The Induction of Steroid ${\Delta}^1$-dehydrogenase from Arthrobacter simplex IAM 1660 (Arthrobacter simplex의 Steroid ${\Delta}^1$-dehydrogenase의 유도와 유도성 스테로이드의 성질)

  • - Since steroid $\Delta^1$-dehydrogenase synthesis has been known to be inducible, the mechanism of the enzyme induction of Arthrobacter simplex IAM 1660 was investigated. Among various steroids tested for inducers, hydrocortisone was the most effective inducer when hydrocortisone was used as a substrate for steroid $\Delta^1$-dehydrogenase. Steroid $\Delta^1$-dehydrogenase synthesis was effectively induced by progesterone, prednisolone and androstenedione, while the enzyme was less induced by cholesterol and not by phytosterols. The results suggest that the presence of 3-keto group and short side chain of steroids are the favorable factors for the induction of the $\Delta^1$-dehydrogenase synthesis. The enzyme was induced at the highest level when hydrocortisone was added at early log phase to the concentration of 0.01% of the culture and the culture was grown for 15 hours.

The Effects of Dangguijakyak-san and Wuelbigachul-tang on Mesangial Cell Proliferation and on ICAM-1 and ${\beta}1-integrin$ Expression (당귀작약산, 월비가출탕이 Mesangial Cell 증식과 ICAM-l 및 ${\beta}1-integrin$ 발현에 미치는 영향)

  • Objectives : This experiment was conducted to investigate the suppressive effects of Dangguijakyak-san and Wuelbigachul-tang on the expression of ICAM-l and ${\beta}1-integrin$, which mediate cell-cell or cell-matrix interaction, and on the proliferation of mesangial cells. Methods : After in vitro culturing of human mesangial cells with the supernatant which was obtained from the monocytes separated from human blood with Con-A, hydrocortisone, Dangguijakyak-san and Wuelbigachul-tang respectively, we evaluated suppressive effects by measuring the mesangial cell surface enzyme immunoassay or flow cytometry. Results : The results are summarized as follows: 1. Dangguijakyak-san and Wuelbigachul-tang induced marked suppressive effects on the mesangial cell proliferation in the 50% and 25% supernatant concentration stimulating experiments, but hydrocortisone had little effect in these experiments. 2. Dangguijakyak-san and Wuelbigachul-tang induced marked suppressive effects on ICAM-l and ${\beta}1-integrin$ expression, but were less effective than hydrocortisone was. Conclusions : Based on these results, Dangguijakyak-san and Wuelbigachul-tang were found to be effective in the suppression of mesangial cell proliferation and in ICAM-1 and ${\beta}1-integrin$ expression. Further in vitro investigations as conducted above, with the in vivo experiments reflected, may prove that Dangguijakyak-san and Wuelbigachul-tang contribute to the prevention of the glomerular disease.

Evaluation of Hydrocortisone Sustained-Release Suppositories Prepared with Eudragit-Polyethylene Glycol Solid Matrix (유드라짓 및 폴리에틸렌글리콜 고체 매트릭스로 제조한 히드로코르티손 좌제의 서방성 평가)

  • Hydrocortisone (HC) sustained-release suppositories were prepared by using a solid matrix of methacrylic acid-methacrylic acid methyl ester copolymer $(Eudragit\;L_{100}^{R}:\;EL)$ as a poorly water soluble carrier and polyethylene glycole 1540 (PEG) as an water soluble carrier. HC release rate was controlled by complexation with ${\beta}-cyclodextrin$ $({\beta}-CyD)$ which was confirmed by X-ray diffractometry, IR-spectroscopy and differential scanning calorimetry. Release rate of HC from the EL-PEG matrix suppositories decreased with increase of EL contents. The release rale from $HC-{\beta}-CyD$ complex decreased in the following order: $HC-{\beta}-CyD/PEG$ > HC/PEG > $HC-{\beta}-CyD/EL_{10%}-PEG$ > $HC/EL_{10%}-PEG$ > $HC-{\beta}-CyD/EL_{15%}-PEG$ > $HC/EL_{15%}-PEG$ > $HC-{\beta}-CyD/EL_{20%}-PEG$ > $HC/EL_{20%}-PEG$. The crystallinity of HC in polymer matrix was identified using X-ray diffractometer and the surface of matrix suppositories after release test was examined by scanning electron microscopy. The sustained release of HC from these matrix suppositories was attributed to the network structure of EL.

Effect of Antifoam Agents on $\Delta^1$-Dehydrogenation of Hydrocortisone (Hydrocortisonee 의 $\Delta^1$-Dehydrogenation 에서 소포제의 영향)

  • Effect of antifoam agents, silicone oil and neolin 302, was investigated on the production of prodnisolone by microbial $\Delta$$^1$-Dehydrogenation of hydrocortisone. The microbial process was conduct-ed by using a pseudo-crystallofermentation. By the hydrophobic-hydrophobic interaction, the steroid crystals aggregated with the antifoam agents. The aggregation resulted in a decrease of total mass transter area of substrate particles which is proportional to the dissolution rate of the solid substrate, and it consequently led to a significant decrease of the bioconversion rate. The bioconversion with neolin proceeded more slowly than with silicone oil. Increase of the concentration of the antifoam agents also yielded a significant decrease of the bioconversion rate.

Influence of Various Cyclodextrins on the Stability of Hydrocortisone 17-Butyrate in Aqueous Solution

  • The stabilizing effects of $\alpha$-$\beta$-$\gamma$- and dimethyl-$\beta$-cyclodextrins $(\alpha$-, $\beta$-, $\gamma$- and DM-$\beta-$-CyDs) on the degradation of hydrocortisone 17-butyrate (HC-17B) in aqueous solution was investigated. Hc-17B underwent a facile hydroxide ion-catalyzed rearrangement to the less active 21-butyrate ester by the apparent first-order kinetics, and maximum stability of HC-17B was obtained at around pH 4.0. The stability of HC-17B was increased by inclusion complexation with $\alpha$-, $\gamma$- and DM-$\beta$-CyD in the pH range of 2.0-8.0 examined, whereas $\beta$-CyD accelerated the degradation of HC-17B at the pH higher than 5.0. The effects of ionic strength, solvent, temperature and CyD concentration were also investigated. Stability constants and apparent degradation rate constants of HC-17B-$\gamma$-CyD and HC-17B-DM-$\beta$-CyD complexes were determined kinetically on the basis of 1:1 complexation. The results suggested that the inclusion complexation with $\gamma$-CyD or DM-$\beta$-CyD was most useful means to enhance the stability of the steroid.

Influences of Hydrocortisone, DHEA, Estradiol and Testosterone on the Hepatic and Intestinal Polyamine Metabolism of Castrated Mice (Hydrocortisone, DHEA, Estradiol 및 Testosterone에 의하여 나타나는 마우스-간 및 소장 Polyamine 대사의 변동에 관한 연구)

  • Hydrocortisone 50 mg/kg (HC), dehydroepiandrosterone 250 mg/kg (DHEA), ${\beta}-estradiol$ 5 mg/kg (E2), and testosterone 20 mg/kg (TS) were subcutaneously injected into the castrated ICR mice at noon for four days, and the animals were sacrificed at 10-12 A.M. of the fifth day. The intestinal DAO activity was significantly decreased by HC, but it was rather increased by E2 and TS, respectively. And DHEA did not change the DAO activity. But the hepatic MAO activity was not affected by anyone of HC, DHEA, E2, and TS. Aminoguanidine 25 mg/kg produced the marked decrease of the intestinal DAO activity and the significant increases of the intestinal PT and SD contents, but it did not change the hepatic polyamine contents. HC and DHEA induced the significant increase of the intestinal PT content. E2 induced the marked increase of the hepatic PT content and the moderate increase of the intestinal PT content. TS little affected the polyamine contents of the liver and intestine. These results suggest that the E2-induced increase of the hepatic PT content is rather ascribed to the greater enhancement of PT synthesis than the inhibition of polyamine catabolism, and that the HC-induced increase of the intestinal PT content is due partly to the inhibition of polyamine catabolism via DAO.

  • Inhibition of over-oxidation of prednisolone in the fermentation has been studied by using vegetative cells as enzyme source. firstly, A. simplex (ATCC 6946) was demonst.ated to degrade p.ednisolone in the vegetative culture of the microorganism. Over 72% of hydrocortisone was transformed into prednisoloneby 3 hours of the fermentation. However, the prednisolone produced was considerably oxidized forming over-oxidation product in 8hours of fermentation period with the intact cells. Secondly, in order to depress the over-oxidation and the breakdown of the steroid skeleton of prednisolone, chelating agents such as $\alpha$$\alpha$'-dipyridyl, o-phenanthroline and 8-hydroxyruinoline were added to the fermentation broth. Conseauently, the breakdown of prednisolone by the iutact cells was able to be remarkably retatded and an intermediate regarded as an oxidized product derived from prednisolone was accumulated, by the addition of $\alpha$$\alpha$'-dipytidyl in the fermentation.

  • In order to investigate the effects of Kaejibokryungwhan, Sobokchugeotang and Dohongsamultang on mesangial cell proliferation and fibronectin synthesis, laboratory study was performed. The results are summarized as follows; 1. Mesangial cell proliferation was significantly decreased in the Kaejibokryungwhan group and the Dohongsamultang group compared with the Control group. In the Sobokchugeotang group, the mesangia1 cell proliferation activity was lesser than Control group, but it was statistically non-significant. In Kaejibokryungwhan group, the Sobokchugeotang group and Dohongsamultang group, mesangial cell proliferation was significantly decreased compared with the Hydrocortisone group 2. In the group, which contained fetal bovine serum, fibronectin synthesis was decreased in the Kaejibokryungwhan group, the Sobokchugeotang group and Dohongsamultang group compared with Control group, but the difference was statistically non-significant. In Kaejibokryungwhan group, Sobokchugeotang group and Dohongsamultang group, fibronectin was less decreased compared with that of Hydrocortisone group. 3. In the group, which contained fetal bovine serum, fibronectin synthesis was significantly decreased in Kaejibokryungwhan group and Sobokchugeotang group than those of Control group. In Dohongsamultang group, the fibronectin synthesis was decreased than Control group, but the difference was statistically non-significant. In Sobokchugeotang group, the fibronectin synthesis was decreased than Hydrocortisone group, but it was statistically non-significant. According to the above results, the mesangial cell proliferation and fibronectin systhesis could be reduced by Kaejibokryungwhan group significantly.
