Effect of Astragalus membranaceus-postbiotics Polysaccharide Changed by Lactic Acid Bacteria on Macrophage (유산균에 의해 변화된 황기-포스트바이오틱스 다당류가 대식세포에 미치는 영향)

  • To increase industrial applicability of Astragalus membranaceus (AM) as immunostimulating materials, hot-water extract (AME) was prepared from AM and fermented with Kimchi-lactic acid bacteria (Lactobacillus sakei & Leuconostoc mesenteroides) to prepare fermented AM-postbiotics (FAME). Although FAME prepared from AM-postbiotics did not show a significant enhancement in macrophage stimulating activity compared to non-fermented AME, crude polysaccharide (FAME-CP) fractionated by EtOH precipitation from FAME showed significantly higher macrophage stimulating activity than AME-CP. Compared to AME-CP, FAME-CP showed dramatic changes in component sugar and molecular weight distribution. FAME-CP was a polysaccharide with a major molecular weight distribution of 113.4 kDa containing Man (44.2%), Glc (19.3%), Gal (10.2%), GalA (10.2%), and Ara (7.4%) as sugar components. FAME-CP with enhanced macrophage stimulatory activity not only increased expression levels of mRNA genes encoding macrophage-activated factors (iNOS, TNF-α, MCP-1, IL-6, and COX-2), but also led the nuclear translocation of activated p65 and c-Jun. In conclusion, crude polysaccharide from AM-postbiotics fermented with lactic acid bacteria could increase industrial applicability as a functional material with enhanced immunostimulating activity than AME-CP.

Preparation and Characteristics of Bread by Medicinal Herb Composites with Immunostimulating Activity (면역활성을 가진 생약복합물을 이용한 빵의 제조 및 특성)

  • In this study, the breads with medicinal herbs (MH) composites showing immunostimulating activity were prepared and their characteristics were examined. Fourteen kinds of medicinal herbs were extracted with hot water and divided into 3 groups (MH-1, MH-2, MH-3) based on their contents. All groups showed immunostimulating activity in terms of macrophage phagocytosis, nitrite production, cytostatic activity and cytokine production. In the preparation of breads containing MH extracts of various contents (0, 30, 50, 70, and 100%), there was no significant difference among dough pHs of all groups after first fermentation, but loaf volume was significantly (p<0.05) increased in 70% added group while decreased in 30%, 50%, and 100% added groups compared to the control. The "a" and "b" values of bread crumb increased with the content of MH extracts while "L" value decreased, but these values of bread crust were similar to the control group. Most improvements in hardness, adhesiveness, gumminess and chewiness of bread were noticed by the addition of 70% MH extracts, but those of springiness, cohesiveness and resilience were mostly by the 50% addition ones. Through the sensory evaluation, it was revealed that mouth feeling, taste and overall preference decreased at breads containing 70% and 100% extracts, although appearance and crumb texture were not significantly (p<0.05) different among all groups.

Pharmacological Activities of a New Glycosaminoglycan, Acharan Sulfate Isolated from the Giant African Snail Achatina fulica

  • Acharan sulfate (AS) is a glycosaminoglycan (GAG) prepared from the giant African snail, Achatina fulica. In this study, some biological activities of AS were evaluated on the basis of structural similarities to heparin/heparan sulfate and the biological functions of GAGs. We demonstrated that it exhibited strong immunostimulating activities as measured by carbon clearance test in mice and in vivo phagocytosis. It also exhibited a significant hypoglycemic activity in epinephrine (EP)-induced hyperglycemia as well as antifatigue effects by weight-loaded forced swimming test. And it showed hypolipidemic activities in cholesterol-rich mixture induced hyperlipidemia in rats. The above results indicate that AS has diverse biological activities and suggest therapeutically important target molecules.

  • Suspension cells of Angelica gigas Nakai was cultivated to produce extracellular polysaccharide(ECP) as immunostimulating agents. In this study, effects of pectinolytic enzymes such as pectinase. Pectinex on cell growth and ECPs production were investigated in suspension cultures. Optimum concentration of pectinase for ECP production ,vas found to be 0.1 U/L. at which ECP production was increased 1.39 fold (0.97 g/L) compared to control culture.

Immunostimulating Effect of Chickent Egg White Derivatives on Chemotactic Activity of Feline Peripheral Blood Polymorphonuclear Cells (고양이 말초혈액 다형핵백혈구의 유주활성에 있어서 계난백유래물질의 면역증강 효과)

  • 고양이 말초혈액 다형핵백혈구 (PMN)의 유주활성에 있어서 계난백유래물질 (EWD)의 면역증강 효과를 검토하였다. PMN의 유주활성은 EWD 그 자체 및 EWD로 배양한 PMN의 배양상층액에서는 활성이 존재하지 않았다. 그러나 EWD로 배양한 MNC 배양상층액에서는 PMN에 대한 유주활성이 증가되었다. 본 활성은 checkerboard assay를 통해 진성 유주활성으로 밝혀졌다. 이러한 고양이 PMN 유주활성은 human recombinant interleukin-8(hr IL-8)에 의해서도 증가되었다. IL-8의 또 다른 특성인 세포 형태변형 실험에서도 EWD로 배양한 MNC의 배양상층액과 hr IL-8 모두에서 높은 수준의 세포 형태변형효과를 나타내었다. 이와 같이 EWD로 배양한 MNC의 배양상층액과 hr IL-8 모두에서 증가된 PMN의 유주활성 및 세포변형효과는 anti-human IL-8mAb에 의해 농도의존적으로 억제되었다. 이상의 결과로부터 EWD는 고양이 PMN의 유주활성에 있어서 면역증강 효과가 있으며, 이것은 EWD에의해 활성화된 MNC에서 분비되는 IL-8양 유주성 인자에 의해 PMN의 유주활성 증가 및 세포 형태 변형이 일어나는 것으로 사료되었다.

Effect of Egg White Derivatives on Phagocytic Response of Feline Peripheral Blood Phagocytes (고양이 말초혈액 탐식세포의 탐식능에 있어서 계난백유래물질의 효과)

  • The immunostimulating effects of egg white derivatives (EWD) on the phagocytic response of feline peripheral blood polymorphonuclear cells (PMN) as well as mono- nuclear cells (MNC) were examined. The phagocytic activity was analyzed by a flow cytometry system. The EWD showed directly an enhanced effect on the phagocytic response of MNC but not PMN. The phagocytic activity of MNC was enhanced by culture supernatant from MNC and PMN treated with EWD, respectively. Similarly, the phagocytic activity of PMN was enhanced by culture supernatant from MNC but not PMN treated with EWD. It was, therefore, indicated that the enhanced phagocytic activity of feline PMN could be mainly mediated by humoral factor(s) released from MNC treated with EWD. These results suggested that EWD could enhance the phagocytosis of feline peripheral blood phagocytes.

  • Three new lectins, MLA-I, MLA-II and MLA-III, have been isolatedand purified from the hemolymph of Meretrix lusoria and reported previously. Biophysicochemical characteristics were investigated with these three MLA lectins. The MLA lectins agglutinated human erythrocytes non specifically and proved as D-galactose group carbohydrate specific. Molecular weight of ML.A-I. II and III were estimated to be 330, 500 and 310KD, respectively, by gel filtration on Sepharose CL-6B column. On SDS-polyacrylamide gel electrophoresis, ML.A-I was dissociated into a single subunit of 42KD, MLA-II was into the twelve subunits of 46, 32, 30, 28, 25, 23. 22, 20. 19, 16, 15, and 14KD, and MLA-III was into the two subunits of 72 and 44KD. The pl of MLA-I, II, III were 4.0. 4.9 and 5.0. Amino acid analysis revealed a high contents of acidic and hydroxy amino acids, and a paucity of sulfur containing amino acids. Proline was not contained in MLA-II.

  • Propolis, a natural resinous compound collected from honey bees, contains many biochemical constituents and has been used for traditional medicines as early as 300 B .C. Recently, it has been reported to possess many biological activities such as antibacterial, antiviral, fungicidal, local anaesthetic, immunostimulating, antiinflammatory and free radical scavenging properties. To investigate activities of chrysin, one of propolis effective compounds for blood coagulation system was injected endotoxin (4000 EU/kg, i.v.) in rats at 1 hr after administered chrysin (20 mg/kg, p.o.). This study was resulted that chrysin has antiplatelet aggregation activity in vitro, delay of blood clotting time and prothrombin time, and reduction of fibrinogen and FDP in vivo. Chrysin has increased SOD activity, GSH content and GST activity, and decreased MDA content in liver. The result suggests that the antithrombosis effect of chrysin is suppressive activity for a blood coagulation system and antioxidative activity.

  • A protein-polysaccharide fraction was prepared from the carpophores of Laetiporus sulphureus. This fraction suppressed growth of sarcoma 180 in A-strain mice when administered i. p. To investigate the mechanism of antitumor action of this fraction, plaque assay was conducted by administrating i. p. to the mise at a dose level of 50mg/kg for five days. Ten days later, the mice were immunized with 1 * 10$^{7}$ sheep red blooc cells. The number of hemolytic plaque forming cells was significantly greater than that of the control mice. Three monosaccharides and fifteen amino acids were identified in the protein-polysaccharide fraction.
