파밤나방(Spodoptera exigua)에 대한 곤충병원세균류 배양액의 곤충면역억제활성 및 비티 생물농약과 혼합효과 (Immunosuppressive Activity of Cultured Broth of Entompathogenic Bacteria on the Beet Armyworm, Spodoptera exigua, and Their Mixture Effects with Bt Biopesticide on Insecticidal Pathogencity)

  • 곤충병원선충으로부터 분리된 곤충병원세균들(Xenorhabdus nematophila, X. sp. and Photorhabdus temperata subsp. temperata)은 곤충혈강에서 높은 살충효과를 보유하고 있다. 이들 세균들은 또한 phospholipase $A_2$ 효소 작용을 억제하여 면역중개반응을 무력화하여 곤충기주의 면역억제를 유도하게 한다. 본 연구는 이들 세균이 성장된 배양액이 이러한 면역억제 활성을 보유하는 지를 분석하였다. 이를 위해 $0.2\;{\mu}m$ 공극 크기의 여과막을 이용하여 세균배양액으로부터 세균을 제거시켰다. 이렇게 멸균된 세균배양액은 배양액 농도에 따라 뚜렷하게 파밤나방(Spodoptera exigua) 5령충의 혈구세포 활착행동을 억제시켰다. 또한 이러한 억제효과는 세균별로 차등을 보였으며, 이 가운데 X. nematophila의 배양이 가장 높았다. 이 세균의 면역억제 활성 성질을 Bacillus thuringiensis (Bt)의 감염력 제고에 적용하였다. 세 세균 모두의 배양액(세균 포함)들은 낮은 농도의 Bt와 혼합하여 섭식 처리할 경우 어린 파밤나방 유충에 대해서 뚜렷하게 Bt 감염력을 제고시켰다. 다시 이 세균을 제거한 멸균된 배양액을 낮은 Bt농도 처리와 혼합하여 섭식 처리하였다. 이때 X. nematophila의 배양액만이 Bt의 감염력을 제고시켰다. 본 연구는 이들 세 곤충병원세균의 배양액에 곤충의 면역억제물질이 포함되었음을 보였으며, 이들 물질을 Bt와 혼용하였을 때 Bt의 살충력을 제고시킬 수 있음을 나타냈다.

Immunosuppressive Effects of Bryoria sp. (Lichen-Forming Fungus) Extracts via Inhibition of CD8+ T-Cell Proliferation and IL-2 Production in CD4+ T Cells

  • Lichen-forming fungi are known to have various biological activities, such as antioxidant, antimicrobial, antitumor, antiviral, anti-inflammation, and anti proliferative effects. However, the immunosuppressive effects of Bryoria sp. extract (BSE) have not previously been investigated. In this study, the inhibitory activity of BSE on the proliferation of $CD8^+$ T cells and the mixed lymphocytes reaction (MLR) was evaluated in vitro. BSE was non-toxic in spleen cells and suppressed the growth of splenocytes induced by anti-CD3. The suppressed cell population in spleen cells consisted of $CD8^+$ T cells and their proliferation was inhibited by the treatment with BSE. This extract significantly suppressed the IL-2 associated with T cell growth and $IFN-{\gamma}$ as the $CD8^+$ T cell marker. Furthermore, BSE reduced the expression of the IL-2 receptor alpha chain ($IL-2R{\alpha}$) on $CD8^+$ T cells and CD86 on dendritic cells by acting as antigen-presenting cells. Finally, the MLR produced by the co-culture of C57BL/6 and MMC-treated BALB/c was suppressed by BSE. IL-2, $IFN-{\gamma}$, and CD69 on $CD8^+$ T cells in MLR condition were inhibited by BSE. These results indicate that BSE inhibits the MLR via the suppression of $IL-2R{\alpha}$ expression in $CD8^+$ T cells. BSE has the potential to be developed as an anti-immunosuppression agent for organ transplants.

개심술 환자에서의 면역기능의 변화;T lymphocyte subset의 변화에 대한 고찰 (Changes in Lymphocyte Subsets following Open-Heart Surgery ; A Study for Changes in Lymphocyte Subsets)

  • Cell mediated immunity is depressed following surgical procedure and the degree of immunosuppression is directly related to the magintude of the procedure, blood transfusion, and length of operation. So we would expect cardiac operations to be highly immunosuppressive, although little is konwn about their immunosuppressive effect. The nearly complete consumption of complement factors and decreased levels of IgM and IgG resulting in an impaired opsonizing capacity. Additionally, peripheral blood mononuclear cell counts including T-and B-lymphocytes and T-cell subsets are reduced. Depression of cell-mediated immunity following open-heart surgery is potentially detrimental because it could increase the susceptability of patients to viral and bacterial infection. We reviewed 20 patients after cardiac operation to search for changes in peripheral blood lymphocyte subsets. Lymphocyte subsets were measured by flow cytometer and the preoperative values of lymphocyte subsets were compared with those from the first, fourth, and seventh days after operation. After cardiac operation, total mumbers of T lymphocyte was severely depressed on the first postoperative day and returned to the preoperative level by the seventh day after operation. CD3, CD4, and CD8 lymphocytes were decreased on the first postoperative day and returned to the preoperative level by the seventh day also. There was four cases of wound infection and these patients had increased CD4 lympocyte and more decreased CD19 lymphocyte compared with the non-infected group. It is concluded from these data that cell-mediated immunity is significantly depressed for at least one week following open-heart surgery and this result was closely related to the postoperative infection.

Immunosuppression-enhancing effect of the administration of allogeneic canine adipose-derived mesenchymal stem cells (cA-MSCs) compared with autologous cA-MSCs in vitro

  • Background: Recently, mesenchymal stem cells therapy has been performed in dogs, although the outcome is not always favorable. Objectives: To investigate the therapeutic efficacy of mesenchymal stem cells (MSCs) using dog leukocyte antigen (DLA) matching between the donor and recipient in vitro. Methods: Canine adipose-derived MSCs (cA-MSCs) isolated from the subcutaneous tissue of Dog 1 underwent characterization. For major DLA genotyping (DQA1, DQB1, and DRB1), peripheral blood mononuclear cells (PBMCs) from two dogs (Dogs 1 and 2) were analyzed by direct sequencing of polymerase chain reaction (PCR) products. The cA-MSCs were co-cultured at a 1:10 ratio with activated PBMCs (DLA matching or mismatching) for 3 days and analyzed for immunosuppressive (IDO, PTGS2, and PTGES), inflammatory (IL6 and IL10), and apoptotic genes (CASP8, BAX, TP53, and BCL2) by quantitative real-time reverse transcriptase-PCR. Results: cA-MSCs were expressed cell surface markers such as CD90+/44+/29+/45- and differentiated into osteocytes, chondrocytes, and adipocytes in vitro. According to the Immuno Polymorphism Database, DLA genotyping comparisons of Dogs 1 and 2 revealed complete differences in genes DQA1, DQB1, and DRB1. In the co-culturing of cA-MSCs and PBMCs, DLA mismatch between the two cell types induced a significant increase in the expression of immunosuppressive (IDO/PTGS2) and apoptotic (CASP8/BAX) genes. Conclusions: The administration of cA-MSCs matching the recipient DLA type can alleviate the need to regulate excessive immunosuppressive responses associated with genes, such as IDO and PTGES. Furthermore, easy and reliable DLA genotyping technology is required because of the high degree of genetic polymorphisms of DQA1, DQB1, and DRB1 and the low readability of DLA 88.

Immunosuppressive Effect of Prodigiosin on Murine Splenocyte and Macrophages

  • Prodigiosin was isolated from marine bacteria Hahella chejuensis which has been recently discovered from Marado, Cheju Island, Republic of Korea. Immunosuppressive properties have been reported for prodigiosin members such as undecylprodigiosin, metacycloprodigiosin, prodigiosin, and its synthetic analogue PNU156804 (PNU). However, the effect of this agent on the function of macrophage and splenocyte has not been characterized in detail. In the present study, we examined the effects of prodigiosin for its ability to alter the function of murine macrophage and NK cell, and the proliferation of splenocytes. When thioglycollate-elicited macrophages pre-exposed to prodigiosin (1-50 ng/ml) were stimulated with LPS/IFN-$\gamma$, pretreatment with prodigiosin resulted in the inhibition of tumoricidal activity of macrophage in a concentration-dependent manner. Tumoricidal activity of NK cell was also inhibited by prodigiosin. Moreover, we found that prodigiosin was able to cause a dose-dependent inhibition of murine lymphocyte responsiveness to Con A and LPS although T-mitogenic response was the more sensitive one. Taken together, the present results point out that prodigiosin has a suppressive effect on the mitogen-induced proliferation of murine lymphocytes and the function of macrophage and NK cell.

Anti-inflammatory Effects of UDCA on Splenocytes Exposed to PMA/Ionomycin

  • Immunosuppressive therapy after organ transplantation is routinely used to prevent rejection of the organ, because this decreases the risk of adverse events, infection, and malignancies. Recently, ursodeoxycholic acid (UDCA), which is isolated from the dried bile of adult Chinese bears, has been shown to reduce the incidence and severity of acute rejection of liver allograft during early phase of liver transplantation. Therefore, in this study, we investigated the effect of UDCA on the proliferation of splenocytes exposed to PMA plus ionomycin. Our results demonstrated that UDCA decreased the splenocytes' proliferation in a dose-dependent manner. The decreased cell proliferation was accompanied with the decreased secretion of cytokines such as IL-2, IFN-${\gamma}$ and TNF-${\alpha}$. In addition, the pretreatment of UDCA on splenocytes stimulated with PMA plus ionomycin decreased the mRNA levels of cytokines (IL-2, IFN-${\gamma}$ and TNF-${\alpha}$) and costimulatory molecules (B7.2 and PD-L1). These results suggest the beneficial effect of UDCA on organ transplantation by decreasing lymphocyte proliferation.

  • As a series of studies to investigate the effect of immunosuppression on Ascaris suum infection in undefinitive hosts, and a delicate relationship between host and parasite, in the present studies, SPF ICR mice were alloted to experiment 1(normal undefinitive host group) and experiment 2(immunosuppressive group treated with prednisolone acetate) and inoculated with a single dose of 1,100 embryonated A suum eggs. In normal group, the infection essentially terminates 4 days after inoculation(DAI) with the attainment of middle third-stage in the liver, although few larvae migrate to the lungs where a few advance to late third stage. In immunosuppressive group, significant numbers developed to late third-stage in liver 8 DAI. In general, increasing of the mast cells and the goblet cells in the jejunum mucosa, of T-cells in the spleen and of activity of peritoneal macrophages followed by expulsion of the worms in the both groups. Considering a series of the results, suitabilities for the host of the worm appeared the highest from rabbit, hamster and mouse in that order. In addition, patent infection of A suum in the mice was also not obviously observed in spite of immunosuppression by prednisolone acetate.

Reticuloendotheliosis virus의 닭에 대한 면역억제효과와 병원성 (Immunosuppressive effects and pathogenicity of a Korean isolate of reticuloendotheliosis virus in chickens)

  • Immunosuppressive effects of reticuloendotheliosis virus (REV) infection in chickens were investigated. Primary antibody responses to Newcastle disease virus (strain B1) and sheep red blood cells were significantly low in chickens inoculated with the local isolate 89-74 of REV compared to those of uninfected chickens. In chickens infected with REV strain T or 89-74, blastogenesis of spleen cells and peripheral blood lymphocytes (PBL) to concanavalin A (Con A) was severely suppressed. When specific pathogen free (SPF) chickens were inoculated with the isolate, the suppressive effect was observed up to 7 weeks of age while, in the contact infected chickens, the suppression was absent. Similar suppressive effects were observed in chickens inoculated with REV strain T at 2, 3 and 4 weeks of age. When spleen cells or PBL from uninfected chickens were co-cultured with spleen cells or PBL from chickens infected with REV at 1 day-old or 2 week-old, the blastogenesis of the normal cells was suppressed. The suppressive effect of PBL from REV-infected chickens on normal lymphocytes was abrogated by the treatment with trypsin. However the suppressive activity of the REV-infected PBL was not influenced at removing machrophage from the cell suspension by incubation in plastic petri dishes. In addition to the immunosuppression, chickens infected with the REV isolate showed abnormal feather development (nakanuke), anemia, paralysis and retarded growth. Three out of 11 chickens inoculated with the isolate at day-old died between 6 and 9 weeks of age by bacterial infections.

Rifampicin 투여 폐결핵 환자의 T-임파구 저하증에 대한 Tuberactin-3 의 효과 (Effects of Tubercin-3 on Rifampicin Induced T-Lymphocytopenia in Tuberculosis Patients)

  • Rifampicin has been widely hailed as the most effective antituberculosis antibiotics since the clinical use of streptomycin, but its immunosuppressive side effect was still annoying problem to be excluded. These studies were carried out to determine the effect of Tuberein-3, tuberculous bacilli extraction with water, on Rifampicin induced T-lymphocytopenia in 5 cases of pulmonary tuberculosis who have never exposed to antimetabolites or steroid compounds. After 2 weeks treatment of Rifampicin, all cases showed T-lymphocytopenia, active $13.0{\pm}2.3$ % and total $43.1{\pm}4.4$%. Followed by another 2 weeks treatment with Rifampicin combined with Tuberein-3, T-lymphocytes in peripheral blood returned to the normal limit, active $21.6{\pm}3.3$% and total $56.3{\pm}1.7$%. Tubercin-3 revealed the restoring activity of suppression of T-lymphocyte rosettes by Rifampicin.

청열약류(淸熱藥類)의 면역약리학적(免疫藥理學的) 고찰(考察) (The immunopharmacologic study on drugs for clearing away heat and detoxicating(II))

  • In the result of investigating traditional chinese medical literatures to understand definite immun opharmacologic effects of drugs for clearing away heat and detoxicating such as Oldenlandiae Diffusae Herba, Fel Ursi, Fraxini Cortex, Pulsatillae Radix, Bruceae Fructus, Portulacea Herba, Patriniae Radix, we could reach conclusions as follows: 1. Oldenlandiae Diffusae Herba can increase voracity of leukocytes and immune function of splen ocytes. 2. Fel Ursi, Patriniae Radix can inhibit acute, chronic inflammation by decreasing voracity of macrophages, monocytes and recover lymphocytes. 3. Fraxini Cortex have anti-inflammatory effect then applied to treat with arthritis. Pulsatillae Radix, Bruceae Fructus, Portulacea Herba have anti-cancer, anti-biotic effects. Above results indicates that drugs for clearing away heat immunosuppressive effect that they can apply to all sorts of inflammatory diseases such as arthritis, DTH, SLE, and cancer.
