쑥부쟁이 추출물의 라디칼 소거활성 평가 (Determination of Radical Scavenging Activity of Aster yomena (Kitam.) Honda)

  • 본 연구에서는 1.1-Diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\cdot}OH$), 그리고 superoxide radical ($O_2{^-}$) 소거능 실험을 통해 쑥부쟁이의 항산화 효과를 평가하였다. 쑥부쟁이는 에탄올로 추출한 뒤, n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc), 그리고 n-butanol (n-BuOH)로 분획하였다. DPPH 라디칼 소거능 실험에서 모든 추출 및 분획물이 $10-100{\mu}g/mL$의 농도에서 농도 의존적으로 라디칼 소거능이 증가하였으며, 특히 EtOAc 분획물은 가장 강한 DPPH 라디칼 소거능을 나타내었다. 또한 쑥부쟁이는 $50{\mu}g/mL$ 농도에서 80% 이상의 $^{\cdot}OH$ 라디칼 소거율을 보였다. 특히 EtOAc 분획물은 $0.03{\mu}g/mL$에서 IC50 값을 나타내어 추출물과 분획물 중 가장 강한 $^{\cdot}OH$ 라디칼 소거능을 보여주었다. 또한, EtOAc 분획물은 $O_2{^-}$ 라디칼 소거능에서도 가장 강한 소거활성을 보였다. 이러한 결과는 쑥부쟁이가 산화스트레스로 인한 질병을 예방하는 천연 항산화제로서의 활용 가능성이 있음을 보여준다.

선박안정수의 해양외래침입생물체 처리 기술 (Technical Treatment on Foreign Invasive Marine Species of Living-things in ship′s Ballast-water)

  • 국제환경기구(GEF)의 해양환경 위해 지정 대상의 하나인 선박안정수 처리기술로써, 선박 안정수에 포함되어 타 지역으로부터 이동되는 유해성외래침입생물 처리방안을 제안하였다 alpha-AL$_2$O$_3$ 유전장벽층 전극에 의한 강 전리방전 기술을 적용하여 주위 산소와 물분자로부터 고농도 수산자유기(OH : hydroxyl radical) 및 활성입자( OH, $O_2$+, O (1D), HO$_2$)를 전리, 발생시켜서 이들을 해수에 신속히 용해 확산시켜 비교적 낮은 ∼20mg/L 정도의 수산기농도 하에서 유해성외래침입생물을 소멸 처리하는 환경 친화적 녹색 청정 처리기술이다. 제안된 기술은 대상물의 처리 후 부수적으로 발생할 수 있는 처리잔류물이 전혀 발생하지 않으며, 인공적 화학성분의 약제를 사용하지 않는 저렴한 처리방법으로 대ㆍ소형 원양선박의 안정수에 포함되어 있는 유해성외래침입생물의 타 지역 해양확산을 방지하고 안전하게 처리할 수 있는 자연치유적 신기술이다.

Anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneata

  • This study evaluated the anti-oxidant activity of avicularin and isovitexin from Lespedeza cuneata using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and hydroxyl (OH) scavenging assays. The results showed that among the four fractions, the highest OH radical scavenging activity was exhibited by the n-butanol fraction. The DPPH radical scavenging activities of avicularin and isovitexin were higher than 50% at a concentration of $100{\mu}g/mL$. Moreover, one hundred micrograms per liter of avicularin and isovitexin exhibited effective anti-oxidant activity, with the OH radical scavenging rates being 87.54 and 91.48%, respectively. These results suggest that Lespedeza cuneata would be useful anti-oxidant agents from natural sources.

울산시 상수원수에서의 오존분해 특성 및 의약물질 분해 거동 연구 (Characteristic behaviors of ozone decomposition and oxidation of pharmaceuticals during ozonation of surface waters in Ulsan)

  • This study demonstrates the oxidative degradation of pharmaceutical compounds during ozonation of surface waters in Ulsan. Diclofenac, carbamazepine, bezafibrate, and ibuprofen were selected as surrogate pharmaceutical compounds, and ozonation experiments were performed using raw waters collected from the Sayeon Dam and the Hoeya Dam in Ulsan. Diclofenac and carbamazepine which have high reactivity with molecular ozone showed higher removal efficiencies than bezafibrate and ibuprofen during ozonation. The addition of tert-butanol, a hydroxyl radical scavenger, increased the removal efficiencies of diclofenac and carbamazepine by increasing the ozone exposure. However, the oxidation of bezafibrate and ibuprofen was inhibited by the presence of tert-butanol due to the suppression of the exposure to hydroxyl radical. The elimination of the selected pharmaceuticals could be successfully predicted by the kinetic model base on the $R_{ct}$ concept. Depending on the experimental condition, $R_{ct}$ values were determined to be $(1.54{\sim}3.32){\times}10^{-7}$ and $(1.19{\sim}3.04){\times}10^{-7}$ for the Sayeon Dam and the Hoeya Dam waters, respectively. Relatively high $R_{ct}$ values indicate that the conversion of $O_3$ into $^{\cdot}OH$ is more pronounced for surface waters in Ulsan compared to other water sources.

Hydroxyl Radical-Generating Function of Horseradish Cu,Zn-Superoxide Dismutase

  • Cu,Zn-superoxide dismutase (SOD) was purified from horseradish by using Mono Q and Superose 12 FPLC column chromatography. The native molecular mass of the purified enzyme was approximately 33 kDa, as determined by gel filtration. The subunit molecular weight, as estimated by SDS-PAGE, was 16 kDa. These results indicated that the native enzyme is a homodimer. We investigated the free radical-generating function of horseradish Cu,Zn-SOD by using a chromogen, 2,2'-azinobis-(3-ethylbenzthiazoline-6-sulfonate) (ABTS) which reacts with ${\cdot}OH$ radicals to form $ABTS^{+{\cdot}}$ The formation of $ABTS^{+{\cdot}}$ was required for both active Cu, Zn-SOD and $H_2O_2$. The optimal pH for the free radical-generating activity of this enzyme was 6.0-8.0, and it retained about $40^{\circ}C$ of its maximum activity when exposed at $40^{\circ}C$ for 15 min. A neutral scavenger, ethanol, inhibited the $ABTS^{+{\cdot}}$ formation by horseradish Cu, Zn-SOD more effectively than that by the mammalian enzyme. These results suggest that the active channel of horseradish enzyme is slightly larger than that of the mammalian enzyme.

Antioxidant and Free Radical Scavenging Potential of Justicia gendarussa Burm. Leaves in vitro.

  • Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.

Application of Chemical Ionization Mass Spectrometry to Heterogeneous Reactions of OH with Aerosols of Tropospheric Interest

  • Studies performed on heterogeneous reactions of hydroxyl radicals (OH) in aerosol materials of tropospheric interest are presented, focusing on the chemical ionization mass spectrometric approach. Kinetic investigations of these reactions reduced deviation in the estimation of OH concentration in the troposphere by atmospheric modeling from field measurements. Recently, OH uptake was investigated under wet conditions to acquire kinetic information under more realistic conditions representative of the troposphere. The information on the mechanism and kinetics of OH uptake by tropospheric aerosol materials will contribute to the updating of atmospheric models, allowing a better understanding of the troposphere.

수삼, 연질 홍삼, 수액 홍삼의 항산화 효과 (Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap)

  • 고려 인삼(Panax ginseng)은 일반적으로 안전성이 우수하고 ginsenosides or panaxosides 같은 많은 생활성적인 물질을 함유하고 있다. 특히 고려 홍삼은 교감 신경계를 안정시키고 인지력을 향상시키고 개인의 의학적 효과를 높이는 데 도움을 줄 수 있다. 새로운 가공 기술로 부드러운 홍삼을 생산하였다. 본 연구는 새로운 가공 기술에 따라 생산된 연질 홍삼이 항산화 효과를 감소시키거나 개선했는지 여부를 조사하는 데 초점을 맞췄다. 연질 홍삼과 시판 홍삼액은 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) 소거활성능에 있어 유의한 차이를 나타내지 않았다(p<0.05). 연질 홍삼은 수삼과 시판 홍삼액에 비해 DPPH (22-Diphenyl-1-pichrylic)와 OH(hydroxyl radical) 소거활성능이 높게 나타났다. OH 소거활성능은 세 그룹(수삼, 연질 홍삼과 시판 홍삼액) 간 유의한 차이를 나타내었다(p<0.05). NO (nitric oxide) 소거활성능 역시 수삼, 연질 홍삼, 홍삼 수액생산품 사이에 유의한 차이가 있었다(p<0.05). 연질 홍삼에서 많은 칼슘 결정체가 전자현미경상에서 관찰되었다. 마그네슘과 칼륨은 연질 홍상과 경질 홍삼 사이에 유의한 차이가 없었다. 연한 홍삼의 추출물은 DPPH와 OH의 존재로 인해 서로 다른 활성산소를 효율적으로 소거할 수 있어 자유 라디칼로 유도된 질환에 도움이 될 수 있다.

Nitrofurantion이 폐장 미크로솜 지질과산화와 반응성 산소 라디칼 생성에 미치는 영향 (Effects of Nitrofurantoin on Lipid Peroxidation and Reactive Oxygen Radical Generation in Porcine Lung Microsome)

  • 항균제 nitrofurantion에 의한 폐독작용의 생화학적 기전을 규명하기 위한 연구 일환으로 in vitro에서 폐장 microsome 지질의 과산화 및 반응성 산소 radical $(O^{-}_{2}{\cdot},\;H_2O_2,\;OH{\cdot},\;^1O_2)$의 생성에 대한 nitrofurantion의 영향과 양자 간의 상호 관련성을 검토하였다. Nitrofurantion은 호기성 반응 조건에서 돼지 폐장 microsome의 NADPH 의존성 지질 과산화를 용량 의존적으로 증가시킬 뿐 아니라 $O^{-}_{2}{\cdot},\;H_2O_2$ 및 두 radical의 상호 작용으로 2차적으로 형성되는 $OH{\cdot}$의 생성 또한 촉진하였으며 $^1O_2$생성은 관찰되지 않았다. 이와 같은 폐장 microsome지질 과산화 증가는 SOD 및 catalase에 의하여 억제될 뿐만 아니라 $OH{\cdot}$ 제거 물질인 mannitol, thiourea에 의하여도 현저히 억제되었으며, $^1O_2$ 제거 물질에 의하여는 영향을 받지 않았던 한편 염기성 반응 조건에서는 nitrofurantoin에 의한 지질 과산화가 관찰되지 않았다. 이상의 결과로 미루어 보아 nitrofurantoin은 폐장 microsome의 NADPH 의존적이 반응성산소 radical $(O^{-}_{2}{\cdot},\;H_2O_2$$OH{\cdot})$의 생성을 증가시키며 이들 중 특히 $OH{\cdot}$ 에 의한 microsome막 지질 과산화를 촉진하는 것으로 결론지었고, 이와 같은 in vivo 현상은 nitrofurantion의 in vitro 폐독작용의 기전을 설명하는 일부가 될 것으로 사료하였다.

Biological Activities of Water and Ethanol Extracts from Two Varieties of Rubus coreanus Miquel Fruits

  • The potential biological activities of water and ethanol extracts from two varieties of Rubus coreanus Miquel fruits produced in the Gochang-gun (GR) and Hoengseong-gun (HR) regions of Korea were examined. The hydroxyl radical ($\cdot$OH) scavenging activity, reducing power, lipid peroxidation inhibitory activity, and antiproliferative activity on cancer cells by these extracts were examined, and $\alpha$-amylase and $\alpha$-glucosidase inhibition assays were also performed. The EtOH extract from GR showed high hydroxyl radical scavenging activity ($EC_{50}=119.47{\pm}5.13\;{\mu}g/mL$), lipid peroxidation inhibitory activity ($EC_{50}=213.45{\pm}3.14\;{\mu}g/mL$) and a concentration dependence, with OD values ranging from 0.15 to 0.56 (50 to 200 ${\mu}g/mL$), for reducing power. The EtOH extract from GR has the highest antiproliferative activities on MDA-MB-231 and HepG2 cancer cells among four extracts. Meanwhile, all extracts showed certain $\alpha$-amylase and $\alpha$-glucosidase inhibition activities. These results indicate that extracts from two varieties of R. coreanus fruits have significant antioxidant, anti-diabetic and anti-tumorigenic activities, which suggest these extracts could be a potential source for pharmaceutical.
