기능성 미량원소 Selenium 화합물에 대한 고찰 (Review on the Selenuium, an Essential Trace Mineral)

  • The trace mineral, selenium (Se), is an essential nutrient of fundamental importance to human health. It is also very toxic and can cause Se poisoning (selenosis) in human and animals when its intake exceeds a suitable amount. Se functions within mammalian systems primarily in the form of solenoprotein. About 35 selenoproteins have been identified, though many have not yet been fully elucidated. Selenoproteins contain Se as selenocyseine (Sec) and perform variety of structural and enzymic roles; the enzymic roles are best-known as the antioxidants for hydrogen peroxides and lipid peroxides, and the catalysts for production of activity thyroid hormone. Glutathione peroxidases ($\textrm{GP}_X$) among the selenoproteins prevent the generation of free radicals and decrease the risk of oxidative damage to tissues, as does thioredoxin reductase (TR). TR also provides reducing power for several biochemical processes. Selenoproteins P and W are involved with oxidant defense in plasma and muscle, respectively, A selenoprotein is also required for sperm motility and may reduce the risk of miscarriage. Some epidemiological studies have revealed an inverse correlation between Se status and cardiovascular disease, and there is considerable evidence 1mm population com-parison data and animal studies that Se is anticarcinogenic. It is also suggested that Se should be needed for the proper functioning of the immune system, and appear to be a key nutrient in counteracting the development of virulence and inhibiting HIV progression to AIDS. As research continues, the role of selenium in the etiology of chronic diseases like appropriate medical nutrition therapy can be delivered and its effectiveness assessed. Se status in individuals is affected by diet and the availability of the Se. The Se content of plants is affected by the content and availability of the element in the soil in which they are grown, and so greatly varies from country to country, while the Se composition of meat reflects the feeding patterns of livestock. This paper provides an overview on Se as an essential trace mineral for human.

액상 효소반응을 이용한 Membrane Strip 형 Cholesterol 측정시스템의 개발 (Development of Membrane Strip Assay System for Lipoprotein Cholesterol Based on Liquid-Phase Enzyme Reactions)

  • A sensitive membrane strip assay for plasma lipoprotein cholesterol that can be performed without handling reagents has been investigated. We previously developed an assay system with immobilized enzymes (cholesterol esterase and cholesterol oxidase) on the surfaces of nitrocellulose membrane(1). In such a case, the amount of enzymes present on the membrane was limited by its surface area and, thus, the detection capability was relatively poor (> 50 mg/dL cholesterol). To overcome this problem, we devised a new system with non-immobilized enzymes by placing them within interstitial spaces of a celullose membrane pad in a dry state. Upon contact with sample medium, the enzymes were immediately dissolved and participated in the reactions with cholesterol in a liquid phase. We constructed a user-friendly system consisting of four membrane pads fro sample application, cholesterol decomposition, color development as signal, and medium absorption to invoke a continuous flow (sequential location from the bottom). A sample containing lipoproteins was added into the application pad by capillary action and transferred to the next pad for decomposition. The decomposition pad (namely, enzyme pad) contained a detergent (sodium cholate) for the destruction of lipoprotein particles, the two enzymes for cholesterol decomposition, and a chromogen (3,3'-diaminobenzidine). As a consequence of the enzyme reactions, hydrogen peroxide was produced, and then reacted in the presence of the chromogen with horseradish peroxidase immobilized on the signal generation pad. Finally, a colorimetric signal directly proportional to the cholesterol concentration was produced. The detection limit determined from this system under optimal conditions was at least 2 times lower than of the enzyme-immobilized system.

HMDSO와 산소를 이용한 PECVD 증착 $SiO_xC_y$필름의 특성연구 (Characterization of $SiO_xC_y$ films deposited by PECVD using BMDSO and Oxygen)

  • 폴리카보네이트 시트의 내마모성을 향상시키기 위하여 HMDSO 모노머와 산소를 사용하여 플라즈마 기상증착시킨 $SiO_xC_y$ 필름의 특성을 분석하였다. RF출력, 산소투입량, 수소투입량을 변화시키면서 각 증착조건에 따른 생성된 필름의 화학결합구조, 원소조성, 표면조도, 헤이즈 특성에 미치는 영향을 FTIR, XPS, AFM, Hazemeter를 이용하여 알아보았다. HMDSO와 산소를 사용한 박막의 증착은 100 nm/min이상의 높은 증착속도를 가졌고,증착실험에서 얻은 증착필름의 원소조성을 XPS를 이용하여 구한 결과, 종전의 다른 유기실리콘계 모노머를 사용했을때보다 박막에 존재하는 탄소잔류물을 효과적으로 감소시키는 것을 확인하였다. 또한, RF출력 200 Watt에서 산소가 100 sccm투입되었을 때 가장 우수한 헤이즈 특성을 보이는 막을 얻을 수 있었다. 본 연구로부터 HMDSO/$O_2$시스템이 탄소함량이 낮은 박막을 형성시키고 내마모도가 좋은 박막을 증착시키는데 효과적인 것을 알 수 있었다.

ECR-PECVD 방법으로 증착한 Diamond-Like carbon 박막의 광 특성 (Optical properties of diamond-like carbon films deposited by ECR-PECVD method)

  • ECR-PECVD 방법을 이용하여 ECR power, $CH_4/H_2$ 가스 혼합비와 유량, 증착시간을 고정 시켜놓고 기판 bias 전압을 변화 시켜가면서 유리 기판위에 DLC 박막을 제작하였다. Raman, FTIR 및 UV/Vis 스펙트럼을 측정하여 기판 bias 진압에 따른 이용 충돌이 박막의 특성 변화에 미치는 영향을 조사하였다. FTIR 분석결과로부터 기판 bias 전압을 증가시킬수록 이용충돌 현상이 두드러져 탄소와 결합하고 있던 수소원자들의 탈수소화 현상을 확인할 수 있었고, 박막의 두께는 bias 전압을 증가시킬수록 감소되었다. 그리고 Raman 스펙트럼으로부터 Gaussian curve fitting을 통하여 sp3/sp2의 결합수에 비례하는 D/G피크의 면적 강도비(ID/IG)는 기판 bias 전압을 증가시킬수록 증가하였다. 그리고 광 투과율은 증착시간을 길게 할수록, 기판 bias 전압을 크게할수록 감소하였으나, 박막의 밀도가 증가하고 더 매끄러운 DLC 박막이 형성되었다. 이 결과로부터 DLC 박막은 기판 bias 전압의 크기를 증가시킬수록 특성이 더 향상됨을 알 수 있었다.

  • 고연료 중 대부분이 수소와 탄소의 결합물이다. 연료들은 엔진 내부에서 완전 연소되었을 경우는 수분인 증기와 $CO_2$가 발생된다. 그러나 대개 불완전 연소가 일어나 $NO_x$나 Smoke가 발생되어진다. 그리고 이 물질은 인해에 매우 해로운 물질들이다. 본 논문에서는 EGR과 상용주파수를 이용한 플라즈마 장치를 조합하여 배기가스 유해물질 제거 특성을 연구하였다. $NO_x$의 경우에는 전압과 EGR율이 증가할수록 유해물질 제거 특성이 우수하였고, Smoke의 경우에는 EGR 율이 증가하게 되면 Smoke의 발생율이 증가하였으나 전압이 증가하게 되면 발생율이 감소하였다.

Identification of Oligosaccharides in Human Milk Bound onto the Toxin A Carbohydrate Binding Site of Clostridium difficile

  • The oligosaccharides in human milk constitute a major innate immunological mechanism by which breastfed infants gain protection against infectious diarrhea. Clostridium difficile is the most important cause of nosocomial diarrhea, and the C-terminus of toxin A with its carbohydrate binding site, TcdA-f2, demonstrates specific abolishment of cytotoxicity and receptor binding activity upon diethylpyrocarbonate modification of the histidine residues in TcdA. TcdA-f2 was cloned and expressed in E. coli BL21 (DE3). A human milk oligosaccharide (HMO) mixture displayed binding with TcdA-f2 at 38.2 respond units (RU) at the concentration of 20 μg/ml, whereas the eight purified HMOs showed binding with the carbohydrate binding site of TcdA-f2 at 3.3 to 14 RU depending on their structures via a surface plasma resonance biosensor. Among them, Lacto-N-fucopentaose V (LNFPV) and Lacto-N-neohexaose (LNnH) demonstrated tight binding to TcdA-f2 with docking energy of −9.48 kcal/mol and −12.81 kcal/mol, respectively. It displayed numerous hydrogen bonding and hydrophobic interactions with amino acid residues of TcdA-f2.

Quantitative Speciation of Selenium in Human Blood Serum and Urine with AE- RP- and AF-HPLC-ICP/MS

  • Various separation modes in HPLC, such as anion exchange (AE), reversed-phase (RP), and affinity (AF) chromatography were examined for the separation of selenium species in human blood serum and urine. While RP- and AE-HPLC were mainly used for the separation of small molecular selenium species, double column AF-HPLC achieved the separation of selenoproteins in blood serum efficiently. Further, the effluent of AF-HPLC was enzymatically hydrolyzed and then analyzed with RP HPLC for selenoamino acid study. The versatility of the hybrid technique makes the in-depth study of selenium species possible. For quantification, post column isotope dilution (ID) with $^{78}Se$ spike was performed. ORC ICP/MS (octapole reaction cell inductively coupled plasma/mass spectrometry) was used with 4 mL $min^{-1}$ Hydrogen as reaction gas. In urine sample, inorganic selenium and SeCys were identified. In blood serum, selenoproteins GPx, SelP and SeAlb were detected and quantified. The concentration for GPx, SelP and SeAlb was $22.8{\pm}3.4\;ng\;g^{-1}$, $45.2{\pm}1.7\;ng\;g^{-1}$, and $16.1{\pm}2.2\;ng\;g^{-1}$, respectively when $^{80}Se/^{78}Se$ was used. The sum of these selenoproteins ($84.1{\pm}4.4\;ng\;g^{-1}$) agrees well with the total selenium concentration measured with the ID method of $87.0{\pm}3.0\;ng\;g^{-1}$. Enzymatic hydrolysis of each selenium proteins revealed that SeCys is the major amino acid for all three proteins and SeMet is contained in SeAlb only.

The Leaf of Diospyros kaki Thumb Ameliorates Renal Oxidative Damage in Mice with Type 2 Diabetes

  • Diabetic kidney disease is the most common and severe chronic complication of diabetes. The leaf of Diospyros kaki Thumb (persimmon) has been commonly used for herbal tea and medicinal purposes to treat a variety of conditions, including hypertension and atherosclerosis. However, the effect of persimmon leaf on kidney failure has not been investigated. This study aimed to examine the role of persimmon leaf in protecting the diabetes-associated kidney damage in a mouse model of type 2 diabetes. Mice were fed either a normal chow diet with or without powered persimmon leaf (5%, w/w) for 5 weeks. In addition to kidney morphology and blood markers of kidney function, we assessed levels of oxidative stress markers as well as antioxidant enzymes activities and mRNA expression in the kidney. Supplementation of the diet with powered persimmon leaf not only decreased the concentration of blood urea nitrogen in the plasma but also improved glomerular hypertrophy. Furthermore, the persimmon leaf significantly decreased the levels of hydrogen peroxide and lipid peroxide in the kidney. The activities of superoxide dismutase, catalase, and glutathione peroxidase and the mRNA expression of their respective genes were also increased in the kidney of persimmon leaf-supplemented db/db mice. Taken together, these results suggest that supplementation with the persimmon leaf may have protective effects against type 2 diabetes-induced kidney dysfunction and oxidative stress.

RF 마그네트론 스퍼터링법을 이용하여 사파이어 기판과 ZnO 박막 위에 증착한 AlN 박막의 특성분석 (Growth of AlN Thin Film on Sapphire Substrates and ZnO Templates by RF-magnetron Sputtering)

  • 먼저 RF 마그네트론 스퍼터링법을 이용하여 사파이어 기판 위에 AlN 박막을 증착하였다. AlN 공급원으로는 분말소결된 AlN 타겟을 적용하였다. 플라즈마 파워를 50에서 110 W로 증가시켰을 때 AlN 층의 두께는 선형적으로 증가하였다. 그러나 동작압력을 3에서 10 mTorr로 증가시켰을 때는 동작기체인 아르곤 양이 증가함에 따라 AlN 타겟으로부터 스퍼터링되어 나온 AlN 입자들의 평균자유행정의 거리가 감소하기 때문에 AlN 층의 두께는 약간 감소하였다. 질소 기체를 아르곤과 섞어주었을 때는 질소의 낮은 스퍼터링 효율에 의해서 AlN의 두께는 크게 감소하였다. 다음으로는 ZnO 형판 위에 AlN를 증착하였다. 그러나 700도 이상의 열처리에 의해서 AlN와 ZnO의 계면이 약간 분리되어 계면의 열적 안정성이 낮다는 결과를 얻었다. 게다가 스퍼터링으로 증착한 AlN 박막의 나쁜 결정성으로 인하여 700도에서 MOCVD의 반응기 기체인 수소와 암모니아에 의해서 AlN 밑의 ZnO 층이 분해되는 현상도 관찰하였다. 그리고 900도 이상에서는 ZnO가 완전히 분해되어 AlN 박막이 완전히 분리되었다.

반사방지 특성을 통일시킨 실리콘 질화막 간의 패시베이션 특성 비교 (Comparison of Passivation Property on Hydrogenated Silicon Nitrides whose Antireflection Properties are Identical)

  • Silicon nitride ($SiN_x:H$) films made by plasma enhanced chemical vapor deposition (PECVD) are generally used as antireflection layers and passivation layers on solar cells. In this study, we investigated the properties of silicon nitride ($SiN_x:H$) films made by PECVD. The passivation properties of $SiN_x:H$ are focused on by making the antireflection properties identical. To make equivalent optical properties of silicon nitride films, the refractive index and thickness of the films are fixed at 2.0 and 90 nm, respectively. This limit makes it easier to evaluate silicon nitride film as a passivation layer in realistic application situations. Next, the effects of the mixture ratio of the process gases with silane ($SiH_4$) and ammonia ($NH_3$) on the passivation qualities of silicon nitride film are evaluated. The absorption coefficient of each film was evaluated by spectrometric ellipsometry, the minority carrier lifetimes were evaluated by quasi-steady-state photo-conductance (QSSPC) measurement. The optical properties were obtained using a UV-visible spectrophotometer. The interface properties were determined by capacitance-voltage (C-V) measurement and the film components were identified by Fourier transform infrared spectroscopy (FT-IR) and Rutherford backscattering spectroscopy detection (RBS) - elastic recoil detection (ERD). In hydrogen passivation, gas ratios of 1:1 and 1:3 show the best surface passivation property among the samples.
