Genetically Engineered Mouse Models for Drug Development and Preclinical Trials

  • Drug development and preclinical trials are challenging processes and more than 80% to 90% of drug candidates fail to gain approval from the United States Food and Drug Administration. Predictive and efficient tools are required to discover high quality targets and increase the probability of success in the process of new drug development. One such solution to the challenges faced in the development of new drugs and combination therapies is the use of low-cost and experimentally manageable in vivo animal models. Since the 1980's, scientists have been able to genetically modify the mouse genome by removing or replacing a specific gene, which has improved the identification and validation of target genes of interest. Now genetically engineered mouse models (GEMMs) are widely used and have proved to be a powerful tool in drug discovery processes. This review particularly covers recent fascinating technologies for drug discovery and preclinical trials, targeted transgenesis and RNAi mouse, including application and combination of inducible system. Improvements in technologies and the development of new GEMMs are expected to guide future applications of these models to drug discovery and preclinical trials.

EFFECT OF GINGER RHIZOME EXTRACT ON TENDERNESS AND SHELF LIFE OF PRECOOKED LEAN BEEF

  • The inclusion of crude ginger rhizome (zingiber officinale) extract at 0.5 to 1.0% (v/w) in the marination of marginally acceptable lean beef improved meat tenderness by 20-30% in the absence of 2% salt and by 35-45% in the presence of 2% salt. Ginger extract also retarded the development of rancidity and increased shelf-life of precooked lean beef two-fold in saran-wrap (no vacuum) storage at $4^{\circ}C$. Ginger rhizome improves the palatability and acceptability of lean beef from carcasses of marginal quality. It is particularly beneficial for the preparation of pre-cooked ready-to-eat beef products that are not vacuum-packaged.

  • Blood lipid levels in rats with high fat-fed hyperlipemia were determined after intraperitoneal administration of a methanolic extract of Prunus davidiana Fr. stems. Administration of the methanolic extract for 6 days produced a significant decrease of blood triglyceride and total cholesterol, and the atherogenic index was also improved. In addition to the hypolipemic effect, the methanolic extract was also shown to be effective in reducing an elevated level of glucose in rats with hyperlipemia resulting from high-fat feeding. On the other hand, blood triglyceride and total cholesterol in rats fed with stock diet were not affected by administration of the methanolic extract even if there was a tendency to decrease. No significant change was also found in the level of glucose. Thus, it is suggested that this methanolic extract probably may increase the metabolic utilization only when fed with excess fat.

  • The trend towards ageing populations has been observed over many years in Europe and the US but has accelerated significantly in developed countries in Asia including Japan and South Korea. In the latter country the elderly population (65+) has increased 5-fold between 1960 and 2000 and this group will comprise 40% of the population by 2050. This creates a new socio-economic group with specific demands and considerable spending power. As ageing occurs a range of changes occur in the body that can be moderated by adjustments in nutrition. A significant body of evidence points to changes in the balance of glutathione synthesis and utilisation as people age. Glutathione is tile most important natural anti-oxidant of the body and the amounts present can become limited by available cysteine in the diet. A cysteine-enriched peptide product, Cysteine Peption$^{TM}$ has been developed by DMV International for dietary supplement and food applications. A qualitative consumer trial has indicated benefits including improved sleep and more energy. Animal and clinical trial will be described that provide indications on bioavailability and possible mechanisms of action of Cysteine Peption$^{TM}$ with particular focus on the ageing population.

  • Hypoglycemic efficacy of natural functional mixture(FM) and level of the diabete related hormones in streptozotocin(STZ)-induced diabetic rats were investigated. Male Sprague-Dawley rats were divided into three groups (normal, diabetic fed diets with/without FM). Supplement of FM did not affect the body weight and feed intake of STZ-induced diabetic rats. The increase in the weight of liver of STZ-induced diabetic rats was weakened by supplement of FM, whereas the weight of kidney and heart was not affected. Blood glucose level was slightly, and glucose tolerance of post-feeding was significantly improved by functional mixture. The mixture significantly reduced the elevated HbA1C level of diabetic rats by 15%, and it increased the level of insulin and C-peptide in blood and decreased glucagon level. Therefore, we conclude that FM in this study has a potency of prevention and treatment of diabetes mellitus.

  • This study aimed at identifying and analyzing consumption of leisure services in silver industry and factors that had effects on them. This focused on the consumption of leisure services and its influence on the variables related to change of elderly's attitude, because socio-economic environment structure is improved and average life span is prolonged. The main findings of the research were as follows: 1. Totally, the percentage of leisure services consumption not so high and kinds of leisure were not various. Also, the reason why they use leisure was just for self-development and their indifference to leisure made them not to use services. 2. There were significant differences in individual characteristics and family-environmental conditions relating the consumption of leisure services in the silver industry. 3. Among variables which influence on the consumption of leisure of the elderly, job was the most significant factors. Also average monthly income, the changable inclination in food, clothing and shelter, social support of the family, status of residing, status of health and source of income were influential variables.

  • With the increasing need for environmental protection, it is particularly important to improve the energy saving and reliability of refrigerators. Generally, the cold air flowing into the freezer compartment transits to the bottom of the refrigerating compartment, which can lead to uneven temperature distribution. This paper proposes two design solutions for improving the temperature distribution problem. Of these, the optimal refrigeration design was selected and tested using Computational Fluid Dynamics (CFD) modeling and simulation. The results showed improved uniformity of the temperature distribution inside the refrigerator, thus benefitting food storage while reducing energy consumption.

  • The effects of mixtures of each of rice hull charcoal and wood charcoal with pyroligneous acid on the cultivation and antioxidant activities of rice were investigated. Results of the total phenolic compounds, DPPH radical electron-donating ability and xanthine oxidase stress activity analyses revealed that the environment-friendly materials increased the total antioxidant capacity in rice. In the case of palatability of rice, total amylose blue value and starch-I2color reaction analyses, data showed that environment-friendly materials generally decreased the amylose content of rice. Also, the environment-friendly materials improved the rice endosperm quality.

Natural Dyeing of Silk Fabric using Purple-Fleshed Sweet Potato (자색 고구마를 이용한 견직물의 염색)

  • The natural dyeing of silk fabric with Purple-Fleshed Sweet Potato (PSP) was investigated. The colorant was extracted with distilled water, and the color difference (${\Delta}E^*$) was increased with increasing the amount of PSP in extraction. The proper temperature and time for the extracting of colorant with PSP were $60^{\circ}C$ and 60 minutes. The optimum temperature, time and pH for the dyeing of silk with extracted PSP were $60^{\circ}C$, 60 minutes and pH 4 respectively. In various mordanted methods, the color difference values of post-mordanted silk fabric were higher than those of pre- and simultaneous-mordanted method. And the wide range of colors( GY, Y, YR, R, RP) were obtained according to various mordants, mordanting methods and mordant concentrations. Light colorfastness of the mordanted silk fabric was improved. Laundering colorfastness, dry cleaning colorfastness and perspiration colorfastness were shown to be good.

  • Using genetic engineering, the Vitreoscilla (bacterial) hemoglobin gene (vgb) was integrated stably into the chromosomes of and Burkholderia cepacia. Similar to previous results, the wild type VHb improved growth for Burkholderia cepacia and degradation of benzoic acid under both normal and low aeration conditions. The stable expression of VHb enhanced these parameters. The results demonstrate the possibility that the positive effects provided by VHb may be augmented by protein engineering.
