  • Isoflavone found in Leguminosae is one of natural phytoestrogens and its effect on bone remodeling is one of key investigational interests in terms of estrogen replacement therapy (ERT). As commonly known, osteoporosis is one of hormonal deficiency diseases, especially in menopausal women. When estrogen is blocked, local factors such as 1L-1$\beta$ and IL-6 that are related in bone resorption are increased and enhance osteoclastogenesis, which is responsible for bone resorption. In the present study, we investigated the effect of isoflavones (Isocal) extracted from Sophorae Fructus on bone resorption in vitro. (omitted)

  • Isoflavones have been a central subject in natural phytoestrogens found in Leguminosae. Their effects on bone formation and remodeling are an important turning point in that they can act like estrogen by binding on estrogen receptors on target cell surface. We, therefore, believed that isoflavones may be applied in estrogen deficiency disease such as osteoporosis in terms of estrogen replacement therapy (ERT).   • Purpose. To investigate the effect of different terpene enhancers on skin penetrations of prostaglandin El (PGE1) and its ethyl ester (PGE1-EE), a therapeutic agent for erectile dysfunction, external gel systems were formulated with the specific enhancers having different values in their lipophilicity (log P was ranged in 2.23-4.58). Methods. Topical gels containing PGEl (0.5 %) and PGEl-EE (0.1 %) were formulated with ethanol and propylene glycol as a vehicle, selective terpenes as a penetration enhancer, and HPC-H as a thickening agent.   • An association between drug treatment for viral infections and severe cutaneous adverse reactions has been noted. We investigated six patients diagnosed with Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) after being prescribed acetaminophen for suspected viral illnesses. Multiplex analysis was performed to measure cytokine levels in sera before and after treatment. IL-2Ra levels significantly decreased during the convalescence phase. Although acetaminophen is relatively safe, the drug can trigger SJS/TEN in patients with suspected viral infections. T-cells and monocytes may be key components of the link between viral infection and acetaminophen-induced SJS/TEN.

  • Oncoprotein Bcl-3 is perceived as an unusual member of $I{\kappa}B$ family since it can both stimulate and suppress NF-${\kappa}B$ activation. Aberrant Bcl-3 results in increased cell proliferation and survival, suggesting a contribution to malignant potential and elevated levels of Bcl-3 have been observed in many HTLV-1-infected T cell lines and ATL cells. To investigate the specific roles of Bcl-3 in HTLV-1-infected cells, we knocked down Bcl-3 expression using shRNA and then examined the consequences with regard to DNA damage and cell proliferation, as well as NF-${\kappa}B$ activation. The HTLV-1 encoded protein Tax promotes Bcl-3 expression and nuclear translocation. In HTLV-1-infected cells, Bcl-3 knockdown obviously induced DNA damage. Cell growth and NF-${\kappa}B$ activation were reduced in HTLV-1-infected or Tax positive cells when Bcl-3 expression was decreased. Together, our results revealed positive roles of Bcl-3 in DNA stabilization, growth and NF-${\kappa}B$ activation in HTLV-1-infected cells.

Consanguinity Protecting Effect Against Breast Cancer among Tunisian Women: Analysis of BRCA1 Haplotypes

  • The purpose of this study is to assess the effect of consanguinity on breast cancer incidence in Tunisia. We conducted a case-control study to evaluate the involvement of heterozygote and homozygote haplotypes of BRCA1 gene SNPs according to consanguinity among 40 cases of familial breast cancer, 46 cases with sporadic breast cancer and 34 healthy controls. We showed significant difference in consanguinity rate between breast cancer patients versus healthy controls P=0.001. Distribution of homozygous BRCA1 haplotypes among healthy women versus breast cancer patients was significantly different; p=0.02. Parental consanguinity seems to protect against breast cancer in the Tunisian population.

  • The intestine represents the largest and most elaborate immune system organ, in which dynamic and reciprocal interplay among numerous immune and epithelial cells, commensal microbiota, and external antigens contributes to establishing both homeostatic and pathologic conditions. The mechanisms that sustain gut homeostasis are pivotal in maintaining gut health in the harsh environment of the gut lumen. Intestinal epithelial cells are critical players in creating the mucosal platform for interplay between host immune cells and luminal stress inducers. Thus, knowledge of the epithelial interface between immune cells and the luminal environment is a prerequisite for a better understanding of gut homeostasis and pathophysiologies such as inflammation. In this review, we explore the importance of the epithelium in limiting or promoting gut inflammation (e.g., inflammatory bowel disease). We also introduce recent findings on how small RNAs such as microRNAs orchestrate pathophysiologic gene regulation.

  • Shrimp has efficient non-specific defense mechanisms based on activities of the hemocytes against pathogens. Up to now, it has been known that one of the non-specific immune reactions is related to mutual association among types of hemocyte, granular cell, semi-granular cell, and hyaline cell. In this study, we tried to know the effectiveness of feed-additives such as 5-aminolevulinic acid(ALA), chitosan, and hot-water extract of herb on immunity of shrimp (Penaeus chinensis) by hemocytic observation and SDS-PAGE analysis. Finally, we suggest a principle of the examination system for effects of various feed-additives.
