Plant Growth Promotion and Antagonistic Activities Against Anthracnose of Burkholderia sp. LPN-2 Strain

  • A rhizobacterium LPN-2, which showed strong antifungal activity and auxin producing ability, was isolated from a farmland in North Gyeongsang Province, South Korea. Based on analysis of the 16S rDNA sequence, strain LPN-2 was identified as a novel strain of Burkholderia and was designated as Burkholderia sp. LPN-2. In vitro experiments showed that the isolated stain LPN-2 significantly produced auxin within 48 hr incubation. In order to check for PGPR function we performed in vivo growth promoting test in different crops, including mung bean, pea and cabbage. Application of Burkholderia sp. LPN-2 showed dramatic growth promoting effect on all the tested plants. We also confirmed siderophore and cellulase productions by Burkholderia sp. LPN-2 using CAS blue agar and CMC plate test. Further treatment with LPN-2 and the crude culture broth was effective in suppressing anthracnose in vitro test and also reduced incidence and severity of anthracnose in apple and pepper. Taken together, we conclude that Burkholderia sp. LPN-2 might be used as organic fertilizer for effective crop production in organic farming.

A STUDY OF THE CYTOTOXICITY OF ROOT CANAL SEALER IN VITRO (생체외 실험을 이용한 근관충전용 Sealer의 세포독성에 관한 연구)

  • The purpose of this study was to evaluate the cytotoxicity of four root canal sealers(Tubliseal, AH26, Apatite Root Canal Sealer I, Apatite Root Canal Sealer II) in Vitro. The root canal sealers were mixed and filled in molds which were $14{\times}1.25mm$ in diameter, in height to use for cell counting and agar overlary method, and $7{\times}1.25mm$ for millipore filter method and set for 7 days to use for experiment. Silicone and copper plate were used for negative and positive control respectively. Using the culture of L929 fibroblast, total cell number and vital cell number were counted and the ratio of vital cell number to total cell number was calculated on 2 nd, 4 th, 6 th experimental day, and the change of cell membrane permeability was tested by agar overlay method, and the succinate dehydrogenase activity was tested by millipore filter method. The obtained results were as follows. 1. In ail experimental groups, the mitotic activity of fibroblast was reduced when compared with that of negative control group, so ail experimental groups showed cytotoxicity. Apatite Root Canal Sealer I group exhibited mild cytotoxicity, and Tubliseal, AH26, Apatite Root Cenal Sealer II groups exhibited severe cytotoxicity. 2. In the test of the change of cell membrane permeability by agar overlay method, all experimental groups showed cytotoxicity. AH26 group exhibited mild cytotoxicity, and Apatite Root Canal Sealer I group exhibited moderate cytotoxicity, and Tubliseal and Apatite Root Canal Sealer II group exhibited severe cytotoxicity. 3. In the test of SDH activity by millipore filter method, there was no cytotoxicity in Apatite Root Canal Sealer I and Apatite Root Canal Sealer II group, but Tubliseal and AH26 group showed mild cytotoxicity.

Inhibitory Effect of Salvia miltiorrhiza Extract on Growth of Some Cancer Cells (단삼(Salvia Miltiorrhiza) 추출물의 암세포 증식 억제 효과에 관한 연구)

  • This study was performed to evaluate the antitumor activities of water and ethanol (EtOH) extract of Salvia miltiorrhiza in vitro and in vivo. The proliferation of the human hepatoma (HepG2), rectum cancer (HRT-18) and colon cancer (HT-29) cells was inhibited by administration of extracts in a dose-dependent manner. Particularly, EtOH extract inhibited proliferation of the cells more effectively than water extract did. The morphology of cells induced by EtOH extract was characterized by reduction of cell size and deformatin. Oral administration of the EtOH extract (3 mg/head) to tumor-bearing mice inhibited the tumor (sarcoma-180) growth by 35% and prolonged their survival rate by 61%. The EtOH extract was shown to be nontoxic at 37.5% mg/head/day on the acute toxicity test. These studies suggest that the EtOH extract of Salvia miltiorrhiza may have antitumor activity in vitro and in vivo.

The Problem and Improvement Solution Regarding the Inquiry of Sample (검체 문의에 관한 문제점 및 개선방안)

  • Purpose: The work flow of international hospital's laboratory consists of rapid test and result report at the present day. However, the frequent inquiry of sample, which cause affairs to delay and efficiency to Lower, affects medical examination. In order to promote work's efficiency, we should improve the problem and make work smooth between a laboratory, outs and ward. Materials and Methods: This study runs as follows. First, Investigating test result, test schedule, test receipt, quick result, etc through the activity required from September to November 2007 about the inquiry of sample. After analysis of the problem in December, remaking the test schedule for improvement solution and reporting it to outs and ward. When the result is retest and dilution, we directly fill in a result space with the result situation of the patient to let them know beforehand. We also, prevent the omission of the result through checking the sample list and discriminate in vivo from in vitro by changing the laboratory's telephone number. We have improved the problem about the inquiry of sample through valuation and analysis since the improvement activity from January to March 2008. Result: The case about the frequent inquiry of sample has reduced by 57.8%. this improvement activity indicated that p-value<0.05 was statistically significant through paired t-test. This activity make study smooth and we rapidly report the result. Conclusion: By reducing the case regarding the inquiry of sample, work discontinuation, and concentration reduction, the work efficiency was increased.

In vitro assay for osteoinductive activity of different demineralized freeze-dried bone allograft

  • Purpose: Various bone graft materials have been used for periodontal tissue regeneration. Demineralized freeze-dried bone allograft (DFDBA) is a widely used bone substitute. The current widespread use of DFDBA is based on its potential osteoinductive ability. Due to the lack of verifiable data, the purpose of this study was to assess the osteoinductive activity of different DFDBAs in vitro. Methods: Sarcoma osteogenic (SaOS-2) cells (human osteoblast-like cells) were exposed to 8 mg/mL and 16 mg/mL concentrations of three commercial types of DFDBA: Osseo+, AlloOss, and Cenobone. The effect of these materials on cell proliferation was determined using the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay. The osteoinductive ability was evaluated using alizarin red staining, and the results were confirmed by evaluating osteogenic gene expression using reverse transcription polymerase chain reaction (RT-PCR). Results: In the SaOS-2 cells, an 8 mg/mL concentration of Osseo+ and Cenobone significantly increased cell proliferation in 48 hours after exposure (P<0.001); however, in these two bone materials, the proliferation of cells was significantly decreased after 48 hours of exposure with a 16 mg/mL concentration (P<0.001). The alizarin red staining results demonstrated that the 16 mg/mL concentration of all three tested DFDBA induced complete morphologic differentiation and mineralized nodule production of the SaOS-2 cells. The RT-PCR results revealed osteopontin gene expression at a 16 mg/mL concentration of all three test groups, but not at an 8 mg/mL concentration. Conclusions: These commercial types of DFDBA are capable of decreasing proliferation and increasing osteogenic differentiation of the SaOS-2 cell line and have osteoinductive activity in vitro.

Antioxidative Effect of Pimpinella brachycarpa Ethanol Extract. (참나물 에탄올 추출물의 항산화 효과)

  • This study was performed to investigate the antioxidant effect of 80% ethanol extracts from Pimpinella brachycarpa in vitro. The extraction yields of 80% ethanol extract was 9.23 g/100 g. The extract was further fractionated subsequently by n-hexane, chloroform, ethylacetate, n-butanol and water. Water fraction showed the highest extraction yield among fractions. Antioxidative activities of different fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical generation, Rancimat test, thiobarbituric acid (TBA) value, nitrite scavenging activity, inhibition of lipid peroxidation and peroxide value in linoleic acid in comparison with the commercial antioxidant butylated hydroxytoluene (BHT). Antioxidant activities of n-hexane fraction of Pimpinella brachycarpa ethanol extract were the highest among fractions. Furthermore, the antioxidative capacity of the n-hexane fraction was similar to that of BHT.

Isolation and Antifungal Activity of the Chitinase Producing Bacterium Serratia sp. 3095 as Antagonistic Bacterium against Fusarium sp. (Chitinase를 생산하는 길항미생물 Serratia sp. 3095의 선발과 Fusarium 속에 대한 항진균성)

  • For the selection of an effective antagonistic biocontrol agent, we have isolated an antagonistic bacterium which produced extracellular chitinase, from a local soil of Kyongju, Korea. The selected strain was identified as Serratia proteamaculans 3095. The chitinase produced from Serratia sp. 3095 showed antifungal activity which can attack the hypha surface of Fusarium oxysporum and F. solani. The carbon and nitrogen sources for chitinase production were 0.15% colloidal chitin and 0.1% ammonium sulfate, respectively. Glucose in the chitinase production medium might inhibit the production of chitinase by feed back repression. The antagonistic Serratia sp. 3095 also showed a powerful biocontrol activity against F. oxysporum through in vitro test and in vivo pot test.

Screening of Korean Marine Plants Extracts for Inhibitory Activity on Protein Tyrosine Phosphatase 1B

  • Crude extracts of 69 marine organisms (27 salt marsh plants and 42 seaweeds) were screened for the inhibitory activity against the protein tyrosine phosphatase 1B (PTP1) in vitro. The most active extracts were methanol extracts from Derbesia marina (80.6% in inhibitory activity) and Symphycladia latiscula (85.6%) at the concentration of $15{\mu}g/mL$. Methanol extracts of Codium adhaerens and Hisikia fuziformis were moderately inhibitory with 71.2 and 69.1% inhibition, respectively. It was peculiar that only the extracts from seaweeds show inhibitory activity where those from salt marsh plants do not show any significant effect.

  • The water extracts of 83 oriental herbal medicines (Hanbang) which have been clinically used to treat bacterial infections in Korea were screened for in vitro antibacterial activity by the paper disc assay method. Two Gram positive bacteria, Staphylococcus aureus SG 511, Bacillus subtilis ATCC 6633, and two Gram negative bacteria, Escherichia coli 055, Pseudomonas aeruginosa 9027 were used as test organisms. Among the extracts tested, MeOH extract of Baenongtang showed remarkably potent antibacterial activity. Activity-guided chromatographic fractionations of the $CH_2Cl_2$ extract of Baenongtang afforded seven antibacterial compounds.

  • For the synthesis of new antifungal agents, We have synthesized four new ketoconazole derivatives were synthesized by the reaction of cis-[2-(2,4-dichlorophenyl)-2-(1H-imidazol-1-yl)-1,3-dioxolan-4-yl]methane sulfonate with isolated fig compounds. These compounds were showed strong antifungal activity against C, albicans ATCC 10231. C, utilis. S, cerevisiae ATCC 9763. A and niger ATCC 9029. Among them, sample No.(13) showed potent inhibition activity. Generally, other samples showed biological activity in vitro test. The above results showed the possibility of the development of new antifungal agents.
