Reactions with Hydrazidoyl Halides (V): Synthesis of Some Amidrazones, Hydrazides, Pyrazoles and Pyrazolo [3, 4-d] pyridazine Derivatives

  • 2-Bromo (2'-benzofury)gloxal-2-aryhydrazones I reacted with nucleophiles displacing the bromide. Treatment of I with active methylene compounds yield the pyrazole derivatives VIII-XI. Compounds XII-XIV reacted with hydrazine to give pyrazolo (3, 4-dipyridazine derivative XIV-XIV. The structures of the products were assigned and confirmed on the basis of their elemental analysis and spectral data.

  • Various long chain aliphatic acid hydrazides react with aromatic and heterocyclic aldehydes in alcoholic medium in refluxing conditions to give corresponding 2-hydroxy benzylidene and 1H-indol-3-ylmethylene hydrazides, a newer class of mosquito para-pheromones. We describe here synthesis of various novel long chain aliphatic acid (2- hydroxy benzylidene and 1H-indol-3-ylmethylene) hydrazides by conventional as well as microwave irradiation techniques. The structures of these compounds have been confirmed by spectroscopic techniques (FTIR, NMR & MS). Some of the interesting features of the electron impact mass spectral fragmentation pattern of these compounds have also been discussed.

  • Five acid hydrazides as precolumn fluorescence derivatization reagents for carboxylic acids in HPLC, which have the benzofuran or benzothiazole moiety conjugated to a furan, thiophene or oxazoline ring, were synthesized and examined in view of reactivity, separability and sensitivity. Of these hydrazides, 2-(5-hydrazinocarbonyl-2-oxazolyl)-5,6-dimethoxybenzothiazole (BTOH) was most favorable. The detection limit of lauric acid as a model acid was 0.1 pmol per $10-{\mu}l$ injection volume at S/N=3, which was roughly equal to that of an analogous compound, 2-(5-hydrazinocarbonyl-2-furyl)-5,6-dimethoxybenzothiazole. The reagent allowed rapid assays of carboxylic acids ($C_{12:0}-C_{20:4}$) within 20 min with satisfactory scparability. The method was applied to the determination of fatty acids in human sera from healthy volunteers as well as from patients with diabetes or thyroid dysfunction.

  • 1-Aryl-4-arylidene-2-oxazoline-5-ones (I) and the corresponding unsaturated amidoesters (II) and arylidene hypuric acid hydrazide (III) were obtained. The hydrazides and 1-aryl-1,3-dithiophenol-2-amidopropan-3-ones derivatives (IV) were also obtained. The derivatives were tested for their antibacterial activities.

  • Reaction of propane-1, 3-sultone with amines gave N-substituted aminosulphonnic acids 2a-i, Dehydration of 2a-c with $POCI_3$ gave the corresponding sultams 3a-c. Propane-1, 3-sultone 1 reacted with tertury amines to give the betaiene salts 4-11. 2-4-Dimethyl-1, 3-butadiene-1, 4-sultone 12 condensed with amines to give N-substituted-2, 4-dimethyl-1, 3-butadiene-1, 4-sultames 13a and 13b. The reaction of 3a, 13a with hydrazine hydrate gave acid hydrazides 3d or 13c. Compounds 3d, 13c reacted with isocyanates to yield urea derivatives 14a-c, 15a-c.

Biological Screening of Novel Derivatives of Valproic Acid for Anticancer and Antiangiogenic Properties

  • Background: Valproic acid (VPA) is a potent anticancer and antiangiogenic agent. However, design and synthesis of chemical derivatives with improved antiangiogenic and anticancer activities are still necessary. In this study a library of novel derivatives of VPA was synthesized and tested. Methods: A human liver cancer cell line (HepG2) and a human normal embryonic kidney cell line (HEK 293) were exposed to various concentrations of VPA derivatives for 24 hours and cell viability was checked by MTT colorimetric assay. Anti-angiogenic properties were evaluated in transgenic zebrafish embryos. Results: N-valproylglycine derivatives suppressed survival almost 70% (p value 0.001) in HepG2 cells but only 10-12% in HEK 293 cells (p value 0.133). They also suppressed angiogenic blood vessel formation by 80% when used between $2-20{\mu}M$ in zebrafish embryos. Valproic acid hydrazides showed moderate level of anticancer activity by affecting 30-50% (p value 0.001) of cell viability in HepG2 cells and 8-10% in HEK293 cells (p value 0.034). Conclusion: The majority of compounds in this study showed potent and stronger antiangiogenic and anticancer activity than VPA. They proved selectively toxic to cancer cells and safer for normal cells. Moreover, these compounds inhibited developmental angiogenesis in zebrafish embryos. Based on the fact that liver is a highly vascularized organ, in case of liver carcinoma these compounds have the potential to target the pathological angiogenesis and could be an effective strategy to treat hepatocellular carcinoma.

  • 3-Chloro-1-benzothiophene-2-carbonyl chloride 1 was reacted with glycine in acetone to give 3-chloro-1-benzothiophen-2-yl-carbonylaminoacetic acid 2. Various aldehydes on treatment with compound 2 in acetic anhydride to gave 1,3-oxazol-5-ones 3a-d. These oxazolones was treated with aromatic amines or hydrazides to get various imidazol-4-ones 4a-t or 5a-l. Oxazolones 3a-d was also treated with aromatic hydrazines, expansion of five member oxazole ring to six member triazine ring occurs to yield 1,2,4-triazin-6-ones 6a-h. The structures of all the synthesized compounds were confirmed by spectral data and had been screened for antibacterial activity.

  • 88 selective COX-2 inhibitors belonging to three chemical classes (triaryl rings, diaryl cycloalkanopyrazoles, and diphenyl hydrazides) were studied using comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). Partial least squares analysis produced statistically significant models with q values of 0.84 and 0.79 for CoMFA and CoMSIA, respectively. The key spatial properties were detected by careful analysis of the isocontour maps. The binding energies calculated from flexible docking correlated with inhibitory activities by the least-squares fit method. (omitted)

  • The reaction of the sodium salts of 4-methoxy and 4, 7-dimethoxy 6-hydroxy benzofuran-5-carboxylic acid with ethyl chloroformate yields the corresponding dicarbethoxy derivatives. The N-substituted amides were obtained by treating the latter compounds with amines. The corresponding hydrazides were synthesized by the reaction of hydrazine hydrate on the dicarbethoxy derivatives which spontaneously cyclized to 5-substituted-2, 3- dihydro-1, 3, 4, -oxadiazol-2-one. Also the reaction with phenyl hydrazine has been studied. The dicarbethoxy derivatives and N-substituted amides were tested against Gram positive and Gram negative bacteria in vitro. Most of the compounds posses moderate or slight activity against Gram positive bacteria.
