Lactic Acid Bacteria from Gamecock and Goat Originating from Phitsanulok, Thailand: Isolation, Identification, Technological Properties and Probiotic Potential

  • From independent swab samples of the cloaca of indigenous gamecocks (CIG), anus of healthy baby goats (AHG), and vagina of goats (VG) originating from Phitsanulok, Thailand, a total of 263 isolates of lactic acid bacteria (LAB) were collected. Only three isolates, designated C707, G502, and V202, isolated from CIG, AHG, and VG, respectively, exhibited an excellent inhibitory zone diameter against foodborne pathogenic bacteria when evaluated by agar spot test. Isolates C707 and G502 were identified as Enterococcus faecium, whereas V202 was identified as Pediococcus acidilactici, based on 16S rRNA sequence analysis. When foodborne pathogenic bacteria were co-cultured with chosen LAB in mixed BHI-MRS broth at 39℃, their growth was suppressed. These LAB were found to be capable of surviving in simulated stomach conditions. Only the isolate G502 was able to survive in the conditions of simulated intestinal juice. This research suggests that selected LAB could be used as a food/feed supplement to reduce foodborne pathogenic bacteria and improve the safety of animal-based food or feed.

  • The purpose of this work was to examine the antibacterial activity derived from a lactic acid bacterium isolated from korean rice wine, called makgeolli. Various physiological and biochemical properties of this strain were characterized. Both the BIOLOG system and phylogenetic analysis using 16S rRNA sequencing were utilized for identification, and the strain was designated as Lactobacillus casei HK-9, and registered in GenBank as [JQ951606]. Growth rate, production of organic acids (e.g., lactic acid and acetic acid), and pH changes during growth were monitored. The maximum concentrations of lactic acid and acetic acid were approximately 576 mM and 199 mM, respectively, and pH was changed from 7.00 to 3.74 after 72 h of incubation. HPLC was used to confirm the production of lactic acid and acetic acid. Significant antimicrobial activity of the concentrated supernatant was demonstrated against various food-poison causing bacteria (e.g., Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, methicillin-resistant Staphylococcus aureus, Escherichia coli, Salmonella enteritidis). Ethanol tolerance of L. casei HK-9 showed up to 12% of ethanol within the culture.

Isolation and Identification of Lactic Acid Bacteria Isolated from a Traditional Jeotgal Product in Korea

  • Seventeen lactic acid bacterial strains (LAB) were isolated using MRS agar medium from Jeotgal, a Korean fermented food, purchased at the Jukdo market of Pohang. To identify the strains isolated, they were tested by examining their cell morphologies, gram-staining, catalase activity, arginine hydrolase activity, D-L lactate form and carbohydrate fermentation. According to the phenotypic characteristics, three strains were tentatively identified as Lactobacillus spp., ten were Enterococcus spp. (or Streptococcus spp., or Pediococcus spp.) and the rest were Leuconostoc spp. (or Weissella spp.). Five strains among 17 were chosen by preliminary bacteriocin activity test. Four bacterial strains which inhibited both indicator microorganisms were identified by 16S rRNA sequencing. The results are as follows; Leuconostoc mesenteroides (HK 4), Leuconostoc mesenteroides (HK 5), Leuconostoc mesenteroides(HK 11), Streptococcus salivarius(HK 8). In order to check LAB which are showing a high survival rate in gut, we investigated three strains inhibiting both indicator microorganisms in artificial gastric acid and bile juice -all except HK8. The three strains mentioned above grew in extreme low acid conditions.

김치에서 분리한 저온성 젖산균의 동정 (Identification of Psychrotrophic Lactic Acid Bacteria Isolated from Kimchi)

  • $5{\sim}7^{\circ}C$의 저온에서 20일 및 50일간 각각 발효된 A 및 B의 두 김치시료에서 저온성 젖산균 각각 30주씩을 무작위로 분리하고 동정을 하였다. 김치 A에서 분리한 30주 중 14주는 Leu. mesenteroides subsp. mesenteroides로, 12주는 Leu. mesenteroides subsp. dextranicum으로, 4주는 Lac. bavaricus로 각각 동정되었고, 김치 B에서 분리한 30주 중 20주는 Lac. bavaricus로, 3주는 Leu. mesenteroides subsp. mesenteroides로, 3주는 Leu. lactis로, 2주는 Leu. paramesenteroides로, 2주는 Lac. homohiochii로 각각 동정되었다. 비록 본 균주들이 위와 같이 동정되었지만 당류발효 pattern과 arginine에서 $NH_3$의 생성능이 Bergey's Manual of Systematic Bacteriology에 기재된 내용과 일치하지 않는 균주가 많았고, Leu. mesenteroides subsp. mesenteroides와 Leu. mesenteroides subsp. dextranicum으로 동정된 균주들 중의 일부는 당류발효 pattern이 동일 subspecies의 균주들 사이보다 다른 subspecies와 더욱 일치하는 모순까지 드러내었다. 이들 저온성 젖산균들을 기존의 체제에 따라 분류함에는 어려움이 따랐으며 이의 해결을 위한 후속연구가 요구되었다.

시판김치로부터 젖산균의 분리 및 동정 (Isolation and Identification of Lactic Acid Bacteria from Commercial Kimchi)

  • 본 연구에서는 시판 김치로부터 분리된 젖산균을 동정하였다. 제조 후 4일 동안 실온에서 배양된 배추김치에서 12주, 2일 동안 실온에서 배양한 총각김치에서 6주, 5일 동안 실온에서 배양한 깍두기에서 15주, 그리고 4일 동안 배양한 파김치에서 23주의 젖산균이 분리되었다. 배추김치에서 분리된 12주의 젖산균 중 8주는 Lactobacillus plantarum로, 1주는 Leuconostoc lactis로, 2주는 Lactobacillus casei subsp. pseudoplantarum으로, 그리고 1주는 Lactobacillus sake로 동정되었다. 총각김치에서 분리된 6주의 젖산균 중 3주는 Leuconostoc paramesenteroides로, 2주는 Leuconostoc mesenteroides subsp. mesenteroides로, 그리고 1주는 Lactobacillus plantarum로 동정되었다. 깍두기에서 분리된 15주 중 2주는 Leuconostoc lactis로, 3주는 Leuconostoc mesenteroides subsp. dextricum로, 4주는 Lactobacillus casei subsp. pseudoplantarum로, 그리고 4주는 Lactobacillus coryniformis subsp. torquens로 동정되었다. 파김치에서 분리된 23주 중 22주는 L. plantarum으로 그리고 1주는 Lactobacillus sake로 동정되었다. 이상의 결과로부터, 배추김치에서는 L. plantarum이, 총각김치에서는 L. paramesenteroides이, 깍두기에서는 L. casei subsp. pseudoplantarum과 L. coryniformis subsp. torquens이, 그리고 파김치에서는 L. plantarum이 가장 많이 동정되었다. 특히 파김치에서는 선발된 23주 중 1주를 제외하고는 모두 L. plantarum으로 동정되었다.

Characterization of L-(+)-Lactic Acid Producing Weizmannia coagulans Strains from Tree Barks and Probiogenomic Evaluation of BKMTCR2-2

  • This study aimed to isolate and identify L-(+)-lactic acid-producing bacteria from tree barks collected in Thailand and evaluate the potential strain as probiotics. Twelve strains were isolated and characterized phenotypically and genotypically. The strains exhibited a rod-shaped morphology, high-temperature tolerance, and the ability to ferment different sugars into lactic acid. Based on 16S rRNA gene analysis, all strains were identified as belonging to Weizmannia coagulans. Among the isolated strains, BKMTCR2-2 demonstrated exceptional lactic acid production, with 96.41% optical purity, 2.33 g/l of lactic acid production, 1.44 g/g of lactic acid yield (per gram of glucose consumption), and 0.0049 g/l/h of lactic acid productivity. This strain also displayed a wide range of pH tolerance, suggesting suitability for the human gastrointestinal tract and potential probiotic applications. The whole-genome sequence of BKMTCR2-2 was assembled using a hybridization approach that combined long and short reads. The genomic analysis confirmed its identification as W. coagulans and safety assessments revealed its non-pathogenic attribute compared to type strains and commercial probiotic strains. Furthermore, this strain exhibited resilience to acidic and bile conditions, along with the presence of potential probiotic-related genes and metabolic capabilities. These findings suggest that BKMTCR2-2 holds promise as a safe and effective probiotic strain with significant lactic acid production capabilities.

이란 발효 유제품에서 분리한 유산균의 특성 (Characterization and Identification of Lactic Acid Bacteria Isolated from Fermented Milks in Iran)

  • This study aimed to identify lactic acid bacteria isolated from eight fermented milk products in Iran. We enumerated Lactobacillus species using De Man-Rogosa-Sharpe (MRS)-maltose and MRS agar with pH adjusted to 5.2, as well as assessment at 37℃ for 48 hr, studied Streptococcus spp. using M17 agar at 43℃ for 24 hr, and assessed Bifidobacterium species using nalidixic acid, paromomycin sulfate, neomycin sulfate, and lithium chloride (BL-NPNL) agar at 37℃ for 48 hr. The total viable Streptococcus spp. cell in fermented milk varied at 4.73-8.83 log CFU/mL. However, Bifidobacterium spp. were not detected in any of the tested samples. Lactobacilli were not detected in four of the eight samples, and viable Lactobacilli cells in the remaining four samples ranged 2.48-3.85 log CFU/mL. The pH of the tested samples ranged 3.53-4.19, and soluble solids (Brix measurement) ranged 7.5%-17.9%. A total of 130 isolates of gram-positive catalase-positive bacteria were characterized at the species level using 16S rRNA sequencing. Sequence analysis identified six species: Streptococcus thermophilus, Lactobacillus delbrueckii subsp. sunkii, Lactobacillus delbrueckii subsp. indicus, Lactiplantibacillus plantarum, Lacticaseibacillus rhamnosus, and Levilactobacillus brevis.

국내 액상 발효유용 유산균 스타터 미생물의 동정 및 생리적 특성 (Identification and Characterization of Lactic Acid Bacteria Starters Isolated from the Commercial Drink-Yogurt Products)

  • Starters of lactic acid bacteria(LAB) were isolated from the commercial yoghurt products and the four isolates have been studied on their identification and some physiological characteristics. For the purpose of identification, microscopic examination, API test, and 16s rRNA gene sequencing were conducted. Isolate A from a yogurt product of local dairy company A was shown to be Gram-positive rod-shaped bacterium. All strains isolated were turned out to be as Lactobacillus paracasei by using a API 50 CHL kit. In contrast, isolate A was identified as a strain of Lactobacillus helveticus based on the 16S rRNA sequencing data, and L. casei ssp. casei for both B and D and L. paracasei for C. All the isolates survived the simulated gastric juice, pH 2.0 within 3 hours and sharply decreased in viability so that no viable cell was observed after 4.5 hours incubation. In addition, the four isolated strains were almost identical in antibiotic susceptibility to six different kinds of antibiotics including erythromycin ($15\;{\mu}g$), ampicillin ($10\;{\mu}g$), gentamycin ($10\;{\mu}g$), neomycin ($30\;{\mu}g$), but rather resistant to colistin ($10\;{\mu}g$) and streptomycin ($10\;{\mu}g$). It was noteworthy that four isolates were confirmed to produce antibacterial substance against foodborne pathogens of Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli 0157:H7 as test organisms based on the inhibitory zones on an MRS soft agar medium. At presence, the inhibitory factor is unknown so that further studies are required to ascertain the active factor responsible for the inhibitory activities.

  • 김치발효 중 Gram 양성세균 또는 유산균의 분리와 동정을 시도하였다. 종의 다양성은 분리배지와 온도에 영양을 받았고, 다양성은 온도가 낮아질수록 감소하는 경향이었다. MRS는 세균의 분리에 KM(김치재료로 만든 자연배지)은 종의 수를 파악 하는데 각각 적합하였다. 분리균의 동정은 Bergey's manual of Systematic Bacterio]ogy (1986)를 기초로 하여 작성한 이 분농생표(dichotomous Identification scheme)에 의하여 선행하였다. 각 온도(5, 15, $25^{\circ}C$)에서 동정된 Gram 양성세균은 Leuconostoc 5종, Streptococcus 4종, Pediococcus 3종, Bacillus 2종 그리고 Ltobacillms 18종이였다. 각 온도에서 출현 빈도가 높은 종은 $25^{\circ}C$에서 LactobaIlus ptantanmz, Streptococcµs faffinolactis, Leuconostoc maιnteroid, subsp mlsentιroides 이었고, $15^{\circ}C$에서 L. mesenteroides Lactobacillus fructosus, L. maen teroid,l.I subsp. mesent,roid, 이였고, $5^{\circ}C$ 에서 Leuconostoc sp.(65.2 빈도)에 의하여 이루어졌다. 그리고 각 온도에 따른 김치발효 중 지금까지 알려진 Pediococcus cerevisiae 와 Streptococcus faecalis는 분리되지 않았다.

Lactic Acid Bacteria in Total Mixed Ration Silage Containing Soybean Curd Residue: Their Isolation, Identification and Ability to Inhibit Aerobic Deterioration

  • We investigated the effects of the predominant lactic acid bacteria (LAB) on the fermentation characteristics and aerobic stability of total mixed ration (TMR) silage containing soybean curd residue (SC-TMR silage). The SC-TMR materials were ensiled in laboratory silos for 14 or 56 days. LAB predominant in SC-TMR silage were identified (Exp. 1). Lactobacillus fermentum (L. fermentum) and Streptococcus bovis (S. bovis) were found in the untreated materials, Leuconostoc pseudomesenteroides (L. pseudomesenteroides) in 14-day silage and Lactobacillus plantarum (L. plantarum) in all silages. Pediococcus acidilactici (P. acidilactici), Lactobacillus paracasei (L. paracasei), and Lactobacillus brevis (L. brevis) formed more than 90% of the isolates in 56- day silage. Italian ryegrass and whole crop maize were inoculated with P. acidilactici and L. brevis isolates and the fermentation and aerobic stability determined (Exp. 2). Inoculation with P. acidilactici and L. brevis alone or combined improved the fermentation products in ryegrass silage and markedly enhanced its aerobic stability. In maize silage, P. acidilactici and L. brevis inoculation caused no changes and suppressed deterioration when combined with increases in acetic acid content. The results indicate that P. acidilactici and L. brevis may produce a synergistic effect to inhibit SC-TMR silage deterioration. Further studies are needed to identify the inhibitory substances, which may be useful for developing potential antifungal agents.
