Roles of T Lymphocytes in Early Human Reproduction (착상과 임신 초기 면역반응에서 T 림프구의 역할)

  • 착상, 태반생성 및 임신 유지 등 생식과정에서 semi-allograft인 배아 및 태아가 생존하기 위해서는 모체 면역계의 면역관용이 요구된다. 면역관용은 분자 생물학적으로 염증반응과 항염증반응의 적절한 균형을 유지하는 인식되고 있으며, 생식과정에서 모체 면역계의 T 림프구, 자연살해세포, 수지상 세포, 대식세포 등 여러 면역세포의 유기적인 협조하에 이루어진다. 면역세포들은 특정 항원 및 사이토카인의 자극에 따라 정반대의 성질을 가진 사이토카인을 생산, 분비할 수 있는 특성을 가지고 있어 각각을 염증성 또는 항염증성 면역세포로 명확히 구분할 수 없으며 면역세포의 이러한 특성에 의해 생산 및 분비되는 사이토카인의 종류에 따라 Th0 형 (Th 0 cell, Tc 0 cell, NK 0 cell), Th1형 (Th 1 cell, Tc 1 cell, NK 1 cell), Th2 형 (Th 2 cell, Tc 2 cell, NK 2 cell), Th3 형 세포 (Th 3 cell, Tc 3 cell, NK 3 cell)로 분류하기도 한다. 즉, 착상 시기에 혈관신생 및 영양막의 자궁 내 침투를 위한 적절한 염증성 사이토카인(inflammatory cytokine)의 분비 및 임신의 지속을 위한 항염증성 (anti-inflammatory) 사이토카인의 분비 등 생식과정에서 수반되는 염증성과 항염증성 면역반응의 적절한 균형을 유지하는 기전은 특정 면역세포만의 작용으로 결론 지을 수 없으며 면역 세포간 network의 산물이라 할 수 있다 (Figure 5). 면역세포 중 최근 그 존재가 알려진 면역조절 T림프구 (T reg cell)는 여러 연구자들에 의해 면역관용에 관여함이 일관되게 보고되고 있어 자궁 내모체-태아간 접촉면에서 면역세포들 간의 network에 중추적인 역할을 할 것으로 인식되고 있으나 작용기전으로 제시되고 있는 가설들을 뒷받침 할 만한 객관적인 연구가 필요한 실정이다. 본 고찰에서는 착상과 임신 유지 등 생식과정에 수반되는 면역세포 및 그 세포들의 작용기전중 T 림프구의 역할에 중점을 두고 그 분류 및 기능에 대해 정리해 보았다. 결론적으로 착상과 임신의 유지 등 생식과정에서 T 림프구는 면역관용과 거부에 적극적으로 작용하며 착상부전, 습관성유산 등 면역학적 병인이 유사한 생식장애 (poor reproductive performance)들의 발병에 중요한 역할을 하는 것은 의심할 여지가 없다. 하지만 향후 T 림프구 및 그와 연관된 면역세포들의 작용에 대한 확실한 분자학적 규명이 요구된다.

  • Immune system provides defense integrity of body against external invaders. In order to accomplish the important defending role immune system is composed of many different components which are regenerated continuously during lifespan. The key components are professional killing cells such as macrophage, neutrophil, natural killer cell, and cytotoxic T cell and professional blocking molecule, antibody, which is produced by plasma cell, the terminal differentiated B cell. Immune response is orchestrated harmoniously by all these components mediated through antigen presenting cells such as dendritic cells. Immune responses can be divided into two ways: innate immune response and adaptive immune response depending on induction mechanism. Aging is a broad spectrum of physiological changes. Likewise other physiological changes, the immune components and responses are wane as aging is progressing. Immune responses become decline and dysregulating, which is called immunosenescense. Immune components of both innate and adaptive immune response are affected as aging progresses leading to increased vulnerability to infectious diseases. Numbers of immune cells and amounts of soluble immune factors were decreased in aged animal models and human and also functional and structural alterations in immune system were reduced and declined. Cellular intrinsic changes were discovered as well. Recent researches focusing on aging have been enormously growing. Many advanced tools were developed to bisect aging process in multi-directions including immune system area. This review will provide a broad overview of aging-associated changes of key components of immunity.

Oral Administration of Poly-Gamma-Glutamic Acid Significantly Enhances the Antitumor Effect of HPV16 E7-Expressing Lactobacillus casei in a TC-1 Mouse Model

  • The conventional prophylactic vaccines for human papillomavirus (HPV) efficiently prevent infection with high-risk HPV types, but they do not promote therapeutic effects against cervical cancer. Previously, we developed HPV16 E7-expressing Lactobacillus casei (L. casei-E7) as a therapeutic vaccine candidate for cervical cancer, which induces antitumor therapeutic effects in a TC-1 murine cancer model. To improve the therapeutic effect of L. casei-E7, we performed co-treatment with poly-gamma-glutamic acid (${\gamma}-PGA$), a safe and edible biomaterial naturally secreted by Bacillus subtilis. We investigated their synergistic effect to improve antitumor efficacy in a murine cancer model. The treatment with ${\gamma}-PGA$ did not show in vitro cytotoxicity against TC-1 tumor cells; however, an enhanced innate immune response including activation of dendritic cells was observed. Mice co-administered with ${\gamma}-PGA$ and L. casei-E7 showed significantly suppressed growth of TC-1 tumor cells and an increased survival rate in TC-1 mouse models compared to those of mice vaccinated with L. casei-E7 alone. The administration of ${\gamma}-PGA$ markedly enhanced the activation of natural killer (NK) cells but did not increase the E7-specific cytolytic activity of $CD8^+$ T lymphocytes in mice vaccinated with L. casei-E7. Overall, our results suggest that oral administration of ${\gamma}-PGA$ induces a synergistic antitumor effect in combination with L. casei-E7.

Probiotic supplementation has sex-dependent effects on immune responses in association with the gut microbiota in community-dwelling older adults: a randomized, double-blind, placebo-controlled, multicenter trial

  • BACKGROUND/OBJECTIVES: Probiotics have been suggested as potent modulators of age-related disorders in immunological functions, yet little is known about sex-dependent effects of probiotic supplements. Therefore, we aimed to investigate sex-dependent effects of probiotics on profiles of the gut microbiota and peripheral immune cells in healthy older adults. SUBJECTS/METHODS: In a randomized, double-blind, placebo-controlled, multicenter trial, healthy elderly individuals ≥ 65 yrs old were administered probiotic capsules (or placebo) for 12 wk. Gut microbiota was analyzed using 16S rRNA gene sequencing and bioinformatic analyses. Peripheral immune cells were profiled using flow cytometry for lymphocytes (natural killer, B, CD4+ T, and CD8+ T cells), dendritic cells, monocytes, and their subpopulations. RESULTS: Compared with placebo, phylum Firmicutes was significantly reduced in the probiotic group in women, but not in men. At the genus level, sex-specific responses included reductions in the relative abundances of pro-inflammatory gut microbes, including Catabacter and unclassified_Coriobacteriales, and Burkholderia and unclassified Enterobacteriaceae, in men and women, respectively. Peripheral immune cell profiling analysis revealed that in men, probiotics significantly reduced the proportions of dendritic cells and CD14+ CD16- monocytes; however, these effects were not observed in women. In contrast, the proportion of total CD4+ T cells was significantly reduced in women in the probiotic group. Additionally, serum lipopolysaccharide-binding protein levels showed a decreasing tendency that were positively associated with changes in gut bacteria, including Catabacter (ρ = 0.678, P < 0.05) and Burkholderia (ρ = 0.673, P < 0.05) in men and women, respectively. CONCLUSIONS: These results suggest that probiotic supplementation may reduce the incidence of inflammation-related diseases by regulating the profiles of the gut microbiota and peripheral immune cells in healthy elders in a sex-specific manner.

Radiation-Induced Chromosome Aberration in Human Peripheral Blood Lymphocytes In Vitro : RBE Study with Neutrons and $^{60}Co\;{\gamma}-rays$. (KCCH cyclotron neutron 및 $^{60}Co\;{\gamma}-ray$에 의한 인체 말초혈액 임파구의 염색체 이상측정)

  • The frequencies of KCCH cyclotron neutron (30 cGy/min) or $^{60}Co\;{\gamma}-rays$ (210 cGy/min)-induced asymmetrical interchanges (dicentrics and centric rings) and acentric fragments (deletion) at several doses were measured in the normal human peripheral blood lymphocytes Chromosome aberrations were scored at the first nitosis after stimulation with phytohemagglutinin. The neutron and y-ray data were analysed on linear, power-law, quadratic and linear-quadratic model . When the dicentrics and centric rings of ${\gamma}-rays$ datas were pooled and fitted to these model, good fits were obtained to power-law $[Y=(5.81{\pm}1.96){\times}10^6D^{1.93+0.06},\; P=0.931]$, quadratic $[Y=(3.91{\pm}0.09){\times}10^{-6}D^2,\;P=0.972]$ an linear-Quadrati model $[Y=(6.55{\pm}6.83){\times}10^{-5}D+(3.72{\pm}0.22){\times}10^{-6}D^2\; P=0.922]$, except for linear model (P=0.067) As in the case of neutron data, the best fit was obtained to the linear model $(Y=(6.12{\pm}0.17){\times}10^{-3}\;D-0.22,\;P=0.987]$ and good fits were obtained to power-law$[Y=(5.36{\pm}3.02) {\times}10^{-4}D^{1.42+0.11},\; P=0.601]$ and linear-quadratic model$[Y=(2.43{\pm}0.70){\times}10^{-3}D+(1.21{\pm}0.39){\times}10^{-7}D^2$, \;P=0.415], except for quadratic model (P<0.005). The relative biological effectiveness (RBE) of neutron compared with y-ray was estimated by best fitting model. In the asymmetrical interchanges range between 0.1 and 1.5 per cell, the RBE was found to be $2.714{\pm}0.408$.

The Physicochemical Stabilities and Biological Activities of Pigment Extracts from Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14 (Pseudoalteromonas sp. Ju11-1과 Pseudoalteromonas sp. Ju14의 색소 추출물의 물리화학적 안정성과 기능성)

  • We investigated the physicochemical stabilities and biological activities of ethanol- extracted pigment from marine bacteria Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14. The bacterial pigment of strain Ju11-1 was very stable at pH 5.0 below $25^{\circ}C$. The stability of the pigment showed higher stability in the presence of metal ions such as $Cu^{2+}$ and $Mg^{2+}$. The pigment has activity of free-radical scavenging ($IC_{50}$ $95.2{\mu}g$/ml) and the protective antioxidant effect ($ED_{50}$ $82.3{\mu}g$/ml) against DNA damage in human lymphocytes. The bacterial pigment of strain Ju14 was very stable at pH range between 4.0 and 8.0 below $40^{\circ}C$. In the presence of light, the pigment was also very stable, showing more than 90 percent of remaining absorbance during 14 days at $25^{\circ}C$. The stability of the pigment, when metal ions were present, showed higher stability in all examined metal ions except for $Fe^{2+}$, $Al^{3+}$, and $Cu^{2+}$, especially in the presence of $Na^+$. The pigment has activity of freeradical scavenging ($IC_{50}$ $208.6{\mu}g$/ml) and the protective antioxidant effect ($ED_{50}$ $ 96.4{\mu}g$/m) against DNA damage in human lymphocytes. The result indicates that the bacterial pigments from marine bacteria, Pseudoalteromonas sp. Ju11-1 and Pseudoalteromonas sp. Ju14 showed higher physicochemical stability and significant effects for reduction in oxidative DNA damage. Therefore, the results suggest that these bacterial pigments could be used as a natural colorant having the advantages of antioxidant.

Effects of lymphocyte DNA damage levels in Korean plant food groups and Korean diet regarding to glutathione S-transferase M1 and T1 polymorphisms (건강한 성인의 glutathione S-transferase M1과 T1 유전자 다형성에 따른 한식에서의 식물성 식품군과 한식의 DNA 손상 감소 효과)

  • Purpose: GST (glutathione S-transferase) M1 and T1 gene polymorphisms are known to affect antioxidant levels. This study was carried out to evaluate genetic susceptibility by measuring the effect of DNA damage reduction in the Korean diet by vegetable food according to GST gene polymorphisms using the ex vivo method with human lymphocytes. Methods: Vegetable foods in the Korean diet based the results of the KNHANES V-2 (2011) were classified into 10 food groups. A total of 84 foods, which constituted more than 1% of the total intake in each food group, were finally designated as a vegetable food in the Korean diet. The Korean diet applied in this study is the standard one-week meals for Koreans (2,000 Kcal/day) suggested by the 2010 Dietary Reference Intakes for Koreans. Ex vivo DNA damage in human lymphocytes was assessed using comet assay. Results: In the Korean food group, the DNA damage protective effect of GSTM1 and GSTT1 was found to be greater in mutant type and wild-type, respectively. and the DNA damage protective effect according to the combined genotype of GSTM1 and GSTT1 was different depending on the food group. On the other hand, in Korean Diet, the DNA damage protective effect appeared to be larger in GSTM1 wild-type than in mutant type and was found to not be affected by GSTT1 genotype. Conclusion: These results can be used as basic data to demonstrate the superiority of the antioxidant function of Korean dietary patterns and food groups. Furthermore, it may be a starting point to begin research on customized antioxidant nutrition according to individual genes.

Assessment of DNA damage and Chromosome aberration in human lymphocyte exposed to low dose radiation detected by FISH(fluorescence in situ hybridization) and SCGE(single cell gel electrophoresis) (FISH기법 및 단세포전기영동기법을 이용한 저선량 방사선에 의한 DNA 상해 및 염색체이상 평가)

  • Comparative study was performed for the assessment of DNA damage and Chromosomal aberration in human lymphocyte exposed to low dose radiation using fluorescence in situ hybridization(FISH) and single cell gel electrophoresis(SCGE). Chromosomal aberrations in human lymphocytes exposed to radiation at doses of 5, 10, 30 and 50cGy were analysed with whole chromosome-specific probes by human chromosome 1, 2 and 4 according to PAINT system. FISH with chromosome-specific probe has been used to be a valid and rapid method fer detection of chromosome rearrangements induced by low dose radiation. The frequencies of stable translocation per cell equivalents were 0.0116, 0.0375, 0.040f, 0.0727 and 0.0814 for 0, 5, 10, 30 and 50cGy, respectively, and those of dicentric were 0.00, 0.0125, 0.174, 0.0291 and 0.0407 respectively. Radiation induced DNA damage in human lymphocyte in a dose-dependent manner at low doses from 5cGy to 50cGy, which were analysed by single tell gel electrophoresis(SCGE). From above results, FISH seemed to be useful for radiation biodosimetry by which the frequencies of stable aberrations in human lymphocyte can be observed more easily than by conventional method and SCGE also seemed to be sensitive method f9r detecting DNA damage by low dose radiation exposure, so that those methods will improve our technique to perform meaningful biodosimetry for radiation at low doses.

Selenium Effect on the Frequency of SCEs Induced by Heavy Metals in Human Lymphocytes (Selenium이 mercury, cadmium 및 chromium에 의한 자매염색분체교환(姉妹染色分體交換)의 빈도(頻度)에 미치는 영향(影響))

  • The protective effect of sodium selenite($Na_2SeO_3$) against the cytogenetic toxicity of heavy metals was investigated on human whole-blood cultures in relation to induction of sister chromatid exchange (SCE) in secondary metaphase chromosome. Methylmercury chloride($CH_3HgCl$), cadmium chloride($CdCl_2$), potassium dichromate($K_2Cr_2O_7$), and sodium selenite caused to the typically dose-dependent increase in sister chromatid exchanges (SCEs) by the concentrations ranging from $0.3{\mu}M\;to\;10{mu}M$. However, the inductions of sister chromatid exchanges by methylmercury chloride or cadmium chloride were inhibited by the simultaneous addition of sodium selenite $1.2{mu}M$. The frequencies of SCE were decreased to the level of control in the molar ratios as 2:1, 1:1, 1:2, and 1:4 of selenium selenite vs. methylmercury chloride, and as 1:1 and 1:2 of selenium selenite vs. cadmium chloride, while the frequencies of SCE induced by potassium dichromate were not changed by the addition of sodium selenite in culture condition. Mitotic indices were decreased in the higher concentrations of chemicals and not significantly changed by the simultaneous addition of sodium selenite to the culture condition containing each chemicals.

Antitumoral, Antioxidant and Antimicrobial Activities of Solvent Ftactions from Grifola umbllatus (저령추출물의 항암, 항산화 및 항균효과)

  • Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.
