Electron Tomography and Synapse Study

  • Electron tomography (ET) is a useful tool to investigate three-dimensional details based on virtual slices of relative thick specimen, and it requires complicated procedures consisted of image acquisition steps and image processing steps with computer program. Although the complicated step, this technique allows us to overcome some limitations of conventional transmission electron microscopy: (1) overlapping of information in the ultrathin section covering from 30 nm to 90 nm when we observe very small structures, (2) fragmentation of the information when we study larger structures over 100 nm. There are remarkable biological findings with ET, especially in the field of neuroscience, although it is not popular yet. Understanding of behavior of synaptic vesicle, active zone, pooling and fusion in the presynaptic terminal have been enhanced thanks to ET. Some sophisticated models of postsynaptic density with ET and immune labeling are introduced recently. In this review, we introduce principles, practical steps of ET and some recent researches in synapse biology.

Vero-E6 세포에서 한탄바이러스의 증식에 관한 전자현미경적 연구 (Electron Microscopic Study on the Replcation of Hantaan Virus in Vero-E6 Cells)

  • Results of the studies on the morphologic and molecular biologic characteristics of Hantaan virus (HTNV), one of the etiologic agents of Hemorrhagic fever with renal syndrome (HFRS), revealed that HTNV was a member of Family Bunyaviridae and its RNA divided into three segments. And the nucleotide sequences of these segments also were known and the differences in nucleotide sequences of HTNV from other members of genus Hantavirus were clearly evaluated. But the morphorgenesis, pathogenesis of HFRS and the replication time had not been clearly determined. In this study, to estimate the replication time of HTNV in Vero E-6 cells, Vero cells were infected with HTNV 76/118 strain, and cells were harvested from two hours post-infection up to 24 hours at two hours-intervals. Harvested cells were treated with ordinary techniques for electron microscopy and immune-electron microscopy. And then thin sections were observed under transmission electron microscope. HTNV particles were not found in the cytoplasm and in the extracellular space between $2{\sim}8$ hours after inoculation of virus, but virus particles were observed in extracellular space near the cell membrane of Vero-E6 cells 10 hours after infection. In immune electron microscopy, mature HTNV particles in extracellular spaces and immature virus labelled with gold particles in the cytoplasm of Vero E-6 cell 10 hours after infection of HTNV could be seen. This results suggest that the replication time of HTNV might be about 10 hours.

Localization of cytoskeletal proteins in Pneumocystis carinii by immuno-electron microscopy

  • Pneumocystis carinii causes serious pulmonary infection in immuno-suppressed patients. This study was undertaken to observe the cytoskeletal proteins of P. carinii by immune-electron microscopy. P. carinii infection was experimentally induced by immunosuppression of Sprague-Dawley rats for seven weeks, and their lungs were used for the observations of this study. The gold particles localized actin, tropomyosin, and tubulin. The actin was irregularly scattered in the cytoplasm of the trophic forms but was much more concentrated in the inner space of the cell wall of the cystic forms called the inner electron-lucent layer No significant amount of tropomyosin was observed in either trophic forms or cystic forms. The tubulin was distributed along the peripheral cytoplasm and filopodia of both the trophic and cystic forms rather than in the inner side of the cytoplasm. Particularly, in the cystic forms, the amount of tubulin was increased and located mainly in the inner electron-lucent layer of the cell wall where the actin was concentrated as well. The results of this study showed that the cell wall of P carinii cystic forms is a structure whose inner side is rich in actin and tubulin. The location of the actin and tubulin in P. carinii suggests that the main role of these proteins is an involvement in the protection of cystic forms from the outside environment by maintaining rigidity of the cystic forms.

Ricin의 독성이 생쥐의 간. 비장, 흉성, 폐 및 심장에 미치는 영향에 관한 조직학적 연구 (Histological effects of ricin on liver, spleen, thymus, lung and heart of mouse)

  • The pathological aspects of purified ricin from the seeds of the castor oil plant, Ricinus communis, were examined, using light and transmission electron microscopy. ICR mice were exposed to ricin by peritoneal injection with 100 ng/1 $m\ell$ PBS(pH 7.0) mouse and histological observations on the liver, spleen, thymus, lung and heart were carried out at intervals up to 48 h after exposures. All the organs examined were damaged by ricin. Among the organs, the spleen and thymus; immune organs were the most sensitive to ricin, whereas the effect delayed in the liver, lung and heart. Furthermore, the immune cells in each organ were the most sensitive to ricin. Accordingly, the effect of ricin on the organs seems to be affected by the immune cells existed in each organ, In each organ, the immune cells showed apoptotic changes, while the capillary endothelial and parenchymal cells showed necrotic changes.

Pathology of C3 Glomerulopathy

  • C3 glomerulopathy is a renal disorder involving dysregulation of alternative pathway complement activation. In most instances, a membranoproliferative pattern of glomerular injury with a prevalence of C3 deposition is observed by immunofluorescence microscopy. Dense deposit disease (DDD) and C3 glomerulonephritis (C3GN) are subclasses of C3 glomerulopathy that are distinguishable by electron microscopy. Highly electron-dense transformation of glomerular basement membrane is characteristic of DDD. C3GN should be differentiated from post-infectious glomerulonephritis and other immune complex-mediated glomerulonephritides showing C3 deposits.

이질(痢疾)아메바(Entamoeba histolytica)의 면역혈청(免疫血淸) 내(內)에서의 미세구조(微細構造) 변화(變化) (Ultrastructural Changes of Entamoeba histolytica in the Immune Serum)

  • Ultrastructural changes of Entamoeba histolytica, a concomitant strain YS-9, which was treated in the immune serum was examined. The amoebae in the serum became immobilized state from about 30 minutes of the treatment and recovered at about 60-90 minutes. In the cells of control group, helix structures were scattered throughout the cytoplasm. The particles comprising the helix structure averaged 20 nm in diameter. At the beginning stage of the immobilization, helical aggregates(chromatoid body) which associated with vacuoles appeared abundantly in the cytoplasm, but gradually tended to aggregate along peripheral region of the cell, specially in intactly immobilized state. Each parallel array of aggregates measured about 45 nm in width. When the cells became remobilize, pseudopodia appeared again, but helical aggregates disappeared and numerous helix structures were observed in the cell periphery. Distribution of glycogen particles showed no change, and acid phosphatase activities were seen in both the immobilized and the control group. The reaction was markedly noticed in the vacuoles.

STUDIES ON THE VIRUSES OF RADISH MOSAIC

  • A mosaic diseased radish collected from the suburb of Seoul, in November, 1961 was used for studing the host range, physical properteis, purificaitiion, insect transmission, and electron microscopy. A Japanese strain of radish mosaic(RPV) was also used with Korean strain of radish mosaic (KRMV) for a comparative study. The two viruses, KRMA and RPV, were identified by the difference in host range, insect transmission and electron microscopy. The KRMA was severely infective on tobacco and Nicotiana glutinosa, while on Gomphrena globsa was immune to the virus. RPV produces necrotic local lesions on Gomphrena globosa but did not infect tobacco and N. glutinosa. Among varieties of radish, Seoul, Akamaroo, Akanagea, Koong-Joong showed more severe symptoms than Simoo, Minong, Paek-soo, which appeared to be fainly resistant. In a number of tests, it was found that the virus KRMA retained its infectivity until to a dilution of 1:2,000, heating at $58^{\circ}$ for 10 minutes, adn aging in vitro for 7 days at room temperature. The RPV was not inactivated until it was diluted to 1:2,000, heated to $56^{\circ}$, and aged for 6 days. The KRMV was readily transmitted by the aphid(Myzus persicae Sulz). The virus RPV was not transmitted by the aphid in a number of tests. Partialy purified viruses using ammonium acetate buffer, salting-out by ammonium sulfate and centrifugation of high and low speed were highly infective. Electron micrographs showed that the KRMV paticles are of spherical particles whereas the RPV particles are rod-shaped.
