Conversion of water-insoluble components of the basidiocarps of ganoderma lucidum to water-soluble components by hydrolyzing with chitinase

  • We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.

찰흑미와 일반찰벼의 열수가용성 물질 (Hot-water soluble on Waxy Black Rice and Waxy Rice Flours)

  • 가용성 탄수화물 함량은 찰흑미와 일반찰벼 쌀가루를 $90^{\circ}C$에서 10분, 20분 및 30분 동안 열처리한 결과 찰흑미는 17.1%, 21.5%, 24.7%이였으며, 일반찰벼는 18.6%, 21.7%, 23.4%의 함량을 보여 두 시료 모두 비슷한 양상을 보였다. 찰흑미와 일반찰벼 모두 가용성 탄수화물 함량의 대수값은 가열시간이 증가할수록 증가하여 직선적인 관계를 보였다. 쌀가루를 가열온도($80^{\circ}C,\;90^{\circ}C,\;95^{\circ}C$$98^{\circ}C$)에 따라 일정 시간(10분, 20분 및 30분) 가열하여 얻은 가용성 탄수화물의 Sepharose CL-2B 컬럼에 의한 용출양상은 찰흑미와 일반찰벼 모두 void volume($V_0$) 부근에서 대부분 용출되었으며, 가열시간이 증가할수록, 가열온도가 높아질수록 가용성 탄수화물의 용출양상은 증가하였다. 또한 $180{\sim}210\;mL$ 부근에서도 일부가 용출되었다.

  • 일반계와 통일계 쌀의 가열에 따른 가용성 물질과 밥의 텍스처와의 관련성을 연구하였다. 가열에 의한 쌀 또는 쌀가루의 가용성 물질 또는 가용성 아밀로오스 함량은 일반계는 품종간에 큰 차이가 없었으나 통일계는 품종간에 차이를 보였다. 같은 가열조건에서 일반계는 통일계 보다 가용성 물질의 함량이 높았고 가용성 아밀로오스함량도 높았다. 쌀 또는 쌀가루의 가용성 물질과 아밀로오스 함량은 밥의 경도와는 부의 상관을, 부착성과는 정의 상관을 보였다.

향나무 심재 열수추출물의 염색성과 기능성 (Dyeing Properties and Functionality of Hot-water Extract from Juniperus chinensis Heartwood)

  • The purpose of this study is to investigate dyeing properties and functionality of cotton and silk fabrics dyed with hot-water extract from Juniperus chinensis heartwood. Water-soluble dye of Juniperus chinensis heartwood extracted with distilled water was expressed YR color series. Dye uptake of cotton continued to increase according to the increase of the dye concentration. Its variation trends were shown to be similar to the isothermal absorption curve of the Freundlich. Dye uptake of silk was better than cotton, increased depending on increasing concentration of the dye, dyeing time and temperature. Its variation trends were shown to be similar to the isothermal absorption curve of the Langmuir. As the pH increased, the dye uptake of fabrics reduced and showed increased on red tinge. Colors of the dyed fabrics were various, depending on dye concentration and the mordant type and mordant concentration. Washing fastness, light fastness and perspiration fastness were not good. However, rubbing and dry cleaning fastness showed relatively good grade. Dyed fabrics of ultraviolet-cut ability and deodorant ability were improved. And dyed fabrics showed antimicrobial abilities of 99.9% against Staphylococcus aureus.

담금 방법을 달리한 오이지의 숙성 중 펙틴질의 변화 (Changes in Pectic Substances of Korean Pickled Cucumbers with Different Preparation Methods)

  • This investigation was undertaken for the purpose of studying the changes of pectic substances in alcohol insoluble solids(AIS) extracted from cucumbers pickled in salt. The preparation methods were brining the cucumbers in 10% NaCl solution (bolied, method A), 20% NaCl solution(bolied, method B) and 20% NaCl solution(cool to $25^{\circ}C$ after boiling, method C). The composition ratio of hot water soluble pectin(HWSP) in AIS decreased during brining, while that of 0.4% sodium hexametaphosphate soluble pectin(NaSP) increased remarkably. The contents of Ca and Mg in AIS increased regardless of preparation methods used. By DEAE-cellulose chromatography, HWSP from fresh and brined cucumbers were divided into six fracitons. 0.2M peak was the main fraction in fresh cucumber, whereas 0.05M and 0.2M peak were the main fraction in pickled cucumbers. On the other hands, NaSP from fresh and brined cucumbers were divided into five fractions and 0.4M peak was the main fraction. However, the composition ratios of galacturonic acid and neutral sugar of 0.4M fraction in brined cucumbers were higher than those of fresh cucumber.

Trichoderma 속에 대한 낙엽송 및 미송 추출물의 항균활성 (Antifungal Activity of Wood Extracts of Larix leptolepis and Pseudotsuga menziesii againstTrichoderma spp.)

  • 본 연구는 국내산 침엽수 톱밥추출물을 이용하여 버섯의 푸른곰팡이병 방제약제 원료를 개발하고자 시도 되었으며, 푸른곰팡이병의 원인균의 하나인 Trichoderma 속 곰팡이를 대상으로 항균활성을 시험하였다. 1,000 ppm 농도의 낙엽송 열수추줄물은 Trichoderma 속 균주에 대하여 최고 20.6%의 항균활성을 나타냈다. 1,000 ppm 농도의 미송 열수추출물은 T. aggressivum에 대하여 60.3%, T. harzianum 52.2%, T. atroviride 46.8%, T.viride 46.1%의 높은 항균활성을 나타냈으며 T. koningii에서는 36.2%의 항균활성을 나타냈다. 미송 열수출물이 낙엽송 열수출물보다 Trichoderma 속 균주에 대하여 보다 효과적인 항균화합물을 함유하고 있다고 판단되었다. 낙엽송 열수출물로부터 분리된 n-hexane 분획물은 Trichoderma 속에 대하여 68.5% ~ 79.9%의 높은 항균활성을 나타냈다. 열수출물로부터 분리된 미송의 n-hexane 분획물은 T. aggressivum에 대하여 68.5%, T. atroviride에 대하여 71.4%, T. harzianum에 대하여 71.9%, T. koningii에 대하여 75.7%, T.viride에 대하여 82.3%의 높은 항균활성을 나타냈고, ethyl acetate 분획물이 n-hexane 분획물 다음으로 높은 항균활성이 나타내었다. Trichoderma 속 균주에 대한 낙엽송 및 미송 열수추출물의 항균활성은 미송 추출물이 낙엽송 추출물보다 높게 나타났지만, 낙엽송과 미송의 열수추출물로부터 분리된 n-hexane 분획물에 의한 항균활성 차이는 거의 없었다. 미송 열수추출물과 낙엽송 및 미송의 열수출물로부터 획득된 n-hexane 분획물은 Trichoderma 속에 대한 항균원료로서 사용가능성을 확인할 수 있었다.

Fabrication of Water-Soluble CuInS2 Quantum Dots by Hot-injection Method and Phase Transfer Strategy

  • Here we report an optimized hot-injection method and a phase transfer strategy for the synthesis of water-soluble $CuInS_2$ QDs with desired properties. The structure and morphology studies demonstrate that the resulting QDs are $CuInS_2$ tetragonal phase with well-defined facets. It is also found that the crystal size gradually increases with the increase of reaction temperature, while the surface of QDs with pre- and post-phase transfer is functionalized with hydrophobic and hydrophilic ligands, respectively. Spectroscopy measurements reveal the size-dependent optical properties of $CuInS_2$ QDs, demonstrating the quantum confinement effect in this system.

Neuritogenic activity of hot water extract from Hericium erinaceus

  • Hot water soluble extract was prepared from Hericium erinaceus and its neuritogenic activity on PC12h cells was analyzed, which is a clone originating from a rat pheochromocytomon. The moisture content of freeze dried hot water extract was 12.08%. The extract was mainly composed of carbohydrate (51.24%) followed by crude protein (24.04%), crude fat (0.26%), dietary fiber (5.09), and ash (12.18%). Fatty acids, glucan and inorganic constituents were found as minor components. The neuritogenic activity of hot water extract was evaluated under microscopic observation of neurite outgrowth in PC12h cells and by measuring the neurite length of induvidual cell. The extract exhibited strong effect of neurite outgrowth in a dose-dependent manner from 0.01 mg/mL to 1 mg/mL, in which longer neurite outgrowth was observed as the treatment dose increased.

한국산 다시마로부터 추출된 열수추출물, 수용성알긴산 및 알칼리가용성 알긴산의 물리적 성상과 겔 형성능 (A Comparative Study on Physical Properties and Gel formation abilities of Hot- water Extractable Material, Water-Soluble Alginate and Alkali-soluble Alginate Extracted from Laminaria japonica in East Sea, Korea)

  • 동해산 다시마 중의 알긴산의 고부가가치적 산업화 이용의 기초자료를 얻고자, 열수추출물 (HWEM),수용성 알긴산(WSA) 및 알칼리가용성 알긴산 (ASA)을 추출해 내어 그 각각의 수율, 겔화능, 겉보기점도, 용해성 (가용한계), 녹는점 등을 비교하여 보았다. 동해산 다시마 중 HWEM, WSA 및 ASA의 함량은 각각 8.3, 2.5 및 $19.1\%$이었다. 이 중 HWEM과 WSA는 전혀 겔화능이 없었고, 반면에 ASA는 $1.5\%$이상 농도에서 강한 점성과 함께 $Ca^{++}$이온에 의한 강한 겔화능도 지녔다. HWEM, WSA 및 ASA의 녹는점은 각각 $31.0^{\circ}C$, $24.0^{\circ}C$$14.3^{\circ}C$이었고, 그 각각의 녹는 온도에서의 가용한계는 2.55, 2.59 및 $2.31\%$이었다. 가용한계농도에는 큰 차이가 없으나, 녹는점면에서는 ASA가 가장 낮아 바람직하였다.

생산조인(生酸棗仁)과 초산조인(炒酸棗仁)의 추출방법에 따른 효능비교연구 (Study on the Comparison of Effects by Extraction Methods of Roast and Raw Semen Zizyphi Spinosae)

  • To observe the difference and change of the efficacy and ingredient appearing according to whether semen zizyphi spinosae has been roasted and its extraction method, locomotor activity, anticonvulsant activity, sleeping effect of each hot water and UMPM low temperature extracts of roast and raw semen zizyphi spinosae were measured through animal tests. Roast semen zizyphi spinosae showed superior pharmacological efficacy than raw semen zizyphi spinosae. Also, hot water extraction method showed superior pharmacological efficacy than low temperature extraction method(UMPM) that uses ultrasonic waves and microwaves. In short, it was shown that herbal medicine's extraction method needs to maximize medicinal effects by trying optimum extraction methods according to efficacy by considering characteristics of each herbal medicine. Together, there's also the need for in-depth studies on water-soluble ingredients of semen zizyphi spinosae that may be extracted through full hot water extraction method.
