대기부유분진추출물로 야기된 DNA 손상에 대한 Quercetin, Vitamin C 및 Trolox 의 보호효과 (Chemopreventive Effect of Quercetin, Vitamin C and Trolox Against the Organic Extract of Airborne Particulate Matter Induced Genotoxicity in A549 Human Lung Carcinoma Cells)

  • In order to evaluate the genotoxicity of airborne particulate matter extracted with dichloromethane (APE), the rat microsome mediated (S-9) or DNA repair enzyme treated Comet assays were performed using the single cell gel electrophoresis in A549 human lung carcinoma cells. It was found that the cells interacting with APE showed more DNA single-strand breaks relative to untreated cells. The genotoxicity of APE was increased with the treatment of S-9 mixture. Microsome mediated DNA damage was inhibited by CYP1Al inhibitor, quercetin. The APE also showed oxidative DNA damage evaluated by endonuclease III treatment. Oxidative DNA damage of APE was inhibited by antioxidants such as vita- min C and Trolox. We also found that the vegetables or fruits extract may reduce APE-induced genotoxicity by their anti- oxidant activity and CYP1A1 inhibition.

  • The effectiveness of N-acetyl-L-cysteine (NAC) to protect blood cells against Mitomycin C (MMC) induced genotoxicity was investigated in human chronic myeloid leukemia cells (KCL22) using the alkaline comet assay. The comet assay was selected as sensitive and rapid method for analysis of DNA damage and repair in individual cells. NAC treatment alone did not produce any damage in KCL22 cell. But NAC was found to be effective in reducing genotoxic damage in KCL22 cells exposed to MMC. These results confirm the literature data that, given the safety and ability to reduce DNA damage. NAC may be useful to prevent drug-mediated genotoxicity.

모발의 탈색 및 손상에 미치는 천연 식물 트리트먼트제의 영향 (Effects of Natural Vegetable Treatment Agent on the Bleaching and Damage of Human Hair)

  • In this study, we investigated the effect of natural vegetable treatment agent on the bleaching and damage of human hair during permanent wave formation. The vegetable treatment agent was prepared by fermenting the mixture of tea leaves (10g), black rice (100g), black bean (100g), corn (100g), malt (50g), and water(500g) at room temperature for 1 week. The degree of bleaching and damage was checked by Munsell value 'L' and morphological changes of hair in FE-SEM. When the hair was bleached, the Munsell value 'L' was almost same regardless of adding the vegetable treatment to the bleaching solution. But when the hair was permanently waved, the curl formation ability was high and the damage was low in the case of adding the vegetable treatment. This means that the vegetable treatment covers the scale of hair and protects the hair from the chemicals.

Protection of the Flowers of Prunus persica Extract from Ultraviolet B-Induced Damage of Normal Human Keratinocytes

  • For an attempt to develop safe materials protecting solar ultraviolet (UV)-induced skin damage, plant extracts were evaluated for their inhibitory activities of free radical generation and arachidonic acid/metabolites release from UVB-irradiated normal human keratinocytes. From the results of these screening procedures, the ethanol extract of the flowers of Prunus persica (Ku-35) was selected for further study. It was found that Ku-35 (100-1,000 ${u}g/m\ell$) inhibited the amount of $^{14}C$-arachidonic acid/metabolites release from UVB-irradiated keratinocytes. It was also demonstrated that Ku-35 possessed the protective activity against UV-induced cytotoxicity of keratinocytes and fibroblasts. In addition, Ku-35 was revealed to protect UVB-induced erythema formation using guinea pigs in preliminary in vivo study. All these results indicate that the flowers of P. persica extract may be beneficial for protecting UV-induced skin damage when topically applied.

  • Forest fires in Korea usually start in the fall and occur every year until spring. Most wildfires are human resources that combine topographical characteristics and carelessness, and failure to respond in the initial stage and lack of cleanup are spreading to large-scale wildfires. In order to prevent these wildfires, active cooperation from the public is essential. As can be seen from recent wildfires, the attention of the public is needed above all else because large-scale wildfires in Korea are occurring due to the people's negligence. If a wildfire spreads and becomes large, it causes damage to life and property, and the damage is irreversible. In this study, various methods were used to prevent forest fires and improve initial suppression ability. In order to minimize damage, the model analyzed by the 119 Special Rescue Team in Gangwon, Chungcheong and nearby forest fires was analyzed on the combustion progress and wind direction by time period. The propagation speed by the wind direction was simulated. Until now, most of the wildfires have been extinguished by firefighting, but I hope that the Forest Service will take the lead and maintain coordination with related organizations.

  • The purpose of this study was to examine the effect of supplementary highly purified chitosan (HPC) on blood alcohol concentration in healthy human. The human study was performed with two sections. Each section of the study was conducted by two-phase cross-over design with a week wash-out period. All volunteers took HPC in one phase, and took a placebo in the next phase. Blood alcohol concentrations were different between in those taking HPC and in those taking the placebo in the human. And the concentration of serum aspartate aminotransferase (AST, GOT) and alanine aminotransferase (ALT, GPT), the indicator of liver cell damage, was lowered in those taking HPC, compared to those taking the placebo. In conclusion, taking HPC prior to drinking alcohol can somewhat reduce alcohol concentration in human blood and liver cell damage.

염.탈색 및 펌 처리에 따른 모발의 손상과 웨이브 형상 (Hair Damage and Wave Shape according to Dyeing, Bleaching, Permanent Wave Treatment)

  • In this paper, the damage of hair and wave shape in process of the dyeing, bleaching, and permanent hair wave are simulated. The virgin hair that do not process the dyeing, bleaching, and permanent hair wave becomes stable in the scale type. On the other hand, the hair that deals with the dyeing, bleaching, and permanent hair wave is heavily damaged in the scale type. It is observed that the higher pH is decided, the higher the hair is damaged because the scale damage in the bleaching hair is more heavily damaged than the dyeing hair. In case of executing the permanent hair wave, the processing of the plaine rinsing becomes less in the scale damage of the hair and better in the wave form. Therefore, it is found that the processing of the plaine rinsing to improve the wave form and to prevent the hair damage is needed.

Protective Effects of Prunus persica Flesh Extract (PPFE) on UV-Induced Oxidative Stress and Matrix Metalloproteinases Expression in Human Skin Cells

  • In our continuous efforts to procure the active materials from natural products in the protective effects of oxidative stress or UV damage to skin cells we found the Prunus persica flesh extract (PPFE) is considerable to meet the demand to protect the skin damage. PPFE attenuated cell damage induced by hypoxanthine-xanthine oxidase in cultured human keratinocytes, indicating that PPFE has the potential of the scavenging effect of reactive oxygen species (ROS) in human skin cell. Moreover, PPFE significantly suppressed UVA-induced ROS production determined by the oxidation of 2,7-dichlorodihydrofluorescein diacetate (DCFH) using FACS analysis. Additional study revealed that UVA irradiation of HaCaT human keratinocytes increased the gelatinolytic activities of matrix metalloproteinase-2, and -9 (MMP-2, -9) and mRNA expression of MMP-9 analyzing by a real-time reverse transcriptase-polymerase chain reaction (RT-PCR), and these events were significantly suppressed by the treatment with PPFE. These results suggest that PPFE might be applicable as natural ingredients for skin antiaging agents via UV-induced ROS scavenging activity and suppression of MMP expression in the skin cells.

PCNA Modifications for Regulation of Post-Replication Repair Pathways

  • Stalled DNA replication forks activate specific DNA repair mechanism called post-replication repair (PRR) pathways that simply bypass DNA damage. The bypassing of DNA damage by PRR prevents prolonged stalling of DNA replication that could result in double strand breaks (DSBs). Proliferating cell nuclear antigen (PCNA) functions to initiate and choose different bypassing pathways of PRR. In yeast, DNA replication forks stalled by DNA damage induces monoubiquitination of PCNA at K164, which is catalyzed by Rad6/Rad18 complex. PCNA monoubiquitination triggers the replacement of replicative polymerase with special translesion synthesis (TLS) polymerases that are able to replicate past DNA lesions. The PCNA interaction motif and/or the ubiquitin binding motif in most TLS polymerases seem to be important for the regulation of TLS. The TLS pathway is usually error-prone because TLS polymerases have low fidelity and no proofreading activity. PCNA can also be further polyubiquitinated by Ubc13/ Mms2/Rad5 complex, which adds an ubiquitin chain onto monoubiquitinated K164 of PCNA. PCNA polyubiquitination directs a different PRR pathway known as error-free damage avoidance, which uses the newly synthesized sister chromatid as a template to bypass DNA damage presumably through template switching mechanism. Mammalian homologues of all of the yeast PRR proteins have been identified, thus PRR is well conserved throughout evolution. Mutations of some PRR genes are associated with a higher risk for cancers in mice and human patients, strongly supporting the importance of PRR as a tumor suppressor pathway.

Buplueri Radix 의 1,2,4-benzentriol에 의해 유발된 DNA Damage에 대한 보호효과에 대한 연구 (Protective Effect of Buplueri Radix (BR) Against 1,2,4-benzentriol Induced DNA Damage in Human Lymphocytes)

  • Objectives : Buplueri Radix (BR), used medical plant in Korea traditional medicine, contains various compounds, including a series of triterpene saponins known as saikosaponins. We performed this study for the protective effect of BR against oxidative damage induced by 1,2,4-benzentriol(BT) in human lymphocytes. Methods : In order to investigate the protective effect of BR against carcinogens, genotoxicity induced by benzene metabolite, BT were performed using cytokinesis-block micronucleus(CBMN) assay and comet assay. Results : The frequency of micronucleus at 25, 50 and $100{\mu}M$ concentration of BT were $8{\pm}2.36$, $23{\pm}2.31$, $35{\pm}4.17$ respectively. In addition of BR with concentration of 25 and $50{\mu}g/mL$, MN frequencies were significantly decreased. According to comet assay, BT induced DNA damage in a dose-dependent manner at concentration of 10 and 50 while BT with BR treatment decreased DNA breakage. No genotoxicity was observed by BR($25{\sim}50{\mu}g/mL$) treatment alone on DNA breakage. Since BT can induce DNA damage through the generation of reactive oxygen species(ROS), we examined the level of ROS in human lymphocytes treated with BT and/or BR using DCF-DA, ROS-sensitive probe. The generation of ROS in BT-treated cells was also observed, and BR addition inhibited the level of BT-induced DNA damage. Conclusions : From above results it is suggested that BR could protect the cell and DNA from pro-oxidant effect by ROS by BT
