호기성 생물막을 이용한 HRT 변화에 따른 오염물질 제거 (Pollutant Removal in Variable HRT Using the Aerobic Biofilm)

  • In this study, an experiment was conducted on influent water with low concentrations of organic matter, such as river water or secondary treatment water of a sewage treatment plant, according to HRT changes by using aerobic biofilm. In the biofilm process, as the biofilm increases in thickness, the inner membrane can be low in oxygen transfer rate and become anaerobic conditions, while the detachment of biomass from biofilm occurs. To overcome these limitations in the detachment of microorganisms in biofilm, the yarn, which was made from poly propylene(PP), was weaved and manufactured into a tube. Then, a test was carried out by injecting air so that the interior of the biofilm could create aerobic conditions. The results of the experiment showed that the removal efficiency of $TCOD_{cr}$ reached 66.1~81.2% by HRT 2hr, and 50.9 ~61.8% after HRT 1 hr. The removal efficiency of $SCOD_{cr}$ was 45.9 to 55.1% by HRT 1hr, and 26.1% in HRT 0.5hr, showing the highest removal efficiency in HRT 1hr. The SS removal efficiency was at 81.8 to 94.6%, and the effluent SS concentration was very low, indicating less than 2.2 mg/L in all HRT's. As a result, the $SCOD_{cr}$ and $NH_4{^+}$-N that were removed per specific surface area and attached to microbial biofilm showed the highest efficiency in HRT 1hr with 8.37 $gSCOD_{cr}/m^2{\cdot}d$, 2.93 $gNH_4{^+}-N/m^2{\cdot}d$. From the result of reviewing the characteristics of biofilm growth, microorganisms were found to be attached, and increased by 36 days. Later, they decreased in number through detachment, but showed a tendency to increase again 41 days later due to microbial reproduction.

Cytochalasin B Modulates Macrophage-Mediated Inflammatory Responses

  • The actin cytoskeleton plays an important role in macrophage-mediated inflammatory responses by modulating the activation of Src and subsequently inducing nuclear factor (NF)-${\kappa}B$ translocation. In spite of its critical functions, few papers have examined how the actin cytoskeleton can be regulated by the activation of toll-like receptor (TLR). Therefore, in this study, we further characterized the biological value of the actin cytoskeleton in the functional activation of macrophages using an actin cytoskeleton disruptor, cytochalasin B (Cyto B), and explored the actin cytoskeleton's involvement in morphological changes, cellular attachment, and signaling events. Cyto B strongly suppressed the TLR4-mediated mRNA expression of inflammatory genes such as cyclooxygenase (COX)-2, tumor necrosis factor (TNF)-${\alpha}$, and inducible nitric oxide (iNOS), without altering cell viability. This compound also strongly suppressed the morphological changes induced by lipopolysaccharide (LPS), a TLR4 ligand. Cyto B also remarkably suppressed NO production under non-adherent conditions but not in an adherent environment. Cyto B did not block the co-localization between surface glycoprotein myeloid differentiation protein-2 (MD2), a LPS signaling glycoprotein, and the actin cytoskeleton under LPS conditions. Interestingly, Cyto B and PP2, a Src inhibitor, enhanced the phagocytic uptake of fluorescein isothiocyanate (FITC)-dextran. Finally, it was found that Cyto B blocked the phosphorylation of vasodilator-stimulated phosphoprotein (VASP) at 1 min and the phosphorylation of heat shock protein 27 (HSP27) at 5 min. Therefore, our data suggest that the actin cytoskeleton may be one of the key components involved in the control of TLR4-mediated inflammatory responses in macrophages.

  • A total of 109 heads of horses and ponies from eight horseback riding clubs nearby Central Taiwan were investigated to evaluate the blood parameters and the biochemical compositions of serum for the documentation of clinical pathological diagnosis and exercise physiology. Blood samples were collected from the jugular vein of resting horses. The sex difference in the blood traits were compared both in horses and in ponies. Results shows that total plasma proteins (PP) and hematocrit (PCV) were found higher in male horses than in females (p < 0.05). The sexual effect also exertes a significant influence on the leucocyte (WBC) count, but not on the erythrocyte (RBC) concentration. According to the differential counts of leucocytes, the number of monocytes and lymphocytes was higher in the male pony than that of in male horse. A close relationship Between the erythrocyte sedimention rate (ESR) and the other blood parameters were found especially in PCV, RBC concentration, and plasma protein level. The average ESR observed at 60 minutes were $21.80{\pm}21.87mm$, $39.50{\pm}18.90mm$ and $43.73{\pm}17.89mm$ in stallions, geldings, and mares, respectively. Most of the biochemical components of horse serum detected were distributed in normal ranges, although some of the items show a great variation in such a large sample size.

Anti-inflammatory effect of methanol extract from Erigeron Canadensis L. may be involved with upregulation of heme oxygenase-1 expression and suppression of $NF{\kappa}B$ and MAPKs activation in macrophages

  • BACKGROUND/OBJECTIVES: In this study, we determined the anti-inflammatory activities and the underlying molecular mechanisms of the methanol extract from Erigeron Canadensis L. (ECM) in LPS-stimulated RAW264.7 macrophage cells. MATERIALS/METHODS: The potential anti-inflammatory properties of ECM were investigated by using RAW264.7 macrophages. We used western blot assays and real time quantitative polymerase chain reaction to detect protein and mRNA expression, respectively. Luciferase assays were performed to determine the transactivity of transcription factors. RESULTS: ECM significantly inhibited inducible nitric oxide synthase (iNOS)-derived NO and cyclooxygenase-2 (COX-2) derived PGE2 production in LPS-stimulated RAW264.7 macrophages. These inhibitory effects of ECM were accompanied by decreases in LPS-induced nuclear translocations and transactivities of $NF{\kappa}B$. Moreover, phosphorylation of mitogen-activated protein kinase (MAPKs) including extracellular signal-related kinase (ERK1/2), p38, and c-jun N-terminal kinase (JNK) was significantly suppressed by ECM in LPS-stimulated RAW264.7 macrophages. Further studies demonstrated that ECM by itself induced heme oxygenase-1 (HO-1) protein expression at the protein levels in dose-dependent manner. However, zinc protoporphyrin (ZnPP), a selective HO-1 inhibitor, abolished the ECM-induced suppression of NO production. CONCLUSIONS: These results suggested that ECM-induced HO-1 expression was partly responsible for the resulting anti-inflammatory effects. These findings suggest that ECM exerts anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of Erigeron Canadensis L.

플로팅 게이트형 유기메모리 동작특성 (Operating characteristics of Floating Gate Organic Memory)

  • 유기메모리 제작을 위해 플라즈마 중합법에 의해 절연박막, 터널링 박막을 제작하였고, Au 메모리박막을 이용하여 플로팅게이트형 유기메모리를 제작하였다. 플로팅 게이트형 유기메모리의 메모리층의 전하충전 및 방전에 따른 유기메모리 동작특성을 생각해 보았고, 이를 증명하고자 게이트전압에 따른 히스테리전압 및 메모리전압을 측정하였다. 그 결과 게이트 전압의 인가에 따른 메모리층의 동작 이론을 증명하고자 게이트전압이 증가함에 따른 소스-드레인 전류의 히스테리시스 현상이 심해지는 것을 확인하였고, -60~60[V]전압 인가시 26[V]의 큰 히스테리시스 전압값을 보였다. 또한 게이트 전극에 쓰기전압인가에 따른 현상을 본 결과, 60[V]의 쓰기 전압을 인가하였을 시 13[V]의 memory 전압을 나타내었고, 80[V]의 쓰기전압을 인가하였을 시 18[V]로 memory 전압이 약 40[%] 향상된 수치를 보였다. 이로부터 메모리층의 전하 충전 및 방전에 따른 메모리 동작특성 이론을 실험적으로 검증하였다.

Identification of a Mature form and Characterization of Thermostability of a Serine-type Protease from Aquifex pyrophilus

  • Aquifex pyrophilus, a hyperthermophilic bacterium, has a serine-type protease that is located at the cell wall fraction with a mature size of 43 kDa. Molecular cloning of the protease gene revealed that it has an ORF of 619 amino acids with homologous catalytic site of serine-type proteases [Choi, I.-G., Bang, W.-K., Kim, S.-H., Yu, G. Y., J. Biol. Chem. (1999), Vol. 274, pp. 881-888]. Constructs containing different regions of the protease gene, including a alanine-substituted mutant at the active site serine, were constructed, and the factors affecting the expression level of the cloned protease gene in E. coli were examined. The presence of the C-terminus hydrophobic region of the protease hindered over-expression in E. coli. Also, the proteolytic activity of the expressed protein appeared to toxic to E. coli. An inactive form that deleted both of the N-terminal signal sequence and the C-terminal polar residues was over-expressed in a soluble form, purified to homogeneity, and its thermostability examined. The purified protein showed three disulfide bonds and three free sulfhydryl group. The thermal denaturation temperature of the protein was measured around $90^{\circ}C$ using a differential scanning calorimeter and circular dichroism spectrometry. The disulfide bonds were hardly reduced in the presence of reducing agents, suggesting that these disulfide bonds were located inside of the protein surface.

Stable Secretion Vector Derived from the RCR (rolling-circle replication) Plasmid of Bacillus mesentericus

  • The 5.8 kb pMMH1, rolling-circle replication (RCR) plasmid of the wild type soil Bacillus mesentericus was developed into a novel secretion vector system in Bacillus subtilis. The pMMHl turned out to have a replication origin and two open reading frames (ORFs) of the putative γ-GTP and type I signal peptidase (sipP). To characterize the regions necessary for plasmid stability and high copy number, five vectors (pPS, pPP, pEN, pMN, pME) were constructed by disruption or deletion of each region in pMMH1. Like pMMHl all constructed vectors were stable over 100 generations In a non-selective medium. Since pPS was the smallest (2.3 kb)of all, it was selected for the construction of a navel secretion vector, Using the $\alpha$-amylase promoter/signal sequence of B. subtilils the novel plasmid pJSN was constructed. When $\beta$-glucosidase was expressed using pJSN, we found $\beta$-glucosidase activity in the medium. This result strongly suggested that plasmid pJSN can be used for the production of bioactive peptides in B. subtilis.

폐수처리시설에서 발생된 유기성 슬러지에 함유된 다환방향족탄화수소의 농도 특성 (Polycyclic Aromatic Hydrocarbons in Industrial Organic Sludge from Wastewater Treatment Facilities in Korea)

  • 이 연구에서는 우리나라에서 유기성 슬러지의 배출량이 많은 6개 주요 산업 업종의 54개 사업장에서 채취한 98개 폐수처리 슬러지에 대하여 PAHs 함량을 조사하였다. 슬러지에 대한 해양배출허용기준과 비교시 C업종의 슬러지는 Nap에 대하여 제1기준과 제2기준을 모두 초과하는 시료가 1건, PC업종의 슬러지에서 Ant와 Fla에 대하여 제2기준을 초과하는 시료가 1건 있었다. ${\Sigma}_{16}PAHs$는 PC> C > CD> PP >WS > FB업종의 순으로 높았다. 슬러지의 육상 이용시 ${\Sigma}_{16}PAHs$ 함량농도를 규제하는 EU와 덴마크의 기준과 비교하였을 때 EU의 기준은 PC업종에서 1건, 덴마크 기준은 C업종과 PC업종에서 각 1건의 시료가 초과하였다. 7종 발암성 PAHs에 대한 ${\Sigma}_7PAHs$은 상대적으로 PC, CD, C업종에서 높게, PP, WS업종에서 낮았으며, FB업종에서는 검출되지 않았다. 슬러지에서의 PAHs의 농도에 대한 ANOVA 분석결과 발생업종과 채취시기에 의한 영향은 없는 것으로 나타났다. 개별 PAHs간 상관분석결과 4-ring 이상의 HMW PAHs 농도간에 상관성이 높았다. ${\Sigma}_{16}PAHs$와는 0.7 이상의 높은 상관성을 갖는 PAHs는 Acy, Ant, Fla, B(a)A, B(f)F, B(a)P 이었다. 주성분분석의 결과 98개의 슬러지는 전체 데이터의 약 70%의 설명력을 갖는 두 개의 주성분에 의해 세 개의 그룹으로 특성별로 분리될 수 있었다. 국내/외 선행연구와의 비교 결과 ${\Sigma}_{16}PAHs$는 스위스의 연구 결과와 비슷하였다.

Induction of Heme Oxygenase-1 by Traditional Herb Mix Extract Improves MKN-74 Cell Survival and Reduces Stomach Bleeding in Rats by Ethanol and Aspirin in vivo

  • Chinese herb medicines have traditionally been used to treat or alleviate the symptom of various diseases. The rationale for use of certain herbs to certain disorder is now getting unveiled by modern technology. In the present study, we investigated whether herb mix extract(HMX), which is alleged to be useful for gastric ulcer, protects stomach from oxidative stress. Rats were allowed to normal diet with and without HMX (1, 5, 10 mg/kg) for 30 days. To induce gastric ulcer, ethanol (75%, 1.5 ml) or acidified aspirin (100 mg/kg in 0.2 N HCl) was administered by oral route in 24 h-fasted rats and examined the gastric ulceration(bleeding) by measuring the size 1 h after the treatment. Results indicated the area of gastric bleeding was significantly less in HMX fed rats than in normal diet fed ones, and it was dependent on the duration and amount of HMX. To investigate the underlying mechanism by which HMX protects stomach from oxidative stress, expression of enzymes like heme oxygenase (HO), cyclooxygenase (COX), and inducible nitric oxide (iNOS) were investigated in MKN-74 cells, where aspirin or H. pylori was introduced. The results were compared with RAW 264.7 cells to check if there's cell specificities exist. The expression of HO-1 but not COX-2, iNOS was significantly increased by HMX. Furthermore, HO-1 inhibitor, SnPP IX reduced the HO-1 activity and reversed the survival rate in HMX-treated MKN-74 cells. There's no difference between RAW 264.7 cells and MKN-74 cells. We, thus, concluded that HMX is beneficial for protection from oxidative injury, and induction of HO-1 by HMX in gastric cells is, at least, responsible for protection from oxidative stress such as ethanol, aspirin and possibly H. pylori infection.

Inhibitory Effect of Sageretia theezans against the Production of Pro-Inflammatory Mediators through the Inhibition of NF-κB and MAPK, and Activation of Nrf2/HO-1 Signaling Pathways in LPS-Stimulated RAW264.7 cells

  • In this study, we evaluated the anti-inflammatory effect of extracts of leaves (ST-L) and branches (ST-B) from Sageretia theezans in LPS-stimulated RAW264.7 cells. ST-L and ST-B significantly inhibited the production of the pro-inflammatory mediators such as NO, iNOS, COX-2, $IL-1{\beta}$ and IL-6 in LPS-stimulated RAW264.7 cells. ST-L and ST-B blocked LPS-induced degradation of $I{\kappa}B-{\alpha}$ and nuclear accumulation of p65, which resulted to the inhibition of $NF-{\kappa}B$ activation in RAW264.7 cells. ST-L and ST-B also attenuated the phosphorylation of ERK1/2, p38 and JNK in LPS-stimulated RAW264.7 cells. In addition, ST-L and ST-B increased HO-1 expression in RAW264.7 cells, and the inhibition of HO-1 by ZnPP reduced the inhibitory effect of ST-L and ST-B against LPS-induced NO production in RAW264.7 cells. Inhibition of p38 activation and ROS elimination attenuated HO-1 expression by ST-L and ST-B, and ROS elimination inhibited p38 activation induced by ST-L and ST-B. ST-L and ST-B dramatically induced nuclear accumulation of Nrf2, but this was significantly reversed by the inhibition of p38 activation and ROS elimination. Collectively, our results suggest that ST-L and ST-B exerts potential anti-inflammatory activity by suppressing $NF-{\kappa}B$ and MAPK signaling activation, and activating HO-1 expression through the nuclear accumulation of Nrf2 via ROS-dependent p38 activation. These findings suggest that ST-L and ST-B may have great potential for the development of anti-inflammatory drug to treat acute and chronic inflammatory disorders.
