Identification and Functional Analysis of LsMNPV Anti-apoptosis Genes

  • Three anti-apoptosis genes, Ls-iap2, iap3 and p49 were found in Leucania separata multiple nuclear polyhedrovirus. Amino acid sequence homology of Ls-IAP2 and Ls-IAP3 with Op-IAP2 and Op-IAP3 from Orgyia pseddotsugata MNPV were 20% and 42%, while that of Ls-P49 is 28% with Sl-P49 from Spodoptera littorolis MNPV. Ls-IAP2 contains one baculoviral IAP repeat (BIR) domain followed by a RING domain, while Ls-IAP3 contains two BIRs and a RING. Ls-P49 contains a reactive site loop, predicted cleavage site (KKLD$^{74}{\downarrow}$G) that is different from Sl-P49 (TVID$^{94}{\downarrow}$G). Expressed Ls-iap3 or Ls-p49 under presence of actinomycin D in SF9 cells, DNA ladder assayrevealed that Ls- IAP3 or Ls-P49 could block the apoptosis of SF9 cells induced by actinomycin D. Replication of p35 deficient-mutant Autographa californica MNPV in SF9 cells was also rescued when Ls-iap3 or Ls-p49 was expressed transiently. No anti-apoptotic activity was observed for Ls-IAP2. The results showed that both of Ls-IAP3 and Ls-P49 were functional apoptotic suppressors in SF9 cells.

  • IAP (In App Purchase) 컨텐츠는 스마트 기기의 앱 내부에서 진행되는 컨텐츠 구매로 최근 앱 스토어의 주요 수입원으로 떠오르고 있다. 많은 앱 개발사들이 사용자들이 쉽게 앱을 접할 수 있도록 앱 자체는 무료로 판매하고 주요한 기능이나 추가적인 기능을 IAP를 통해서만 구입하여 사용할 수 있도록 유도하고 있다. 이런 방식을 통해 사용자가 불법으로 앱을 다운로드 하더라도 추가적인 기능이나 컨텐츠를 이용하기 위해서는 IAP를 사용하여 대가를 지불하도록 유도하는 것이다. 그러나 앱 자체가 클라이언트 기능만을 수행하는 경우는 문제가 없지만 full-time으로 서버와 동기화를 해가며 동작하는 방식이 아닌 앱의 경우 IAP를 불법적으로 해킹하여 취득하는 것이 가능하다. 본 논문에서는 이러한 불법적인 IAP 컨텐츠 획득을 방지하기 위하여 full-time으로 서버와 동기화를 하지 않으면서도 IAP 컨텐츠를 보호하기 위한 메커니즘을 제안한다.

Clinical Effects of Intra-Abdominal Pressure in Critically Ill Trauma Patients

  • Purpose: There is increasing interest in intra-abdominal pressure (IAP) and intra-abdominal hypertension (IAH) in critically ill patients. This study investigated the effects and outcomes of elevated IAP in a trauma intensive care unit (ICU) population. Methods: Eleven consecutive critically ill patients admitted to the trauma ICU at Pusan National University Hospital Regional Trauma Center were included in this study. IAP was measured every 8-12 hours (intermittently) for 72 hours. IAP was registered as mean and maximal values per day throughout the study period. IAH was defined as $IAP{\geq}12mmHg$. Abdominal compartment syndrome was defined as $IAP{\geq}20mmHg$ plus ${\geq}1$ new organ failure. The main outcome measure was in-hospital mortality. Results: According to maximal and mean IAP values, 10 (90.9%) of the patients developed IAH during the study period. The Sequential Organ Failure Assessment (SOFA) score was significantly higher in patients with $IAP{\geq}20mmHg$ than in those with IAP <20 mmHg (16 vs. 5, p=0.049). The hospital mortality rate was 27.3%. Patients with a maximum $IAP{\geq}20mmHg$ exhibited significantly higher hospital mortality rates (p=0.006). Non-survivors had higher maximum and mean IAP values. Conclusions: Our results suggest that an elevated IAP may be associated with a poor prognosis in critically ill trauma patients.

Intestinal Alkaline Phosphatase: Potential Roles in Promoting Gut Health in Weanling Piglets and Its Modulation by Feed Additives - A Review

  • The intestinal environment plays a critical role in maintaining swine health. Many factors such as diet, microbiota, and host intestinal immune response influence the intestinal environment. Intestinal alkaline phosphatase (IAP) is an important apical brush border enzyme that is influenced by these factors. IAP dephosphorylates bacterial lipopolysaccharides (LPS), unmethylated cytosine-guanosine dinucleotides, and flagellin, reducing bacterial toxicity and consequently regulating toll-like receptors (TLRs) activation and inflammation. It also desphosphorylates extracellular nucleotides such as uridine diphosphate and adenosine triphosphate, consequently reducing inflammation, modulating, and preserving the homeostasis of the intestinal microbiota. The apical localization of IAP on the epithelial surface reveals its role on LPS (from luminal bacteria) detoxification. As the expression of IAP is reported to be downregulated in piglets at weaning, LPS from commensal and pathogenic gram-negative bacteria could increase inflammatory processes by TLR-4 activation, increasing diarrhea events during this phase. Although some studies had reported potential IAP roles to promote gut health, investigations about exogenous IAP effects or feed additives modulating IAP expression and activity yet are necessary. However, we discussed in this paper that the critical assessment reported can suggest that exogenous IAP or feed additives that could increase its expression could show beneficial effects to reduce diarrhea events during the post weaning phase. Therefore, the main goals of this review are to discuss IAP's role in intestinal inflammatory processes and present feed additives used as growth promoters that may modulate IAP expression and activity to promote gut health in piglets.

Softening Related Changes in Cell Wall Polysaccharides of Hot Pepper Fruit (고추과실 세포벽 다당류의 연화에 따른 변화)

  • Various cell wall polysaccharides such as IAP(ionically associated pectin), CBP(covalently bounded pectin), HF(hemicellulosic fraction) and CF(cellulosic fraction) were fractionated by chemical method from hot pepper fruit cell wall during ripening and softening. And then, the composition of the polysaccharides were determined. The IAP and CBP were composed of $65{\sim}88%$ polyuronide, $4{\sim}29%$ hemicellulose and $0.3{\sim}2.1%$ cellulose. And hemicellulose also contained $8{\sim}13%$ polyuronide and $0.1{\sim}1.1%$ cellulose. The containing ratio of hemicellulose associated with the CBP was higher than IAP. All of the polysaccharides except CF decreased by the turning stage showed softening. The polyuronide made up IAP changed little but, that of the CBP increased during softening owing mainly to decrease of the hemicellulose that was bounded to the CBP and was mostly linked to galactose.

NF-${\kappa}B$ Activation and cIAP Expression in Radiation-induced Cell Death of A549 Lung Cancer Cells (A549 폐암세포주의 방사선-유도성 세포사에서 NF-${\kappa}B$ 활성화 및 cIAP 발현)

  • Background : Activation of the transcription factor NF-${\kappa}B$ has been shown to protect cells from tumor necrosis factor-alpha, chemotherapy, and radiation-induced apoptosis. NF-${\kappa}B$-dependent cIAP expression is a major antiapoptotic mechanism for that. NF-${\kappa}B$ activation and cIAP expression in A549 lung cancer cells which is relatively resistant to radiation-induced cell death were investigated for the mechanism of radioresistance. Materials and methods : We used A549 lung cancer cells and Clinac 1800C linear accelerator for radiation. Cell viability test was done by MTT assay. NF-${\kappa}B$ activation was tested by luciferase reporter gene assay, Western blot for $I{\kappa}B{\alpha}$ degradation, and electromobility shift assay. For blocking ${\kappa}B$, MG132 and transfection of $I{\kappa}B{\alpha}$-superrepressor plasmid construct were used. cIAP expression was analyzed by RT-PCR and cIAP2 promoter activity was performed using luciferase assay system. Results : MTT assay showed that cytotoxicity even 48 hr after radiation in A549 cells were less than 20%. Luciferas assay demonstrated weak NF-${\kappa}B$ activation of $1.6{\pm}0.2$ fold compared to PMA-induced $3.4{\pm}0.9$ fold. Radiation-induced $I{\kappa}B{\alpha}$ degradation was observed in Western blot and NF-${\kappa}B$ DNA binding was confirmed by EMSA. However, blocking NF-${\kappa}B$ using MG132 and $I{\kappa}B{\alpha}$-superrepressor transfection did not show any sensitizing effect for radiation-induced cell death. The result of RT-PCR for cIAP1 & 2 expression was negative induction while TNF-${\alpha}$ showed strong expression for cIAP1 & 2. The cIAP2 promoter activity also did not show any change compared to positive control with TNF-${\alpha}$. Conclusion : We conclude that activation of NF-${\kappa}B$ does not determine the intrinsic radiosensitivity of cancer cells, at least for the cell lines tested in this study.

Change in intestinal alkaline phosphatase activity is a hallmark of antibiotic-induced intestinal dysbiosis

  • Objective: Intestinal alkaline phosphatase (IAP) maintains intestinal homeostasis by detoxifying bacterial endotoxins and regulating gut microbiota, and lipid absorption. Antibiotics administered to animals can cause gut dysbiosis and barrier disruption affecting animal health. Therefore, the present study sought to investigate the role of IAP in the intestinal environment in dysbiosis. Methods: Young male mice aged 9 weeks were administered a high dose of antibiotics to induce dysbiosis. They were then sacrificed after 4 weeks to collect the serum and intestinal organs. The IAP activity in the ileum and the level of cytokines in the serum samples were measured. Quantitative real-time polymerase chain reaction analysis of RNA from the intestinal samples was performed using primers for tight junction proteins (TJPs) and proinflammatory cytokines. The relative intensity of IAP and toll-like receptor 4 (TLR4) in intestinal samples was evaluated by western blotting. Results: The IAP activity was significantly lower in the ileum samples of the dysbiosis-induced group compared to the control. The interleukin-1 beta, interleukin-6, and tumor necrosis factor-alpha concentrations were significantly higher in the ileum samples of the dysbiosis-induced group. The RNA expression levels of TJP2, claudin-3, and claudin-11 showed significantly lower values in the intestinal samples from the dysbiosis-induced mice. Results from western blotting revealed that the intensity of IAP expression was significantly lower in the ileum samples of the dysbiosis-induced group, while the intensity of TLR4 expression was significantly higher compared to that of the control group without dysbiosis. Conclusion: The IAP activity and relative mRNA expression of the TJPs decreased, while the levels of proinflammatory cytokines increased, which can affect intestinal integrity and the function of the intestinal epithelial cells. This suggests that IAP is involved in mediating the intestinal environment in dysbiosis induced by antibiotics and is an enzyme that can potentially be used to maintain the intestinal environment in animal health care.

Identification of a Variant Form of Cellular Inhibitor of Apoptosis Protein (c-IAP2) That Contains a Disrupted Ring Domain

  • Among the members of the inhibitor of apoptosis (IAP) protein family, only Livin and survivin have been reported to have variant forms. We have found a variant form of c-IAP2 through the interaction with the X protein of HBV using the yeast two-hybrid system. In contrast to the wild-type c-IAP2, the variant form has two stretches of sequence in the RING domain that are repeated in the C-terminus that would disrupt the RING domain. We demonstrate that the variant form has an inhibitory effect on TNF-mediated $NF-{\kappa}B$ activation unlike the wild-type c-IAP2, which increases TNFmediated $NF-{\kappa}B$ activation. These results suggest that this variant form has different activities from the wild-type and the RING domain may be involved in the regulation of TNF-induced $NF-{\kappa}B$ activation.

A study on reflective spectrum between In-Ceram alumina core and IPS Empress 2 core (In-Ceram Alumina Core와 IPS Empress 2 core의 분광반사율 연구)

  • A study on reflective spectrum between In-Ceram alumina plate(IAP) and IPS empress 2 plate were carried out in order to determine the difference of reflective spectrum rates between materials and between thicknesses of materials(0.8mm, 1.0mm, 1.2mm) by visible wave. The rates are measured by spectrophotometer(Top scan model TC-1800). And an analysis of ANOVA and paired sample t-test were carried out. The results are summarized as follows. 1. The reflective rates of IAP and IPS increased slowly as visible wave spectrum increased. 2. The difference of reflective rates of IAP between 0.8mm and 1.0mm is significant but a little(less than 1%). The differences of reflective rates of IAP between 0.8mm and l.2mm and between 1.0mm and l.2mm are significant(8%, 10% ). 3. The differences of reflective rates of IPS between 0.8mm and 1.0mm and between 0.8mm and 1.2mm and between 1.0mm and 1.2mm are significant(0.06 %, 0.01 %, 2 %). 4. The differences of reflective rates of 0.8mm, 1.0mm between IAP and IPS are significant but a little(less than 0.9%, 0.5%). The difference of reflective rates of l.2mm between IAP and IPS is significant and larger than the other thicknesses relatively(7 %).

Changes in the Pectic Substance during Maturity of Apple (사과의 성숙에 따른 Pectin질의 변화)

  • The changes of methoxy content. compositional sugars. degree of esterification (DE) and pectic substances such as LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin) and CBP (covalently bounded pectin) of Fuji apple were investigated during maturity. The contents of LMP+HMP and IAP+CBP in alcohol insoluble solid(AIS) were 70% and 20%. respectively. These pectic substances were increased slightly during maturity. The reductions of hexose and uronic acid were Iron 20-50% and from 2-4%, respectively, but the inclosed of pentose was from 1-9% in LMP, HMP, IAP and CBP during maturity of Fuji apple. The galactose was decreased significantly among hexose. Whereas the relative ratio of uronic acid in HMP and CBP was increased.
