The Growth Inhibitiory Effect of New Pyrrolo[1,2-${\alpha}$]benzimidazole Derivatives on Human Gastric Cancer Cells

  • In the course of screening synthetic compounds to inhibit tumor cell growth, pyrrolo[1,2-.alpha.] benzimidazole (PBI), an intermediate of azamitosene, was found to inhibit a proliferation of gastric cancer cell lines. Despite a potential cytotoxic activity against solid tumor cells as opposed to that against rapidly-doubled leukemic cells, there has been no report on the inhibition of gastric cancer cell line by PBI and its' derivatives. The present experiment was designed to determine if PBI derivatives can effectively inhibit the cellular proliferation of gastric cancer cells by using in vitro as well as in vivo chemosensitivity system (MTT assay, clonogenic assay and human tumor xenografted assay). Of the tested PBI derivatives, PBI (18) and PBI (20), displayed the effective growth inhibition of cultured gastric cancer cells or even in the xenografted nude mouse model.

Hesperidin Induces Apoptosis in SNU-668, Human Gastric Cancer Cells

  • Hesperidin, known as a flavonoid constituent of citrus, has been known to reduce the proliferation of several cancer cells. We investigated whether hesperidin-induced cell death on SNU-668, human gastric cancer cells. The cytotoxicity of hesperidin on SNU-668 cells was determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay at the concentration of 1, 10, 50, and 100 ${\mu}M$. Cell viability by hesperidin was 53.18$\pm$2.85% of control value at 100 ${\mu}M$. The cell death by hesperidin showed apoptotic features, which were confirmed using a combination of 4, 6-diamidino-2-phenylindole (DAPI) staining and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) assay. In the apoptosis-regulating genes, treatment of hesperidin decreased mRNA expression of B-cell CLL/lymphoma 2 (BCL2), whereas expression of BCL2-associated X protein (BAX) was increased. The mRNA expression and the activity of caspase3 (CASP3), a major apoptotic factor, was significantly increased by hesperidin treatment. These results suggest that hesperidin could induce apoptosis through CASP3 activation on SNU-668, human gastric cancer cells.

위암에서 정량적 역전사 중합효소연쇄반응을 이용한 다중 표지자 분석 (Multiple Genetic Marker Analysis with Using Quantitative RT-PCR in Gastric Cancer)

  • 목적: 위암 세포주 및 조직에서 다중 표지자 mRNA 발현을 정량적 RT-PCR 검사를 통해 확인함으로써 이들 표지자를 이용하여 위암의 복강내 미세전이 진단이 가능한가를 평가하고자 본 연구를 시행하였다. 대상 및 방법: 12개의 인체 위암 세포주와 10개의 위암 조직을 대상으로 Carcinoembryonic antigen (CEA), Cytokeratin 20 (CK20), Dopa decarboxylase (DDC), L-3-phosphoserine phosphatase (L3PP)의 네 가지 mRNA를 이용한 정량적 RT-PCR 다중 표지자 분석을 시행하였다. 결과: 12개의 인체 위암 세포주 중 CEA는 4개(33%), CK-20는 1개(8%), DDC는 6개(50%), L3PP는 12개 세포주 모두(100%)에서 과발현되었다. 10개의 위암 조직 중 CEA는 9개, CK20은 3개, DDC는 9개, L3PP는 10개 조직 모두에서 과발현되었다. L3PP는 모든 위암 세포주와 조직에서 과발현을 나타내었으나 과발현 정도는 비교적 낮게 측정된 반면, CEA와 DDC는 일부 위암 세포주 및 조직에서만 과발현을 나타내었지만 파발현 시 충분한 발현도를 나타내었다. 결론: 위암 환자에서 하나 이상의 암 특이적 유전자를 이용한 다중 표지자 분석은 단일 표지자 분석이 가지는 단점을 보완할 수 있을 것으로 예상되며, CEA, DDC, 및 L3PP의 세 가지 mRNA가 후보 유전자로 사용될 수 있을 것으로 생각한다.

인체 위암세포주에서 반지련(半枝蓮)의 항암 효능에 관한 연구 (Anti-cancer Effects of Scutellaria barbata in AGS Human Gastric Adenocarcinoma Cells)

  • The aim of the study is to investigate the anti-cancer effects of Scutellaria barbata in AGS human gastric adenocarcinoma cells. MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay and caspase 3 or 9 activity assay were carried out to examine cell death with Scutellaria barbata. To elucidate the inhibitory effects of Scutellaria barbata, cell cycle (sub-G1) analysis and mitochondrial membrane potential were performed in AGS cells after 24 h incubation with Scutellaria barbata. Scutellaria barbata induced apoptosis in AGS cells by using the MTT assay, the sub-G1 analysis and mitochondrial membrane potential assay. The stronger inhibition effects of AGS cell growth was observed by application of Scutellaria barbata combined with several anti-cancer drugs (paclitaxel, 5-fluorouracil, cisplatin, ectoposide, doxorubicin and docetaxel) in comparison to the application of Scutellaria barbata or anti-cancer drugs. Our findings provide insight into unraveling the effects of Scutellaria barbata in human gastric cancer cells and developing therapeutic agents against gastric cancer.

Function of hepatocyte growth factor in gastric cancer proliferation and invasion

  • Cancer incidence has been increasing steadily and is the leading cause of mortality worldwide. Gastric cancer is still most common malignancy in Korea. Cancer initiation and progression are multistep processes involving various growth factors and their ligands. Among these growth factors, we have studied hepatocyte growth factor (HGF), which is associated with cell proliferation and invasion, leading to cancer and metastasis, especially in gastric cancer. We explored the intercellular communication between HGF and other surface membrane receptors in gastric cancer cell lines. Using complimentary deoxyribonucleic acid microarray technology, we found new genes associated with HGF in the stomach cancer cell lines, NUGC-3 and MKN-28, and identified their function within the HGF pathway. The HGF/N-methyl-N'-nitroso-guanidine human osteosarcoma transforming gene (c-MET) axis interacts with several molecules including E-cadherin, urokinase plasminogen activator, KiSS-1, Jun B, and lipocalin-2. This pathway may affect cell invasion and metastasis or cell apoptosis and is therefore associated with tumorigenesis and metastasis in gastric cancer.

Inonotus obliquus (차가버섯) 추출물의 EBV 양성 인간위암에 대한 in vivo 항종양 효능 연구 (Anti-tumor effect of Inonotus obliquus in xenograft animals with EBV+human gastric carcinoma)

  • 차가버섯(Inonotus obliquus)은 다양한 생리활성을 가진 약용버섯으로 항암, 항산화, 항염효능 등을 가진 것으로 보고되었다. EBV 양성 위암은 EBV관련 암 중 가장 빈번하게 나타나는 형태로 EBV 잠복감염이 그 원인이다. 본 연구에서는 차가버섯 주정추출물의 경구투여를 통해 EBV 양성 인간위암(SNU719) 세포주를 면역결핍 쥐에 주입 후 생기는 고형암 생성억제에 대한 효능을 연구하였다. 또한, 실험종료 후, 각각의 종양조직을 절제하여 항종양 억제기전을 탐구하였다. In vivo 종양생성억제 실험에서 차가버섯은 유의적으로 고형암 생성억제 효능을 보였다. 차가버섯이 투여된 동물유래 종양조직에서 세포자멸사와 관련된 p53, p21 및 Bax의 발현이 크게 증가하였으며, 이는 cleaved caspase-9와 cleaved Parp 발현의 상승과 동반하여 항종양 효능이 세포자멸사를 통해 나타남을 제시하였다. 또한, 이러한 항종양 효능은 세포주 내 잠복되어 있는 EBV 바이러스 유전자인 BZLF-1 및 LMP-2의 발현에도 영향을 미치는 것으로 밝혀졌다.

Corticotropin-Releasing Hormone (CRH)에 의한 인간 위암 세포(MKN-28)의 Migration 증가 (Enhancement of Cell Migration by Corticotropin-Releasing Hormone (CRH) in Human Gastric Cancer Cell Line, MKN-28)

  • Background: Corticotropin-Releasing Hormone (CRH), an important regulator of stress response, has a potent immunoregulatory effect with the ability to promote the growth of various cancer through CRH receptor type 1 under stress. Although the metastasized cancers through cell migration are more aggressive than the primary cancers, little is known about the effect of CRH on cell migration. Gastric cancer is prone to metastasize to other tissues and it is reported that gastric cancer is response to various stresses such as oxidative stress. Herein, we studied the relationship between CRH and gastric cancer cell migration. Methods: We used gastric cancer cell line, MKN-28 and tested the CRH receptor type 1 expression on MKN-28 by RT-PCR. To examine the change in the ability of migration by CRH in MKN-28, cells were incubated with CRH and then migration ability was measured using a cell migration assay. Results: We confirmed that CRH receptor type 1 was expressed in MKN-28 and HaCaT cells. The migration ability of MKN-28 cells was increased by CRH in a time-, dose- dependent manner. Conclusion: These data suggest that CRH increases migration ability in gastric cancer cell line and that CRH may be a critical regulator in the metastasis of gastric cancer cell.

8-Methoxypsoralen Induces Apoptosis by Upregulating p53 and Inhibits Metastasis by Downregulating MMP-2 and MMP-9 in Human Gastric Cancer Cells

  • Furanocoumarin 8-methoxypsoralen (8-MOP) is the parent compound that naturally occurs in traditional medicinal plants used historically. 8-MOP has been employed as a photochemotherapeutic component of Psoralen + Ultraviolet A (PUVA) therapy for the treatment of vitiligo and psoriasis. Although the role of 8-MOP in PUVA therapy has been studied, little is known about the effects of 8-MOP alone on human gastric cancer cells. In this study, we observed anti-proliferative effect of 8-MOP in several human cancer cell lines. Among these, the human gastric cancer cell line SNU1 is the most sensitive to 8-MOP. 8-MOP treated SNU1 cells showed G1-arrest by upregulating p53 and apoptosis by activating caspase-3 in a dose-dependent manner, which was confirmed by loss-of-function analysis through the knockdown of p53-siRNA and inhibition of apoptosis by Z-VAD-FMK. Moreover, 8-MOP-induced apoptosis is not associated with autophagy or necrosis. The signaling pathway responsible for the effect of 8-MOP on SNU1 cells was confirmed to be related to phosphorylated PI3K, ERK2, and STAT3. In contrast, 8-MOP treatment decreased the expression of the typical metastasis-related proteins MMP-2, MMP-9, and Snail in a p53-independent manner. In accordance with the serendipitous findings, treatment with 8-MOP decreased the wound healing, migration, and invasion ability of cells in a dose-dependent manner. In addition, combination treatment with 8-MOP and gemcitabine was effective at the lowest concentrations. Overall, our findings indicate that oral 8-MOP has the potential to treat early human gastric cancer, with fewer side effects.

ER membrane protein complex subunit 6 (EMC6) is a novel tumor suppressor in gastric cancer

  • The endoplasmic reticulum (ER) membrane protein complex subunit 6 (EMC6) is a novel human autophagy-related molecule. Here, using tissue microarray and immunohistochemistry, we report that EMC6 protein is lost or reduced in glandular cells of patients with gastric adenocarcinoma, compared to normal stomach mucosa. Overexpression of EMC6 in gastric cancer cells inhibited cell growth, migration, invasion, and induced apoptosis and cell cycle arrest at S-phase. Further investigation suggested that EMC6 overexpression in BGC823 human adenocarcinoma gastric cancer cells reduced tumorigenicity in a xenograft model, demonstrating that EMC6 has the characteristics of a tumor suppressor. This is the first study to show that EMC6 induces cell death in gastric cancer cells. The molecular mechanism of how EMC6 functions as a tumor suppressor needs to be further explored.

배추김치 분획물의 in vitro 항암효과 (Invitro Anticancer Effect of Chinese Cabbage Kimchi Fractions)

  • In vitro anticancer effect of Chinese cabbage kimchi fractions was investigated by using human cancer cells, AGS human gastric adenocarcinoma cells and HT 29 human colon adenocarcinoma cells. The Chinese cabbage kimchi(fermented for 4 days at 15oC) was fractionated into 7 groups, methanol extract, hexane fraction(fr.), methanol soluble fr., dichloromethane fr., ethylacetate fr., butanol fr. and aqueous fr.. Chinese cabbage kimchi fractions inhibited the growth of AGS and HT 29 cancer cells as dose dependent. In particular, the dichloromethane fr. showed the highest inhibitory effect among other fractions. When the dichloromethane fr.(0.2mg/ml) was treated, the number of AGS and HT 29 survival cancer cells reduced to 12$\times$104/ml and 11$\times$104/ml compared to 166$\times$104/ml and 50$\times$104/ml of the controls, respectively. Chinese cabbage kimchi fractions also inhibited the DNA synthesis of the cancer cells. They inhibited the DNA synthesis of AGS human gastric adenocarcinoma cells more efficiently than that of HT 29 human colon adenocarcinoma cells. These results indicate that Chinese cabbage kimchi fractions show in vitro anticancer activity and the dichloromethane fr. among them reveals the highest effect.
