As a component for a recirculating aquaculture system, a new strain of denitrifying bacterium was isolated from municipal sewage. The isolate was motile by means of one polar flagellum, catalase-positive, and a Gram-negative rod-shaped cell measuring $0.5-0.6{\mu}m$ in width and $1.3-1.9{\mu}m$ in length. The isolate was identified as Pseudomonas fluorescens and produced dinitrogen gas via the reduction of nitrate. The optimal growth conditions (pH, temperature, carbon source, and C/N ratio) of the isolate were found to be 6.8, $30^{\circ}C$, malate, and 3, respectively. Under optimal growth conditions of P. fluorescens, dinitrogen gas was first detected in the exponential growth phase, then a small amount of nitrite was developed and converted to dinitrogen gas in the stationary phase. Pseudomonas fluorescens cells were immobilized in modified polyvinyl alcohol (PVA) gel beads, and the maximum denitrification rate was measured as $36.6 {\mu}lN_2h^-1$ per bead with an optimum cell loading of $20mg {\mu}l^-1$ and $2\%$ sodium alginate added to the PVA gel. The operating stability of the modified PVA gel beads remained unchanged for up to 43 repeated batches.
Aspergillus terreus NRRL 1960 was immobilized on various alginate gel beads, Celites, and polyurethane foam cubes, and the comparisons were made for the production of itaconic acid according to the types and sizes of each carrier. The levels of itaconic acid produced from Ca- alginate and Sr-alginate were similar, and the addition of bentonite to Ca- and Sr-alginate resulted in an increase of itaconic acid. The addition of 1.67% bentonite and 0.33% starch to Sr-alginate (SABS bead) produced higher level of itaconic acid (11.59 g/1) than other gel beads. A decrease in the size of Celite increased the itaconic acid production, and the smallest size of Celites, R- 634, produced 6.37 g/l of itaconic acid. Among various types of polyurethane foam cubes, HR 08 (2X2X2 cm) produced about 19 g/l of itaconic acid, which was more efficient than other carriers. In a repeated batch culture using immobilized cells on polyurethane foam cubes (HR 08, 2X2X2 cm), the stability of itaconic acid production was maintained up to 4 batches. Also, the possibility of itaconic acid production by continuous culture was shown in a packed-bed reactor.
생물고분자 생산을 위하여 오니에서 slime을 생산하는 Zoogloea ramigera 115를 선택하였다. 추출한 생물고분자는 카드뮴, 아연 같은 금속에 대해서 높은 흡착성능을 나타냈다. 특히 발효조 broth는 높은 흡착성능을 나타냄으로써 부가적인 분리공정없이 생물흡착제로서 사용 가능할 것이다. 생물고분자는 calcium alginate bead 형태로 고정화되어 유가금속 회수를 위하여 충전층 반응기에 사용되었다. 생물흡착제는 구리, 카드뮴, 망간, 아연에 대하여 80% 또는 그이상의 높은 제거율을 나타냈으며 흡착-탈착-재흡착 실험에서 높은 안정성을 나타냈다. 고정화된 생물고분자 시스템은 이온교환수지와 같은 다른 중금속 제거 시스템과 경쟁력 및 잠재적인 산업상 응용 가능성을 보였다.
For the selection of peptide specifically binding to $Pb^{2+}$, the biopanning with the commercially available Ph.D.-7 phage displayed heptapeptide library was carried out against $Pb^{2+}$ immobilized on a metal-chelating IDA (iminodiacetic acid) resin. After four rounds of screening against $Pb^{2+}$-IDA including negative selections against charged bead with metal ions other than $Pb^{2+}$ and uncharged bead, several candidate lead-binding phage peptides were initially determined based on the order of frequency from the screened phage clones. Of the selected phage peptide sequences, the peptide of the highest frequency, CysSerIleArgThrLeuHisGlnCys (CSIRTLHQC) also exhibited the strongest affinity for $Pb^{2+}$ in binding assays for individual phage clones. However, there was not a significant difference in $Pb^{2+}$ affinity between selected peptides when using synthetic heptapeptides corresponding to the displayed peptide sequences of phage clones.

We designed an effective screening method for double strand DNA (dsDNA) binders using DNA-modified magnetic particles. Hairpin DNA was immobilized on the surface of magnetic particle for a simple screening of dsDNA binding materials in a solution containing various compounds. Through several magnetic separation and incubation processes, four DNA-binding materials, DAPI, 9AA, AQ2A, and DNR, were successfully screened from among five candidates. Efficiency of screening was demonstrated by HPLC analysis using a C2/18 reverse-phase column. In addition, their relative binding strengths were verified by measuring the melting temperature ($T_m$). If hairpin DNA sequence is modified for other uses, this magnetic bead-based approach can be applied as a high-throughput screening method for various functional materials such as anti-cancer drugs.

The objective of this study was to investigate microbial removal using layered double hydroxides (LDHs) and iron (hydr)oxides (IHs) immobilized onto granular media. Column experiments were performed using calcium alginate beads (CA beads), LDHs entrapped in CA beads (LDH beads), quartz sand (QS), iron hydroxide-coated sand (IHCS) and hematite-coated sand (HCS). Microbial breakthrough curves were obtained by monitoring the effluent, with the percentage of microbial removal and collector efficiency then quantified from these curves. The results showed that the LDH beads were ineffective for the removal of the negatively-charged microbes (27.7% at 1 mM solution), even though the positively-charged LDHs were contained on the beads. The above could be related to the immobilization method, where LDH powders were immobilized inside CA beads with nano-sized pores (about 10 nm); therefore, micro-sized microbes (E. coli = 1.21 ${\mu}m$) could not diffuse through the pores to come into contact with the LDHs in the beads, but adhere only to the exterior surface of the beads via polymeric interaction. IHCS was the most effective in the microbial removal (86.0% at 1 mM solution), which could be attributed to the iron hydroxide coated onto the exterior surface of QS had a positive surface charge and, therefore, effectively attracted the negatively-charged microbes via electrostatic interactions. Meanwhile, HCS was far less effective (35.6% at 1 mM solution) than IHCS because the hematite coated onto the external surface of QS is a crystallized iron oxide with a negative surface charge. This study has helped to improve our knowledge on the potential application of functional granular media for microbial removal.
양어장 순환수 속의 암모니아성 질소의 제거를 위한 Ba-alginate 와 Ca-alginate 반응기는 0.6시간의 수력학적 체류시간에서 51.0, 52.6 g $NH_3-N/m^3/day$의 거의 유사한 암모니아성 집소의 제거속도릎 나타내었다. Ca-alginate를 이용한 합성양어장수 속의 암모니아성 짖소의 제거 실험에서 본 반응기의 암모니아성 짐소의 제거속도는 수력학적 체류시간이 0.3시간인 지점에서 82.0 g $NH_3-N/m^3/day$로 서 최고의 재거속도릎 나타냈으며 이때 암모니아 제거율은 48% 이었다. 수중의 암모니아의 농도가 2mg/L 정도의 범위에서는 반응기 내에 주입되는 공가량을 0.1 vvm 으로 공급하더라도 용존산소 농도를 7.0 - 5.6 g/m3로 유지 할 수 있었으므로 질화에 필요한 용존산소량을 충분히 유지할 수가 있는 것으로 나타났으며 pH 는 7.2 - 7.9의 범위플 유지할 수가 있어서 pH 변화에 따른 위 해 요소는 없는 것으로 나타났다.
돼지감자 분말을 원료로 alginate에 고정화된 Kluyveromyces marxianus FO43의 에탄올 생산성을 향상시키기 위한 연구를 수행하였다. 15% 돼지감자 배지에서 발효시킨 고정화 효모에 의한 에탄올 농도와 이론치에 대한 에탄올 수율은 4일 후에 각각 3.38%(w/v), 54.20%로 이것은 고정화하지 않은 효모의 3.76%(w/v), 71.13% 보다 낮았다. Cellulase의 첨가는 $15{\sim}20%$ 돼지감자 배지의 점조성을 크게 낮추어 고정화 효모의 에탄올 생산성을 증가시켰다. 그리하여 20% 배지에서도 효소를 처리하여 고정화 효모를 4일 발효 시키면 에탄올 농도를 5.57%(w/v), 이론치에 대한 에탄올 수율을 68.86%까지 얻을 수 있었다. Repeated batch culture를 실시한 결과 bead의 활성이 22일 동안 저하되지 않고 유지되었다.
본 연구에서는 미세조류의 고밀도 배양을 위한 경제적인 공정을 개발하는 차원에서 3종의 미세조류 즉, Chlorella vulgaris, Dunaliella salina 및 Porphyridium purpureum를 Ca-alginate 로 microencapsulation 한 후 air-bubble column형 배양조에서 고밀도 배양을 시도하였다. $2\%\;CaCl_2$ 농도로 capsule을 제조할 때 capsule의 안정성이 가장 좋았으며, capsule 막의 두께 및 $CO_2$ 공급유무에 따른 미세조류 (Chlorella vulgaris)의 생산성은 거의 차이가 없었다. 한편, 3종의 미세조류 모두 capsule로 고정화시킨 것이 bead로 고정화한 미세조류나 free cell 보다 농도가 높았으며, 특히Dunaliella salina가 microencapsulation에 의한 고밀도 배양 효과가 가장 뛰어나 free cell 보다 약 5배 정도의 생산성을 나타내었다.
다공성 실리카표변에 폴리에틸렌이민과 글루타르 알데히드로 전화당 효소를 고정화시키고 자당과 에 탄올로부터 메틸 프룩토시드를 합성한 결과 다음과 같은 결론을 얻었다. 다공성 설리카를 폴리에틸렌이 민으로 표면처리하여 얻어진 고정화 담체는 그 다공 성 구조와 표면화학적 특성이 효소의 고정화에 적합 하여 전화당 효소에 대한 고정화 함량 120mg/g, 활 성도 lOOD/mg을 얻어 배당체의 합성반응에서 필요 로 하는 고정화 촉매를 제조할 수 었었으며 고정화 담체 표변에 형성된 폴리에틸렌이민 피막은 효소 부근에 친수성 분위기를 유지하여 높은 효소활성을 나 타내었고 에틸 프룩토시드가 효소에 접근하여 가수 분해되는 반응을 억제하여 메틸 프룩토시드의 수율 과 농도를 크게 증가시켰다. 자당과 메탄올 수용액 으로부터 메틸 프룩토시드를 합성한 결과, 자당 농도 O.291mol/l , 메탄올 농도 30%(v/v), 반응온도 $25^{\circ}C$, pH 4.8, 효소활성도 2U/ml일 때 자당의 전화 율 91.2%, 에틸 프룩토사드 농도 27.0g/l , 그 평균 수율 55.9%을 얻었으며, 위의 연구결과로부터 고정화 효소에 의한 배당체의 합성반응이 연속생산공정 으로 연구개발 될 수 있을 것으로 기대된다.
