Expression of major piroplasm protein(p33)of Theileria sergenti (Korean isolate) and its immunogenicity in guinea pigs

  • To investigate the development of a subunit vaccine against theileriosis in cattle, the DNA fragments encoding piroplasm surface protein (p33) of Theileria sergenti of a Korean isolate were expressed in baculoviruses. The expressed p33 was characterized by indirect fluorescent antibody (IFA) and western blotting analysis. The expression of p33 was mainly detected on the surface of infected Sf21 cells by IFA. The immunoblotting analysis revealed the presence of a same molecular weight protein band of p33. The antigenicity of expressed polypeptide was further examined through the inoculation of a guinea pig. The sera of guinea pigs immunized with p33 expressed cell Iysate showed similar fluorescent antibody patterns and reacted with the same molecular weight protein of T. sergenti in immunoblotting analysis, thus indicating that this protein can be a promising candidate for a subunit vaccine in the future.

  • 한국인 383명의 Compiement Component 6(C6)의 유전적 다형을 등전점 전기영동과 immunoblotting으로 조사하였다. 5가지 allotypes, C6 A, C6 B, C6 B2, C6 Ml, C6 M11이 나타났고, 이들의 대립유전인자 빈도는 C6*A=0.4399, C6*B=0.5144, C6*B2=0.0392로 산출되어 이는 동아시아인과 비슷하였다.

  • 콩은 우유, 계란과 더불어 우리나라에서 과민성 알레르기를 일으키는 대표적인 식품중의 하나로 손꼽힌다. 이에 본 연구에서는 콩에 알레르기 반응을 일으키는 환아의 혈청과 콩 단백질을 토끼에 면역하여 얻은 다클론 항체를 이용하여 immunoblotting 및 ELISA 방법을 통하여 가열 처리한 콩과의 반응성을 조사 함으로서 가열처리가 콩의 항원성 내지는 알레르기성에 미치는 영향을 조사하였다. 물중탕으로 1 시간까지 가열처리 후 SDS-PAGE에 의해 분리된 콩 단백질은 열처리에 따른 콩 단백질 band의 변화를 관찰할 수 없었다. 또한 다클론 항체와의 반응성 확인을 위한 immunoblot 방법에서도 열처리에 따른 반응성의 차이는 나타나지 않았으며 콩 알레르기 환자의 혈청 역시 가열 처리에 따른 콩과의 반응성의 차이가 ELISA법을 이용한 측정에서 관찰되지 않았다. 이상의 결과로 부터 콩 알레르겐의 항원성 및 알레르기성은 가열처리에 의해 변화되지 않음을 확인할 수 있었다.

Assessment of the Potential Allergenicity of Genetically Modified Soybeans and Soy-based Products

  • A comprehensive safety evaluation was conducted to assess the potential allergenicity of newly introduced proteins in genetically modified (GM) crops. We assessed the allergenicity of CP4 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) in GM soybeans. This assessment was performed by IgE immunoblotting with soy-allergic children's sera, amino acid sequence homology with known allergens, and the digestibility of CP4 EPSPS. No differences in IgE-antigen binding by immunoblotting were found between GM soy samples and the corresponding non-GM samples. Based on the comparison of EPSPS amino acid sequence homology with current allergen databases, no known allergen was found. In addition, CP4 EPSPS protein was rapidly digested by simulated gastric fluid (SGF). Taken together, these results indicate that GM soybeans have no allergenicity in children and are as safe as conventional soybeans.

Chronic Exposure of Nicotine Modulates the Expressions of the Cerebellar Glial Glutamate Transporters in Rats

  • Rats were given nicotine (25 ppm) in their drinking water at the start of their mating period in order to study the expressions of glutamate transporter subtypes in cerebellar astrocytes following the chronic exposure of nicotine after mating. After the offspring were delivered, each group was divided into two subgroups. One group, the control group, was given distilled water only and the other group, the experimental group, was given distilled water containing nicotine. The cerebellar astrocytes were prepared from 7 day-old pups at each group. Ten days after the cells were cultured, the expression of the glutamate transporter subtypes (GLAST and GLT-1) was determined using immunochemistry and immunoblotting. During the continuous treatments, the developmental expression patterns of the GLAST and GLT-1 in the cerebellum were also determined from 2, 4 and 8 week-old rats. The expression levels of GLAST in cultured astrocytes of both the pre- or post-natally exposed groups were higher than those of the control group. However, these expression levels of the continuously exposed group were lower than those of the control group. Compared to those of the control group, the GLT-1 expression levels of all the nicotine-treated groups were higher, particularly in the continuously treated group.. According to the results from the immochemistry procedure, the cerebellar GLAST and GLT-1 expression levels of all nicotine-treated groups were lower than those of the control group at each age. However, the immunoblotting procedure showed that the cerebellar GLT-1 expression levels of all the nicotine-treated groups were higher than those of the control group, except for the rats that were continuously exposed for 8 weeks using immunoblotting. These results suggest that the expression of the glial GLAST and GLT-1 are altered differently depending on the initial exposure time and the partcicular period of nicotine exposure. In addition, nicotine exposure during gestation has persistent effects on glial cells.

The Effect of L-Ornithine on the Phosphorylation of mTORC1 Downstream Targets in Rat Liver

  • A non-protein amino acid, L-ornithine (Orn), has been shown to stimulate the urea cycle and tissue protein synthesis in the liver. The purpose of the current study was to assess whether Orn affects the mammalian target of rapamycin (mTOR) complex 1 (mTORC1) pathway, which is involved in protein synthesis. Primary cultured cells isolated from Wistar rat liver were incubated in an amino acid-free medium, followed by addition of Orn for 3 h. The cell lysate was subjected to immunoblotting to evaluate the phosphorylation of downstream targets of mTORC1, including p70S6K, S6, and 4EBP1. To assess the involvement of mTORC1 for the effect of Orn, the cells were pretreated with the mTOR inhibitor rapamycin before the addition of Orn and the cell lysate was subjected to immunoblotting. We next examined whether the effects of Orn were exerted in vivo. Orn was orally administered to 18 h food-deprived rats, the blood and the livers were collected at 1 and 3 h after administration for immunoblotting. Orn treatment for primary cultured cells for 3 h enhanced the phosphorylation of p70S6K, S6, and 4EBP1. In addition, rapamycin blocked the effects of Orn completely (p70S6K and S6) or partially (4EBP1). The oral administration of Orn to the rat also augmented the phosphorylation of mTORC1 downstream targets notably in S6 at 1 h. Our findings demonstrate that Orn has the potential to induce the phosphorylation of downstream targets of mTORC1 in the rat liver. This may be mediated by the augmentation of mTORC1 activity.

치근단 병소가 있는 환자에서 Porphyromonas endodontalis 항원에 대한 혈청 특이 항체의 면역 반응 연구 (IMMUNE REACTION OF SPECIFIC SERUM ANTIBODIES TO PORPHYROMONAS ENDODONTALIS ANTIGEN IN PATIENTS WITH PERIAPICAL LESION)

  • Porphyromonas endodontalis is a black-pigmented anaerobic Gram-negative rod which is associated with endodontal infections and this microorganism possesses a potential for pathogenicity. The purpose of this study was to compare the membrane components of Porphyromonas endodontalis and Porphyromonas gingivalis and to study the immune reaction patterns of Porphyromonas endodontalis with patients with periapical lesion. Porphyromonas endodontalis (ATCC 35406), Porphyromonas gingivals serotypea (381), serotype b(W50), serotype c(A7A1-28) were cultured in anaerobic condition. Rabbit antisera were prepared by intravenous injection of formalized whole cells and human sera were obtained from patients and dental students. Indirect immunofluorescence method was used to study on the cross reaction between Porphyromonas endodontalis and Porphyromonas gingivalis serotype a, b, c antigen. Total membrane protein profiles of Porphyromonas endodontalis antigen were studied by sodium dodecyl sulfate polyacrylamide gel electrophoresis and the reactivity of antigenic components of Porphyromonas endodontalis against sera of patients and rabbit anti-Porphyromonas endodontalis antisera were assessed by Immunoblotting method. The following results were obtained : 1. Antigens of Porphyromonas endodontalis has multiple antigenic components, and both patients with periapical lesion and normal healthy individual showed immune response to this. 2. Patients group and healthy individual group showed a diversity of immune reaction pattern but they showed immune response against 43kd protein. 3. Patients with periapical lesion showed more diverse immune response than healthy individual and in some patients, much more bands appeared to lower molecular weight protein. 4. According to indirect immunofluorescence and Immunoblotting study, Porphyromonas endodontalis did not share common antigen with Porphyromonas gingivalis serotype a, b, c.

물리적 처리에 의한 강력분 밀가루 Gliadin의 항원성 변화 (Changes in Allergenicity of Gliadin in Strong Wheat Flour by Physical Treatments)

  • 본 연구에서는 가압가열 및 microwave 처리가 gliadin의 항원성에 미치는 영향을 살펴보기 위해 강력분에 가압가열과 microwave를 단독 또는 병행으로 처리하여 Ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였다. 가압가열 처리의 경우 시간이 길어질수록 IgG와의 결합력이 감소하였으며, 특히 50분 처리구에서 약 87%로 가장 낮은 결합력을 보였다. 또한 SDS-PAGE와 immunoblotting 결과에서도 무처리구에서 강하게 보였던 gliadin band가 가압가열 처리에 의해 거의 소실되고 항체와 반응하지 않았다. 가압가열 및 microwave를 병행 처리 시도 마찬가지로 gliadin의 결합력이 감소하였으며, 처리구 중 가압가열 50분, microwave 5분 처리구에서 약 93%로 가장 낮은 결합력을 보였다. 반면 microwave를 단독으로 처리한 경우에는 일부 단백질의 변화는 관찰되었으나, 항원성 감소에는 효과가 없음을 확인하여 단백질 변화가 항원성에는 큰 효과를 준 것 같지 않다. 이상의 결과를 통해 가압가열 단독 처리 및 가압가열 및 microwave의 병행처리 시 gliadin의 항원성이 감소함을 확인하였다.

요로 감염환자에서 혈청학적 방법을 이용한 P-pili특이혈중 항체의 조사 (Serological Studies on the Specific Antibodies Against P-pili of Uropathogenic Escherichia coli)

  • 비뇨기계 병원성 대장균의 중요한 병원성 인자 중의 하나로 인정되고 있는p-fimbriae의 subtype의 분포를 확인하기 위하여 요로감염증으로 확진된 환자의 혈청을 이용하여 immunoblotting 을 실시하였고, 이와 동시에 효소면역 측정법을 실시하여 p-fimbriae특이 항체 보유를 확인하였다. Immunoblotting 결과 우리나라 요로감염증환자에서 높은 빈도로 확인되는 p-fimbriae subtype의 분포는 $F7_1$34(56.7%), $F7_2$28(46.7%), F13 30(50%)등이 높게 나타났으며, 이와 같은 결과는 효소면역측정법에서도 동일하게 나타났다. 그러나 P-pili를 순수분리하지 않고 whole cell을 이용한 효소면역 측정법은 교차반응 때문에 비뇨기 감염증의 혈청학적인 진단에 적합하지 않는 것으로 나타났다. 또 우리 나라의 요로감염 환자에서 항체 양성율이 높은 $F7_1$, $F7_2$, F13만을 혼합하여 항원으로 이용한 효소면역측정법의 특이도와 민감도가 각각 92.6%, 90%로 나타나, 이와 같은 방법을 임상진단에 응용할 수 있을 것으로 판단되었다.

열성질환 환자에서 라임병균 Borrelia burgdorferi 감염증 진단을 위한 혈청학적인 분석 (Analysis of Antibodies Against Lyme Disease Agent, Borrelia burgdorferi, in Sera from Patients with Unknown Fever)

  • 현재 라임병을 진단을 위해 국내에서 이루어지고 있는 연구 방법은 원인균인 B. burgdoferi 항원에 대한 항체를 검출하는 혈청학적인 방법이다. 하지만 대부분의 경우 항원으로 사용되는 균주가 다른 나라에서 분양받은 균주이므로 이때 얻어진 결과를 유추 해석하는데 어려움이 수반되고 있다. 본 실험에서는 국내 분리균주에 대한 항체를 검출하는 조건을 확립하고, 이를 토대로 하여 불명열 환자의 항체가 항원에 반응하는 양상을 파악함으로써 국내 분리균주를 이용한 혈청학적인 분석방법으로 국내 라임병 진단에 보다 적합한 조건을 제시하고자 하였다. 본 실험 결과 병원에 의뢰된 불명열 환자의 혈청이 B. burgdoferi sensu lato 중 3가지 균종의 항원에 대해 평균 약 8% 정도의 양성 반응을 보이는 것으로 미루어 볼 때 이 들 중 B. burgdorferi에 의한 열성 질환 감염증이 존재할 것으로 사료된다. 특히 국내 분리균주 중 가장 많이 분리된 바 있는 B. afzelii에 대한 양성반응이 14.3%나 나타난 것으로 미루어 아직 밝혀지지 않은 국내 발생 라임병의 원인균은 주로 B. afzelii일 가능성이 가장 크다고 할 수 있을 것이다. 또한 효소결합면역 측정법 (ELISA)을 이용하여 양성으로 판별된 혈청에 대해 immunoblotting법을 실시한 결과 주로 41kDa (ftagellin), 27kDa, 31kDa (OspA), 34kDa (OspB)에서 band가 나타났다. 본 실험의 결과 국내 열성질환 환자들 중 라임병균에 의한 감염증이 있을 것으로 사료되므로 국내 분리균주를 항원으로 사용한 효소결합면역 측정법 (ELISA), immunoblotting법과 같은 혈청 학적 진단이 이루어져야 한다고 사료된다.
