Stabilization of Bioluminescence of Immobilized Photobacterium phosphoreum and Monitoring of Environmental Pollutants

  • Stability of bioluminescence was investigated with Photobacterium phosphoreum immobilized on the strontium alginate in order to develope continuous real time monitoring of pollutants. The stability of bioluminescence emission was improved by prolonged aging time. The aging time of ${\geq}40$ min and the cell concentration of ${\leq}0.6\;of\;OD_660$ were selected for the immobilization of P. phosphoreum to give linearity between cell concentrations and bioluminescence intensity. In sensitivity tests using phenol, it was found that this compound quenched bioluminescence proportional to the concentration without lowering of cell growth. The lower value for maximum quenching ($q_s$) and higher dissociation constant ($K_s$) were observed with strontium-alginate immobilized cells compared to free cells. The response of bioluminescence to toxicants was evaluated with the immobilized luminescent bacteria. The sensitivity of the immobilized cells was found to be good in response to toxicants, 4-nitrophenol, salicylate and cadmium, when evaluated with a specific rate of bioluminescence quenching.

Photosynthetic Characteristics of Polyvinylalcohol-Immobilized Spinach Chloroplasts (Polyvinylalcohol에 고정화한 시금치 엽록체의 광합성특성에 대한 연구)

  • Photoxynthetic properties of polyvinylalcohol (PVA)-immobilized chloroplast especially regarded to stability of photosynthetic electron transport and the fluorescence induction pattern were studied. When isolated spinach chloroplasts were immobilized with PVA, it showed good preservation of photosynthetic electron transport activity, especially PS II activity, during storage at -15$^{\circ}C$, 4$^{\circ}C$ and 2$0^{\circ}C$. And immobilized chloroplasts revealed similar thermostability of whole chain electron transport to free chloroplsts. And the absorption peak of red band of chloroplasts showed the blue-shift of 2-4 nm after immobilization. Fv/Fm ratio of chlorophyll fluorescence slightly decreased after immobilization. White light pulse after continuous light do not induced the additional fluorescence rise. This means chlorophyll fluorescence at room temperature reached to Fmax under continuous light in the immobilized chloroplasts. It seems that PVA may be a good candidate for immobilization matrix for the preservation of photosynthetic function of thylakoids and for the continuous use of chloroplast membranes of higher plants for solar energy storage and conversion.

Preparation and Characteristics of Immobilized Sludge by the PAA Entrapment Method (PAA 포괄법에 의한 고정화 슬러지의 제조 및 특성에 관한 연구)

  • This study was conducted to evaluate the feasibility and characteristics of poly acrylamide (PAA) immobilized sludge as a microbial entrapment bead for wastewater treatment. In the PAA method of immobilized sludge, it was found that the optimum acrylamide concentration for actual wastewater treatment was to be 12%. When the sequencing batch reactor (SBR) was operated during 30 days, removal efficiencies of TOC and phosphate was 95% and 70ft, respectively. From this research, repeated cycle of anaerobic and aerobic conditions is required to enhance the removal of TOC and phosphate. During the operation, immobilized cells could be used without being disrupted.

  • The biosorption performances of copper were investigated by the immobilized biomass of nonliving marine brown algae Undaria pinnatifida by each of the Ca-alginate method(Ca-ALG), Ba-alginate method(Ba-ALG), polyethylene glycol method(PEG), and carrageenan method (CARR). The copper removal performance increased but the copper uptake decreased as the biomass amount was increased. However, the copper uptake by the immobilized biomass increased with increasing initial copper concentration. Among the immobilization methods, the copper uptake decreased in the following sequence: Ca-ALG > Ba-ALG > PEG > CARR. The pattern of copper uptake by the immobilized biomass fitted the Langmuir isotherm better than the Freundlich isotherm. Desorption of deposited copper with 0.05 ~0.5M HCI, resulted in no changes of the copper uptake capacity of the immobilized biomass by the immobilization methods except for PEG, through five subsequent biosorption/desorption cycles. There was no damage to the immobilized biomass which retained its macroscopic appearance in repeated copper uptake/elution cycles.

Immobilization of Keratinase from Aspergillus flavus K-03 for Degradation of Feather Keratin

  • Extracellular keratinase isolated from Aspergillus flavus K-03 was immobilized on calcium alginate. The properties and reaction activities of free and immobilized keratinase with calcium alginate were characterized. The immobilized keratinase showed proteolytic activity against soluble azo-casein and azo-keratin, and insoluble feather keratin. Heat stability and pH tolerance of keratinase were greatly enhanced by immobilization. It also displayed a higher level of heat stability and an increased tolerance toward alkaline pHs compared with free keratinase. During the durability test at $40^{\circ}C$, 48% of the original enzyme activity of the immobilized keratinase was remained after 7 days of incubation. The immobilized keratinase exhibited better stability, thus increasing its potential for use in industrial application.

Immobilization Imparts Stability to Watermelon Urease to Work in Water Miscible Organic Media

  • The behaviour of alginate immobilized and soluble watermelon (Citrullus vulgaris) urease in water miscible organic solvents like, acetonitrile, dimethylformamide (DMF), ethanol, methanol, and propanol is described. The organic solvents exhibited a concentration dependent inhibitory effect on both the immobilized and the soluble urease in the presence of urea. Pretreatment of soluble enzyme preparations with organic solvents in the absence of substrate for 10 min at $30^{\circ}C$ led to rapid loss in the activity, while similar pretreatment of immobilized urease with 50% (v/v) of ethanol, propanol, and acetonitrile was ineffective. Time-dependent inactivation of immobilized urease, both in the presence and in the absence of urea, revealed stability for longer duration of time even at very high concentration of organic solvents. The soluble enzyme, on the other hand, was rapidly inactivated even at fairly lower concentrations. The results suggest that the immobilization of watermelon urease in calcium alginate make it suitable for its application in organic media. The observations are discussed.

Production of Laccase and Bioremediation of Pentachlorophenol by Wood-Degrading Fungus Trichophyton sp. LKY-7 immobilized in Ca-Alginate Beads

  • The lignin-degrading fungus Trichophyton sp. LKY-7 was immobilized in ca-alginate beads for laccase production and PCP remediation. The immobilized Trichopphyton sp. LKY-7 enabled the repeated use of this fungus for laccase production and produced high amount of laccase throughout 5 cycles incubation. As a laccase inducer oak wood meal(Quercus variabilis) seemed to be effective laccase inducer for Trichophyton sp. LKY-7, and the optimum addition amount was 1%(W/W) in glucose-peptone medium. Biotransformation of pentachlorophenol by immobilized Trichophyton sp. LKY-7 reached an efficency of up to 90% without toxic inhibition. Immobilized Trichophyton sp. LKY-7 might thus be applicable for semicontinuous laccase production and bioremediation to serve inoculum for reactor system.

A Study on the Design of Denitrification Reactor and the Characteristics (탈질화 반응기의 설계 및 특성에 관한 연구)

  • Removal of nitrogen compound from waste water is essential and often accomplished by biological process. Deni-trification bacterium. Paracoccus denitrificans(KCTC 2350) is employed to estimate the ability and the characteristics of denitrification. In the immobilized biological reactor system, the measurement of absolute amount of active strain in the reactor is comparatively difficult or impossible. In this study, strain immobilized denitrification reactor was designed with the unwoven texture wrapped peeped hole plastic tube to calculated the absolute amount of active strain by comparing the activity of the immobilized reactor adn the free cell reactor. The reactor system was continuous stirred tank reactor and the rate of substrate consumption was assumed to be Michaelis-Menten equation. As a result, we found that the amount of immobilized active strain was the half of the total active strain in the reactor and the time required to reach in the equilibrium state in the immobilized reactor system was shorter than that of the free cell reactor system.

  • In order to enhance the selectivity, productivity and yield of methyl fructoside, which was synthesized by enzymatic glycosylation of sucrose and methanol solution, controlling of surface property of solid support using different immobilization procedures optimized microenvironment of immobilized invertase. Silanization and polyethylene imine coating methods were adopted to give a hydrophobic and hydrophilic environment of immobilized invertase. As a result, polyethyleneimine coating method gave higher loading of enzyme, effective activity, and relative activity than silanization method, because it brought on increasing the functional density of amino group and enhancing the conservation of activity by regulating of hydrophilicity. And then, hydrophilic environment was possible to restraint the assessing of methyl fructoside molecule, which was more hydrophobic than sucrose, fructose, and glucose molecule in the reaction mixture, into .the active site of immobilizedinvertase. Consequently, hydrophilic microenvironment of immobilized invertase by polyethyleneimine coating obtained higher yield and productivity with increasing conversion than silanized and native invertase. Thus, this procedure optimized the microenvironment of immobilized invertase suitable for the enzymatic synthesis of methyl fructoside.

Ethanol Production from Lactose by Immobilized Reactor System Using a Fusant Yeast Strain of Saccharomyces cerevisiae and Kluyveromyces fragilis

  • Yeast cells of a fusant strain constructed by protoplast fusion of Saccharomyces cerevisiae and Kluyveromyces frugilis were immobilized on calcium alginate beads. The increment of the ethanol tolerance of this strain to 8.0%, when compared with the parent K, fragilis, was confirmed. Based on the results from jar fermentation, a packed-bed reactor of theh immobilized yeast cells was operated. The optimal performance of the immobilized yeast reactor for ethanol production was achieved when supplying 10% lactose (suplemented 1.0% yeast extract) at a temperature of 30.deg.C. The maximal ethanol productivity was obtained as 13.3 g/I/hr at a dilution rate of $0.76 hr^{-1}$.
