Overexpression of Periostin Protein in Non-Small Cell Lung Carcinoma is Not Related with Clinical Prognostic Significance

  • Background: Periostin is preferentially expressed in periosteum, indicating a potential role in bone formation. Recently, there have been emerging controversies about its role in invasion and metastasis of human malignancies. We attempted to determine the clinicopathological significance of periostin expression in non-small cell lung carcinoma (NSCLC). Methods: Immunohistochemical staining of periostin protein from 91 cases of NSCLCs was performed using tissue microarray blocks. The results were correlated with clinicopathological parameters. Results: Positive reaction to periostin was predominantly noted in the tumor stroma. The strongest reaction presented as a band-like pattern just around the tumor nests. Non-neoplastic lung tissue and most in-situ carcinomas did not show a positive reaction in their stroma. With respect to tumor differentiation, moderate to poor differentiated tumors (47/77) revealed even higher periostin expression than the well-differentiated ones (4/14) (p=0.024). High periostin expression was positively correlated with E-cadherin and p53 expression, but was not related with patient age, sex, tumor type, PCNA index, b-catenin, cyclin D1, pTNM-T, pTNM-N, stage, and patient survival (p>0.05). Conclusion: These results suggest that periostin might play a role during the biological progression of NSCLC, but may not be related to the clinical prognostic parameters.

오가피의 NO 생성과 PDE-5 억제를 통한 음경해면체 이완효과 (Relaxing Effects of Acanthopanacis Cortex through NO Production and PDE-5 Inhibition in Corpus Cavernosum)

  • This study was aimed to examine relaxing effects of Acanthopanacis cortex(AC) through nitric oxide(NO) production and phosphodiesterase type 5(PDE-5) inhibition in corpus cavernosum. In order to define the relaxation effects of AC extract, rabbit corpus cavernous tissues were prepared in $2{\times}2{\times}8mm$ sized strip. AC extract ($0.01-3.0mg/m{\ell}$) were treated in contracted strips induced by phenylephrine(PE) and $N{\omega}$-nitro-L-arginine (L-NNA) was treated before AC extract-treated. And calcium chloride($Ca^{2+}$) 1 mM was infused into precontracted strips after pretreatment of AC extract in $Ca^{2+}-free$ krebs-ringer solution. When AC extract was applied to human umbilical vein endothelial cell(HUVEC), cell viability was measured by MTT assay, and NO concentration was measured by Griess reagent system. Ratio of smooth muscles to collagen fibers and eNOS, PDE-5 positive reaction were measured by histochemical and immunohistochemical process on mice corpus cavernosum. AC extract significantly affected relaxion of the cavernous strips, and the pretreatment of L-NNA inhibited AC extract-induced relaxation. Contraction induced by the addition of $Ca^{2+}$ was inhibited by treatment with the AC extract in $Ca^{2+}-free$ solution. In AC group, NO concentration, ratio of smooth muscle to collagen fibers, and eNOS positive reaction were increased, PDE-5 positive reaction was decreased compared to PE group. As a result of the above experiment, it was thought that AC extract inhibits the inflow of extracellular $Ca^{2+}$ by activating cGMP through the increase of eNOS / NO and the decrease of PDE-5 which inhibits cGMP activity, in the corpus cavernosum.

  • Recently, available interests concerning the biologic significance of the extracellular matrix and proliferating cells associated with periodontal disease has been increased. The distribution or expression of cellular proliferation by PCNA, macrophage detection by ${\alpha}$-l-antichymotrypsin, fibronectin playing a important role in host defence mechanisms indirectly, and transglutaminase that cross linked to fibronectin and stimulate fibrin stabilization were studied in inflammed and healthy gingiva. The excised tissue samples were fixed neutral formalin for 24 hours, embedded with paraffin, sectioned at 4-61lffi in thickness, and immunohistochemically processed by LSAB method. The positive reaction to PCNA was localized in the suprabasal and basal layer of inflammed gingiva and an increasing reactivity was observed than healthy gingiva. ${\alpha}$-I-antichymotrypsin positive cells were localized in the basal layer of inflammed gingiva, and there was no or rare positive cells in healthy gingiva. The positive reaction to fibronectin in inflammed gingiva was more than healthy gingiva,"and shown in the connective tissue subjacent to basement membrane of epithelium and in the periphery of the collagen fiber bundles. The positive cells by transglutaminase in inflammed gingiva were noted in suprabasal, spinous, and keratin layer of epithelium, and slightly increased in the capillaries of connective tissues. But the results of this study demonstrated in vitro reaction. Therefore, the role of PCNA,${\alpha}$-l-antichyrnotrypsin, transglutaminase, fibronectin and coefficient with other growth factor and extracellular matrix were further investigated in vivo.

  • 저자들은 마우스를 실험모델로 하여 간흡충의 항원을 투여 했을 때 비장조직에 대한 CD3, CD4 및 CD8 모노클로날 항체의 반응 여부를 알아보고자 하였다. 즉, 간흡충에 대한 세포면역학적인 특성을 규명고자 하였으며 특히 비장 조직에 대한 phenotype을 관찰한 결과 다음과 같은 결과를 얻었다. 간흡충의 조항원을 면역증강제와 함께 복강 투여한 다음 일정 기간 후에 비장조직을 Avidin-biotin complex 면역조직염색을 실시한 결과 CD3에서 강한 양성 반응을 나타냈고 CD4와 CD8에서는 약한 반응을 나타냈다. 조직부위를 보면 피막, 혈관, 임파관, 백수부위와 림프구 및 대식 세포의 세포막에서 양성반응을 보였다.

가미신기환(加味腎氣丸)이 수컷 노화 생쥐의 생식기능에 미치는 영향 (Effects of Gami-Shinkiwhan on the Reproductive Function of Male Aged Mice)

  • This study was performed using histochemical and immunohistochemical methods to investigate the effect of Gami-Shinkiwhan(GS) on reproductive function of male aged mice. 8-weeks-old ICR mice were used as control group, without any treatment, and 15-month-old ICR mice were used as aging elicited group(AE) and Gami-Shinkiwhan treatment group(GS). AE group didn't restrict diets and drinking for 6 months without any treatment. GS was administered 0.56g/kg/day for 6 months. Compared with AE group, the cell division of sertoli cells, spermatids, and spermatogonial cells was increased and the apoptosis of sertoli cells was decreased on GS group. Androgen receptor positive reaction and $17{\beta}$-HSD positive reaction were significantly increased in the GS group compared to AE group. In addition, the DJ-1 positive reaction was significantly increased and the HDAC3 positive response was significantly decreased in the GS group compared with AE group. Based on the above results, GS prevented the apoptosis of sertoli cells in the tubules, and increased the production of sertoli cells, spermatozoa and testosterone. Based on this, it is thought that it improves male reproductive dysfunction caused by late-onset hypogonadism.

  • 효율적인 바이오칩을 개발을 위해 칩 표면에 생체 물질들의 상호반응이 효과적으로 일어날 수 있는 센싱 표면의 조성과 항원-항체 반응과 같은 생체인식 반응을 정량적 신호로 전환하는 방법에 대해 연구하였다. 전기화학식 센서의 표면을 개선하기 위해 폴리아미도아민 덴드리머를 가교 물질로 도입하였다. 생체 분자들의 인식작용을 정량적인 신호로 전환하기 위해 전형적인 면역조직화학분석에서 사용된 반응들을 바이오센서에 적용한 방법론을 사용하였다 효소에 의해 촉매되는 신호화 방법은 면역반응들에 대하여 광학식 센서와 전기화학식 센서에서 공히 수행되었으며, 매우 정량적인 신호로 측정되었다. 측정된 신호들로부터 단백질 농도에 비례하는 검량곡선을 획득할 수 있었으며 다양한 면역 샘플에 대한 적용 가능성을 제시하였다.

폐 유상피 혈관내피종 3예 (Three cases of Pulmonary Epithelioid Hemangioendothelioma)

  • 국내에서 폐 유상피 혈관내피종이 악성으로 진행하여 사망한 예는 1예만 보고되어 있다. 단일 폐결절이 있는 환자에서 개흉 폐절제술후 병리조직학적으로 종양세포의 유사분열과 괴사를 동반하여 악성 유상피 혈관내피종으로 진단하고 예후가 불량할 것으로 예상되는 1예와 단순 흉부 촬영상 양측 다발성 폐결절이 있는 무증상 환자에서 비디오 흉강경 폐생검으로 폐 유상피 혈관내피종을 진단 후 조직학적 및 임상적으로 악성 소견 없이 경과를 관찰 중인 1예 및 양측 폐의 다발성 결절성 병변과 늑막액을 동반하여 개흉 폐생검으로 폐 유상피 혈관내피종을 진단 후 호흡부전으로 사망한 1예의 폐 유상피 혈관내피종 3예를 경험하였기에 문헌 고찰과 함께 보고하는 바이다.

간흡충 충체의 부위별 항원성에 대한 면역조직화학적 연구 (Immunohistochemical study on the antigenicity of each organ structure of Clonorchis sinensis)

  • 간흡충의 충체 부위별 항원성을 알아보고자 면역조직화차적 방법을 이용하여 간흡충의 소화기관, 생식기관, 배설기관, 흡반, 표피 등의 염색강도를 비교 관찰하였다. 간흡충 실험 감염 후 2~8주 된 토끼에서 담관 내 충체를 포함하는 간 조직을 얻어 중성 포르말린 용액에 고정하고 파라핀으로 포매한 다음 4 $\mu\textrm{m}$두께로 잘라 항원으로 이용하였다. 항-간흡충 항체(1차 항체)는 실험 감염 10주 된 고양이 혈청을 희석하여 사용하였고 peroBidaseconjugated goat anti-cat IgG를 2차 항체로 하여 간접 면역효소 염색을 시행하였다. 가장 적합한 1차 항체의 희석 농도는 1 : 1,000~1 : 2,000, 2차 항체의 희석 농도는 1 : 1,000이었다. 충체의 장 상피세포, 장 내용물 및 배설낭은 1차 항체 희석 농도 1 : 1,000~1 : 2,000에서 강한 양성 반응을 보인 반면, 자궁 및 일부 자궁 내 충란, 난관선, 웅성 생식기관 등은 1차 항체 회석 농도 1 : 1,000에서 미약한 양성 반응을 나타내었다. 한편, 흡반, 표피, 표피하 세포, 충체 실질 등은 1차 항체 회석 농도 1 : 1,000에서도 음성 반응을 나타내었다. 이상의 결과를 종합하면, 간흡충 감염시 숙주의 항체 반응은 충체의 소화-분비 기관에서 유래된 이른바 분비-배설 항원에 의해 가자 강력하게 유발되는 것으로 추측된다.

비침습식 미세전류자극이 토끼 경골의 골절 후 BMP-4 발현에 미치는 영향 (Effects of Non-Invasive Constant Microcurrent Stimulation on Expression of BMP-4 After Tibia Fracture in Rabbits)

  • 골절 후 골 유합에 대한 미세직류전류 자극의 효과를 관찰하기 위하여 체중 2.5~3 ㎏내외의 6개월 령 뉴질랜드 웅성토끼 24마리를 대상으로 경골 골절 후 미세전류 자극을 적용한 실험군과 비적용군인 대조군으로 나누어 3일, 7일, 14일 및 28일 후 BMP-4에 대한 면역조직화학적 염색을 실행하여 다음과 같은 결론을 얻었다. BMP-4의 발현은 비침습식 미세전류를 적용한 실험군과 자연치유군인 대조군 모두 시간이 경과함에 따라 통계학적으로 유의하게 증가하다가 감소되었다. 그러나 실험군에서 경골 골절 3일 후 대조군은 하버씨계의 간질층판을 중심으로 약한 갈색의 면역양성반응(+)을 보였으나 실험군의 경우 중등도의 면역양성반응(++)을 보였다. 경골 골절 7일 후 대조군은 하버씨계의 동심원과 간질층판을 중심으로 중등도의 갈색의 면역양성반응(++)을 보였으나 실험군의 경우 바깥층판을 포함하여 매우 강한 갈색의 면역양성반응(++++)을 보였다. 경골 골절 14일 후 대조군은 하버씨계의 간질층판과 동심원을 중심으로 강한 갈색의 면역양성반응(+++)을 보였으며 실험군 또한 강한 면역양성반응(+++)을 보였다. 그 후 점차 감소하여 경골 골절 28일 후 대조군은 하버씨계의 간질층판을 중심으로 약한 갈색의 면역양성반응(+)을 보였으나 실험군의 경우 중등도의 면역양성반응(++)을 보였다. 위의 결과로 보면 골절 후 미세전류를 비침습적으로 적용할 때 치유과정 초기에 골형성단백질인 BMP-4의 발현을 증가시켜 골절 치유를 촉진시킴을 알 수 있다.

오약순기산(烏藥順氣散)이 허혈성 뇌손상 흰쥐의 인지 및 운동기능회복에 미치는 효과 (The Effects of Ohyaksungi-san(Wuyaoshunqi-san) on Cognition and Motor Function Recovery after Ischemic Brain Injury in Rat)

  • Objectives : Ohyaksungi-san(Wuyaoshunqi-san) has been used for many years as a treatment for cerebrovascular diseases in Oriental medicine. This study was designed to evaluate the effects of Ohyaksungi-san(Wuyaoshunqi-san) on cognition and motor function recovery after ischemic brain injury, and also the expression of BDNF in hippocampus. Methods : This study was designed with 4 subgroups to evaluate the effects of Ohyaksungi-san(Wuyaoshunqi-san). As control groups, group I has no treatment during 1 week after ischemic brain injury and group II has no treatment during 2 weeks after ischemic brain injury. As experimental groups, group III has been treated with Ohyaksungi-san(Wuyaoshunqi-san) during 1 week after ischemic brain injury and group IV have treated with Ohyaksungi-san(Wuyaoshunqi-san) during 2 week after ischemic brain injury. Each group has been examined by tests as follows, neurological motor behavioral tests, cognitive motor behavior test and histological test. Neurological motor behavior tests consisted of limb placement test, beam-walking test and horizontal wire test. Cognitive motor behavior test was performed by using Morris water maze. In the histological test, TTC(2,3,5-triphenylteterazolium chloride) staining, hematoxylin & eosin staining, and immunohistochemical staining were used. Results : 1. The tests for motor function recovery change had significantly good result in the experimental groups as compared with control groups(p<.05). 2. The Morris water maze test on cognition also had significantly good result in the experimental groups as compared with control groups(p<.05). 3. In the immunohistochemical staining for the expression of BDNF in hippocampus, more immune reaction was investigated in the experimental groups as compared with control groups. Especially group IV has the greatest immune reaction. Conclusions : Ohyaksungi-san(Wuyaoshunqi-san) has good effects on cognition and motor function recovery after ischemic brain injury, and also the expression of BDNF in hippocampus.
