(-)-Hydroxycitrate의 식이 투여가 흰쥐의 식이 섭취량, 체중, 지방대사 및 합성에 미치는 영향 (Influence of (-)-Hydroxycitrate on food Intake, Body Weight and Lipogenesis in Rats)

  • The influence of (-0-Hydroxycitrate(HCA), shown to be a competitive inhibitor of adenosine 5-triphosphate(ATP) citrate lyase, on food intake and body weight, serum triglyceride and cholosterol level, in vivo rates of fatty acid and cholesterol synthesis, and fat cell number and size was investigated. 3 groups of female, 5 weeks old Sprague Dawley rats, 8 animals each, were ad libitum meal-fed or pair-fed(3 hours from 10 : 00 to 13 : 00) AIN based high glucose diet for a total period of 8 weeks. Providing normolipidemic rats orally with 400mg of HCA formula containing approximately 20mg of HCA 1 hour prior to daily feeding schedule significantly depressed in vivo hepatic rates of fatty acid and cholesterol synthesis in the liver and adipose tissue. Serum triglyceride and cholesterol levels were significantly reduced by HCA. At the end of treatment period, the rats administered with HCA resulted in a significantly reduction in body weight gain. The reduction in weights was attributable to a significant decrease in fat cell size with a smaller extent, but not significant, reduction in fat cell number. Rats receiving HCA demonstrated less food intake than the controls ; however, this decreased caloric intake was not fully responsible for the HCA induced depression of hepatic and adipocytic lipogenesis, since experiment using pair-fed cojntrol rats showed, less magnitude but similar results. Both a anorectic and an antilipogenic properties of HCA seem to be responsible for this weight reduction activity of HCA. The outcome of this study suggest that metabolic regulation may be a feasible approach to the control of obesity and hyperlipidemia.

과체중인 여성에서 체중 감소 보조제를 이용한 체중 및 내장지방의 감소 효과 (The Effect of a Potential Antiobesity-Supplement on Weight Loss and Visceral Fat Accumulation in Overweight Women)

  • Chitosan, hydroxycitrate and L-carnitine have been known to be antiobesity components. The purpose of this study was to evaluate the combined effects of chitosan, hydroxycitrate and L-carnitine mixture as a potential antiobesity supplement in overweight women. Pre-menopausal healthy females who were overweight (percent ideal body weight > 110) were included in this study. Forty-nine subjects randomly received a placebo (n = 25) or antiobesity-supplement (n = 24), which was a mixture of chitosan, hydroxycitrate, and L-carnitine. Before and after the eight-week experimental period, anthropometric parameters, blood components and computerized tomography were measured. At baseline, the two groups were well matched in terms of age, body mass index and lipid profile. After the eight weeks of potential antiobesity supplementation, the subjects' body fat percent had decreased significantly (p < 0.001) by 5.6% (39.1 $\pm$ 1 vs 36.9 $\pm$ 1%) while lean body mass increased (p < 0.01). Vsceral fat area at the L4 vertebra decreased significantly (p < 0.01) by 8.6% in the supplemented group and the total fat area at the L4 vertebra showed a tendency to decrease (p = 0.051) by 2.4%. Also, in the group given the antiobesity-supplement rather than the placebo, the fasting triglyceride level decreased significantly (p < 0.05) by 10.0%. In addition, serum total cholesterol levels in the antiobesity-supplement group showed a tendency to decrease (p=0.159) by 2.7% (194 $\pm$ 6 vs 189 $\pm$ 6 mg/dl). No side effects were found in either group during the intervention. In conclusion, the present study demonstrated that taking a mixture of chitosan, hydroxycitrate, and L-carnitine as a potential antiobesity supplement for eight weeks produced advantageous changes in the weight and visceral fat accumulation of overweight women without any side effects. Anti-Obesity Effect of a New Dietary Supplement Consisting of Hydroxycitrate, Carnitine and Red Pepper (3D-Relax Diet) in High-Fat Fed Rats

  • Anti-obesity effect of a new dietary supplement (3D-relax) in high-fat fed rats. The aim of this study was to assess the effects of 3D-relax; a proprietary formulation containing hydroxycitrate (233 mg/g), carnitine (150 mg/g) and red pepper (150 mg/g); on body weight, body fat, and serum lipids levels in rats fed a high-fat diet. Male SD 7-wk-old rats (n=8) were fed a high fat diet [52% total dietary energy (E%) from fat, 15.4 E% protein, 32.6E% carbohydrate] with or without 3D-relax administration (1 g/kg body weight/day) for 3 weeks. Administration of 3D-relax significantly reduced the increase in body weight compared to the group fed high fat without 3D-relax. Food efficiency ratio (FER) tended to be decreased with administration of 3D-relax, but was not significant. The perirenal and epididymal fat pad weights of vats administered 3D-relax were significantly lower than those of the high fat group that did not ingest 3D-relax during the 3 weeks. The oral administration of 3D-relax significantly increased HDL-cholesterol level and lowered total cholesterol level compared to those of high fat alone group. These results suggest that 3D-relax reduced body weight and fat gains, and those effects are presumably linked to its inhibitory effects on lipogenesis.

(-)-Hydroxycitrate를 이용한 비만 여성의 체중 조절 프로그램 실시 및 효과 평가에 관한 연구(I) (The Effect of (-)-Hydroxycitrate on Weight Control Program in Obese Women -I : Effect on Anthropometric Parameters-)

  • This study investigated the effect of weight control by using the commercial appetite suppressant ((-)-hydroxycitric acid(HCA) formula) and nutrition education on 72 obese women over a period of 8 weeks. During the study it conducted nutritional education for women to control their weight, thus analyzed their changes in anthropometric variables. All obese women were randomized in a double-blind method to consume either HCA(HCA group : experimental group) or placebo(placebo group : control group). Two groups were also divided randomly into 2 groups combined with commercial formula diet in 1 meal a day(HD group and PD group : HCA + gormula diet and placebo + formula diet) or not(HO group and conducted with 4 groups(HD, HO, PD, and PO group). All subjects were assigned to consume 800-1500kcal/d balanced diet which is 500kcal less than their usual energy requirement. To evaluate the effect of the weight control program, weight, percent of body fat, waist and hip circumferences, and 5 skinfold thickness were measured up to 5 times in all obese women. The mean weight of the subjects at the onset of the study was 76.5$\pm$10.6kg. The mean body mass index(BMI) wa 30.1$\pm$3.8 and it was in the upper 5 percentile of mean BMI of Korean women. At the end of the program, mean weight loss was 3.5kg after 2 weeks(p<0.001), and 5.8kg after 4 weeks(p<0.001). The waist, hip ratio(WHR) and skinfold thickness measurements of biceps, triceps, subscapular, suprailiac and abdominum showed significant reduction over the entire study period(p<0.05). These outcome were evaluated by effect of nutritional education and counselling. The reduction of % of body fat was significantly different among the 4 groups. Women who administrated HCA demonstrated more change in weight, BMI than the placebo group. There was also significant reduction in body composition (% of body fat, WHR, and skinfold thickness) than the other groups. The HD group which was administrated HCA combined with formula diet was more decreased than other groups. It showed that this program using commercial HCA and formula diet induced not only a change in weight but also a change in body composition. The outcome of this study suggests that HCA has a more effctive change on weight control which is carried out with nutritional education and counselling.

비만여성에서 저열량식사와 체중감량제 섭취에 의한 체중 및 체지방 감소 효과 (The Effects of Low Calorie Meal and Weight Control Preparation on the Reduction of Body Weight and Visceral Fat in Obese Females)

  • 저열량 처방을 위한 식사대체제와 식이섬유ㆍ탄수화물 가수분해 억제제ㆍHCA를 주 성분으로 하는 체중감량제의 섭취 효과를 판정하기 위하여 20대∼50대의 BMI 25이상의 여성 자원자를 모집하여 9주간 시험을 실시하였다. 시험기간 동안 1일 1끼 식사는 식사대체제로 대치하고 1일 2회 체중감량제를 섭취하게 한 후 신체계측과 체조성, 배변 상황, 식이섭취 실태, 자각 증상 등의 변화를 조사하여 효과를 판정하였다. 시험 결과는 9주 동안 지속적으로 체중과 체지방, BMI, 허리 및 엉덩이 둘레가 감소한 것으로 나타났으며 체중감소율은 영양학적으로 안전한 범위에 속해 있었다. 혈청 지질에는 유의적인 변화가 없었으며 혈당과 인슐린이나 C-peptide 함량에도 유의적인 차이는 없었으며 BUN이나 ALP는 감소하였고 헤모글로빈은 유의적으로 증가하였으나 모두 정상범위에 있었다. 배변 시간은 단축되었으며 건강에 대한 부정적인 자각증상도 감소되었다. 1일 섭취 열량은 1300 kcal 정도로 유지되었으며 비타민 A, 비타민 $B_2$, 엽산, 칼슘, 철, 아연등의 섭취량은 권장량의 80% 미만으로 나타났다. 그러므로 본 연구의 저열량식사와 체중감량제의 섭취는 체중 및 체지방 감소와 배변에 효과적이었으며 부정수소를 감소시킬 수 있었다.
