In situ Hybricization of White Spot Disease Virus Experimentally Infected Penaeid Shrimp

  • Abstract White spot disease (WSD), resulting in more than 90% mortality of aquacultured penaeid shrimp, has been reported off the southern and western coasts of Korea since 1993. The pafuogen of WSD has been identified as being a virion wifu an envelope around a central nucleocapsid, and with an average size of 167 nm in diameter and 375 nm in length. In the present study, an in situ hybridization technique was developed as a rapid. sensitive, and specific diagnostic assay for the WSD viros infection in shrimp. Furthermore. the pathological changes ofWSD, in shrimp experimentally infected with WSD viroses. were investigated. Using a biotinylated 643 bp probe obtained from a peR using primers specific to the rod-shaped virus of Penaeus japonicus (RV-PJ), positive signals were detected in both naturally and experimentally infected shrimps. The in situ hybridization revealed positive reactions in the nuclei of the stromal matrix cells in the lymphoid organ, epithelia of the gills, foregut. epidermis, and hematopoietic cells of the interstitial tissues, suggesting the presence of WSD virus. Tills result indicates that the in situ hybridization method can be useful for a rapid and sensitive detection of WSD viruses in shrimp.shrimp.

Evaluation of Applicability of CMD-SOIL using the Deep Mixing Method in Ulsan Area for the Construction of Coastal Structure Foundation (해안구조물 기초의 건설을 위해 울산지역에서의 심층혼합공법을 사용한 CMD-SOIL의 적용성 평가)

  • As global trade and maritime port environments change, the need to respond to larger and faster ships is increasing. Accordingly, new ports are being built around metropolitan cities such as Busan and Ulsan. In general, a compaction method using sand or gravel is applied to the construction of a new port. However, due to the lack of sand or gravel and the difficulty in securing economic feasibility due to the increase in unit price, the deep mixing method has recently been used. Therefore, in this study, CMD-SOIL using circulating resources was applied to the Ulsan area, and the applicability was determined by analyzing the laboratory mixing test and boring results at in-situ. As a result of the test, it was analyzed that it showed more than the design mixing strength, and it was possible to secure the similar performance as blast furnace slag cement. In addition, it was analyzed that the design standard strength can be sufficiently secured as a result of in-situ boring. Therefore, considering the field applicability in the Ulsan, it is judged that the use of CMD-SOIL is possible.

In situ Recovery of hGM-CSF in Transgenic Rice Cell Suspension Cultures (형질전환 벼 현탁세포 배양에서 hGM-CSF의 in situ Recovery 연구)

  • Production of foreign proteins by transgenic plant cell cultures has several advantages such as post-translational modification, low risk of product contamination and low-cost production and purification. However, target proteins are degraded by extracellular proteases existing in the media. A solution to this problem is the use of perfusion culture and ion exchange chromatography for the application of integrated bioprocess using in situ recovery. With this method, production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in this study. First, optimization of cell concentration during the induction phase for the production of hGM-CSF was examined. As cell concentration increased, the level of hGM-CSF was decreased due to the presence of extracellular proteases. Induction using sugarfree media produced 33% more hGM-CSF. The effects of pH on the binding of hGM-CSF to cationic and anionic exchange resins were also investigated. In terms of stability, optimal pH was found to be 5~7. In the case of using buffer exchange when CM-Sepharose was used as a cationic exchange resin, optimal pH for binding was 4.8 and adsorption yield was 77%. When DEAE-Sepharose was used as an anionic exchange resin, it was 5.5 (74%). Without buffer exchange, optimal pH was 4.6 (84%). From these results, an integrated bioprocess using in situ recovery with simultaneous production and separation of foreign protein in transgenic plant cell suspension cultures was found to be feasible.

  • In this study, the in-situ stress, strength and stress-strain characteristics with shear parameters (UU, CU, ${\bar{CU}}$) are analytically evaluated and the stability analyses are carried out under loading/unloading conditions. The in-situ stress and the stress-strain behaviour may become different according to input shear parameters in finite element analyses with construction step, Especially, if the internal friction angle in Mohr-Coulomb model is set to zero, the in-situ stress and the stress-strain behaviour might not be properly predicted. The results from CU parameter of total stress analysis have no significant difference with the results from CU of effective stress analysis. Therefore, in the numerical analysis for soft ground, CU parameters can be applied to predict in-situ stress and stress-strain behaviors. In addition, the calculation method was proposed to determine the shear parameter of Mohr-Coulomb model, which is corresponding to the shear strength equivalent to that of in-situ soil.

  • In-situ leaching could be one of the promising mining methods to extract the minerals from deep fractured rock mass. Constrained by the low permeability at depth, however, the performance does not meet the expectation. In fact, the rock mass permeability mainly depends on the pre-existing natural fractures and therefore play a crucial role in in-situ leaching performance. More importantly, fractures have various characteristics, such as aperture, persistence, and density, which have diverse contributions to the promising method. Hence, it is necessary to study the variation of fluid rate versus fracture parameters to enhance in-situ leaching performance. Firstly, the subsurface fractures from the depth of 1500m to 2500m were mapped using the discrete fracture network (DFN) in this paper, and then the numerical model was calibrated at a particular case. On this basis, the fluid flow through fractured rock mass with various fracture characteristics was analyzed. The simulation results showed that with the increase of Fisher' K value, which determine the fracture orientation, the flow rate firstly decreased and then increased. Subsequently, as another critical factor affecting the fluid flow in natural fractures, the fracture transmissivity has a direct relationship with the flow rate. Sensitive study shows that natural fracture characteristics play a critical role in in-situ leaching performance.

Preparation of GdBCO Thin Film by Ex-situ Process using Nitrate Precursors (질산염 전구체 원료로 Ex-situ 공정에 의한 GdBCO 박막 제조)

  • Many research groups have been manufacturing coated conductor by various processes such as PLD, MOD, and MOCVD, but the methods with production cost suitable for wide and massive application of coated conductor did not develop yet. Spray pyrolysis method adopting ultrasonic atomization was tried as one of the possible option. GdBCO precursor films have been deposited on IBAD substrate by spray pyrolysis method at low temperature and converted to GdBCO by post heat treatment. Ultrasonic atomization was used to generate fine droplets from precursor solution of Gd, Ba, and Cu nitrate dissolved in water. Primary GdBCO films were deposited at $500^{\circ}C$ and oxygen partial pressure of 1 torr. After that, the films were converted at various temperatures and low oxygen partial pressures. C-Axis oriented films were obtained IBAD substrates at conversion temperature of around $870^{\circ}C$ and oxygen partial pressures of 500 mtorr ~ 1 torr in a vacuum. Thick c-axis epitaxial film with the thickness of 0.4 ~ 0.5 ${\mu}m$ was obtained on IBAD substrate. C-axis epitaxial GdBCO films were successfully prepared by ex-situ methods using nitrate precursors on IBAD metal substrate. Converted GdBCO films have very dense microstructures with good grain connectivity. EDS composition analysis of the film showed a number of Cu-rich phase in surface. The precursor solution having high copper concent with the composition of Gd : Ba : Cu = 1 : 2 : 4 showed the better grain connectivity and electrical conductivity.

Detection of bovine viral diarrhea virus by In situ hybridization (In situ hybridization에 의한 소 바이러스성 설사증 바이러스의 검출)

  • Detection and distribution of bovine viral diarrhea virus(BVDV) was studied in formalin-fixed, paraffin-embedded tissues from two naturally infected cattle by in situ hybridization with a non-radioactive biotinylated probe. A 600 base pair cDNA probe from BVDV B-25 strain was used for probe. The whole procedure of ISH to diagnose was carried out within 1~2 hours in $Microprobe^{TM}$ capillary action system. The biotin-labelled probe was demonstrated after hybridization under standard conditions by the application of streptoavidin and biotinylated alkaline phosphatase. Alkaline phosphatase was visualized using a fast red TR/naphthol phosphatase and the sections were counterstained with hematoxylin. We have obtained the result of positive reactions in digestive tract(sm1.all intestine and colon) and epidermis of tongue in the state of the intact tissues. The result suggested that in situ hybridization method can be considered as a useful diagnostic technique for detection of specific nucleic acid sequences of BVDV.

Synthesis of CO2 Adsorbent with Various Aminosilanes and its CO2 Adsorption Behavior (다양한 아미노실란을 이용한 이산화탄소 흡착제 합성 및 흡착 특성)

  • The carbon dioxide adsorption behavior of silica with a large specific surface area and pore volume functionalized with aminosilane compounds via in-situ polymerization and functionalization method were investigated. The organosilanes include amino functional group capable of adsorbing carbon dioxide. Elemental analyzer, in situ FT-IR and thermogravimetric analyzer were used to characterize the sorbents and to determine their $CO_2$ adsorption behavior. Comparison of different aminosilane loading in the support revealed that polyaminosilane functionalization of 70% of the pore volume in the support was better in terms of the adsorption capacity and amine efficiency than that of 100% of the pore volume of the support. Furthermore, the sorbents showed a higher adsorption capacity at an adsorption temperature of $75^{\circ}C$ than at $30^{\circ}C$ due to the thermal expansion of synthesized polyaminosilanes inside the pore of silica. The N-[3-(trimethoxysilyl)propyl]ethylenediamine (2NS) sorbent with 70% of the pore volume functionalized showed the highest adsorption capacity of 9.2 wt% at $75^{\circ}C$.

  • Background : A presumptive histopathologic diagnosis of tuberculosis is commonly based on the finding of acid-fast bacilli upon microscopic examination of a diagnostic specimens. Although this traditional histochemical staining method is satisfactory, it is time-consuming and not species-specific. For more specific assessment, in situ hybridization assay with oligonucleotide probes is introduced. Methods : The human surgical specimens were obtained from tuberculosis patients, and experimental specimens were made by injecting cultured M. tuberculosis organisms into fresh rat liver. Oligonucleotide probes complementary to ribosomal RNA portion were synthesized and labeled with multiple biotin molecules. For a rapid detection, all procedures were carried out using manual capillary action technology on the Microprobe staining system. Results : The in situ hybridization assay produced a positive reaction in experimental specimens (80-90% sensitivity) after pepsin-HCl pre-treatment for a good permeabilization of probes, but reliable result was not obtained from human surgical specimens. Conclusion : It is, therefore, suggested that biotin-labeled oligonucleotide probes have considerable potential for identification and in situ detection of M. tuberculosis but, there are some barriers to overcome for the diagnostic use of this method.

Methods of in situ PCR to Retain the Amplification Products Inside the Cells (원위치 중합효소 연쇄반응에서 증폭산물의 세포내 보존을 위한 방법들)

  • Highly effective polymerase chain reaction (PCR) often brings about false positivity caused by contamination of the sample with target nucleic acids. To solve this problem, in situ PCR (ISPCR) has been developed and applied onto various tissue sections and suspension cultures. With combination of PCR and in situ hybridization, this method amplifies the nucleic acid targets in situ and detect the amplified products inside the cells over the background of various cell types. In order to amplify the nucleic acid targets inside the cells, permeabilisation of a sample is required for the entry of amplification reactants into a cell. Treatments of a sample for the purpose allow not only the entry of reactants into the cell but also the exit of amplification products out of the cell. As a means to reduce the leakage of the amplification products, two methods were applied to suspension cultures of HIV-infected Molt/LAV and U 1.1 cells, in which modified, tailed primers produced long linear amplificants whereas biotinylated dUTP instead of dTTP did bulky products.
