Objectives : This study was conducted to evaluate the efficacy of a complex mixture of natural substances of ginseng and baeknyeoncho on the arthritic rats. Methods : In vitro experiments were conducted to ensure the stability of the complex. After setting toxicity and concentration by MTT assay, the antioxidant effect was measured through DPPH and ABTS radical scavenging activity. To confirm the anti-inflammatory effects of the complex, levels of nitric oxide (NO) and pro-inflammatory cytokines (IL-1β, TNF-α) were measured in LPS-treated macrophage cell lines (RAW264.7). We injected monosodium iodoacetate (MIA) 50 μl (60 mg/ml) into knee joints of rats to induce osteoarthritis. The rats were divided into three groups (normal (n=5), control (n=5), and OR (n=5) group). The control group consumed 2 mg/kg of physiological saline once a day for 4 weeks, and the OR group was mixed at a concentration of 416.5 mg/kg of Baengnyeoncho (O) and 208.25 mg/kg of red ginseng (R) and ingested 1 mL each 5 days a week. Results : This complex increased the DPPH and ABTS radical scavenging rate. The complex decreased NO production and pro-inflammatory cytokine production of macrophages. In the OR group, the secretion of cytokine in serum was decreased. In histopathological examination, the joint tissue of the composite showed less damage to the synovial membrane, cartilage, and fibrous tissue than the control group. Conclusions : As a result of this study, natural complexes have antioxidant, anti-inflammatory and cartilage protection effects. Therefore, we expect the complex to be effective in treating osteoarthritis.
Deer antler velvet is widely used in traditional medicine for its anti-aging, antioxidant, and immunity-enhancing effects. However, few studies have reported on the discovery of probiotic strains for deer antler fermentation to increase functional ingredient absorption. This study evaluated the ability of probiotic lactic acid bacteria to enhance the concentrations of bioactive molecules (e.g., sialic acid and gamma-aminobutyric acid [GABA]) in extracts of deer antler velvet. Seventeen strains of Lactobacillus spp. that were isolated from kimchi and infant feces, including L. sakei, L. rhamnosus, L. brevis, and L. plantarum, and those that improved the life span of Caenorhabditis elegans were selected for evaluation. Of the 17 strains, 2 (L. rhamnosus LFR20-004 and L. sakei LFR20-007) were selected based on data showing that these strains increased both the sialic acid and GABA contents of deer antler extract after fermentation for 2 d and significantly improved the life span of C. elegans. Co-fermentation with both strains further increased the concentrations of sialic acid, GABA, and metabolites such as short-chain fatty acids and amino acids. We evaluated the biological effects of the fermented antler velvet (FAV) on the antibacterial immune response in C. elegans by assessing worm survival after pathogen infection. The survival of the C. elegans conditioned with FAV for 24h was significantly higher compared with that of the control worm group fed only normal feed (non-pathogenic E. coli OP50) exposed to E. coli O157:H7, Salmonella typhi, and Listeria monocytogenes. To evaluate the protective effects of FAV on immune response, cyclophosphamide (Cy), an immune-suppressing agent was treated to in vitro and in vivo. We found that FAV significantly restored viability of mice splenocytes and immune promoting-related cytokines (interleukin [IL]-6, IL-10, inducible nitric oxide synthase [iNOS], interferon [IFN]-γ, and tumor necrosis factor [TNF]-α) were activated compared to non-fermented deer antlers. This finding indicated the protective effect of FAV against Cy-induced cell death and immunosuppressed mice. Taken together, our study suggests that immune-promoting antler velvet can be produced through fermentation using L. rhamnosus LFR20-004 and L. sakei LFR20-007.
Background: Lung inflammation occurs in many lung diseases, but has limited effective therapeutics. Ginseng and its derivatives have anti-inflammatory effects, but their unstable physicochemical and metabolic properties hinder their application in the treatment. Panaxadiol (PD) is a stable saponin among ginsenosides. Inhalation administration may solve these issues, and the specific mechanism of action needs to be studied. Methods: A mouse model of lung inflammation induced by lipopolysaccharide (LPS), an in vitro macrophage inflammation model, and a coculture model of epithelial cells and macrophages were used to study the effects and mechanisms of inhalation delivery of PD. Pathology and molecular assessments were used to evaluate efficacy. Transcriptome sequencing was used to screen the mechanism and target. Finally, the efficacy and mechanism were verified in a human BALF cell model. Results: Inhaled PD reduced LPS-induced lung inflammation in mice in a dose-dependent manner, including inflammatory cell infiltration, lung tissue pathology, and inflammatory factor expression. Meanwhile, the dose of inhalation was much lower than that of intragastric administration under the same therapeutic effect, which may be related to its higher bioavailability and superior pharmacokinetic parameters. Using transcriptome analysis and verification by a coculture model of macrophage and epithelial cells, we found that PD may act by inhibiting TNFA/TNFAR and IL7/IL7R signaling to reduce macrophage inflammatory factor-induced epithelial apoptosis and promote proliferation. Conclusion: PD inhalation alleviates lung inflammation and pathology by inhibiting TNFA/TNFAR and IL7/IL7R signaling between macrophages and epithelial cells. PD may be a novel drug for the clinical treatment of lung inflammation.
Rosmarinic acid (RA) is a phenolic ester that protects human keratinocytes against oxidative damage induced by ultraviolet B (UVB) exposure, however, the mechanisms underlying its effects remain unclear. This study aimed to elucidate the cell signaling mechanisms that regulate the antioxidant activity of RA and confirm its cyto-protective role. To explore the signaling mechanisms, we used the human keratinocyte cell line HaCaT and SKH1 hairless mouse skin. RA enhanced glutamate-cysteine ligase catalytic subunit (GCLC) and glutathione synthetase (GSS) expression in HaCaT cells in a dose- and time-dependent manner. Moreover, RA induced nuclear factor erythroid-2-related factor 2 (NRF2) nuclear translocation and activated the signaling kinases protein kinase B (AKT) and extracellular signal-regulated kinase (ERK). Treatment with the phosphatidylinositol 3-kinase (PI3K) inhibitor LY294002, the ERK inhibitor U0126, and small interfering RNA (siRNA) gene silencing suppressed RA-enhanced GCLC, GSS, and NRF2 expression, respectively. Cell viability tests showed that RA significantly prevented UVB-induced cell viability decrease, whereas the glutathione (GSH) inhibitors buthionine sulfoximine, LY294002, and U0126 significantly reduced this effect. Moreover, RA protected against DNA damage and protein carbonylation, lipid peroxidation, and apoptosis caused by UVB-induced oxidative stress in a concentration-dependent manner in SKH1 hairless mouse skin tissues. These results suggest that RA protects against UVB-induced oxidative damage by activating AKT and ERK signaling to regulate NRF2 signaling and enhance GSH biosynthesis. Thus, RA treatment may be a promising approach to protect the skin from UVB-induced oxidative damage.
본 실험 결과, EF 열수 추출물이 항산화 활성뿐만 아니라, MAPK 신호 경로의 활성을 조절하고 전 염증성 사이토카인의 수준을 낮추는 것으로 나타났으며, 특히 EF 열수 추출물의 낮은 농도보다 높은 농도에서 더 뛰어난 효과를 나타내었다. 이는 EF 열수 추출물이 NADPH 산화효소, 항산화 단백질 및 염증성 단백질 발현을 조절하여 위 조직 내 급성 위염을 완화시킬 수 있음을 시사한다.
말라카이트 그린과 메틸렌 블루 같은 기본 염료는 1930년대부터 물 곰팡이 감염을 통제하는 소독제로 사용되었다. 하지만 이 염료의 발암성 및 생체 내부 축적이 보고된 후 공중보건을 위해 이 염료의 사용이 금지되었다. 본 연구에서는 천연유황수의 Saprolegnia parasitica의 감염에 대한 치료 효과와 어류에서의 안전성을 평가하고자 하였다. 천연유황수의 항진균성 시험에서 S. parasitica 균사체의 성장이 50 ppm 또는 그 이상의 농도에서 억제되었다. 미꾸라지의 치어와 미성어에 대한 천연유황수의 급성 독성 시험에서 무 영향농도(NOEC)는 100 ppm, 최저 영향농도(LOEC)는 125 ppm 그리고 반수치사농도($LC_{50}$)는 치어에서 125 ppm, 미성어에서 250 ppm으로 측정되었다. 또한, 물벼룩을 이용한 천연유황수의 생태 독성 시험에서는 100 ppm 이하의 농도에서 물벼룩의 유영저해가 나타나지 않았다. 결론적으로, 천연유황수는 50 ppm 농도에서 미꾸라지에 대한 S. parasitica의 감염을 방지하였을 뿐만 아니라 100 ppm 농도까지 미꾸라지에 급성독성 그리고 물벼룩에 생태독성을 나타내지 않았다. 그러므로 미꾸라지 양식에서 많은 문제를 일으키는 수생균을 제거하기 위하여 말라카이트 그린 및 메틸렌 블루 같은 발암성 소독제를 대신하여 천연유황수를 유용하게 사용할 수 있으며 잠재적으로 다른 어종 양식에서도 천연유황수를 친환경 소독제로 사용할 수 있을 것으로 기대된다.
한방에서 건강골(建强骨)시키는 효능이 있는 5가지 약물(골쇄보(骨碎補), 상기생(桑寄生), 김모구척(金毛狗脊), 별갑(別甲), 법파고지(法破古紙))로 구성된 처방(CEDR로 약칭)으로 실험을 하였다. 생쥐의 calvarial osteoblast를 분리하고 배양한 후 실험을 행하였는데, 골 재흡수 인자인 PTH, $1,25(OH)_2D_3$, $TNF-\alpha$, IL-1에 자극받은 osteoblasts는 활동적인 gelatinase를 생산하므로서 collagen 용해를 증가시켰다. 암컷 생쥐의 long bone organ을 culture하여 골 재흡수를 자극하는 IL-1를 실험한 결과, IL-1은 골의 재흡수를 자극하였으며 동시에 존재해있을 때에는 뚜렷한 재흡수를 보였다. 더욱이, indomethacin과 dexamethasone이 $IL-1{\alpha}$의 투여량 증가에 대한 영향을 관찰하였을 때에는 indomethacin과 dexamethasone은 IL-1의 투여량에 대한 곡선 그래프를 오른쪽으로 이동시켰다. 시험관내에서 세포 독성에 대하여 관찰하였을 때, CEDR추출물은 $1-60{\mu}g/ml$의 농도에서 아무런 세포 독성이 나타나지 않았으며, 뿐만 아니라 생쥐의 두개골 세포에서는 $120{\mu}g/ml$ 농도에서 전혀 세포독성이 관찰되지 않았다. CEDR 추출물은 생쥐의 calvarial cells에서 PTH (5 units/ml), $IL-1{\alpha}$ (1 ng/ml), $TNF-\alpha$, $1,25(OH)_2D_3$ (20 ng/ml) 및 $IL-1{\alpha}$, $IL-1{\beta}$에 유도되는 collagen 용해를 보호하는 효과가 있었다. CEDR 추출물을 1시간동안 전처리하였을 때, 그 자체로는 세포 생존에 영향이 없었으며, collagen 용해를 증가시키지도 않았으며 전처치로 collagen 용해를 유의성있게 감소시키지도 않았다. 게다가, 추출물은 $IL-1{\alpha}$와 $IL-1{\beta}$에 의해 유도되는 collagen 용해를 방지하는 효과가 있었다. 1시간 동안 전처리로 한 결과, collagen 용해를 유의성있게 감소시켰다. 흥미로운 것은 CEDR 추출물이 gelatinase 활성과 PTH, $1,25(OH)_2D_3$, $TNF-\alpha$, $IL-1{\beta}$ 및 $IL-1{\alpha}$의 재흡수 인자에 유도되어 진행되는 활성을 억제하였으며, CEDR 전처리시에는 강력한 보호 효과를 나타내었다. CEDR 추출물은 $IL-1{\alpha}$와 $IL-1{\beta}$에 자극되는 골 재흡수를 억제하는 효과를 나타내였으며, 또한 농도를 다양하게 한 CEDR의 전처리시에는 유의성이 있었다. Indomethacin과 dexamethasone의 비 스테로이드성 항 염증 인자에 의한 IL-1에 자극받은 골 재흡수를 억제하는 정도와 현상은 CEDR 추출물을 생쥐의 두개골 배양 시스템에 적용시켜 얻은 결과와 유사하였다. 이러한 결과들로 보건데, CEDR 추출물은 임상적으로 골다공증의 치료에 매우 안정적으로 적용할 수 있음을 제시하고 있다.
본 연구에서는 식물자원으로부터 생리활성을 보유한 새로운 기능성 소재를 찾고자 하였다. 이를 위해 수종의 식물 추출물을 분석하여 강한 항산화능을 보유한 5종(Apios fortunei Maxim., Colubrina arborescens Sarg., Croton caudatus Geiseler, Osmanthus matsumuranus Hayata 그리고 Schima noronhae Reinw.)을 선별하고, 각 추출물의 항산화 활성을 시험관 분석법, 세포실험 모델계, 대상 단백질 발현 분석 등을 통해 평가하였다. 먼저 각 추출물의 항산화 활성을 DPPH 라디칼 소거능을 통해 분석한 결과 모두 양성 대조군으로 사용한 아스코르빈산과 유사한 정도의 높은 활성을 보여 각 소재가 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주에서 $H_2O_2$에 의해 유도된 ROS에 대한 각 추출물의 소거능을 분석한 결과에서도 농도의존적인 강한 소거능을 보였다. 뿐만 아니라 대표적인 항산화효소 중 하나로 항산화능 보유 천연물에 의해 발현이 유도되는 HO-1 및 그 전사인자인 Nrf2의 단백질 발현이 각 추출물의 처리에 의해 증가됨을 보였다. 이러한 결과를 통해 5종 식물 추출물의 강한 항산화 활성을 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.
The purpose of this study was to estimate the effect of a bleaching agent on tooth surfaces and to evaluate the resin bond strength according to different surface treatments on bleached teeth. To prepare for the experimental samples, first, extracted human third molars were used and the body portions of the crowns were cut into four equal-sized specimens. Next, each specimen was mounted in an plastic bottle with self-cured resin and highly polished to have them reveal the enamel or dentin surfaces. Then, the enamel(E) and dentin(D) specimens were divided into four ; 1) non-bleached, laser-treated(NBLA) group 2) bleached, laser-treated(BLLA) group 3) non-bleached, acid-treated(NBAC) group and 4) bleached, acid-treated(BLAC) group. Here, $opalescence^{(R)}$ (10% carbamide peroxide) was used for bleaching agent. The treated specimens were observed by confocal laser scanning microscopy and bonded with composite resin for shear bond test. The following results were obtained from this experiment : 1. Compared with the ENB group, the EBL group was shown be dyed about $20{\mu}m$ deeper with rhodamine B. The DBL group appeared to be caved in at the entry part of the dentinal tubules, was dyed about $20{\mu}m$ deeper and $5{\mu}m$ wider in diameter, compared with the DNB group. 2. In comparison with the EBLAC group, the ENBAC group looked evenly bonded with the resin, while the DNBAC group, compared to DBLAC group, was observed to have its resin tags penetrated about $50{\mu}m$ deeper. Other than those, however, no observable differences between ENBLA and EBLLA group or between DNBLA and DBLLA group were found. 3, At the shear bond test, the ENBAC group was shown to have statistically significant higher shear bond strength than the EBLAC group(p<0.05). No statistically significant differences between the ENBLA and the EBLLA groups were observed(p>0.05). 4. At the shear bond test, the DNBAC group was shown to have statistically significant higher shear bond strength than the DBLAC group(p<0.05). No statistically significant differences between the DNBLA and the DBLLA groups were observed(p>0.05). The in vitro observations above suggest that tooth-bleaching procedure brings a certain change on enamel and dentin surfaces and it weakens the shear bond strength with composite resin when the bleached tooth was acid-treated.
경제적으로 중요한 가치가 있는 제주 자생 왕벚나무를 조직배양을 통하여 대량 증식하고자 동아를 이용할 때 분화 및 생장에 미치는 기내 조건을 구명하고자 하였다. 동아의 채취시가, 배지의 $NH_4NO_3$의 농도, 생장조절물질의 종류(BAP, $GA_3$, IBA) 및 농도에 따른 영향을 조사하였다. 동아의 채취시기는 2월이 적합하였으며 그 중 $1,200mg/{\ell}$$NH_4NO_3$의 농도에서 잎 전개율이 70%로 가장 높았다. 생장조절물질 처리구 중 BAP $3.0mg/{\ell}$ 첨가 배지에서 채취시기에 관계없이 잎의 전개율이 높았다. 반면 IBA 처리구에서는 전혀 잎이 전개되지 않아 동아의 전개를 저해하는 것으로 나타났다. BAP 첨가 배지에서 전개된 동아는 줄기로 자라지 못하고 다발 모양을 이루었다. BAP와 함께 $GA_3$를 처리한 결과 $GA_3$의 농도가 BAP보다 높을 때 줄기 선장에 효과가 있는 것으로 나타났으며, BAP $1.0mg/{\ell}$와 $GA_3$$2.0mg/{\ell}$ 혼합 처리구에서 줄기의 생장이 가장 양호하였다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
