인삼약침(人蔘藥鍼)이 glucocorticoid투여 Mouse의 면역반응에 미치는 영향 (Effects of aqua-acupuncture with Panax Ginseng on immune response induced by Glucocorticoid in mouse)

  • In order to investigate the effect of aqua-acupuncture solution with Panax Ginseng into $Qihai(CV_6)$ and $Shenshu(BL_{23})$ on immune response induced by glucocorticoid in mouse, Panax Ginseng aqua-acupuncture solution was injected into $Qihai(CV_6)$ and $Shenshu(BL_{23})$ for seven days after injection with glucocorticoid. And then MA-HRP (methamphetamine-horseradish peroxidase) induced antibody production, numbers and lysozyme activity in macrophage were measured. The results were as follows: 1. The antibody production in mouse immunized with MA-HRP was decreased in control group as compared with normal group. Although $Qihai(CV_6)$ group showed slight increasement, $Shenshu(BL_{23})$ group indicated great increasement compared with normal group. However in Aa-BL group, antibody production was almost increased to normal group. 2. In control group, the numbers of macrophage were decreased about 14% as compared with normal group. And in the pretreated groups of $Qihai(CV_6)$ and $Shenshu(BL_{23})$ were respectively increased 3.0 times and 2.9 times as compared with normal group. 3. Effect of Panax Ginseng-aqua acupuncture solution on the lysozyme activity in macrophage was increased gradually in the pretreated groups of $Qihai(CV_6)$ and $Shenshu(BL_{23})$ as compared with control group. These results suggest that Panax Ginseng aqua-acupuncture at $Qihai(CV_6)$ and $Shenshu(BL_{23})$ may increase antibody production and lysozyme activity in macrophage that is suppressed by glucocorticoid, and Panax Ginseng aqua-acupuncture will have immuno adjuvant effects on the cells which concerned with immunomechanisms.

Development of Rapid Diagnostic Kit for Identification of Hanwoo (Korean Native Cattle) Brand Meat by Detecting BIO-TAG

  • This study was performed to develop a rapid immuno-assay kit, by using a specific antigen to detect Hanwoo brand meat. We selected a synthetic antigen specific to our target antibody, named BIO-TAG (Tyr-D-Ala-Phe), by utilizing a computer-based analysis and literature review. BIO-TAG tagged with adjuvant was subcutaneously injected in sheep and Hanwoo. The serum and meat juice of the immunized or non-immunized animal were then analyzed, to measure the titer of antibody by ELISA and Western blot. The amount of antibodies against the BIO-TAG increased (p<0.05) in serum by vaccination. Furthermore, meat juice from the immunized Hanwoo showed greater (p<0.05) antibody titer, compared with those from non-immunized groups. To optimze the dilution factor, we performed dot-ELISA, with various combination levels of BIO-TAG. Results from dot-ELISA showed that 2 mg/mL BIO-TAG was sufficient to distinguish the immunized meat from non-immunized groups. These results support our hypothesis that simple immunization of Hanwoo generates a sufficient amount of antibodies to be detectable in the meat juice by means of the immune-assay. Therefore, specific Hanwoo brand meat can be more precisely identified by our rapid diagnostic kit. This technology can deter possible fraud of counterfeit meat brands in the Korean domestic market with ease and rapidity; and offers a new tool that guarantees consumers high quality Hanwoo brand beef.

한국산 겨우살이 유산균 발효 추출물의 독성 및 면역증강 효과 (In vivo Toxicity and Immunoadjuvant Activity of Korean Mistletoe (Viscum album coloratum) Extract Fermented with Lactobacillus)

  • Lactobacillus casei KCTC 2180 및 Lact. plantarum KCTC 3103으로 발효된 한국산 겨우살이 추출물인 FKM-110의 렉틴 성분의 함량 변화, 독성 및 면역증강 효과를 검토하여 다음과 같은 결과를 얻었다. 두 가지 유산균을 이용하여 KM-110을 1-3일간 발효시킨 결과, 렉틴 성분은 발효 전에 비하여 53-71% 정도의 함량을 나타냄으로서 감소되는 경향을 보였다. 마우스에 대한 시료의 독성시험을 위하여 각 시료를 피하 주사하고 $LD_{50}$ 값을 측정한 결과, KM-110은 50-100 mg/kg인 반면 유산균으로 발효된 겨우살이 추출물인 FKM-110은 150-200 mg/kg인 결과를 보임으로 발효에 의하여 in vivo 독성효과는 감소하는 경향을 보였다. 각 시료를 macrphage에 직접 자극하여 염증성 cytokine인 IL-$1{\beta}$ 및 TNF-${\alpha}$의 생산능력을 조사한 결과, FKM-110이 KM-110에 비하여 낮은 cytokine의 생산양식을 나타냈다. 그러나 FKM-110의 항원 KLH에 대한 면역증가 효과를 항체 생산능으로 조사한 결과, KM-110과 차이 없이 항원 단독 투여군에 비하여 높은 역가의 항체가 생산되었다. 따라서 겨우살이 추출물을 유산균으로 발효시킨 결과, in vivo 독성은 감소하였으나 면역증강 활성은 KM-110과 동일하게 유지되는 결과를 얻었다. 따라서 겨우살이의 면역증강 활성에 관여하는 성분으로 렉틴 이외에 물질이 관여할 수 있음을 강하게 시사하였다.
