Characterization of Azospirillum spp. Isolated from Korean Paddy Roots (우리나라 수도근권에서 분리된 Azospirillum spp.의 특성)

  • Nitrogen fixing activity associated with 40 varieties of rice was assayed at heading stage by an in situ acetylene reduction method. The in situ acetylene reduction activity and population of nitrogen fixing bacteria obtained on nitrogen-free malate medium for Azospirillum spp. enrichment showed positive correlation. Six Azospirillum spp. with high nitrogenase activity were isolated from the rice roots, from which five spp. were identified as A. lipoferum and one was A. brasilense. The physiological characteristics of the six Azospirillum isolates, that is, carbon source utilization, biotin requirement, antibiotic resistance, indole acetic acid excretion, plasmid profile and protein patterns were compared.

  • At construction sites, due to the reason of inconvenience and difficulties of producing and using curing equipment and when it comes to measuring compression strength of the actual structure, strength of structure concrete according to general standards which are suggested in concrete standard specification are assessed. However, this method does not consider various variables of the sites such as kinds concretes and sizes of frame works so that it is not easy to measure proper curing period and strength. Thus, this study reviews description of strength development according to In-situ temperature system and analyzes and compares properties of strength development of the existing curing methods such as sealing curing so that it provides basic materials for period of removal of molds.

  • Self-Boring Pressuremeter Test(SBPT) is known to be the most effective in-situ test method which can reliably determine consolidation characteristics as well as deformation modules and untrained shear strength. In order to derive the coefficient of consolidation using SBPT results it is necessary to obtain the dissipation behavior from the pore pressure change with time during constant radial strain(generally 10%) and to derive the reliable time factor(Τ) from the analytical method which considers the real in-situ conditions. As previous studies on time factor are based on the assumptions of plane strain condition that the membrane of SBP is infinite, of untrained condition during the expansion of the probe and of elastic soil behavior during consolidation, these analyses can't consider the real boundary conditions and the real soil behaviour. In this study, consolidation analysis similar to real in-situ conditions including test procedure is conducted using finite element program which employs MCC model and Biot theory. Time factor considering the effects of finite membrane length, the total pressure change during consolidation and partial drainage is proposed and compared with previous results.

Electrochemical Performance of Activated Carbons/Mn3O4-Carbon Blacks for Supercapacitor Electrodes

  • In this work, manganese dioxide ($Mn_3O_4$)/carbon black (CB) composites (Mn-CBs) were prepared by an in situ coating method as electrical fillers and the effect of the Mn-CBs on the electrical performance of activated carbon (AC)-based electrodes was investigated. Structural features of Mn-CBs produced via in situ coating using a $KMnO_4$ solution were confirmed by XRD and TEM images. The electrical performances, including cv curves, charge-discharge behaviors, and specific capacitance of the ACs/Mn-CBs, were determined by cyclic voltammograms. It was found that the composites of $Mn_3O_4$ and CBs were successfully formed by in situ coating method. ACs/Mn-CBs showed higher electrical performance than that of AC electrodes fabricated with conventional CBs due to the pesudocapacitance reaction of manganese oxides in the aqueous electrolyte. Consequently, it is anticipated that the incorporation of $Mn_3O_4$ into CBs could facilitate the utilization of CBs as electrical filler, leading to enhanced electrochemical performance of AC electrodes for supercapacitors.

Application of Handheld Raman Spectroscopy for Pigment Identification of a Hanging Painting at Janggoksa Temple(Maitreya Buddha) (장곡사 미륵불 괘불탱의 채색 재료 분석을 위한 휴대용 라만 분광기의 적용성 연구)

  • The purpose of this study is to apply the handheld Raman spectrometer to identify the coloring materials used in a large Buddhist painting (of Maitreya Buddha) at Janggoksa Temple through cross-validation with HH-XRF. An in situ investigation was performed together with use of a digital microscope and HH-XRF analysis to verify the properties of pigments used in the gwaebul ("large Buddhist painting") via a non-destructive method. However, the identification of coloring materials composed of light elements and mixed or overlaid pigments is difficult using only non-destructive analysis data. Unlike in situ investigation, laboratory analysis often required samples yet the sampling is restricted to a small quantity due to the cultural heritage characteristic. Thus, it is necessary to develop a non-destructive in situ method to supplement the HH-XRF data. The large Buddhist painting at Janggoksa Temple was painted mainly using white, red, yellow, green, and blue colors. The Raman spectroscopy provides molecular information, while XRF spectroscopy provides information about elemental composition of the pigments. Analysis results identified various coloring materials: inorganic pigment, such as lead white, minium, cinnabar, and orpiment, as well as organic pigment such as gamboge and indigo. Therefore, it is possible to obtain more information for the identification of pigments; organic pigment and mixed or overlaid pigments, while at the same time minimizing the collection sample and simplifying the analysis procedure compared to previously used methods. The results of this study will be used as basic data for the analysis of painting cultural heritage through a non-destructive in situ method in the future.

Solvothermal Synthesis and Gas Permeation Properties of Nanoporous HKUST-1 Membranes (용매열합성법에 의한 나노기공 HKUST-1 막의 제조 및 기체투과 특성)

  • In this study, nanoporous HKUST-1 membranes were synthesized by solvothermal method. It is very difficult to prepare uniform and crack-free HKUST-1 layer on macroporous alumina support by in-situ solvothermal method. In this study, continuous and crack-free HKUST-1 membranes could be obtained by spraying solvothermal precursor solution on the heated alumina support, followed by solvothermal synthesis. HKUST-1 membranes were characterized by XRD, FE-SEM and single gas permeation experiments.

  • A coil-type capacitive sensor on which the deterioration of the car engine oil can be in situ measured, has been developed. The sensor was designed to get over 10 pF at the limited space on the drain hole of the oil pan. The design factors for stable capacitance measurement such as coil diameter and winding condition, materials and configuration of the coil former, and shielding method, etc., were known by both computer simulation and experimental investigations. The dielectric properties measured by several sensors for an used sample oil were consisted within 0.25%. The sensor installed on the car having severe vibration, temperature and humidity fluctuation, even electromagnetic noise, has shown very distinguishable results.
