용각산의 면역조절 및 백혈병세포의 아폽토시스에 미치는 효과 (Effect of Yonggak-san on the Immuno-regulatory and Apoptosis of Leukemia cells)

  • The purpose of this research was to investigate the effect of Yonggak-san (YGS) on the immune reaction and apoptosis of leukemia cells. Administration of YGS(500 mg/kg) enhanced proliferation of splenocytes, thymocytes and mesenteric lymph node cells, and also YGS accelerated subpopulation of splenic Band T, thymic T and mesenteric lymph node-T lymphocytes, especially significantly increased CD4+-TH cells in BALB/c mice. YGS accelerated phagocytic activity and production of nitric oxide in peritoneal macrophages. YGS induced apoptosis of transplanted-L1210 cells in vivo, increased apoptotic cell death of cultured-L1210 and/or Molt4 human leukemia cells, decreased of mitochondrial transmembrane potential of both cells in vitro. These results suggest that YGS have an immune-regulatory effect and anti-cancer property.

Epigallocatechin-3-gallate Inhibits Tax-dependent Activation of Nuclear Factor Kappa B and of Matrix Metalloproteinase 9 in Human T-cell Lymphotropic Virus-1 Positive Leukemia Cells

  • Epigallocatechin-3-gallate (EGCG) is the most abundant polyphenol molecule from green tea and is known to exhibit antioxidative as well as tumor suppressing activity. In order to examine EGCG tumor invasion and suppressing activity against adult T-cell leukemia (ATL), two HTLV-1 positive leukemia cells (HuT-102 and C91-PL) were treated with non-cytotoxic concentrations of EGCG for 2 and 4 days. Proliferation was significantly inhibited by 100 ${\mu}M$ at 4 days, with low cell lysis or cytotoxicity. HTLV-1 oncoprotein (Tax) expression in HuT-102 and C91-PL cells was inhibited by 25 ${\mu}M$ and 125 ${\mu}M$ respectively. The same concentrations of EGCG inhibited NF-kB nuclearization and stimulation of matrix metalloproteinase-9 (MMP-9) expression in both cell lines. These results indicate that EGCG can inhibit proliferation and reduce the invasive potential of HTLV-1-positive leukemia cells. It apparently exerted its effects by suppressing Tax expression, manifested by inhibiting the activation of NF-kB pathway and induction of MMP-9 transcription in HTLV-1 positive cells.

Albumin and Antioxidants Inhibit Serum-deprivation-induced Cell Adhesion in Hematopoietic Cells

  • Previously, we identified albumin as an inhibitory factor in serum for cell adhesion of T cells such as human Jurkat T and primary cultured human T cells. In the present study, we found that other hematopoietic cell lines including U-937 human monocytes, THP-1 human monocytes, K-562 promyelocytic leukemia cells, and HL-60 human leukemia cells, also adhere to tissue culture flasks when serum is withdrawn, and albumin exerts an inhibitory effect on cell adhesion by those cells, implying that this inhibition is a common phenomenon in hematopoietic cells. Furthermore, we found that cell adhesion is inhibited by antioxidants such as (-)-epigallocatechin- 3-gallate (EGCG), morin, and a-tocopherol. Our results suggest that albumin may inhibit basal cell adhesion of hematopoietic cells and that the oxidative balance in the plasma may be important for cell adhesion of hematopoietic cells in vivo.

  • Spatholobus suberectus belonging the family Leguminosae has been used for promoting blood circulation, removing blood stasis, tonifying the blood, relaxing tendons, stopping internal bleeding and eliminating dampness in oriental traditional medicine. This study investigates whether the water extracts of S. suberectus induce apoptotic cell death in Jurkat T-acute lymphoblastic leukemia (ALL) cells. Jurkat cells were increased inhibitions of cell viability in a concentration-dependent manner by S. suberectus, as measured by cell morphology. The capability of S. suberectus to induce apoptosis was associated with proteolytic cleavage of specific target protein such as poly (ADP­ribose)polymerase protein suggesting the possible involvement of caspases. The purpose of the present study is also to investigate the effect of S. suberectus on cell cycle progression. G1 checkpoin related gene products tested (cyclin D1, cyclin dependent kinase 4, retinoblastoma, E2Fl) were decreased in their protein levels in a dose-dependent manners after treatment of the extract. These results indicate that the increase of apoptotic cell death by S. suberectus may be due to the inhibition of cell cycle progression in wild type p53-lacking Jurkat cells.

  • Apoptosis is a morphologically and biochemically distinct form of cell death that occurs in many different cell types in a wide variety of organisms. Ajbizzia julibrissin belonging the family Leguminosae has been used for the treatment of contusion, sore throat, amnesia, and insomnia in oriental traditional medicine. This study investigates whether the water extract off julibrissin induce apoptotic cell death in Jurkat T-acute lymphoblastic leukemia (ALL) cells. Jurkat cells were increased inhibitions of cell viability in a concentration-dependent manner by A julibrissin. This herbal medicine also caused apoptosis as measured by cell morphology and DNA fragmentation. The capability oft julibrissin to induce apoptosis was associated with proteolytic cleavage of specific target protein such as poly (ADP-ribose) polymerase (PARP) protein suggesting the possible involvement of caspases. Our result skewed that Bcl-2 and Bax protein levels were not changed in all A julibrissin-treated groups compared to control group. These results suggest that A julibrissin-mediated apoptosis is independent with Bcl-2 related signaling pathway in this cells.

  • CALLA 항원에 대한 단일클론 항체는 백혈병의 진단이나 치료에 대한 활용가능성을 잠재하고 있기 때문에 구미, 일본 등지에서는 이에 대한 연구가 활발하다. 이 점에 유의하여 본 연구자들은 CALLA에대한 새로운 단일클론항체 KP-22를 개발하고, 이 단일클론항체를 이용 CALLA의 분포를 여러 세포주에 대하여 조사한 결과 common ALL, Burkitt's lymphoma, T-ALL 세포주는 현저한 CALLA양성 반응을 보였으나 사람의 섬유아세포 계역을 제외한 모든 비백혈병성 암세포주 및 myelocytic leukemia 세포주들은 음성이었다. 막 단백질을 125I 방사능 표지한 후 KP-22를 이용, 면역 침전법으로 백혈병세포주와 사람의 정상 섬유아세포에서 CALLA를 정제하여 전기영동한 결과 각각 분자량이 약 95Kd 및 100Kd인 단일 band로 확인되었으나 이들의 peptide mapping 양상은 같았으므로 분자량에서의 미세한 차이는 posttranslational modification 과정에서 첨가되는 sialic acid에 기인 하는 것으로 사료된다.

Differentiation Induction of Dendritic Cell Phenotypes from Human Leukemic Cell Lines

  • Recent clinical studies have shown that a high proportion of patients with acute promyelocytic leukemia (APL) achieve complete remission after treatment with all-trans retinoic acid (ATRA). However, most patients who receive continuous treatment with ATRA relapse and develop ATRA-resistant leukemia. Dendritic cells (DCs) are important antigen-presenting cells in the development of antileukemic T-cell responses. In this study, we investigated the strategies to overcome ATRA resistance of APL cells by inducing the differentiation of DCs from human leukemic cell lines for the developtment of adoptive immunotherapy. CD83 was used as a mature DC marker in this study and the expression of CD83 mRNA was determined by RT-PCR method. The promyelocytic leukemic cell line HL-60, B lymphoblast cell lines RPMI 7666 and NC-37 could be induced to dendritic cells in vitro. Treatment of HL-60 with phorbol 12-myristate 13-acetate (PMA) resulted in the expression of myeloid-related DC phenotypes, while treatment of RPMI 7666 with fms-like tyrosine kinase 3 ligand (Flt3-ligand, FL) and treatment of NC-37 with PMA and FL led to the expression of lymphoid-related DC phenotypes. In conclusion, myeloid-related DC phenotypes and lymphoid-related DC phenotypes could be generated from HL-60, NC-37 and RPMI 7666 cell lines, respectively. These DC phenotypes can potentially be used to generate antileukemic T cells in vitro for adoptive immunotherapy.

Rapamycin-resistant and torin-sensitive mTOR signaling promotes the survival and proliferation of leukemic cells

  • The serine/threonine kinase mTOR is essential for the phosphoinositide 3-kinases (PI3K) signaling pathway, and regulates the development and function of immune cells. Aberrant activation of mTOR signaling pathway is associated with many cancers including leukemia. Here, we report the contributions of mTOR signaling to growth of human leukemic cell lines and mouse T-cell acute leukemia (T-ALL) cells. Torin, an ATP-competitive mTOR inhibitor, was found to have both cytotoxic and cytostatic effects on U-937, THP-1, and RPMI-8226 cells, but not on Jurkat or K-562 cells. All cells were relatively resistant to rapamycin even with suppressed activity of mTOR complex 1. Growth of T-ALL cells induced by Notch1 was profoundly affected by torin partially due to increased expression of Bcl2l11 and Bbc3. Of note, activation of Akt or knockdown of FoxO1 mitigated the effect of mTOR inhibition on T-ALL cells. Our data provide insight on the effect of mTOR inhibitors on the survival and proliferation of leukemic cells, thus further improving our understanding on cell-context-dependent impacts of mTOR signaling.   • Adult T-cell leukemia/lymphoma (ATLL) is a malignancy of mature T-cells caused by the human T-cell lymphotrophic virus type I (HTLV-D. HTLV-I is endemic in some areas in Japan, the Caribbean basin, and Africa but has low prevalence in South Korea. Patients with ATLL are susceptible to opportunistic infections such as cytomegalovirus (CMV) infection, but CMV infection in chronic ATLL is uncommon. Reported herein is a case involving a 44-year-old woman with chronic ATLL who presented the symptoms of fever and diarrhea. She was suspected to have acute-type ATLL but was later diagnosed with CMV colitis.

Cytotoxic and Anti-oxidant Constituents from the Aerial Parts of Aruncus dioicus var. kamtschaticus

  • Ten compounds (1 - 10), palmitic acid (1), 10-nonacosanol (2), pentacosan-1-ol (3), phytol (4), ${\beta}$-sitosterol (5), ${\beta}$-sitosterol-3-O-${\beta}$-D-glucopyranoside (6), 2,4-dihydroxycinnamic acid (7), hyperoside (8), uridine (9) and adenosine (10), were isolated from the n-hexane and EtOAc-soluble fractions of the aerial parts of A. dioicus var. kamtschaticus (Rosaceae). The structures of these compounds were elucidated on the basis of spectroscopic evidence. All compounds (1 - 10) were isolated for the first time from this plant. Cytotoxicity of 1 - 10 against Jurkat T (T-lymphocytic leukemia cells), HeLa (Human cervical epitheloid carcinoma cells), MCF-7 (Human breast cancer cells), and HL-60 (Human promyelocytic leukemia cells) cell lines was measured. Compound 6 showed good cytotoxicity against HL-60 cell line with $IC_{50}$ value of 8.13 ${\mu}g/mL$. In addition, compounds 7 and 8 exhibited antioxidant activity with $IC_{50}$ values of 16.30 and 12.42 ${\mu}g/mL$, respectively.
