Effects of dietary valine:lysine ratio on the performance, amino acid composition of tissues and mRNA expression of genes involved in branched-chain amino acid metabolism of weaned piglets

  • Objective: The goal of this study was to investigate the effects of dietary standard ileal digestible (SID) valine:lysine ratios on performance, intestinal morphology, amino acids of liver and muscle, plasma indices and mRNA expression of branched-chain amino acid (BCAA) metabolism enzymes. Methods: A total of 144 crossbred pigs (Duroc${\times}$Landrace${\times}$Large White) weaned at $28{\pm}4days$ of age ($8.79{\pm}0.02kg$ body weight) were randomly allotted to 1 of 4 diets formulated to provide SID valine:lysine ratios of 50%, 60%, 70%, or 80%. Each diet was fed to 6 pens of pigs with 6 pigs per pen (3 gilts and 3 barrows) for 28 days. Results: Average daily gain increased quadratically (p<0.05), the villous height of the duodenum, jejunum and ileum increased linearly (p<0.05) as the SID valine:lysine ratio increased. The concentrations of plasma ${\alpha}-keto$ isovaleric and valine increased linearly (p<0.05), plasma aspartate, asparagine and cysteine decreased (p<0.05) as the SID valine:lysine ratio increased. An increase in SID lysine:valine levels increased mRNA expression levels of mitochondrial BCAA transaminase and branched-chain ${\alpha}-keto$ acid dehydrogenase in the longissimus dorsi muscle (p<0.05). Conclusion: Using a quadratic model, a SID valine:lysine ratio of 68% was shown to maximize the growth of weaned pigs which is slightly higher than the level recommended by the National Research Council.

  • Four fibrous feedstuffs from alfalfa meal (AFM), cassava leaf meal (CLM), rubber seed meal (RSM) and leucaena meal (LM) were included in semi-purified diets for growing pig (45 kg body wt.) at 20%, to investigate the effects of these fiber sources and fractions on amino acid digestibility. Cellulose (C), a purified fiber source was included in another diet at 5% level for comparison. The barrows fitted with ileal T-cannula were used in the digestion trials with latin square design. The digestibilities of amino acids were measured at both terminal ileum and fecal level. NDF and hemicellulose content were the highest in AFM-diet whereas LM-diet had the highest ADF and lignin content. RSM-diet contained the highest crude fiber and cellulose content. The digestibilities of amino acids at ileal level were found the highest with CLM-diet, while LM-diet was the least. At fecal level, control diet and CLM-diet were the highest in amino acid digestibility while AFM-diet was the least. The digestibility of amino acids was higher at ileal than fecal level. The digestibility of arginine was not affected with fiber fractions but was found to be the most disestible across all diets. The most depressed amino acid was methionine at both levels; praline and glycine, in the dispensable amino acid group, were depressed at ileal and fecal level, respectively. Lignin did not depress amino acid digestibility in general but specifically depressed methionine, histidine, isoleucine and threonine digestibility. Cellulose content did not affect amino acid digestibility but undesirable factors might be responsible.

Transcriptional Response and Enhanced Intestinal Adhesion Ability of Lactobacillus rhamnosus GG after Acid Stress

  • Lactobacillus rhamnosus GG (LGG) is a probiotic commonly used in fermented dairy products. In this study, RNA-sequencing was performed to unravel the effects of acid stress on LGG. The transcriptomic data revealed that the exposure of LGG to acid at pH 4.5 (resembling the final pH of fermented dairy products) for 1 h or 24 h provoked a stringent-type transcriptomic response wherein stress response- and glycolysis-related genes were upregulated, whereas genes involved in gluconeogenesis, amino acid metabolism, and nucleotide metabolism were suppressed. Notably, the pilus-specific adhesion genes, spaC, and spaF were significantly upregulated upon exposure to acid-stress. The transcriptomic results were further confirmed via quantitative polymerase chain reaction analysis. Moreover, acid-stressed LGG demonstrated an enhanced mucin-binding ability in vitro, with 1 log more LGG cells (p < 0.05) bound to a mucin layer in a 96-well culture plate as compared to the control. The enhanced intestinal binding ability of acid-stressed LGG was confirmed in an animal study, wherein significantly more viable LGG cells (${\geq}2log\;CFU/g$) were observed in the ileum, caecum, and colon of acid-stressed LGG-treated mice as compared with a non-acid-stressed LGG-treated control group. To our knowledge, this is the first report showing that acid stress enhanced the intestine-binding ability of LGG through the induction of pili-related genes.

Effects of Feeding Single or Multiple Probiotics on Performance and Intestinal Microflora of Broiler Chicks (단일 및 혼합 생균제의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향)

  • Two experiments were conducted to investigate the effents of feeding single or combined probiltics on performance and intestinal micreflora of broiler chicks for five weeks. Diets based on corn and soybean meal contained 21.50, 19% CP and 3,100, 3,150kcal/kg ME for starting and finishing period, respectively. Lactobacillus salvarius isolated from chicks intestine(LSC), Lactobacillus salvarius isolated from piglet(LSP), Bacillus polyfermenticus(BP) were fed with alone and mixed ones at the level of 0.21 and 0.1% in experiment 1 and 2, respectively. Three hundred eighty four chicks were randomly assigned to eight treatments with four replicates of 12 chicks each per treatment. Weight gain, feed consumption, feed conversion ratio(FCR) were weekly measured for 5 weeks. The number of intestinal micreflora was examined at the end of experiment. In both experiments, weight gain of chicks fed probiltics tended to be similar or higher than control, but was not in LSC+LSP treatment. Feed intake was not consistent among treatments. In experiment 1, FCR of chicks fed LSC alone was the lowest of all treatments, whereas it was significantly higher in LSC+LSP treatments than control(P〈0.05). In experiment 2, LSP supplemental groups tended to increase FCR compared to the control. The number of ileal E. coli was the lowest in LSP alone treatment of experiment 1, whereas cecal E. coli was higher concentration in probiotics supplemental groups than control. Total Lactobacillus of chicks fed probiotics was decreased in ileum, but was no consestency in cecum. In experiment 2, ileal total yeast tended to be higher in probiotics supplemental groups except LSP alone supplement than control. Total Lactobacillus of chicks fed LSC or LSP alone treatments was significantly higher than control(P〈0.05).

A Comparison of Feeding Lactobacillus and Virginiamycin Influence on Performance and Intestinal Microflora of Broiler Chicks (유산균과 버지니아마이신의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향)

  • This experiment was conducted to investigate the effect of feeding two strains of Lactobacillus and virginiamycin on performance, nutrients digestibility and intestinal microflora of broiler chicks(Abor acres$\times$Abor Acres) were randomly allocated into six treatments with four replications for five weeks. Control(no supplement), 0.05% virginiamycin(VM), Lactobacillus crispatus avibro1(LC), Lactobacillus reuteri avibro2(LR), LC+0.05% VM(LC+VM), LR+0.05% VM(LR+VM) were supplemented into basal diets, which contained ME 3,100kcal/kg and CP 22.0, 20.0% for starting and finishing period, respectively. Weight gain, feed intake and feed conversion(FC) were weekly measured. Nutrients digestibility, intestinal microflora and fecal noxious gas were examined at the end of experiment. Weight gains of chicks fed Lactobacillus or VM was significantly higher than control(P〈0.05). Feed intake increased significantly in those supplemental groups(P〈0.05). FC of chicks fed Lactobacillus or VM significantly lower than control(P〈0.05). Degestibility of crude protein, calcium, and phosphorus improved significantly in alone or combined Lactobacillus treatments(P〈0.05). Whereas DM, crude fat and ash digestibility were not statistically different. Feeding Lactobacilli tended to increase the total Lactobacillus spp. in ileum at one and three weeks of age(WOA) and showed significantly higher in cecum than control at 5 WOA. Total yeast were not shown difference at 1 and 3 WOA, but significantly increased at 5 WOA(P〈0.05). The ileal and cecal anaerobes were started to increase from the first WOA. Fecal NH$_3$gas tended to decrease in Lactobacillus treatments compared to that of other treatments.

Therapeutic Effects of Panax ginseng on the Neurotoxicity Induced by Abuse Drugs

  • Panax ginseng has been useful for the treatment of diverse disease in oriental countries for thousands of years. In addition, a folk medicine prescribed by seven herbal drugs including Panax ginseng has been antinarcotics in the treatment of morphine-dependent patients. Many articles have been reported on these works. Therefore, we review the protective effects of Panax ginseng on the neurotoxicity induced by abuse drugs. Ginseng total saponins (GTS) extracted and isolated by Panax ginseng antagonized Morphine-induced analgesia, and inhibited the development of analgesic tolerance to and physical dependence on morphine. GTS inhibited morphine-6 dehydrogenase, which catalyzes production of mophinone from morphine, and increased hepatic glutathione level responsible to toxicity. Therefore, we hypothesized that these dual actions of ginseng can be associated with the detoxication of morphine. In addition, the inhibitory or facilitated effects of GTS on electrically evoked contraction in guinea pig ileum ($\mu$-receptors) and mouse vas deferens($\delta$-receptors) were not mediated through opioid receptors, suggesting non-opioid mechanisms. On the hand, antagonism of U-50,488H ($\kappa$-agonist)-induced antinociception is mediated by serotonergic mechanisms. GTS also inhibited hyperactivity, reverse tolerance (sensitization) and conditioned place preference-induced by psychostimulants such as methamphetamine, cocaine and morphine. On the other hand, GTS reduced the dopamine levels induced by methamphetamine. Moreover, GTS blocked the development of dopamine receptor activation, showing antidopaminergic effect. We suggest that GTS Prevent the methamphetamine-induced striatal dopaminergic neurotoxicity. In addition, Ginsenoside also attenuates morphine-induced cAMP signaling pathway. These results suggested that GTS might be useful for the therapy of the adverse actions of drugs with abuse liability.

General Pharmacological Properties of the New +/K+ ATPase Inhibitor DBM-819

  • The effects of a newly synthesized $H^+/K^+$ ATPase inhibitor,1-(2-methyl-4-methoxypheny)-4-[(3-hy-droxypropyl)amino] -6-methyl-2,3-dihydropyrrolo (3,2-c) quinoline (DBM-819) , on the central nervous system, isolated smooth muscle, cardiovascular and digestive systems and renal function were investigated in various experimental animals. Oral administration of DBM-819 had no effect on the central nervous system except body temperature of mice slightly decreased at doses of 15 and 50 mg/kg. DBM-819 produced a moderate analgesic effect in acetic acid-induced writhing test in mice at 50 mg/kg (p.o.). In conscious rats, DBM-819 (15 and 50 mg/kg, p.o.) showed a slight increase in blood pressure and a small decrease in heart rate. DBM-819 had an significant effect on agonist-induced contraction of guinea pig ileum at $1.5{\times}10^{-5}g/ml.$ No significant effect of DBM-819 (5 and 15 mg/kg, i.p) on urinary volume or urinary excretion of $Na^+,\;K^+$ and Cl- was observed in rats. DBM-819 had no significant effect on intestinal transport of a semisolid meal in mice at 15 and 50 mg/kg (p.o.). These findings suggest that DBM-819 exerts no significant pharmacological effects on the central nervous system and renal function at 15 mg/kg (p.o.), but produces some effects on the smooth muscle and circulatory system.

A Case of Intestinal Duplication Cyst Identified as Pathological Lead Point in a Child with Recurrent Intussusception (재발성 장중첩증 환아에서 병적 선두로 확인된 장중복 낭종 1예)

  • Intestinal duplication cysts are characterized by the attachment to some part of the gastrointestinal tract with which a blood supply is shared, and have an epithelial lining resembling some part of the alimentary tract. A 15-month-old female was admitted to our hospital with cyclic irritability, vomiting, and blood-tinged stool. The results of an ultrasound showed an ileocolic intussusception and a 1.3 cm cystic mass had double-wall sign and a Y-configuration with an adjacent ileal loop. She had a past history of two ileocolic intussusceptions. The cystic mass was considered to be a pathologic lead point, so resection and end-to-end anastomosis was performed. The gross and histologic evaluation of the specimen demonstrated a $2.4{\times}2.4$ cm cystic mass containing yellow mucoid fluid and the cyst wall was lined with intestinal and gastric mucosa and enclosed by a layer of muscle, which was shared with the adjacent ileum.

Intestinal T cell lymphoma in a cat, Korea (고양이 장에서 발생한 T 세포 림프종)

  • An 11 year-old male Korean short-haired cat was presented to local animal hospital due to weight loss, vomiting, and intestinal hypomotility. After the cat was euthanized by poor clinical outcomes, necropsy was performed at Animal and Plant Quarantine Agency. At necropsy, the stomach was enlarged and had some nearly complete pellet food and the yellow mucous contents. The lumen of the middle and lower parts of the jejunum became narrow. Histopathologically, medium-sized lymphoid cells with hyperchromatic nuclei enclosed by scant cytoplasm were diffusely proliferated from mucosa to serosa of the small intestine. These findings were mainly observed in the jejunum and slightly in the duodenum and ileum. The monomorphous lymphocytes were 1 to 1.5 times larger than red blood cells and had few mitotic figures. Metastasis of the tumor cells to other organs was not observed. In the result of immunohistochemical analysis for identifying the origin of tumor cells, CD3 was expressed, but $CD79{\alpha}$ was not detected in the infiltrated cells. This case was diagnosed as T cell intestinal lymphoma in a Korean short-haired cat based on the clinical signs, gross findings, histopathology, and immunohistochemistry.

A histochemical study of argentaffin endocrine cells in the gastrointestinal tract of ovariectomized rats

  • The regional distributions and frequencies of argentaffin endocrine cells in gastrointestinal (GI) tract of osteoporotic Sprague-Dawley rat induced by ovariectomy were studied by Masson-Hamperl silver stain. The experimental animals were divided into two groups, one is non-ovariectomized group (Sham) and the other is ovariectomized group (OVX). Samples were collected from each part of GI tract (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) at 10th week after ovariectomy or sham operation. Argentaffin cells were detected throughout the entire GI tract with various frequencies regardless of ovariectomy except for the rectum of OVX in which no cells were detected. Most of these argentaffin cells in the mucosa of GI tract were generally spherical or spindle in shape (open type cell) while cells showing round in shape (close type cell) were rarely found in gland regions. Significant decrease of argentaffin cells was detected in OVX compared to that of Sham except for the fundus and jejunum. However, in the fundus and jejunum, argentaffin cells in OVX showed similar frequency compared to that of Sham. In conclusion, the endocrine cells are the anatomical units responsible for the production of gut hormones that regulate gut motility and digestion including absorption, and a change in their density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion. Ovariectomy induced severe quantitative changes of GI argentaffin endocrine cell density, and the abnormality in density of GI endocrine cells may contribute to the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis.
