Role of CD10 Immunohistochemical Expression in Predicting Aggressive Behavior of Phylloides Tumors

  • Background: Phylloides tumors are rare breast neoplasms with a variable clinical course depending on the tumor category. Along with histologic features, the role of immunohistochemical staining has been studied in predicting their behavior. Objectives: Our aim was to evaluate the role of CD 10 immunohistochemical staining in predicting survival, recurrence and metastasis in phylloides tumor. We also evaluated correlations of other clinicopathological features with overall and disease-free survival. Materials and Methods: CD10 expression was studied in 82 phylloides tumors divided into recurrent/metastatic and non-recurrent/non-metastatic cohorts. The Chi-square test was applied to determine the significance of differences in CD10 expression between outcome cohorts. Uni and multivariate survival analyses were also performed using log-rank test and Cox regression hazard models. Results: All 3 metastatic cases, 5 out of 6 (83.3%) recurrent cases and 37out of 73 (50.7%) non-recurrent and non-metastatic cases expressed significant (2+ or 3+) staining for CD10. This expression significantly varied between outcome cohorts (p<0.03). Tumor category and histological features including mitotic count and necrosis correlated significantly with recurrence and metastasis. A significant decrease in overall and disease free survival was seen with CD10 positivity, malignant category, increased mitoses and necrosis. Neither CD10 expression nor any other clinicopathologic feature proved to be an independent prognostic indicator in multivariate analysis. Conclusions: CD10 immunohistochemical staining can be used as a predictive tool for phylloides tumor but this expression should be interpreted in conjunction with tumor category.

Preventive Effects on Porcine Epidemic Diarrhea(PED) Using by PEDV Antiserum II. Clinical Sign, Histopathological Lesion and Immunohistochemical Finding (항혈청 투여에 따른 돼지 유행성 설사병 예방효과 II. 임상증상, 병리조직학적 검사, 면역조직학적 검사)

  • The purpose of this study was to investigate to protective effects against porcine epidemic diarrhea virus (PEDV) infection in piglets by administration of the PEDV antiserum orally at 2 hrs, 24hrs and 36hrs after birth. six piglets administered the antiserum were experimentally infected with PEDV at five-day-old. Control group were four piglets infected with PEDV only. Clinical signs and gross, histopathological lesion and immunohistochemical findings were examined. The results obtained were as follows; 1. In clinical signs, piglets of the control group appeared the typical signs of severe watery diarrhea, depression and anorexia but piglets of the PEDV antiserum treated group recovered progressively. In mortality, control group showed 75%, but PEDV antiserum treated group showed 16.7%, respectively. 2. In gross findings, piglets of the control group appeared the typical findings of congestion, distension of lumen, containing curdes of undigested milk in stomach. But piglets of the PEDV antiserum treated group appeared milder than those of control group. 3. In histopathological findings, piglets of the control group appeared the typical findings of villous atrophy and fusion, congesion, exfoliation, vacuolation, squamation, loss of cilia and proliferation of crypt. But piglets of the PEDV antiserum treated group appeared milder than those of control group. 4. In immunohistochemical findings, piglets of the PEDV antiserum treated group showed more intensive in reaction for IgG and IgG than those of control group. The recation for IgA was stronger than that of IgG. It was concluded that oral administration of PEDV antiserum to piglets was effective in preventing PEDV infection and reduced their mortality.

Comparative histomorphologic study of regenerated bone for dental implant placement in the atrophied posterior maxilla

  • The purpose of this study is to evaluate the regenerative capacity of reconstruction in the atrophied posterior maxilla by comparing bone graft procedures and alveolar distraction osteogenesis (ADO) techniques. We performed the autogenous iliac bone graft (AGB group, 5 specimens in 3 patients), and the combination (Mixed group, 3 specimens in 3 patients) of the autogenous and deproteinized bovine bone ($Bio-Oss^{(R)}$, Geistlich Co., Switzerland) as the ratio of 2:1 in the sinus floor elevation procedures. ADO procedures using $TRACK^{(R)}$ (KLS Martin Co., Germany) were also performed to augment vertical alveolar height in atrophied posterior maxilla (ADO group, 5 specimens in 4 patients). Newly generated bone tissues were obtained with the 2.0mm diameter trephine bur (3i Co., USA) during implant fixture installation after 5-7 months. Routine histolomorphological observation, immunodot blot assay for quantitative evaluation, and immunohistochemical staining with antibodies to MMP-1, -9, -10, TIMP-1, -2, and BMP-2, -4 were all carried out. Lamellar bone formation was well shown in all specimens and new bone formations of ADO group increased than those of other procedures. In immunohistochemical staining, the strong expression of BMP-2 was shown in all specimens, and immunodot blot assay showed that bone formation is accompanied by the good induction of factors associated with angiogenesis and appeared more increased amount of osteogenic and angiogenic factors in ADO group. ADO is the most effective technique for new bone formation compared to sinus floor elevation with autogenous or mixed bone graft in the atrophied posterior maxilla. In the quantitative immunodot blot assay, the regenerated bone after ADO showed more increased products of VEGF, BMP-2, PCNA and MMP-1 than those after the other procedures, and these findings were able to be confirmed by immunohistochemical stainings.

The morphological and immunohistochemical changes of endocrine pancreas by pancreatic duct ligation in chicken (췌관을 결찰한 닭 췌장 내분비부의 형태학적 및 면역조직화학적 변화)

  • To investigate morphological changes in the endocrine pancreas of chicken after pancreatic duct ligation, experimental animals were subdivided to control, 12 hours, 1 day, 2 days, 4 days, 7 days and 10 days groupes and all of three pancreatic ducts of chicken were ligated by surgical procedure and then the morphological changes were observed. In pancreatic islets, the vacuolation and invasion of connective tissue were occurred in all experimental groups and dissociation of pancreatic islets was detected in 4 days after pancreatic duct ligation and hold out to 10 days. The peak of the morphological changes in pancreatic islets was detected in 4 days after pancreatic duct ligation. In the results of immunohistochemical methods against glucagon, insulin, somatostatin and bovine pancreatic polypeptide(BPP), the number of immunoreactive pancreatic islets were decreased but the size increased with time, so the number of immunoreactive cells in each pancreatic islets were increased. Glucagon-immunoreactive cells were not changed but insulin-immunoreactive cells were decreased with time(p<0.05). BPP-immunoreactive cells were increased in 2 days after pancreatic duct ligation and then decreased with time(p<0.05). Somatostatin-immunoreactive cells were increased with time(p<0.05) in dark islets.

Observations of a endometrial stromal sarcoma in the rat uterus wall (Rat의 자궁벽에 발생된 stromal sarcoma의 관찰)

  • The present observation was focussed mainly on the histopathological and immunohistochemical finding of the tumor developing from the outside of rat uterus wall. In gross finding, this tumor were a oval form mass and 0.75cm and 1cm in greatest demension. The sectioned surface showed uniform, solid, gray-white. In histopathological finding, the tumor showed the form of lymphoid nodule-like structure and was composed of the cells with karryorrhexis, karyolysis and necrosis in the central zone, mature cells in the paracentral zone, and concentric layers of compressed, flattened and atrophied smooth muscle fibers by the proliferating of the cells closely resembling endometrial stromal ells in the peripheral zone and a few smaller nodule-like structure in certain areas of the peripheral zone. In immunohistochemical observations, the proliferating cells presents only among the concentric layers of the smooth muscle fibers in the peripheral zone. Labeling index of proliferating cells in the peripheral zone was $4.34{\pm}1.28%$. These tumor cells did not metastasized in another orgns and labeling index was low, So it was concluded that this tumor was benign stomal sarcoma.

LIGHT is Expressed in Foam Cells and Involved in Destabilization of Atherosclerotic Plaques through Induction of Matrix Metalloproteinase-9 and IL-8

  • Background: LIGHT (TNFSF14) is a member of tumor necrosis factor superfamily and is the ligand for TR2 (TNFRSF14/HVEM). LIGHT is known to have proinflammatory roles in atherosclerosis. Methods: To find out the expression pattern of LIGHT in atherosclerotic plaques, immunohistochemical analysis was performed on human carotid atherosclerotic plaque specimens. LIGHT induced atherogenic events using human monocytic cell line THP-1 were also investigated. Results: Immunohistochemical analysis revealed expression of LIGHT and TR2 in foam cell rich regions in the atherosclerotic plaques. Double immunohistochemical analysis further confirmed the expression of LIGHT in foam cells. Stimulation of THP-1 cells, which express TR2, with either recombinant LIGHT or immobilized anti-TR2 monoclonal antibody induced interleukin-8 and matrix metalloproteinase(MMP)-9. Electrophoretic mobility shift assay demonstrated that LIGHT induces nuclear localization of transcription factor, nuclear factor $(NF)-{\kappa}B$. LIGHT induced activation of MMP-9 is mediated by $NF-{\kappa}B$, since treatment of THP-1 cells with the $NF-{\kappa}B$ inhibitor PDTC (pyrrolidine dithiocarbamate) completely blocked the activation of MMP-9. Conclusion: These data indicate that LIGHT is expressed in foam cells in atherosclerotic plaques and is involved in atherogenesis through activation of pro-atherogenic cytokine IL-8 and destabilization of plaque by inducing matrix degrading enzyme.

Immunohistochemical and serological observations on Neospora caninum infection in Korean indigenous cattle and calves of abnormal deliveries (한우 및 이상산 송아지의 Neospora caninum의 감염에 대한 면역조직화학적 및 혈청학적 관찰)

  • The present study was carried out to investigate the prevalence of Neospora caninum infection in Korean indigenous cattle and calves of abnormal deliveries and focus on correlation between malformation and N caninum infection. To determine the prevalence of antibodies to N caninum, sera of 473 Korean indigenous cattle from slaughter house were tested for N caninum antibodies using indirect fluorescence assay. Of the 473 cattle sera, 9.5% (45/473) showed positive against N caninum. Regional seropositive rates of the samples were 16.7% (5/85), 11.0% (11/100), 8.8% (21/240) and 5.9% (5/85) at Kyonggi, Gyeongbuk, Daegu and Kyongnam province, respectively. In female, seropositive rates were 17.5% (25/143) and 6.1% (20/330) in male. During the period from march 2000 to August 2001, 55 abnormal deliveries of Korean indigenous cattle including abortion, stillbirth and congenital malformation were examined by histopathological, immunohistochemical and serological methods for evidence of N caninum infection. Of the 55 abnormal deliveries, only 5 calves showed positive reaction against N caninum in serological test. In microscopical observation, gliosis and nonsuppurative myositis were observed. However, Neospora-like organisms were not detected by either periodic acid-schiff (PAS) reaction or immunohistochemical technique. Taken together all these data, this study indicate that N caninum infection was widespread in breeding farms of Korean native cattle, but correlation between malformation and N caninum infection was not recognized.

  • Immunohistochemical study on the intestinal tissues obtained from the 21 pigs of the 14 terms in Korea in which the clinical and epidemiological features had indicated the possible outbreaks of porcine epidemic diarrhea(PED) was performed using the indirect immunofluorescence test and/or the immunoperoxidase method in order to detect PED viral antigens in the infected cells of the intestines, and histopathological features were described as well. By immunohistochemical analysis, PED viral antigens were detected in the epithelial cells covering the small intestinal villi and recognized slightly in the cells lining the colonic surface epithelium as well. Occasional fluorescence was also seen in a few intestinal crypt epithelium. On light microscopy, the piglets with PED showed marked villous atrophy and fusion, and severe enterocyte degeneration and desquamation. On the other hand, the older pigs more than 4 week old age was mild villous atrophy and fusion, severe villous epithelial cell proliferation, and moderate mononuclear cell infiltration.

Expression of Neuronal Markers, NFP and GFAP, in Malignant Astrocytoma

  • Background: Immunohistochemical markers are considered as important factors in diagnosis of malignant astrocytomas. The aim of the current study was to investigate the frequency of the immunohistochemical markers neurofilament protein (NFP) and glial fibrillary acidic protein (GFAP) in malignant astrocytoma tumors in Firoozgar and Rasool-Akram hospitals from 2005 to 2010. Materials and Methods: In this cross-sectional study, immunohistochemical analysis of NFP and GFAP was performed on 79 tissue samples of patients with the diagnosis of anaplastic and glioblastoma multiform (GBM) astrocytomas. Results: The obtained results demonstrated that all patients were positive for GFAP and only 3.8% were positive for NFP. There was no significant association between these markers and clinical, demographic, and prognostic features of patients (p>0.05). Conclusions: NFP was expressed only in GBMs and not in anaplastic astrocytomas. It would be crucial to confirm the present findings in a larger number of tumors, especially in high grade gliomas.

The Relationship between the Expression of Melanoma Differentiation-Associated Gene-7/Interleukin-24 (MDA-7/IL-24) and Clinicopathological Features in Colorectal Adenocarcinomas

  • The melanoma differentiation-associated gene-7 (MDA-7) protein, also known as interleukin-24 (IL-24), is a novel candidate of tumor suppressor that can induce apoptosis experimentally in a variety of human malignant cells. However, there have been few studies about its role in colorectal cancer. We performed immunohistochemical detection of MDA-7/IL-24 in 399 tissue samples from primary colorectal adenocarcinoma patients using a tissue microarray. Western blotting was then done to confirm the immunohistochemical observations. MDA-7/IL-24 immunoreactivity was observed in 116 (29.1%) of the 399 colorectal adenocarcinoma cases. Analysis of the MDA-7/IL-24 expression by Western blotting confirmed the immunohistochemical results. The tumors with a negative MDA-7/IL-24 expression more frequently showed poor differentiation (P=0004), lymph node metastasis (P=0.001), deep invasion (P=0.008) and high stage (P=0.001). A subset of colorectal adenocarcinoma revealed a decreased expression of MDA-7/IL-24, and this was associated with progressive pathologic features. These findings suggest that loss of MDA-7/IL-24 expression may play a role in tumor growth and progression of colorectal adenocarcinomas.
