Ginsenoside Rg1 is a natural compound with various efficacies and functions. It has beneficial effects on aging, diabetes, and immunity, as well as antioxidant and proliferative functions. However, its effect on porcine embryo development remains unknown. We investigated the effect of ginsenoside Rg1 on the in vitro development of preimplantation porcine embryos after parthenogenetic activation in high-oxygen conditions. Ginsenoside treatment did not affect cleavage or blastocyst formation rates, but did increase the total cell number and reduced the rate of apoptosis. In addition, it had no effect on the expression of four apoptosis-related genes (Bcl-2 homologous antagonist/killer, B-cell lymphoma-extra large, Caspase 3, and tumor protein p53) or two metabolism-related genes (mechanistic target of rapamycin, carnitine palmitoyltransferase 1B), but increased the expression of Glucose transporter 1 (GLUT1), indicating that it may increase glucose uptake. In summary, treatment with the appropriate concentration of ginsenoside Rg1 ($20{\mu}g/mL$) can increase glucose uptake, thereby improving the quality of embryos grown in high-oxygen conditions.
To determine optimal plant density of forage rye in southern areas of Korea, forage rye cv. Paldanghomil, the highest yielding variety among experimental varieties of forage rye, was grown under five different plant density. Yield components such as plant height, and number of leaves were highest at the broad casting. Plants sown at broadcasting also showed highest fresh and dry matter yield. When plants were grown of the broad casting, they showed higher values in content of crude protein and lower values in contents of crude fiber such as neutral detergent fiber(NDF), acid detergent fiber(ADF), cellulose and lignin. There was no relationship between variation of in vitro dry matter digestibility(IVDMD) and plant density. Plants sown at the broad casting showed highest digestible dry matter yields.

Spices can be used as novel supplements to enhance the utilization of low quality forages (LQF) and reduce nutrient wastage by ruminant animals. However, it is essential to characterize these spices alongside LQF before testing their potential use as supplements in ruminant diets. This study characterized four spices (cinnamon, cumin, clove and turmeric) alongside three forages (rice straw, wheat straw and hay) for their chemical components before evaluating their effect at four different doses (0, 10, 30 and 90 mg/g forage DM) on the in vitro rumen degradability of dry matter (DM) (IVD) and organic matter (OM) (IVOMD) of these forages at various incubation times. It appeared that some spices could provide complementary nutrients which could improve the utilization of LQF where hay had better chemical composition than the other two forages. Cumin contained more crude protein (CP), ether extract and mineral contents whereas turmeric contained more soluble sugars than the other spices. Cinnamon was least acceptable as a ruminant supplement due to its higher condensed tannin and saponin and lower CP and mineral contents. The IVD and IVOMD were highest for hay and lowest for wheat straw with all spices at all incubation times (p<0.001). Due to relatively better nutrient profiles, cumin and turmeric had greater effect on IVD and IVOMD of the forages. In contrast, cinnamon had negative effects on IVD and IVOMD. IVD and IVOMD were greater at 10 mg/g than at other levels of most spices suggesting that using certain amounts of spices can increase forage degradability. However, the choice of a spice will depend upon the forage type being offered to ruminants. Further studies will examine the effect of these spices on fermentation profile, methane production and nitrogenous loss by ruminants.

Candida utilis can assimilate fatty acids, so it was hypothesized that the treatment of rice by Candida utilis would improve feed quality by reducing fat content and adding the yeast function that would stimulate rumen microbes. In this study, the oil assimilation ability of Candida utilis IFO1086, 0988, 0626 and the effect of treatment of Candida utilis IFO1086, IFO0626 on the nutrient contents of rice bran were examined. The effect of Candida utilis addition on the in vitro degradability of forage was also investigated. It was found that the oil assimilating ability of IFO1086 and IFO0626 was significantly (p<0.01) higher than that of IFO0988. Candida utilis treatment reduced the EE content and increased the CP, ADF and NDF percentage. The absolute amount of ether extract was decreased by 35.9% in IFO1086 and IFO0626 treatment. The absolute amount of crude protein was not changed by yeast treatment. The ADF and NDF amounts were increased. The addition of Candida utilis increased in vitro forage degradability significantly (p<0.05). Based on these results it can be postulated that treatment of rice bran by Candida utilis may improve feed quality by reducing fat content, increasing the CP content and adding the function of yeast for stimulating rumen microbes.

Objective: Research was conducted to test the effect of including fiber-rich feedstuffs in practical pig diets on nutrient digestibility, nitrogen balance and ammonia emissions from slurry. Methods: Three Vietnamese fiber sources were screened, namely cassava leaf meal (CL), cassava root residue (CR), and tofu by-product (TF). Accordingly, a control diet (Con) with 10% of dietary non-starch polysaccharides (NSP) and three test diets including one of the three fiber-rich feedstuffs to reach 15% of NSP were formulated. All formulated diets had the same level of crude protein (CP), in vitro ileal protein digestible and metabolisable energy, whereas the in vitro hindgut volatile fatty acid (VFA) production of the test diets was 12% to 20% higher than the control diet. Forty growing barrows with initial body weight at $28.6{\pm}1.93kg$ ($mean{\pm}standard$ deviation) were allocated to the four treatments. When pigs reached about 50 kg of body weight, four pigs from each treatment were used for a nitrogen balance trial and ammonia emission assessment, the remaining six pigs continued the second period of the feeding trial. Results: The TF treatment increased fecal VFA by 33% as compared with the control treatment (p = 0.07), suggesting stimulation of the hindgut fermentation. However, urinary N was not significantly reduced or shifted to fecal N, nor was slurry pH decreased. Accordingly, ammonia emissions were not mitigated. CR and CL treatments failed to enhance in vivo hindgut fermentation, as assessed by fecal VFA and purine bases. On the contrary, the reduction of CP digestibility in the CL treatment enhanced ammonia emissions from slurry. Conclusion: Dietary inclusion of cassava and tofu byproducts through an increase of dietary NSP from 10% to 15% might stimulate fecal VFA excretion but this does not guarantee a reduction in ammonia emissions from slurry, while its interaction with protein digestibility even might enhance enhanced ammonia emission.

According to the results from the experiments of selecting a suitable variety of forage rape and comparing productivity and nutrient quality between forage rape and oil seed rape, Velox appeared to be the most suitable variety in terms of productivity and nutrient quality at the southern area of Korea among the varieties used in the experiments. Consequently, Velox was grown under two different cultural methods, individual culture and population culture, and productivity and variation of nutrient quality during the growth period were observed and compared between the cultural methods. The results from the experiment are summarized as follows: 1. Although variations were great in plant fresh weight and plant dry matter weight among plants, individual culture showed significantly higher value in these characters at the 1 % level than population culture. Plant fresh weight and plant dry matter weight were largely dependent upon the amount of branches in case of individual culture. However, in population culture, they were equally dependent upon the amount of branches and main stem. There was no significant difference in dry matter percentage between cultural methods, and main stem showed highest dry matter percentage. 2. Content of crude protein was decreasing gradually as plants continued to grow. Individual culture showed higher content of crude protein than population culture from 90 to 120 days after sowing but vice versa from 180 days after sowing to flowering stage. Contents of fiber such as NDF, ADF, cellulose and lignin was low at the early stage of growth. It was increasing gradually as plants grew older and at the latter stage of growth plants under individual culture showed higher values in contents of fiber. 3. In vitro dry matter digestibility (IVDMD) of both stems and leaves was decreasing gradually as plants grew older. Plants under individual culture showed higher IVDMD of stems than plants under population culture, but no significant difference in IVDMD of leaves was observed between cultural methods.

This study was conducted at the rice field of Livestock Experiment station, Suweon to investigate the effects of harvesting time on forage production and quality of rye (Secale cereale L.) on paddies. The results obtained are summarized as follows; 1. Fresh matter yield was peaked at the 10 days (May 10) after heading, however, dry matter yield was increased in proportion to maturity. 2. Digestibility (in vitro) showed the 83-77% by heading stage (Apr. 25-30) but decreased to 66-58% after heading stage. 3. Digestible yield reached to maximum level, 619.2 kg/lOa at the 10 days after heading (May 10). 4. The content of crude protein and crude fat in forage rye was decreased with advancing the maturity. 5. The lactic acid content of rye silage ranged from 1.3% to 2.0%.
Protein modifications of recombinant pharmaceuticals have been observed both in vitro and in vivo. These modifications may result in lower efficacy, as well as bioavailability changes and antigenicity among the protein pharmaceuticals. Therefore, the contents of modification should be monitored for the quality and efficacy of protein pharmaceuticals. The interface of EPO and its receptor was visualized, and potential amino acids interacting on the interface were also listed. Two different types of modifications on the interface were identified in the preparation of rHu-EPO BRP. A UPLC/Q-TOF MS method was used to evaluate the modification at those variants. The modification of the oxidized variant was localized on the Met54 and the deamidated variants were localized on the Asn47 and Asn147. The extent of oxidation at Met54 was 3.0% and those of deamidation at Asn47 and Asn147 were 2.9% and 4.8%, respectively.

This study was conducted to observe the fermentative quality and anthocyanin content in whole crop colored barley silage during storage periods and anthocyanin stability in in vitro ruminal fluid. Silages of colored barley cultivar "Boanchalbori" and normal barley cultivar "Yuyeonbori" were stored during 0, 2, 4, 6, and 12 months. The in vitro ruminal fluid was fermented for 0, 6, 12, 24, and 48 hrs. For the feed value, crude protein of colored barley silage was slightly increased in the silage compared to that of normal barley silage, and being increased up to 2 months after ensiling and thereafter maintained at the similar level. Neutral detergent fiber (NDF) and acid detergent fiber (ADF) contents of both the barley significantly increased by prolonged storage of 2 months (p<0.05), but they were maintained at the constant level after 2 months of storing silage. Whereas TDN (total digestible nutrients) contents of them were decreased by the prolonged storage of 2 months (p<0.05), then maintained at the constant levels. The fermentative quality and pH values in both the barley silages were slightly decreased during the storage time. Lactic acid and acetic acid contents were increased during prolonged storage period, but not significantly different among treatments. Butyric acid was not detected. In the colored barley silage, pH value showed slightly lower compared to that of the normal barley silage but not significant, and lactic acid content was significantly higher than the normal barley silage (p<0.05). The total anthocyanin content in the whole crop colored barley silage decreased to 42% after 2 months of ensilage, however maintained at the constant level until 12 months of ensilage. In the case of anthocyanin stability on in vitro ruminal fluid digestion, the pH value of the ruminal fluid was slightly lower at 6, 12, 24, 48h incubation time and the content of anthocyanin was at similar levels. These results indicated that the colored barley showed higher fermentation quality, and total anthocyanin content was maintained stable at 42% level of the first value in storing silage. As the anthocyanin had higher stability in the ruminal fluid, the colored barley has a potential as functional feeds for Ruminants.
Cytokines are protein mediators that possess the ability to assist cell-to-cell communication in immune system-related activities. In general, pathogen endotoxins activate the release of inflammatory mediators, and with time, there is an increase in the cytokine levels in the body. Interleukin (IL)-6 mediates the acute-phase inflammatory response, and elevated IL-6 levels have been reported in peritoneal fluids of women with pelvic inflammation and endometriosis, thereby associating it with oocyte quality and infertility. To overcome subfertility or infertility in humans and animals, the present study was done to examine the effect of recombinant IL-6 on porcine oocytes matured in vitro and subsequently to determine the fertilization rate and embryo development. Porcine oocytes were incubated with varying concentrations of IL-6 (0 - 2 ㎍·mL-1) for 44 h followed by in vitro fertilization and culturing of the oocytes. The oocytes or embryos were fixed with 3.7% paraformaldehyde (PFA) and stained with fluorescence dyes, and the meiotic spindle, chromosome organization, fertilization status and embryo development were subsequently assessed under a fluorescence microscope. We observed induction of an abnormal meiotic spindle alignment in the oocytes incubated with IL-6 compared to the control oocytes incubated without IL-6. Moreover, significantly decreased fertilization rates and embryo development were observed for oocytes incubated with IL-6 (p < 0.05). Thus, an increased IL-6 level during oocyte maturation could be associated with fertilization failure due to an aberrant chromosomal alignment and a disruption of the cortical granules. Taken together, our results indicate that successful assisted reproduction can be achieved by controlling the levels of inflammatory cytokines.
