Objectives: Inflammasomes are molecular platforms that are generated inside cytoplasmic compartments. The objective is to mediate immunological responses of the host to cell damage and infection. Caspase-1 is triggered by inflammasome to generate interleukin-1𝛽 (IL-1𝛽), an inflammatory cytokine, and pyroptosis, an inflammatory form of apoptosis. Methods: In the past two decades, scientists have uncovered several inflammasomes. The most research has been conducted on NLRP3 inflamamsomes, whose activity can be stimulated by a variety of induction factors. However, the unregulated activation of NLRP3 inflammasomes is also a role in the etiology of several human disorders. Previous research has demonstrated that NLRP3 inflammasomes have a significant role in the innate and acquired immune systems, as well as in the prevalence of joint illnesses such rheumatoid arthritis. Conclusion: Within the scope of this review, we will present a brief overview of the biological features of NLRP3 inflamamsomes as well as a description of the underlying mechanisms governing activation and regulation. In particular, we explore the function of inflammasomes in the development of rheumatoid arthritis as well as the promise of recently identified medicines that target inflamasomes.
The adjuvant effect of a muramyl dipeptide (MDP) derivative, MDP-Lys(L18), on enhancing of antitumor immunity induced by X-irradiated tumor cells against highly metastatic B16-BL6 melanoma cells was examined in mice. Mice immunized intradermally (i.d.) with a mixture of X-irradiated B16-BL6 cells and MDP-Lys (L18) [Vac+MDP-Lys (L18)] followed by an intravenous (i.v.)inoculation of $10^4$ viable tumor cells 7 days after immunization, showed a significant inhibition of experimental lung metastasis of B16-BL6 melanoma cells. The most effective immunization for the prophylactic inhibition of tumor metastasis was obtained from the mixture of $100{\;}\mu\textrm{g}$ of MDP-Lys (L18) and $10^4$ X-irradiatied tumor vaccine. Furthermore, immunization of mice with Vac+MDP-Lys(L18), 3 days after tumor challenge, resulted in a significant inhibition of lung metastasis of B16-BL6 melanoma cells in an experimental lung metastasis model. Similarly, the administration of Vac+MDP-Lys(L18), 1 or 7 days after tumor removal, markedly inhibited tumor metastasis of B16-BL6 in a spontaneous lung metastasis model. When Vac+MDP-Lys (L18) was i.d. administered 3 days after subcutaneous (s.c.) inoculation of tumor cells ($5{\times}10^5/site$) on the back, mice treated with Vac+MDP-Lys(L18) showed inhibition of significantly tumor growth on day 20. These results suggest that MDP-Lys (L18) is able to enhance antitumor activity induced by X-irradiated tumor vaccine to reduce lung metastasis of tumor cells, and is a potent immunomodulating agent which may be applied prophylactically as well as therapeutically to treatment of cancer metastasis.
Objective : Lonicera japonica contains anti complementary polysaccharides and polyphenolic compound. Among these polyphenolic substances, chlorogenic acid is the major active component of this plant. However, the immunological mechanisms for these activities, have not been elucidated, nor the active components. To clarify immunomodulatory effects of those we examined the relationship between the activity of CD8+ T cell-mediated lysis and the frequency of cytokine profiles in spleen, thymus (especially IFN-${\gamma}$, IL-4, GM-CSF etc.) expressing CD8+ T cells activated by IL-2. Methods : To study immunomodulatory effects ethyl acetate fraction from Lonicera japonica, chlorogenic acid on cytokine gene expression from spleen, thymus cells, RT-PCR was performed after quantitative normalization for each gene by a densitometry using ${\beta}$-actin gene expression. A modified standard $^{51}Cr$-release assay was used to measure cytotoxic activities of cytotoxic T cells. Spleen, thymus cells from NOD mice were stained with CD3, CD4, CD44, CD69 in staining buffer and analyzed by two color flow cytometry. Results : We showed that ethyl acetate fraction from Lonicera japonica in combination with IL-2 resulted in a significant enhancement of PCR products for IFN-${\gamma}$, IL-4, IL-10, GM-CSF, IL-6 and cytotoxtic CD8+ T cell proportion in spleen and thymus T cells in NOD mice. This suggests that IFN-${\gamma}$, IL-6 like IL-4 may be acting as a regulatory rather than proinflammatory cytokine. Conclusions : In conclusion, based on the results of the present study which showed that ethyl acetate fraction from Lonicera japonica and chlorogenic acid upregulating cytokine gene expression in spleen and thymus, we are tempted to speculate that some of the therapeutic efficacies such as anti-diabetic activity of Lonicera japonica are due to the immunomodulatory its ethylacetate fraction and chlorogenic acid.

Sopungchungyoung-tang (SCT) has been widely used in Korea as a treatment of atopic dermatitis. SCT consists of Talcum, Rehmannia glutinosa, Angelica sinensis, Paeonia lactiflora, Cnidium officinale, Ledebouriella divaricata, Schizonepeta tenuifolia, Scutellaria baicalensis, Glycyrrhiza uralensis, Mentha arvensis, Cordyceps cicadae. We examined the immunological effect of SCT in vitro. We studied about the effect of SCT on Th cells' differentiation. In the case of CD4 T cells under neutral condition where there was only rIL-2 stimulus, SCT inhibited IFN-γ secretion by 70-80 %. Likewise, SCT also inhibited the IL-4 secretion of neutral Th cells by 85-90 %. We also experimented with the polarized Th1 cells/ Th2 cells and their production of IFN-γ and IL-4, respectively. There also were inhibitory effects on the polarized cells like there was on neutral cells, they were not as strong on the polarized cells. Under Th1 polarized condition, SCT acted dose-dependently, while in Th2 cells, the IL-4 production was inversely proportional to the doses of SCT. From the current study, it can be concluded that SCT exerts inhibitory effects on cytokine production without interfering with immune cells' activity. The result that SCT inhibits IFN-γ and IL-4 confirms that it does have a effect on immunomodulation.

The enzymatic properties of ${\beta}-amylase$ from soybean and sweet potato were compared. The sweet potato enzyme consists of four identical subsunits whereas soybean enzyme has no subunit $structure^{12,\;15)}$. In the denatured state, both enzymes exhibited the same molecular weight on SDS-gel electrophoresis and on gel-filtration analysis. The spectra of circular dichroism revealed that both enzyme have almost same secondary structure but the environment of aromatic side chains are different. The chemical cleavage of soybean and sweet potato ${\beta}-amylases$ at cysteine residues and methionine residues demonstrated the homology of amino acid sequence between the enzymes. The similarity between soybean and sweet potato ${\beta}-amylase$ was also revealed by immunological method. The antibody for soybean enzyme inhibited the activity of sweet potato enzyme but it did not inhibit the activity of wheat, barley and Japanese-raddish ${\beta}-amylases$.
Brain, heart, liver, lung, kidney and thymus etc. 12 organs were removed and homogenized from Dawley-Sprague rats after suffocation. After fractionation of the tissue cytosols, enzymatic activities of the key enzymes in metabolic inositol phosphates cycle, PLC, IPSK and Ins(1,4,5) P35-phosphatase, were measured respectively. Hybridoma monoclones producing anti-lP3K murine monoclonal antibodies were obtained by the fusion of SP2/Ag 0-14 and spleen cells of mouse immunized with purified 53KDa IPSK, screening and cloning procedures. 18 cloned hybridoma cells were obtained, background due to nonspecific binding was very low with 10 clones. These Abs were purified from ascitic fluids by using affi-gel 15, and determined subtype of Abs. When immunoreactivities for rat tissues IP3K were exercised by adding the mixed Abs of 19Gl and 19G2b, they showed an overall similarity with noncompetitive inhibition. Brain tissue has high sensitivity for anti-lP3K Ab, whereas heart tissue has very low activity. In kinetic parameters Km value was 1.58 mM and Vmx value was 5.41umol/min/ml, respectively Only one form of 40 KDa IPSK was detected in heart tissues, however rat brain contains at least three immunologically distinct IP3K (53, 51 and 40 KDa) in western blot analysis. Of them 53 KDa protein was major enzyme in enzymatic activity. Northern blot analysis with 32P-labeled CDNA probe which encodes 1.8 Kb IPSK gene was performed. These results suggest that IPSK are regulated at transcriptional level during rat tissue development.

Dekkera anomala YAE-1 strain separated from "airag" (Mongolian fermented mare's milk) produces β-glucosidase, which can convert ginsenoside Rb1 from Panax ginseng. Ginseng- derived bioactive components such as ginsenoside Rb1 have various immunological and anticancer activities. Airag was collected from five different mare milk farms located near Ulaanbaatar, Mongolia. YAE-1 strains were isolated from airag to examine the hydrolytic activities of β-glucosidase on Korean Panax ginseng using an API ZYM kit. Supernatants of selected cultures having β-glucosidase activity were examined for hydrolysis of the major ginsenoside Rb1 at 40℃, pH 5.0. The YAE-1 strain was found to be nearly identical at 99.9% homology with Dekkera anomala DB-7B, and was thus named Dekkera anomala YAE-1. This strain exerted higher β-glucosidase activity than other enzymes. Reaction mixtures from Dekkera anomala YAE-1 showed great capacity for converting ginsenoside Rb1 to ginsenoside Rd. The β-glucosidase produced by Dekkera anomala YAE-1 was able to hydrolyze ginsenoside Rb1 and convert it to Rd during fermentation of the ginseng. The amount of ginsenoside Rd was highly increased from 0 to 1.404 mg/ml in fermented 20% ginseng root at 7 days.
Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of a new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on hyperglycemia induced by streptozotocin(STZ) and by epinephrine in rats and in mice as well as on immunological functions in mice were investigated. The 70% methanol extract of the fungus, when administered orally at 100 and 300 mg/kg in STZ-induced hyperglycemic rats, caused a significant decrease in blood glucose level 3hr after sample treatments. The methanol extract, when administered p.o. at the same dose levels in epinephrine-induced hyperglycemic mice, also caused a significant decrease in serum glucose levels as well as a significant reversal of the liver glycogen contents suggesting its hypoglycemic activity might be due to glycogen breakdown in the liver. Treatment of normoglycaemic mice with the methanol extract of the fungus exhibited a significant glucose tolerance up to 3hr after oral glucose load(2.0 g/kg). The methanol extract also showed immuno-stimulating activity as measured by carbon clearance in mice and a significant antifatigue effect as measured by weight loaded forced swimming performance in mice.

CpG-DNA, which contains unmethylated CpG dinucleotides in the context of specific sequences, has remarkable and diverse immunological effects, including induction of proinflammatory cytokine expression and regulation of the Th1/Th2 immune response. Here, we examined the immunostimulatory activities of double-stranded (ds) CpG-DNA in the human B cell line RPMI8226. To investigate whether dsCpG-DNA stimulates immune cells, we constructed a plasmid containing repeated dsCpG-DNA and produced dsCpG-DNA by PCR amplification and EcoR I digestion. PCR-amplified dsCpG-DNA alone did not have immmunostimulatory activity. However, dsCpGDNA encapsulated with lipofectin induced IL-8 promoter activation, HLA-DRA expression, and IL-8 expression in a CG sequence-independent manner. The effects of encapsulated dsCpGDNA were independent of minor endotoxin contamination. These findings suggest the potential use of dsCpG-DNA as a therapy for immune response regulation.

Tyrosinemia I (fumarylacetoacetate hydrolase deficiency) is an autosomal recessive inborn error of tyrosine metabolism that produces liver failure in infancy or a more chronic course of liver disease with cirrhosis, often complicated by hepatocellular carcinoma in childhood or early adolescence. We studied a 37-year-old woman with tyrosinemia I whose severe liver disease in infancy and rickets during childhood were resolved with dietary therapy. From 14 years of age, she resumed unrestricted diet with the continued presence of the biochemical features of tyrosinemia, yet maintained normal liver function. In adult years, she accumulated only a small amount of succinylacetone. Despite this evolution to a mild biochemical and clinical phenotype, she eventually developed hepatocellular carcinoma. Her fumarylacetoacetate hydrolase genotype consists of a splice mutation, IVS6-1G>T, and a novel missense mutation, p.Q279R. Studies of resected liver revealed the absence of hydrolytic activity and immunological expression of fumarylacetoacetate hydrolase in tumour. In the non-tumoral areas, however, 53% of normal hydrolytic activity and immunologically present fumarylacetoacetate hydrolase were found. This case demonstrates the high risk of liver cancer in tyrosinemia I even in a seemingly favorable biological environment. In this study of tyrosinemia I, Case 2 with negative succinylacetone accumulation and the recovery of acute liver failure was compared with Case 1. Diet restriction and NTBC treatment are crucial to prevent hepatocellular carcinoma until liver transplant can take place and cure the condition. Further studies are needed to examine cases where liver cancer did not result despite clinical symptoms/signs of tyrosinemia type I.
