마늘성분의 산화방지작용에 관한 연구 - 제1보 전자공여능 및 과산화지질생성억제효과에 미치는 영향 (Studies on antioxidative action of Garlic components isolated from garlic - Part 1. Effects of garlic components on electron donating ability and inhibitory effect of lipoperoxide formation)

  • In order to study antioxidative acton of garlic, alliin, scordinin, garlic oil, ethanol fraction and non-kaolin fraction which have been discovered from garlic until now were extracted and isolated, and each fraction was utilized as the experimental materials. Antioxidative action of each fraction was compared through the in vitro and in vivo experiments. Electron donatingability on $\alpha$, $\alpha$-diphenyl-$\beta$picrylhydra-$\chi$yl, the inhibitory effect of lipoperoxide formation by TBA and peroxide value were measured and analyzed. RESULTS : 1. When observed antioxidative ability by EDA value, ethanol fraction of garlic components showed the strongest reaction as 15.25. 2. In vitro experiment with TBA value, garlic oil, alliin and ethanol fracton showed distinctive effect on inhibitory effect of lipoperoxide formation. 3. Comparing with the inhibitory effect of lipoperoxide formation with TBA value in vivo, the ethanol fraction was the most effective in the blood or liver by intraperitoneal administration, whereas the ethanol fraction in the blood and non-kaolin fraction in the liver was most effective each other by orally administration. 4. In vitro experiment with peroxide value, garlic oil was distinctive effect on the inhivitory effect of lipoperoxide formation, which was a similar to the trend of TBA value in vitro. 5. Examining the induction time for the first period of lipoperoxide formation in vitro, garlix oil, ethanol fraction and alliin were effective, which was a similar to the trend of TBA value and peroxide value in vitro.

In Vitro법에 의한 식이섬유의 포도당 및 담즙산 흡수 지연 효과 (Retarding Effect of Dietary Fibers on the Glucose and Bile Acid Movement Across a Dialysis Membrane in Vitro)

  • In order to anticipate the physiological function of dietary fibers, glucose and bile acid retarding effects were experimented by using in vitro methods based on dialysis for commercial fibers and dietary fiber residue of food samples. The glucose retarding effect in commercial fibers increased in the order of alginic acid, guar gum, CM-cellulose, citrus pectin > apple pectin > $\alpha$-cellulose and the effect in food fiber residues increased in the order of sea mustard > Korean cabbage, apple > rice bran, barley, soybean, and tangerine. The bile acid retarding effect in commercial fibers increased in the order of citrus pectin, guar gum > CM-cellulose, alginic acid > apple pectin > $\alpha$-cellulose and the effect in food fiber residues increased in the order of barley, rice bran > sea mustard > tangerine > Korean cabbage, soybean > apple. The higher the retarding effect of glucose movement through the dialysis membrane, the more effective the control of the human blood glucose level. As the retarding effect of bile acid movement across the dialysis membrane increased, the human serum cholesterol level correspondingly reduced. Consequently these in vitro methods can be used as a preceding test before undertaking animal and human experiments to predict the physioloical effects of fiber residues from diverse food samples as well as commercially refined fibers.

납의 In Vitro 흡수에 미치는 식이 섬유의 억제효과 (A Suppressive Effect of Dietary Fiber on in Vitro Absorption of Lead)

  • 식이섬유가 중금속인 납의 흡수억제 효과를 가지고 있는지를 알아보기 위하여 반투막을 이용한 in vitro법으로 흡수실험을 실시하였다. 식이섬유중 cellulose는 억제효과를 거의 기대할 수 없었고 guar gum과 carboxymethyl cellulose는 약간 나타났으며 citrus pectin과 sodium alginate는 매우 크게 나타났다. 섬유질 식품중 쌀겨, 밀기울, 배추, 무우, 미역은 납의 흡수억제 효과가 컸으며, 귤, 사과, 김은 효과가 다소 적었다.

Effect of MEM Vitamins Supplementation of In vitro Maturation Medium and In vitro Culture Medium on the Development of Porcine Embryos

  • This study was carried out to examine the influence of minimum essential medium (MEM) vitamins supplementation to in vitro maturation medium and in vitro culture medium on the development of porcine embryos. Porcine embryo development was investigated following cultivation in both in vitro maturation and culture medium with the supplementation of MEM vitamins (0, 0.1, 0.2 and 0.4%) using immature oocytes collected from the ovary of prepubertal gilts. Embryo development was observed and the total cell number in each blastocyst generated under the culture conditions was quantified following supplementation of the medium. The embryonic development rate of the blastocyst and hatched blastocyst was higher, but not significantly so, when 0.4% MEM vitamins were supplemented to the in vitro maturation medium of the porcine oocyte. Interestingly, the total number of cells in the blastocyst was significantly higher in the in vitro maturation MEM vitamins supplemented group compared to either the untreated group or the group which had MEM vitamins supplemented to both in vitro maturation and in vitro culture medium simultaneously (p<0.05). Therefore, the supplementation of 0.4% MEM vitamins to the in vitro mature medium has a beneficial effect on the embryonic development of in vitro produced blastocysts derived from the immature porcine oocytes.

소 체외수정란의 발생배양에 적합한 배양환경 조성 연구 II. 성장인자가 체외수정란의 발생배양에 미치는 효과 (A Study on Culture Environments of In Vitro Matured/In Vitro Fertilized Bovine Embryos II. Effect of Growth Factors on In Vitro Development of Bovine Embryos)

  • This study examined the effects of growth factors in TCM199 on bovine 1-cell embryos development in vitro. After 6 day to 11 day in culture, 15.8%(19/120), 15.3%(20/130), 21.8%(35/160), 27.0%(56/207), 26.3%(53/201) and 30.7%(40/130) of the 1-cell embryos developed into expanding blastocysts in su, pp.ementing TCM199 with control, insulin, IGF-I, IGF-II, FGF and EGF, respectively. Hatching rate of 1-cell embryos in su, pp.ementing TCM199 with FGF, EGF and IGF-II were 21.4%(53/247), 20.3%(42/206) and 16.8%(41/243), respectively. The beneficial effect of growth factors on embryo development in vitro could be duplicated. These data indicate that the presence of FGF, EGF or IGF-II in the culture medium is beneficial for embryo development in vitro and accelerate cell differentiation.

  • The promotive effect of Ca in semi-vitro culture of Oenothera biennis pollen was by and large identical to that of in vitro systems. This promotive effect became additive when boron was supplemented to the media and even more so if K was further supplemented. No such Ca action was observed then performed under the conditions of relatively low temperature. An uncoupling agent of ATP in respiration, DNP, inhibited the promotive action of B, but not that of Ca. The inhibitory effect of DNP was greater at low temperatures. IAA was rather inhibitory on pollen growth in semi-vitro culture, even much greater than it if DNP was supplemented to the IAA-containing media.

  • Several studies have shown that extracts from yellow-green vegetables reveal antitumor activities. In the present study we investigated the effect of phytol in order to elucidate the immunological mechanism of antitumor activity of this substance. The results obtained from the experiment as follows: 1) Phytol showed cytotoxic effect on sarcoma 180 cells in vitro. 2) When phytol was injected into the peritoneal cavity of mice transplanted with sarcoma 180 cells, the average survival time (24.0 days) tended to increase as compared with the nontreated control (19.2 days). 3) When sarcoma 180 cells were injected subcutaneously into the right groin of mice, and then phytol was injected into the peritoneal cavity, the tumor inhibition ratio was 33%. 4) The natural killer(NK) cell activity was significantly augmented by phytol in vitro and in vivo. Similar augmentations of NK cell activity were obtained with culture supernatants of phytol exposed spleen cells and peripheral blood mononuiclear cells. 5) Phytol on the macrophage from peritoneal cavity showed a higher effectiveness in vivo than in vitro. These results indicate that phytol shows the inhibitory effect for growth of sarcoma 180 cells in vitro, also it can augment macrophage and NK cell activities in vivo.

  • This study was conducted to investigate the effects of preincubation time, concentration and exposure time of sperm on in vitro fertilization of porcine follicular oocytes rnatured in in vitro. The results obtained are as follows ; 1. Effect of preincuhation time for porcine sperm capacitation on in vitro fertilization in medium with heparin was investigated. Normal fertilization rate was highest in 15 min(26.4%). However, there were no significant differences among preincuhation times of 5~90 min, 2. Normal fertilization rates of sperm concentrations were 17.0~26.5%, and normal fertilization rate from l$\times$ l05cell /ml concentration was also higher than those of other sperm concentration. 3. Normal fertilization rates of sperm exposure time of 4, 8, 12, 16 and 20 hours were 6.1, 20.8, 27.8, 25.0 and 26.7%, respectively. Normal fertilization rate from sperm exposure time of 12 hours was also higher than that of other sperm exposure times.

  • These experiments were undertaken as a basic study to develop immunocontraceptive vaccine and to understand the role of zona pellucidae in early fertilization process by investigating the effect of monoclonal and polyclonal antibody to porcine zona pellucidae and polyclonal antibody to mouse zona pellucidae on the fertilization of porcine and mouse eggs in vitro. The results obtained in these experiments were summarized as follows : 1. Treatment of porcine and mouse eggs with undiluted anti-zona serum produced intense precipitation layer on the poricne and mouse zonae, respectively, thus resulting in the total inhibition of sperm adherence on surface of zona. 2. In vitro fertilization of eggs pre-treated with 0.3∼10% of various antibodies was examined, and resulting in that 5 and 10% of rabbit polyclonal antibodies to porcine zona inhibited completely both in vitro fertilization and polyspermy of porcine eggs while monoclonal to porcine zona and rabbit polyclonal antibody to mouse zona did not inhibit in vitro fertilization but monoclonal antibody reduced the rate of polyspermy compared to that of control group. Almost the same results were obtained in the study on the effect of anti-zona serum on in vitro fertilization of mouse eggs.

  • The effect of cations on the formation of protein aggregates was examined by in vitro refolding of mutant tryptophan synthase $\alpha$-subunit in which Pro 24 was replaced by Leu. $NH^{4+},; K{^+}; and; Na^{+}$ and no effect, but $Mg^{2+}; and; Ca^{2+}$stimulated the formation of protein aggregates in dose-dependent manner. It is suggested that $Mg^{2+} and Ca^{2+}$ may be implicated in the formation of protein aggregates in vivo.
