Effect of In Vitro Culture conditions on Ex Vitro Sprouting of Bulblets of Oriental Lilium Hybrid cv. Casa Blanca (기내 배양환경이 오리엔탈 백합 '카사블랑카' 의 기외 발아에 미치는 영향)

  • This study was carried out with Lilium oriental hybrid cv. Casa Blanca to observe the effect of in vitro culture conditions on ex vitro sprouting of bulblets. Low temperature (15$^{\circ}C$) inhibited the growth of in vitro bulblets while high temperature ($25^{\circ}C$) enhanced the growth. Bulblets cultured at 15$^{\circ}C$ did not show dormacy while those cultured at 2$0^{\circ}C$ ,$25^{\circ}C$ had a longer dormancy period. High sucrose concentration (9%) induced longer dormancy. Dormancy period was also prolonged in bulblets cultured in vitro at high temperature ($25^{\circ}C$). Dormancy period was more affected by in vitro culture temperature rather than sucrose concentration. Physiological dormancy was released more rapidly when bulblets were cultured at $25^{\circ}C$ for 6 weeks and further transferred at 15$^{\circ}C$ and cultured for another 12 weeks. Treatment of ABA induced the dormancy in Lilium bulblets but when bulblets were subjected to chilling treatment (4$^{\circ}C$ for 8 weeks) nearly 100% sprouting were observed. The medium containing 1.0 mg/L BA or 1.0 mg/L fluridone was also effective to produce non-dormant bulblets.

Effects of Edible Herbs on the Growth of In Vitro Intestinal Microorganisms (산채류가 장내세균의 In Vitro 생육에 미치는 영향)

  • This study was aimed to screen edible herbs which control the composition of intestinal microflora. With in vitro experiments, we screened the water or ethanol extracts of about 60 edible herbs and wild plants in terms of the inhibition activity on the growth of the harmful Clostridium perfringens and growth promoting activity for the beneficial Bifidobacteria. The water extracts of mugwort and small water dropwort inhibited the growth of Cl.perfringens both in agar diffusion method and broth culture. On the other hand, the water extracts of petasites, mugwort, yellow day-lily and bitter cress have shown the promotion effect on the growth of Bifidobacterium longum. In the culture test using human feces as starter, the extracts of the above selected herbs increased the population of Bifidobacteria and Lactobacillus while they reduced the numbers of Cl.perfringens and E.coli.

Effects of Surface Charges on the Retention of Placenta-loaded Liposome Formulations Administered by Intramuscular Route

  • We aimed to optimize the formulation of porcine placental extract (PPE)-loaded liposomes for intramuscular administration and to investigate the effect of surface charges on the muscular retention in mice. PPE-loaded liposomes were formulated to have neutral, anionic, or cationic surface charges. The in vitro release profiles were studied by spectrofluorometry. In vivo distribution patterns at mice were studied using molecular imaging technology. Among the three types of liposomes, 1,2-dioleoyl-3-trimethylammonium-propane and 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine-based cationic liposomes showed the most prolonged in vitro release profile. Consistent with the in vitro results, the in vivo distribution study revealed that the cationic liposomes were retained at the site of administration for the longest period. Our results suggest the potential of cationic PPE-loaded liposomes for sustained release of the components after intramuscular administration.

  • Repellent effcacies of two natural aroma compounds. citronella and citronellal, against mosquitoes were evaluated both in field and in vitro. In vitro, the experiment was conducted with three controlled bands impregnated with $30\%$ citronella extract, $15\%$ citronella extract and $30\%$ citronellal extract, and with band impregnated $30\%$ citronella in field. Raw data was obtained by the means of counting numbers bitten by mosquitoes per unit time, namely human bait method. Comparative repellent efficacies of above three controlled bands were calculated at $86\%$, $73\%$, and $78\%$, respectively in vitro, and $80\%$ in field, showing high repellent effectiveness against mosquitoes. This estimation was also confirmed by t-test compared between control group and each experimental group.

  • For evaluating the cytolytic effects on the mouse thymocytes, four typical antiinflammatory steroids (dexamethasone, triamcinolone acetonide, prednisolone, hydrocortisone) were selected in this study. When steroids were treated to the mouse thymocytes in vitro cytolysis occurred with dose-dependent fashion and the activities were found to be paralle with the known local anti-inflammatory activities. In vivo thymus atrophogenic activities appeared by the treatment of topical and subcutaneous applications of the derivatives were also found to dose-dependent, but not coincided with the thymocyte cytolytic activities in vitro and local anti-inflammaatory activity in the case of triamcinolone acetonide. Triamicinolone acetonide induced potent thymocyte cytolysis in vitro, but showed less thymus atrophy.

Effect of Treatment of In Vitro Matured Pig Oocytes with Calcium Ionophore on Monospermic Penetration In Vitro

  • The present study examined whether treatment of in vitro matured pig oocytes with calcium ionophore (A23187) could prevent polyspermic penetration in vitro. When oocytes cultured for maturation for 33, 36 or 44 h were subsequently treated with $50{\mu}M$ A23187 in medium with fetal calf serum (FCS) for 1, 2 and 3 h and then cultured for 12 h without spermatozoa, virtually no activation occurred. In the absence of FCS, however, 31-42, 45-49 and 56-64% of oocytes were activated, respectively. When oocytes treated with $50 {\mu}M$ A23187 in medium with FCS for 3 h were inseminated in vitro, the penetration rates (14-57%) were lower (p<0.01) with a higher (p<0.01) incidence (35-67%) of monospermy compared with untreated oocytes (69-80% penetration and 15-17% monospermy). However, sperm penetration was completely blocked in all oocytes treated with A23187 in the absence of FCS. When oocytes matured for 33 h were treated with different concentrations of A23187 for 3 h and inseminated in vitro, the penetration rate did not change but there was an increased incidence (p<0.05) of monospermy at $10-20{\mu}M$ and $2.5-5{\mu}M$ A23187 in the presence and absence of FCS, respectively, compared with at $0{\mu}M$ A23187. With these lower concentrations of A23187, treatment of oocytes for at least 60 and 30 min in the presence and absence of FCS, respectively, was required to increase the incidence of monospermy without reducing penetration rate. These results indicate that a high concentration ($50{\mu}M$) of A23187 in medium without FCS, but not in medium with FCS, stimulated in vitro matured pig oocytes to induce parthenogenetic activation and a complete block to sperm penetration in vitro. However, treatment of oocytes with lower concentrations of A23187 ( $10-20{\mu}M$ and $2.5-5{\mu}M$) both in the presence and absence of FCS maintained sperm penetration in vitro and increased the incidence of monospermy.

Verification of Estrogenic Activities in Ethanol Extracts of Oriental Herbal Medicines using In vitro Detection System (In vitro 검출시스템을 이용한 한약재 추출물로부터의 에스트로겐 활성의 검증)

  • In order to evaluate the direct effect of estrogenic compounds in oriental herbal medicines, the estrogenic activity was measured using an in vitro detection system. For this system, human breast cancer cell line MCF7 was transfected using an estrogen responsive CAT reporter plasmid. Estrogenic activities of Platycodi radix, Astragali radix and Glycyrrhizae radix were evaluated using this system. Estrogenic activity of a 500 ㎍/ml ethanol extract of Platycodi radix was as same as that of a 10/sup -8/ M standard solution (17β-estradiol) and activity of a 50 ㎍/ml ethanol extract was between those of a 10/sup -8/ M and a 10/sup -9/ M standard solutions. In addition, estrogenic activity of a 50 ㎍/ml ethanol extract of Platycodi radix was as same as that of a 10/sup -10/ M standard solution. The same activity patterns were observed in the system which was treated by Astragali radix or Glycyrrhizae radix extracts. The most effective activity was observed in a system which was treated by Platycodi radix extract, but the least activity was observed by Glycyrrhizae radix extract. In this result, it was confirmed that Platycodi radix, Astragali radix and Glycyrrhizae radix extracts possess estrogenic compounds.

Effect of Culture Conditions and Freezing Methods on Developmental Competence of Hanwoo Embryos Cultured In Vitro

  • This study was carried out to examine on developmental competence of Hanwoo embryos cultured in vitro according to culture conditions and freezing methods. The in vitro developmental competence to blastocyst stage at Day 8 of culture in SOF was significantly (p<0.05) higher than that in CR1aa (30.3% vs. 18.4%). The in vitro developmental rate of morula and blastocysts cultured in group culture was significantly (p<0.05) higher than that in individual culture (41.4% and 36.0% vs. 21.1% and 10.5%, respectively). The cell number of Day 8 blastocysts in group culture was significantly (p<0.05) higher than that in the individual culture ($120.1{\pm}12.8\;vs.\;94.1{\pm}12.1$, respectively). The survival rates of frozen-thawed balstocysts that were exposed in 1.5 M ethylene glycol or 1.5 M ethylene glycol containing 0.1 M sucrose were 77.5% and 78.7%, respectively. The survival rates of blastocysts cultured for 48 h in slow freezing and vitrification was not significantly different (73.3 and 74.0%). In conclusion, in vitro developmental competence of bovine embryos was influenced on the culture medium (SOF) and culture method (Group culture). Survival rate of frozen-thawed of bovine embryos was not influenced on freezing solutions and freezing methods.

Effect of Pyroligneous Liquor on the Content and Activity of Endogenous Substances of Neofinetia falcata Cultured in vitro (목초액이 기내 배양한 풍란(Neofinetia faicata)의 내생물질 함량 및 활성에 미치는 영향)

  • This experiment was conducted to clarify the effect of pyroligneous liquor on endogenous substances that were GA (gibberelic acid)-like substances, starch, and protein of Neofinetia falcata cultured in vitro. When seedlings of Neofinetia falcata were treated with several concentrations (0, 0.5, 1.0, 2.0, 3.0 ml/L) of pyroligneous liquor, the growth of seedlings was enhanced in 1.0ml/L pyroligneous liquor added media. The activity of GA-like substances was lower in the leaf, but higher in the and root than that of the control. The content of starch in 1.0ml/L pyroligneous liquor was lower in the leaf, but higher in the root than that of the control. And the content of protein in 1.0ml/L pyroligneous liquor was also lower in the leaf, but higher in the root than that of the control.
