Recombinant proteins can be generated quickly and easily in large amounts and at low-cost in silkworm larvae by using Bombyx mori nuclear polyhedrosis virus (BmNPV). We searched for high-permissive silkworm strains that have high production levels of heterologous proteins and are thus suitable for use as biofactories. In this study, we performed the analysis using a BmNPV vector expressing luciferase as a marker, and we confirmed protein expression by evaluating luciferase activity, determined by western blotting and luciferase ELISA, and confirmed transcription expression by semi- and quantitative real time PCR. For the selection of host silkworm strains, we first chose 52 domestic BmNPV sensitive strains and then identified 10 high-permissive and 5 low-permissive strains. In addition, to determine which hybrid of the high-permissive strains would show heterosis, nine strains derived through three-way crossing were tested for luciferase activity by western blotting, and luciferase ELISA. We found a correlation between luciferase activity and luciferase protein expression, but not transcription. There was no noticeable difference in protein expression levels between Jam313 as the high-permissive control strain and the three-way hybrid strains; however, the three-way cross strains showed lower luciferase activity compared with Jam313. In this study, luciferase protein production in the larvae of 52 domestic silkworm strains was elucidated using BmNPV.

Breeding of Lentinula edodes generates a number of hybrid strains that are subject to evaluation for good traits for the mushroom production. As an effort to understand biochemical properties of the hybrid strains, this study tried to develop a fast and easy method for comparison of the ability of producing extracellular laccase among hybrid strains of Lentinula edodes. For this aim, we estimated the effect of media types and chromagenic chemicals on the detection of extracellular laccase in seven hybrid strains of L. edodes. When Remazol Brilliant Blue R (RBBR) dye was used for chromagenic reaction, the detection of the enzyme activity was feasible both in the solid and liquid media containing not potato dextrose but malt extract as a nutrient component. When guaiacol was used for chromagenic reaction, the detection of the enzyme activity was feasible both in the solid and liquid media containing either potato dextrose or malt extract as a nutrient component. Malt extract-based liquid culture with RBBR or guaiacol in 2 ml microfuge tube allowed us to economically and quantitatively detect and compare the enzyme activity within 3 days among the tested hybrid strains of L. edodes.

A flywheel system is an electromechanical energy storage device that stores energy by rotating a rotor. The rotating part, supported by magnetic bearings, consists of the metallic shaft, composite rims of fiber-reinforced materials, and a hub that connects the rotor to the shaft. The delamination in the fiber wound composite rotor often lowered the performance of the flywheel energy storage system. In this work, an advanced hybrid composite rotor with a split hub was designed to both overcome the delamination problem in composite rim and prevent separation between composite rim and metallic shaft within all range of rotational speed. It was analyzed using a three-dimensional finite clement method. In order to demonstrate the predominant perfom1ance of the hybrid composite rotor with a split hub, a high spin test was performed up to 40,000 rpm. Four radial strains and another four circumferential strains were measured using a wireless telemetry system. These measured strains were in excellent agreement with the FE analysis. Most importantly, the radial strains were reduced using the hybrid composite rotor with a split hub, and all of them were compressive. As a conclusion, a compressive pressure on the inner surface of the proposed flywheel rotor was achieved, and it can lower the radial stresses within the composite rotor, enhancing the performance of the flywheel rotor.
Trichoderma spp. cause large crop losses of the cultivated shiitake mushroom, Lentinula edodes. We bred several shiitake strains that are resistant to Trichoderma spp. using di-mon mating to establish a useful method for controlling the greenmold disease. We examined the competitive ability of L. edodes against Trichoderma spp. using a dual culture system to select resistant strains. By screening Trichoderma-resistant strains, we found that among 11 parental strains, 4 strains, including KFRI 36, were confirmed resistant strains. They showed especially strong resistance to T. harzianum, which formed deadlock after mycelial contact and then invaded into the territory of T. harzianum. KFRI 171 also showed resistance to T. atroviride strains. Among 13 strains, which were made by hybridization of shiitake strains, 5 were confirmed to be resistant to Trichoderma, including KFRI 58-1. Their resistance was not correlated to the resistant activity of their parents’ strains. Two strains lose resistance and two strains acquire resistance compared to their parents’ strains. In SEM observation, the mycelium of L. edodes at the interaction zone of Lentinula-Trichoderma was rugged and swollen by T. harzianum.
To develop the new strains of microorganisms having the degradative ability for various aromatic hydrocarbons, the hybrid plasmid pKG2 having the 2,4-Dichlorophenoxyacetic acid(2,4-D) degradative genes, the hybrid plasmid pKG3 containg the naphthalene degradative genes and TOL plasmid were introduced into Pseudomonas putida KUD 12 and P. putida KUP 10 by transformation or conjugation which originally have the degradative ability of the synthetic surfactants and phthalate esters, respectively. From P. putida KUD12, the new strains of P. putida KUD101(pKG2), KUD102(pKG3), KUD103(TOL), and KUD202(pKG3, TOL) were obtained, and KUD106(pKG2), KUD107(pKG3), KUD108(TOL) were originated from the P.putida KUP10. The degradative abilities in P. putida KUD101, KUD102 and KUD107 were similar with those of the original strains. The P. putida KUD103, KUD106 and KUD202 had a little lower and P. putida KUD108 had a better degradative abilitie than those of the original ones. In the case of mixed cultures, the mixed culture of KUD107 and KUD108 had a better degradative abilities than those of the other mixed cultures.
It is necessary to evaluate the resistance to disease among genetic resources for development of disease resistant grapes. This study was conducted to screen the resistance to crown gall in muscadine and Florida hybrid bunch grapes by pathogen inoculation. In order to compare the responses to infection with different pathogen strains, muscadine and Florida hybrid grapes were inoculated with 3 strains of Agrobacterium vitis. Although there were different levels crown gall formation among grape cultivars, there little variation in response to inoculated strains. Among 29 muscadine cultivars tested by inoculation of A. vitis 'C4612', most of them were shown to be susceptible, and 'Gold Isle' and 'Africa Queen' were highly susceptible, and two cultivars, 'Welder' and 'Jumbo' were found to be resistant to crown gall disease. Among Florida hybrid grapes, 'Daytona', 'Stover', and 'Swanee' were susceptible and 'Blanc du Bois' was moderately susceptible to crown gall. Because muscadine grapes have been actively utilized as useful genetic resources for development of new grape varieties by intersub-genus cross, this result from the screening of resistance among muscadine grapes can provide valuable information in breeding programs of grape resistant to crown gall.

Selective breeding and hybridization were attempted to improve shiitake(Lentinus edodes) strains for bed-log cultivation, and the results obtained are as follows. 1. Thirty five strains collected in Korea and abroad were cultivated for one generation(five years) and fruit body yields were estimated by the amounts produced from $1m^3$ of oak tree(Quercus mongolica) logs. Thus, three high-temperature type strains including FRI 177 showing productivity of $139kg/m^3log$, two mid-temperature type strains including FRI 188 of $120kg/m^3log$, and seven low-temperature type strains including FRI 187 of $96kg/m^3log$ were selected as superior strains. 2. Twelve hybrid strains confirmed as independent strains through indoor-test with hybrid strains made by Di-mon mating method were cultivated for five years(one generation). Among them, FRI 182 and FRI 184 were turned out to be superior strains. FRI 182 of high-temperature type showed the yield of 108kg per $1m^3logs$ and FRI 184 of low-temperature type produced $103kg/m^3log$. 3. There were no significant differences in productivity among high-, mid- and low-temperature types, but some differences appeared among strain groups. The correlation was not shown in productivities between bed-log and sawdust cultivations.
Hybrid strains was selected by crossing between dikaryotic strain ASI 0628 (Dagul) bred from ASI 2596 (Suhan No.3) and ASI 2782(Black pileus mutant) and monokaryotic strain ASI 2344-84. The strain 84 that shown the best cultural characteristics was selected to be a new cytoplasmic hybrid variety and named as 'Chunhwashim'. The 'Chunhwashim' shown incompatibility among them forming the confrontation growth line against parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho). Fruiting body produced about $161.4{\pm}4.8g$ per bottle. And also the individual generation of fruit body is multiple than chunchu2ho as 40.9. The 'Chunhwashim' was cytoplasmic hybrid included hybrid DNA bands of parental strains ASI 0628 (Dagul) and ASI 2344 (chunchu 2ho) by Random Amplified Polymorphic DNA (RAPD) primer, and mitochondria DNA band of monokaryotic ASI 2344 (chunchu 2ho) using mitochondria microsatellite DNA marker. This new cytoplasmic hybrid variety 'Chunhwashim' of oyster mushroom is characterized by multiple of individual generation and deeply grey color of pileus.
We determined the effects of different water temperatures (15, 20, and 25℃) and photoperiod cycles (24L:0D, 12L:12D, and 0L:24D) on the oxygen consumption of the offspring of a cultured Japanese strain (JJ), a selected Korean strain (KK), and intraspecific hybrid strains (JK and KJ) of red seabream, Pagrus major, under starvation conditions. The different fish strains, water temperatures, and photoperiod cycles had effects on the mean oxygen consumption of fish. Oxygen consumption increased with increasing water temperatures for all photoperiod treatments (p<0.001). Fish held in continuous darkness (0L:24D) used consistently less oxygen than fish exposed to continuous light (p<0.05). The oxygen consumption of fish exposed to the light phase in a 12L:12D photoperiod was higher than that of fish in the dark phase of the 12L:12D cycle, and differences were significant in three of the strains: JJ (15℃), KK (15 and 20℃), and KJ (25℃). The oxygen consumption of the inbred (JJ and KK) and intraspecific hybrid (JK and KJ) strains varied with differing water temperatures and photoperiod cycles. The JK strain displayed significantly higher oxygen consumption than the other strains under all experimental conditions except 15℃ with a 0L:24D photoperiod. The JK and KJ strains usually showed the highest and lowest oxygen consumption values, respectively, whereas the inbred strains exhibited intermediate values. Oxygen consumption in the JJ and JK strains was usually higher than that of the KK and KJ strains. We propose that differences in the thermal sensitivity and photosensitization properties of the strains contribute to differences in their ability to adapt to changes in water temperature and photoperiod, thus resulting in differences in the amplitude of their metabolic rates.
Intergeneric hybrids formed between Aspergillus oryzae var oryzae and Penicillium verruculosum F-3 were obtained by nuclear transfer technique. Several auxotrophic mutants isolated fromconidio-spores of the two strains mutagenized with ultraviolet and N-methyl-N-nitrosoguanidine. Optimal conditions for formation of intergeneric hybrids were investigated. Frequencies of hybrid formation by nuclear transfer were 3$\times $10$^{-5}$ $~$1$\times $10$^{-5}$. From observation of genetic stability, conidial size, DNA content, nuclear stain, it was suggested that their karyptypes are aneuploid. The hybrid posses the 1.3$~$2.2 fold higher amylase activities than those of parental strains. It was also revealed that some hybids had different isozyme patterns compared to those of parental strains by amylase activity assays.
