A gene (hpkA), encoding a histidine protein kinase homolog, has been identified in the upstream region of the espAB operon in Myxococcus xanthus. It encodes a 333 amino acid (35,952 Da) protein with a histidine protein kinase domain in the region from amino acid 90 to 317. Null mutations in the hpkA gene caused formation of loose irregular fruiting bodies, while wild-type strains developed tight hemispherical fruiting bodies under developmental conditions. Sporulation of the hpkA mutant was delayed by at least 12 h compared to that of the wild-type. It appeared that the hpkA mutation increased the expression of the espAB operon by more than 2-fold compared with the wild-type under developmental conditions. Expression of the hpkA gene was low under vegetative conditions, but was highly induced under developmental conditions.

A new eddy covariance-based flux tower has been established since June 2016 in a dry direct-seeding rice paddy field in southwestern Korea - Haenam Paddy field KoFlux site (HPK). To assess the competitiveness of direct-seeding agricultural technique in the context of climate-smart agriculture, we have been monitoring the $CO_2$, $CH_4$ and energy fluxes continuously. In this communication, we (1) introduce the HPK site and (2) explain the structure and processes of the HPK-specific data processing and quality control. The latter consists of 10-steps data processing and quality control procedures following the KoFlux standardized protocol and explained each step in layman's term. The final data produced during the data processing were stored in NCAM-DAPS (National Center for AgroMeteorology-Data Acquisition and Processing System, http://daps.ncam.kr). We hope that the introduction of new HPK KoFlux site would serve as a platform to facilitate transdisciplinary efforts in the research and education associated with climate-smart agriculture toward sustainability.
pH values in all samples were in the range of 5.04 to 5.32. pH of kanjang prepared with normal pressure(NPK) was higher than that of kanjang prepared with high pressure(HPK) and kanjang prepared with steam(SPK). Titratable acidity(TA) in all samples were found to be 4.26~7.72 mL. TA in HPK(5.99~6.96 mL) was lower than that in SPK(4.26~5.61 mL) and NPK(7.32 mL). Reducing sugar(RS) contents in all samples were in the range of 25.18 to 194.81 mg%. RS contents of HPK and SPK were a similar pattern and were higher than that of NPK. Major free sugars(FS) were glucose, fructose and xylose. In total FS, SPK was higher than that of NPK and HPK. Major organic acids(OA) were succinic acid, oxalic acid, tartaric acid, malic acid and citric acid. In total OA contents, SPK(40.38~76.96 mg%) was higher than that of NPK(55.85 mg%)and HPK(27.78~34.21mg%). OA contents in HPK and SPK were decreased slightly according to the heating time of whole black bean. The highest percentage of fatty acid (FA) in all samples was palmitic acid(33.28~43.51 % as area % of GC chromatogram), followed in a decreasing order by stearic acid(14.15~19.16%) and myristic acid(5.93~19.14%). The composition ratios of three FA to total FA ranged from 55.97 to 72.85&. In all samples, there was myristic acid that was not contained in raw soybean.
Total nitrogen(TN) contents in all samples were in the range of 308.3 to 925.9 mg% and TN value of kanjang prepared with high pressure(HPK)-heated bean was lower than that of normal pressure(NPK) and steam(SPK)-heated bean. TN content was slightly increased according to the heating time of bean. Amino type nitrogen(AIN) contents in all samples were in the range of 133 to 451.5 mg% and AIN value of NPK(451.5 mg%) was higher than that of HPK(133∼171.5 mg%) and SPK(178.9∼224 mg%). Ammonia type nitrogen(AON) contents in all samples were in the range of 23.5 to 142.0 mg% and AON value of HPK was lower than that of HPK and SPK. Free amino acid(FA) contents in all samples were in the range of 133 to 451.5 mg%, and then FA content of NPK was higher than that of SPK(178.9∼224 mg%) and HPK(133 ∼171.5 mg%). Lightness(L) value of Hunter color in all samples were in the range of 45.13 to 49.08 and was similar with each other. Redness(a) and yellowness(b) value were in the range of 25.30∼34.43 and 52.55∼74.13, respectively.

The standardized eddy covariance flux data processing in KoFlux has been updated, and its database has been amended accordingly. KoFlux data users have not been informed properly regarding these changes and the likely impacts on their analyses. In this paper, we have documented how the current structure of data processing in KoFlux has been established through the changes and improvements to ensure transparency, reliability and usability of the KoFlux database. Due to increasing diversity and complexity of flux site instrumentation and organization, we have re-implemented the previously ignored or simplified procedures in data processing (e.g., frequency response correction, stationarity test), and added new methods for $CH_4$ flux gap-filling and $CO_2$ flux correction and partitioning. To evaluate the effects of the changes, we processed the data measured at a flat and homogeneous paddy field (i.e., HPK) and a deciduous forest in complex and heterogeneous topography (i.e., GDK), and quantified the differences. Based on the results from our overall assessment, it is confirmed that (1) the frequency response correction (HPK: 11~18% of biases for annually integrated values, GDK: 6~10%) and the stationarity test (HPK: 4~19% of biases for annually integrated values, GDK: 9~23%) are important for quality control and (2) the minimization of the missing data and the choice of the appropriate driver (rather than the choice of the gap-filling method) are important to reduce the uncertainty in gap-filled fluxes. These results suggest the future directions for the data processing technology development to ensure the continuity of the long-term KoFlux database.
We have shown that the volcanic rocks from the northern part of Cheju Island can be divided into high $P_2O_5/K_2O$(HPK) and low $P_3O_5/K_2O$(LPK) groups, each with distinct geochemical characteristics(Park and Kwon, 1993a and b). This study reports mineral compositions for plagioc-lase, olivine, and clinopyroxene in order to see the dependence of mineral chemistry on the whole rock composition, and discusses equilibrium relationships between crystal and liquid. Plagioclase and olivine phenocrysts show no compositional differences for the two rock group. However, $Al^{ⅵ}/Al^{ⅳ}$ ratios of clinopyroxenes suggest that pyroxenes have fractionated at deeper level, and that the LPK group might have fractionated at higher pressure than the HPK group. These are in good agreement with our previous interpretation based on whole rock chemistry(Park and Kwon, 1993a). Although subhedral or euhedral form and homogenous composition for most plagioclase and clinopy-roxene phenocrysts suggest equilibrium relationship with liquid, the uncertainties associated with liquid, the uncertainites associated with equilibrium constant for these minerals do not allow testing equilibium relationship between mineral and liquid on the basis of chemistry. On the other hand, olivine phenocrysts in hawaiites, for which Kd is well known, show distinct nonequilibrium relationship with calculated liquid composi-tions, while those in other rock compositions are in equilibrium from those for other rocks. We report for the first time as far as we know plagioclase xenocryst and pigeonite inclusion in plagioclase, which indicates assimilation process. In conclusion, these mineralogical observations imply that mag-matic processes underneath the Cheju volcano were not simple.
