Cleaved Amplified Polymorphic Sequence and Amplified Fragment Length Polymorphism Markers Linked to the Fertility Restorer Gene in Chili Pepper (Capsicum annuum L.)

  • Cytoplasmic male sterility (CMS) in plants, which is due to failure to produce functional pollen, is a maternally inherited trait. Specific nuclear genes that suppress CMS, termed fertility restorer (Rf) genes, have been identified in several plants. In this study, Rfl-inked molecular markers in pepper (Capsicum annuum L.) were detected by bulked segregant analysis of eight amplified fragment length polymorphisms (AFLPs). Only AFRF8 was successfully converted to a cleaved amplified polymorphic sequence (CAPS) marker. This was named AFRF8CAPS and genotype determination using it agreed with that obtained with the original AFRF8. A linkage map with a total size of 54.1 cM was constructed with AFRF8CAPS and the seven AFLP markers using the Kosambi function. The AFRF8CAPS marker was shown to be closest to Rf with a genetic distance of 1.8 cM. These markers will be useful for fast and reliable detection of restorer lines during $F_1$ hybrid seed production and breeding programs in pepper.

Identification of Heterosis QTLs for Yield and Yield-Related Traits in Indica-Japonica Recombinant Inbred Lines of Rice (Oryza sativa L.)

  • Supplying sufficient rice to growing populations is a global challenge. Hybrid indica rice varieties exploiting heterosis have increased yields, but inter-subspecific crosses between indica and japonica varieties are hampered by sterility. Examination and genetic understanding of yield heterosis in indica/japonica crosses addressing yield barriers are basic requirements. In this study, QTLs for heterosis of yield traits were identified in indica-japonica recombinant inbred lines (RILs) using a total of 178 RILs originating from Dasanbyeo (indica) ${\times}$ TR22183 (japonica) (DT-RILs) and their backcrossed populations. Nine of sixty-six major quantitative trait loci (QTLs) identified in DT-RILs exhibited heterosis. Heterosis QTLs clustered with other traits on chromosomes 1, 4, and 8, and clusters were conserved between different RILs. The clusters contained several known yield enhancement genes/QTLs. Specific heterotic allele combinations contributed to four major heterosis QTLs, particularly for panicle and spikelet number traits. Heterosis for yield and yield-related traits was explained by the harmonized effects of overdominance, dominance, and epistatic interactions in inter-subspecific breeding populations.

Sources of Resistance to Bacterial Wilt Found in Vietnam Collections of Pepper (Capsicum annuum) and Their Nuclear Fertility Restorer Genotypes for Cytoplasmic Male Sterility

  • Eleven hot pepper accessions collected in Vietnam showed stable resistance to bacterial wilt as well-known resistance sources, MC4 and MC5, in repeated inoculation tests with different Ralstonia solanacearum isolates conducted from 2004 to 2010. Seven of these accessions (specifically KC981, KC1006, KC1021, KC1027, KC1045, KC1050, and KC1055) resulted in stable male sterile F1 plants in the crosses with a cytoplasmically male sterile (CMS) Chilseong (CMS-A, Srfrf ), and therefore, they were maintainers (CMS-B) with a genotype of Nrfrf. The rest (KC980, KC995, KC999, and KC1009) produced stable male fertile F1 plants in the crosses, and therefore, were restorers (CMS-C) with a genotype of N(S)RfRf. Therefore, the maintainer and restorer sources of resistance may be used in preference in breeding maternal (CMS and their maintainers) and paternal parents (restorers) for resistance to bacterial wilt, respectively, in the hybrid breeding system utilizing cytoplasmic male sterility.

  • Androgenesis in silkworm acquires a special significance as along with combined applications of other breeding strategies like parthenogenesis and cloning, it may serve as n valuable tool for sex control in sericulture as well as selection and production of bisexual homozygous androgenetic lines. Production of hybrid silkworm yielding high proportion of male larvae is of immense use to silk industry (Strunnikov, 1975, 1983). In this review, an attempt has been made to assimilate the works carried out on androgenesis, different techniques of induction towards androgenetic development and its role in silkworm breeding.

  • A single cross hybrid, Heukjinjuchal, is a black color waxy corn (Zea mays L.) developed by the maize breeding team at NICS in 2008. Heukjinjuchal was produced by crossing two inbred lines, KBW24 as seed parent and KBW2 as pollen parent. Ear length and ear diameter of Heukjinjuchal was 16.0 cm and 4.4 cm, respectively. The ratio of kernel set length/ear length was 89%, similar with that of a check hybrid, Chalok1. It is resistant to Northern corn leaf blight disease and also lodging tolerant. Fresh ear yield of Heukjinjuchal comparable with those of a check hybrid, Chalok1 in regional yield trials for three years. Seed production of this hybrid is relatively easy because of well overlapping period of silking and pollen dispersal between the two parents. This cultivar would be well adaptable to the whole country.

Genetic Studies of Major Agronomic Traits in Hybrid Populations of Maize Inbred Lines. (옥수수 유망자식계통들의 잡종집단에서 주요 농업형질들의 유전분석)

  • This study was conducted to find out the scientific maize breeding protocol for developing high performing single cross hybrid using introduced U.S. elite inbred lines; the pattern of inheritance, heterosis and heritabilities of six agronomic traits were studied in the progenies derived from five crosses (Mo17/B14A, Va85/B73, C103/ND203, FR35/Oh43, Wf9/A632). Among the five cross combinations, the cross combination of Mo17/B14A showed the highest heterosis for the most agronomic traits. Among 6 agronomic traits, the grain yield showed the highest heterosis effect in most cross combinations. Most of the agronomic traits in this study showed more than 50% heritability for six cross combinations, with an exception of the ear length trait. In conclusion, since Mo17/B14A showed excellent performance for most of the agronomic traits, these inbred lines were desirable combination and regarded as superior germ plasm sources for F1 hybrid development. The results of current studies will be utilized for developing high performing single cross hybrid from maize inbred lines, and will be used for the further genetic analysis of agronomic traits and maize breeding programs.

  • A new single glutinous maize hybrid "Huin Chal 1" was developed at the Genetics and Breeding Lab., Dept. of Agron., College of Agriculture, Chungnam National University. Parents for the hybrid were all derived from Korean indigenous maize lines which were collected in 1980s. One of the parental inbred lines was originated from Bosung, Chonnam province and other inbred line was from Jewon, Chungbuk province. Both inbreds were developed by ear-to-row selection method through selling and named as Bosung and Jewon, respectively. Performace tests for the hybrid which was produced between Bosung and Jewon have been conducted five times including one test at Crop Experiment Station, Rural Development Administration. From the previous performance tests, the hybrid was recognized as high yielding hybrid with good agronomic characteristics and we named the hybrid as Huin Chal 1., and hereby we report the agronomic characters of the newly developed hybrid.

Effect of Male-Sterile Cytoplasm on the Genetic Performance of Agronomic Traits in $F_1$ Hybrid Rices

  • Three different male-sterile cytoplasmic lines and their common maintainer 'Zhenshan 97B' and two elite restorer lines were used to study cytoplasmic effects on agronomic trait manifestation per se under different nitrogen supply levels. The result showed that cytoplasmic effects could be modified by nitrogen environments. The cytoplasmic effect on grain yield under 150 kg N/ha varied depending on crosses, while it was significantly negative in most crosses under both 60 and 330 kg N/ha. The correlation and path-coefficient analyses suggested that it was expected to improve cytoplasmic effects through reducing maximum tillers and increasing the percentage of productive tillers, leading to increased productive tillers and higher yield in hybrid rice by the aid of cultural practice and genetic transformation. This study also revealed that the same cytoplasm in different combinations had differential effect under the same nitrogen environment, indicating that cytoplasmic effect was produced by interaction of nuclear genes with cytoplasm rather than cytoplasm per se. These results indicated the usefulness of evaluating diverse cytoplasmic sources in various nuclear genotypes bred for hybrid rice breeding program. The finding also suggested that negative cytoplasmic effect could be effectively overcome by elite restorer lines through the interaction of nuclear genes with female cytoplasm.

Development of a Robust Polyvoltine Breed $'NP_1'$ of the Mulberry Silkworm, Bombyx mori L.

  • A breeding programme was initiated during 2001 utilizing two polyvoltine silkworm breeds viz. $BL_{69}$, an evolved breed tolerant to high temperature and MAR, comparatively resistant to Bombyx mori nuclear polyhedrosis virus (BmNPV) with the objective to develop robust polyvoltine breeds and hybrids. The breed $NP_1$ was developed by exposing the fifth instar larvae to high temperature $(36{\pm}1^{\circ}C)$, high Relative Humidity ($85{\pm}5%$ R.H.) and inoculating third instar larvae with BmNPV inoculum. At $F_{12}$, the breed was tested for hybrid forming ability utilizing six bivoltine silkworm breeds viz. $CSR_2,\;CSR_4,\;CSR_{17},\;CSR_{18},\;CSR_{19}\;and\;NB_4D_2$. The hybrid $'NP_1{\times}CSR_{17}'$ exhibited its superiority by recording 97.2% survival, 1.892 g cocoon weight, 0.406 g cocoon shell weight, 21.5% cocoon shell ratio, 16.6% raw silk percentage and 890 m filament length whereas the control $(PM{\times}CSR_2)$ has recorded 90.2% survival, 1.599 g cocoon weight, 0.304 g cocoon shell weight, 18.9% cocoon shell ratio, 13.1 % raw silk percentage and 768 m filament length. Commercial exploitation of the new $polyvoltine{\times}bivoltine$ hybrid in sericulture industry has been discussed.
