluazinam were similar to that of benomyl. Because fluazinam, iprodione+propineb, and triflumizole were found effective against the seed pathogens, these were subjected for field-testing. Suppression of pod and seed infection by fluazinam and iprodione+propineb was as high as that of benomyl without any reduction in agronomic characters of soybean. This study shows that fluazinam and iprodione+propineb may be used in combination with benomyl to control seed pathogens, manage resistance, and ensure production of high quality soybean seeds.

Inhibition of Hepatitis B Virus Replication by in vitro Synthesized RNA

  • Human hepatitis B virus (HBV) is a pathogen related to the development of liver diseases including chronic hepatitis, cirrhosis, and hepatocellular carcinoma (HCC). However, the efficient methods to suppress HBV replication have not been developed yet. Therefore, we have used RNA interference (RNAi) as a potential tool for the suppression of HBV replication. Here, we designed a 21 nt small intefering dsRNA (siRNA) against hepatitis B virus X (HBx) RNA with 3' overhanging ends derived from T7 promoter. It has been reported that HBV X protein plays an important role in HBV gene expression and viral replication. The suppression of HBx gene expression by the 21 nt siRNA was investigated by Northern blot analysis and chloramphenicol acetyl transferase (CAT) assay. The level of HBx mRNA was decreased by siRNA in a dose-dependent manner. We also found that the 21 nt siRNA inhibited the HBV replication in hepatocellular carcinoma cell.
