줄기 절간조직 배양에 의한 교잡종 사시나무의 대량증식 (Multiple Shoot Induction from Ex Vitro and In Vitro Derived Stein Node Culture of Populus alba L.$\times$P.grandidentata Michx.)

  • 잡종 사기나무의 기내 대량증식을 목적으로 줄기절간 유래의 기내 및 기외시료를 이용하여 다경줄기 유도에 관여하는 식물생장 조절물질의 효과에 대하여 조사하였다. 기외시료의 측아를 제거한 다음 20에서 30$\mu$M 의 zeatin이 함유된 WPM 배지에 6주간 배양하였을때 각각 11개와 13개의 다경줄기가 유도되었다. 그러나 기외시료의 측아가 붙어있는 상태에서 기내배양하여 2주후에 측아를 제거하고 동일 배지에 배양하였을 경우에는 BA가 1.0에서 2.0 $\mu$M 함유된 배지에서도 13개와 15개의 다경줄기를 유도할 수 있었으며, 특히 이때 생산된 줄기의 상태는 아주 건전한 것으로 나타났다. 기내줄기를 사용하였을때는 20 $\mu$M의 zeatin을 처리하였을때 가장 많은 줄기를 생산할 수 있었으며, 생산된 조직배양묘는 일정기간 순화를 시킨 후 온실이나 포지에 식재가 가능하였다.

In vitro micropropagation of water hyacinth (Eichhornia crassipes)

  • This study was conducted to refine a micropropagation method of water hyacinth (Eichhornia crassipes) in vitro. When young shoots were cultured on media with various concentrations of BA or TDZ alone, LS medium containing $5.0\;mgl^{-1}$ BA was found favorable for shoot proliferation from young shoots with a mean of 4.2 shoots. Using BA together with IAA, more shoots were obtained on LS medium containing $5.0\;mgl^{-1}$ BA and $1.0\;mgl^{-1}$ IAA with a mean of 5.7 shoots. In liquid medium, number of shoots and fresh weight per explant increased significantly. The best shoot proliferation and increasing of fresh weight were achieved on LS liquid medium containing $5.0\;mgl^{-1}$ BA and $1.0\;mgl^{-1}$ IAA with 6.9 shoots and more than 4,000 mg fresh weight. Of the different concentrations of LS salt, double strength of LS medium provided the highest shoot proliferation with 7.3 shoots, and fresh weight with 5,539 mg per explant. Shoot proliferation on LS medium containing $50\;gl^{-1}$ sucrose had better results with 8.7 shoots and 5,979 mg per explant in fresh weight than other conditions. In conclusion, the optimal level for shoot proliferation and biomass increase of water hyacinth was attained with the application of the double strength of LS medium containing $5.0\;mgl^{-1}$ BA, $1.0\;mgl^{-1}$ IAA and $50\;gl^{-1}$ sucrose.

Multiple Shoots Regeneration and In vitro Bulblet Formation from Garlic Callus

  • The leaf segments of garlic (Allium sativum L.) were cultured in vitro and determined optimal concentration of plant growth regulators and sugars for callus induction, multiple shoots regeneration and in vitro bulblet formation. Highest yield of callus was observed in the leaf segment culture on Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-D, 30 g/L sucrose and 8 g/L agar. Regeneration rate of multiple shoots from callus was high in the MS medium supplemented with kinetin 3.0 + NAA 3.0 mg/L or SA 1.0 + NAA 3.0 mg/L, containing 30 g/L sucrose. High rate of bulblet formation was observed as the concentration of jasmonic acid increased from 0.5 to 2.0 mg/L in medium, whereas addition of gibberellic acid significantly suppressed bulblet formation. The rate of in vitro bulbing was as high as $96\%$ in MS medium supplemented with 2.0 mg/L jasmonic acid and 120 g/l sucrose after two month culture at $25{\pm}1^{\circ}C$ under 16 hours day light.

Rapid Micropropagation of Hovenia dulcis Thunb. Through in vitro Stem Nodal Cultures

  • An efficient method for in vitro propagation of the medicinal plant Hovenia duleis, was established. Plantlets for micropropagation of H. dulcis were obtained from in vitro germinated seeds. The effectiveness of various levels of cytokinins (BAP, Kinetin and TDZ) on multiple shoot formation from stem nodes was tested. BAP (1.0 mg/L) treatment induced highest number of multiple shoots. The growth pattern of plantlet on various culture media was undertaken. The shoot elongation was optimal on 2MS basal medium without growth regulators. The in vitro rooting ability of H. dulcis shoots was examined with two-auxins IAA and IBA. The IAA (1.0 mg/L) treatments induced earliest rooting with maximum number of roots and root growth. Rooted shoots were transferred directly to small pots with artificial soil and such established plant exhibited a normal growth pattern similar to wild plantlet.

In vitro Conservation of Coleus forskohlii- an Endangered Medicinal Plant

  • Protocols for in vitro conservation was developed for Coleus forskohlii. Plants maintained both in field served as explant source. Shoot tips and single node cuttings were used to optimize protocols for in vitro multiplication. MS basal medium supplemented with $0.54\;{\mu}M$ naphthalene acetic acid (NAA) and $8.87\;{\mu}M$ benzy-ladenine (BA) induced multiple shoots in shoot tips and nodes. Shoot multiplication was amplified with a gradual decrease of BA concentration, leading to its final omission after 4 months. Concomitant rooting on multiplication media enabled successful establishment extra vitrum. For in vitro conservation studies, experiments were carried out with 2-3 week maintained in vitro plants under standard and reduced culture conditions (SCC, RCC). In vitro plants could be successfully conserved in full strength MS medium (FMS) under SCC for 6 months without subculture with full potential to regenerate, producing viable shoots and nodes. The root production remained unaffected due to conservation, showing high rooting activity in mannitol and low temperature treatments. Preset low temperature (15 and $10^{\circ}C$) and reduction in media constituents does not appear to favour conservation, although the former accomplished conservation levels equal to (FMS) under SCC.

제주(濟州) 자생(自生) 왕벚나무(Prunus yedoensis Matsumura)의 기내(器內) 줄기 증식(增殖)을 위한 배양조건(培養條件) 구명(究明) (Study on the Various Conditions of In Vitro Culture for Mass-propagation of Prunus yedoensis Matsumura)

  • 제주 자생 왕벚나무 동아에서 분화된 줄기를 이용하여 대량증식 하고자 기내배양조건을 구명하였다. 다섯 종류의 배지에서 모두 양호한 생장을 하여 차이가 나타나지 않았으나 sucrose가 첨가되지 않은 배지에서는 생장이 매우 저조하였다. BAP와 $GA_3$를 여러 농도로 처리한 결과 BAP 처리구에서 평균 3.5~9.5개의 줄기가 유도되었다. 그러나 BAP의 농도가 높아질수록 줄기의 길이생장은 저조하였다. $GA_3$ 단독 처리구에서는 줄기가 발생하지 않았으나 BAP와 혼용처리 하였을 때 BAP 단독 처리구보다 더 많은 줄기가 발생하였으며, 길이 생장도 양호하였다. 그 중 BAP $0.5mg/{\ell}$$GA_3$ $4.0mg/{\ell}$ 조합에서 가장 많은 줄기가 발생하였다. 배양기간이 경과할수록 줄기형성은 많아졌으나 12주간 배양된 줄기는 정단부가 고사하였다. 배양 8주째 양호한 줄기를 다량으로 얻을 수 있었다.

액체배지 첨가에 의한 Anthurium andreanum ′Atlanta′의 기내생육 촉진 (Growth Stimulation of In Vitro Shoots by the Post-supplying of Liquid Medium in Anthurium andreanum ′Atlanta′)

  • In order to enhance shoot elongation and rooting of Anthurium andreanum 'Atlanta' in vitro, 15mL of liquid media containing various concentrations of activated charcoal, sucrose and MS salts were added in same vessels after small shoots were induced from the calli on mudium supplemented with 10.0mg/L BA and 0.1mg/L 2.4-D. The post-supplying of 15mL liquid medium containing MS macro and micro elements, 30g/L sucrose and 5.0∼10.0g/L activated charcoal was significantly stimulated the shoot elongation and rooting of regenerated shoots from calli. The medium addition was also resulted in the enhanced soil survival, elongation and rooting of plantlets in cultural soil mixed with perlite and vermiculite(1 : 1)

Optimal Culture Conditions for the Shoot Growth of Freesia 'Shiny Gold' during Bioreactor Culture

  • For rapid production of freesia 'Shiny Gold' shoots by using a bioreactor, several culture conditions were investigated. Young shoots (< 1 cm) obtained from freesia corm section in vitro were used as plant materials for this experiment. As a basic experimental environment, 20 young shoots were inoculated into a 5 L balloon type bubble reactor which contained 1 L 1/2 strength MS medium supplemented with 30 g sucrose (3%), and the aeration was 0.1 vvm (vessel volumes per minute). The bioreactors were placed in a growth room with $23^{\circ}C$ temperature, 60% relative humidity and $60{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ light condition (16 h/8 h, day/night). The concentrations of MS media were set with 1/4, 1/2, 1 strength, medium volume 10, 20, 40%, sucrose concentration 3, 6, 9%, and aeration 0.1, 0.2, 0.4 vvm. After 4 weeks of cultivation, the growth indexes including the fresh and dry weight, and plant height were evaluated. At the same time, the consumption, pH, and EC of medium were estimated 4 weeks after incubating. The best results were achieved when 40 young shoots were incubated in a bioreactor in which 1 L of 1/2 strength MS medium supplemented with 6% sucrose was used for the rapid production of freesia shoots. The shoots were 17 cm in plant height and 1.0 g in fresh weight only 4 weeks after incubation which could be a good plant material suitable for corm enlargement in vitro. No correlation was observed between the growth of freesia shoots and the consumption, pH or EC of medium.

Enhancing in vitro Growth of Bulbs for Mass Propagation of Lily Germplasm

  • Plants regenerated from in vitro cultures carry chromosomal variations, especially in long-term culture. Reducing the duration of plant tissue culture is one of the ways to reduce genetic and epigenetic changes. In this study, we reduced the duration of long-term culture and repeat subculture using small bulblets derived from bulb scales in two lily cultivars. The adventitious bulblets derived from bulb-scale tissue were cultured on three different media containing Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. About seven weeks later, the number of newly propagated multiple shoots in the two media, A and B media, showed little differentiation. Compared to both media, the number of propagated multiple shoots increased 5-fold in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal (C medium). The number of propagated multiple shoots ranged from 5 to 6 and 4 to 6 with an average of 5 in TropicalPink and GreenStar cultivars, respectively. The flow cytometric measurements indicated no variation in the ploidy level between control and in vitro propagated plants.

Micropropagation of Echinosophora koreensis Nakai, a Korean Endemic Species in Danger, Using Axillary Buds

  • To establish in vitro axillary bud culture conditions of Echinosophora koreensis Nakai, one of Korean endemic endangered species famous for beautiful flowers, we tested the influence of plant growth regulators (PGRs) in shooting and rooting stage from in vitro plants. In shoot multiplication, addition of 6-benzylaminopurine (BA) to the media induced 2.5 to 3 shoots per bud during 4 weeks of culture. And media including 0.5 mg L-1 thidiazuron (TDZ) produced 3 to 4 shoots per bud. However, zeatin and isopentenyl adenine (2-ip) were not successful to increase shoot number, and the combination treatments of BA with other PGRs were also not effective. Shoots were smaller than 2 cm in length, in most of the treatments. In rooting, naphthalene acetic acid (NAA) treatments in the range of 0.5 to 4.0 mg L-1 appeared to increase rooting rate by 10% to 60% approximately when compared with the control but roots developed with callus clusters. Indole butyric acid (IBA) addition had little effect on rooting (below 10%), while some roots were longer than in NAA treatments and some shoots were longer on high IBA concentrations (4.0 to 8.0 mg L-1). It is suggested that micropropagation is a highly applicable and promising to multiplication and conservation of rare and endangered endemic species.
